The Effects of Morinda Officinalis Radix on Thyroid function in Hypothyroidism Rat Model induced by 6-propyl, 2-thiouracil (PTU) (6-propyl, 2-thiouracil로 유발된 갑상선 기능저하증 동물모델에서 파극천이 갑상선 호르몬 및 갑상선 조직에 미치는 효능)

  • Objectives : Hypothyroidism is the most common pathological disease of the endocrine system. Morinda Officinalis Radix has been used in treatment of Yang deficiency. In this study, we investigated the therapeutic effects of Morinda Officinalis Radix on a rat model of hypothyroidism induced by 6-propyl, 2-thiouracil (PTU). Methods : Two-month-old Sprague-Dawley male rats were used with injection of PTU which induced rats into hypo-thyroidism. During 2 weeks, rats were treated with Morinda Officinalis Radix and thyroxine. Body weights were checked every week, the after 4 weeks, biochemical analyses were performed. The levels of T3, T4 and TSH was measured by EILSA. Results : In the Morinda Officinalis Radix group, we could observe the level of T4 significantly increased and TSH level significantly decreased. Conclusions : Morinda Officinalis Radix could help the thyroid to produce thyroid hormones under PTU suppression. There is no harm effect on liver and kidney function, or other metabolism. According to these results it could help to treat hypothyroidism patients and relieve their symptoms.

Long Term Effect of Morinda Officinalis How on Muscular Strength & Stamina and Muscle Fatigue (장기간(長期間)의 파극(巴戟) 투여(投與)가 생쥐의 근력(筋力)과 근(筋) 지구력(持久力) 및 근(筋) 피로(疲勞)에 미치는 영향(影響))

  • Purpose: This study was conducted to investigate the muscular strength & stamina improvement and muscle anti-fatigue effects of Morinda officinalis How after long term administration. Methods: 4-6 weeks old ICR mice were used in this study and we administered the water soluble extracts of Morinda officinalis How in the concentration of 1, 10. 100/mg/0.3ml to each 5 mice (ATS group) and same volume of normal saline was administered to 5 mice (control group) once a day for 90 or 120 days. After the administration we performed the swimming exercise test and the grip strength test. And we measured the concentration of glucose and the activity of lactate dehydrogenase in plasma regarded as biochemical indicator related with the muscular fatigue. Results: In swimming exercise test, all ATS groups showed increased time compared with relative control group after 90 days administration. After 120 days adminstration 100/mg/0.3ml ATS groups showed increased results. In grip strength test, all ATS groups showed increased grip strength compared with relative control group after 90 days administration. After 120 days adminstration 10/mg/0.3ml ATS groups showed increased result. All the results of plasma glucose didn't showed statistically significant difference after 90 and 120 days administration. In the plasma concentration of activity level of lactate dehydrogenase, all ATS groups didn't showed statistically significant difference compared after 90 days administration. Conclusion: This study show that Morinda officinalis How can increase the muscular strength & stamina and anti-fatigue effect.

  • Morinda officinalis How. (Rubiaceae) has been used as tonic, warming, sex impulse and anti-inflammatory agents. Two known anthraquinones, rubiadin-1-methyl ether (I) and rubiadin (II) were isolated from root of M. officinalis. Their structures were identified using NMR and literature comparisons. The contents of I in eighteen M. officinalis were evaluated by HPLC-PDA. Chromatography was performed using a reversed-phase system with Luna $C_{18}$ (2) column and acetonitrile-water (50:50, v/v) with a flow rate of 1.0 mL/min under UV 280 nm. Under these conditions, the content of rubiadin-1-methyl ether was 0.013%.

  • Purpose: This study was conducted to investigate the muscle anti-fatigue effects of Morindae officinalis Radix liquid extract. Methods: 4-6 weeks old ICR rats are used in the study, and we administered the extracts powder of Morinda officinalis concentration of 1, 10, 100mg/0.3 ml to each to rats(MO group) once a day for each 30 and 60 days. After 30 and 60 days, we examined the plasma concentraion of lactate dehydrogenase, glucose. And we measured the persistent time of swimming exercise test and conducted grip strength test Results: We found that there are no significantly differences between the control and the treatment group in the plasma concentration of activity level of lactate dehydrogenase. And we found that in 30days group, level of glucose concentration is significant. In swimming exercise test, as administration time and concetration growed up, swimming time is increased. And in grip strength test, as concentraion is growed up, grip strength is increased. Conclusion: This study show that Morinda officinalis is effective against muscle fatigue.

Effects of Autoclaved Morinda officinalis Root Extract on the Suppressive Efficacy of MMP-1 Enzyme (고온고압 처리된 파극천 추출물의 MMP-1 발현 억제 효능 증진)

  • Morinda officinalis (Rubiaceae) is a medicinal herb that has traditionally been used for the treatment of skin inflammation. The present study was to investigate the inhibitory efficacy of matrix metalloproteinases-1 (MMP-1) of the extracts of the root of M. officinalis, which was autoclaved at $132^{\circ}C$ and $1.2kgf/cm^2$ for 15 min using an autoclave. The composition of the extracts were compared with that prepared without autoclaved treatment. Total phenol and flavonoid contents were analyzed for the autoclaved M. officinalis root extract (AME) and M. officinalis root extract (ME). Results showed that the autoclaved AME contained total phenol and flavonoid contents 1.5-fold times more than those from ME. AME showed DPPH and superoxide radical scavenging activities as 79.25% and 94.5%, respectively, at the concentration of $500{\mu}g/mL$. In anti-inflammatory assay, AME inhibited the activity of COX-2 and 5-LOX metabolites. In addition, AME showed higher an inhibition rate in MMP-1 expression than ME in UVA-irradiated human dermal fibroblast (HDF) without any significant cytotoxicity. UVB-induced cytotoxicity and cell death were effectively suppressed by AME. In conclusion, autoclaving the M. officinalis root increased the phenol and flavonoid contents. The extracts of the autoclaved M. officinalis enhanced the antioxidant, anti-inflammatory and anti-MMP-1 effects. Thus, the extracts could be an useful active ingredient in cosmetics.

  • The aim of this study was to explore the potent phosphodiesterase type 5 (PDE %) inhibitor from various herbal medicines for erectile dysfunctions. In this study, 61 herbal medicines, which were extracted with ethanol, have been investigated with PDE 5 assay using enzyme inhibitory activity on 22 species of herbal medicines. Of these, 5 species of herbal medicines, Cnidium monieri, Cuscuta chinensis, Epimedium koreanum, Morinda officinalis, and Tribulus terrestris were exhibited stronger inhibitory effect against phosphodiesterase 5 (PDE 5) among 61 species; Cnidium monieri ($IC_{50}=33.7{\mu}g/ml$), Cuscuta chinensis ($IC_{50}=65.7{\mu}g/ml$), Epimedium koreanum ($IC_{50}=90.3{\mu}g/ml$), Morinda officinalis ($IC_{50}=48.7{\mu}g/ml$) and Tribulus terrestris ($IC_{50}=32.5{\mu}g/ml$).

Study for the Standardization and Comparison by Processed Morindae Radix (파극천(巴戟天) 포제방법(?製方法)에 따른 품질표준화 연구)

  • Objectives : We have been used many herbal medicines after processing to improve the effect, decrease toxicity and side-effect, and change property. We have studied the physico-chemical change and HPLC pattern of Morindae Radix by means of processing method. Methods : This study was investigated the contents of loss on drying, residue on ignition, residue on acid insoluble ignition, 50% ethanol extract and HPLC pattern of Morindae Radix(Morinda officinalis How.) by processed and non-processed. We have conducted Morindae Radix and Damnacanthi Radix which is circulated in herbal medicine market by forgery. Processed Morindae Radix was prepared by heating of added to salt(SP), liquor(LP) and Glycyrrhizae Radix solution(GP) for 20-40 minutes. Results and Conclusions : From this analysis, we found that the content of 50% ethanol extract was increased by processing method. And we were detected distinguishable marker of processed and non-processed from Morindae Radix(Morinda officinalis How.) by HPLC pattern analysis.

Effects of Morinda officinalis (MDOF) on Inhibition of Impairment of Learning and Memory, and Acetylcholinesterase in Amnesia Mice (파극천(巴戟天)이 치매병태모델에 미치는 영향(影響))

  • Alzheimer's disease(AD) is progressive neurodegenerative disease, which is pathologically characterized by neuritic plaques and neurofibrillary tangles associated with the acetylcholinesterase, apolipoprotein E and butylcholinesterase, and by mutations in the presenilin genes PS1 and PS2, and amyloid precursor proteins (APP) overexpression. The present research is to examine the inhibition effect of MDOF on PS-1, PS-2 and APP overexpression by detected to Western blotting. To verify the effects of MDOF on cognitive deficits further, we tested it on the scopolamine-induced amnesia model of the mice using the Morris water maze tests, and there was ameliorative effects of memory impairment as a protection to scopolamine. MDOF only partially blocked the increase in blood serum level of acetylcholinesterase and Uric acid induced by scopolamine, whereas blood glucose level was shown to attenuate the amnesia induced by scopolamine and inreased extracelluar serum level compared with only scopolamine injection. In conclusion, studies of MDOF that has been know as anti-choline and inhibition ablilities of APP overexpression, this could also be used further as a important research data for a preventive and promising symptomatic treatment for Alzheimer's disease.

Anti-epilepsy Effect of Methanol Extract of Morinda officinalis Augments Pentylenetetrazol-Induced Convulsion Behaviors (파극천 추출물이 펜틸렌테트라졸로 유도된 실험동물에서 항 뇌전증 작용)

  • Morinda officinalis (MO) is a oriental medicinal herb which has been used traditionally for the treatment of impotence, anti-inflammatory, menstrual irregularity action and various brain diseases including antidepressant and anti-stress. In order to examine the mechanism of anticonvulsive effect, we treated the methanol extract of MO (100, 200 mg/kg, P.0) to the sleeping time and pentylenetetrazol (PTZ)-induced convulsive mice. The methanol extract of MO prolonged sleep time by pentobarbital. Dose-dependent of methanol extracts of MO were effected the concentration of GABA and GABA-T activity in the brain of PTZ-induced mice. Methanol extracts of MO significantly inhibited the convulsion state as well as the level of lipid peroxidation in the brain. The butanol and dichloromethane fraction of methanol extracts among the others effectively inhibited in vitro lipid peroxidation dose dependently ($5.0{\times}10^{-2}{\sim}20.0{\times}10^{-2}g/ml$).
