Comparative proteomics of the mixotrophic dinoflagellate Prorocentrum micans growing in different trophic modes

  • Protein profiles of a common mixotrophic dinoflagellate, Prorocentrum micans, growing autotrophically and mixotrophically (fed on the cryptophyte Rhodomonas salina) were compared using two-dimensional gel electrophoresis (2-DE) to determine if they vary in different trophic modes. Approximately 2.3% of the detected proteins were differentially expressed in the different trophic modes. Twelve proteins observed only in the mixotrophic condition had lower pI value (<5) than the fifteen proteins observed only in the autotrophic condition (>5). When the internal amino acid sequences of five selected proteins differentially expressed between autotrophic and mixotrophic conditions were analyzed using matrix-assisted laser desorption time-of-flight (MALDI-TOF) mass spectrometry, two proteins that were specifically expressed in the autotrophic condition showed homology to glyceraldehyde-3-phosphatase dehydrogenase (GAPDH) and a bacterial catalase. Three mixotrophy-specific proteins showed homology to certain hypothetical proteins from an insect and bacteria. These results suggested the presence of certain gene groups that are switched on and off according to the trophic mode of P. micans.

Influence of Organic Carbon Sources on Growth and Lipid Content of Marine Green Alga Dunaliella tertiolecta

  • This study investigated the potential use of various organic carbon sources (glucose, glycerol and acetate) and different concentrations of $CO_2$ for culturing marine microalga Dunaliella tertiolecta. Cell growth and lipid production were monitored under heterotrophic, mixotrophic and photoautotrophic modes of cultivation. D. tertiolecta showed the ability to grow under mixotrophic (acetate and glucose), heterotrophic (glucose) and photoautotrophic condition under high $CO_2$ concentration (15%). With all the organic carbon sources (glucose, glycerol and acetate) tested in this study, 1~5% acetate enhanced cell growth rate and lipid content, while higher concentrations of acetate (10% and 15%) were inhibitory and resulted in cell death.

유가식 배양에서 배양조건에 따른 Chlorella minutissima의 생육 및 지질생산 (Cell Growth and Lipid Production from Fed-batch Cultivation of Chlorella minutissima according to Culture Conditions)

  • 본 연구는 다양한 조건에서의 실험을 위한 C. minutissima의 500 mL flask의 종 배양을 통해 고농도의 균주 획득 후, 가장 효율적인 배양 방식을 보인 fed-batch를 통한 autotrophic, heterotrophic, mixotrophic 조건하에서 바이오디젤용 지질 생산의 최적 배양 조건을 찾아야했다. 먼저, fed-batch의 Heterotrophic 배양 시 유기 탄소 원으로 glucose의 가장 효율적인 안정한 초기농도가 10 g/L임을 확인했다. 이 후, 20 L 배양기를 이용한 batch, fed-batch의 heterotrophic 배양에서 fed-batch배양의 경우 지질 생성양이 1.62배 정도 더 높았으며, 최대 균체량 역시 4 (g-dry wt./L)정도 높으며, 또한 디젤용 지질로 적합한 $C_{18}$의 조성 역시 49%이상 차지하여, 옥외 대량 배양 시 유기탄소 원으로 적절한 glucose의 이용을 통한 효율적인 배양방법임을 알 수 있다. 이 결과를 바탕으로, glucose를 포함한 배지를 0.5 L/day로 공급하여 유가 식 배양을 각 배양 조건에 따라 비교 실시했다. 그 결과 mixotrophic의 경우 다른 배양 조건에 비해 각각 24 (g-dry wt./L)의 최대 군체 농도와 함께 43 (%, w/w)의 높은 지질 생성량을 보였다. 또한 생성되는 지질의 조성이 균체 당 바이오 디젤용 양질의 지질 생성이 가능하였다. 또한 fed-batch 배양 이후 0.047 (% lipid/g-dry wt./day)의 비 생산 속도를 유지하며, 일정 균체 농도를 유지하는 경우 지속적인 지질 생성이 가능함을 보여주었다. 이러한 세 가지 배양 조건하에서 지질 추출을 통한 지방산 조성을 살펴본 결과 mixotrophic 배양 하에 디젤용으로 잘 알려진 $C_{17}$, $C_{18}$의 지방산 조성은 각각 18%와 49%의 생성을 보여주었다. 이 결과들을 바탕으로 이 균주를 이용해 옥외 배양이나 배양 조에서 대량 배양이 가능함을 확인했으며, 특히, 옥외배양을 위한 혼합영양 배양조건하에 배양 공정의 scale-up에 대한 추가적인 연구를 통해 C. minutissima 미세조류로부터 바이오 디젤의 보다 경제적 생산이 가능하도록 하고자 한다.

  • Microalgae are functional foods because they contain special anti-aging inhibitors and other functional components, such as ecosapentaenoic acid (EPA), docosahexaenoic acid (DHA), and omega-3 polyunsaturated fatty acids. Many of these functional dietary components are absent in animals and terrestrial plants. Thus, microalgae are widely utilized in human functional foods and in the feed provided to farmed fish and terrestrial livestock. Many marine organisms consume microalgae, often because they are in an appropriate portion of the cell size spectrum, but also because of their nutritional content. The nutritional requirements of marine organisms differ from those of terrestrial animals. After hatching, marine animals need small live forage species that have high omega-3 polyunsaturated fatty acid contents, including EPA and DHA. Euglena cells have both plant and animal characteristics; they are motile, elliptical in shape, 15-500 μm in diameter, and have a valuable nutritional content. Mixotrophic cell cultivation provided the best growth rates and nutritional content. Diverse carbon (fructose, lactose, glucose, maltose and sucrose) and nitrogen (tryptone, peptone, yeast extract, urea and sodium glutamate) supported the growth of microalgae with high lipid contents. We found that the best carbon and nitrogen sources for the production of high quality Euglena cells were glucose (10 g L–1) and sodium glutamate (1.0 g L–1), respectively.

Optimal Temperature and Light Intensity for Improved Mixotrophic Metabolism of Chlorella sorokiniana Treating Livestock Wastewater

  • Mixotrophic microalgal growth gives a great premise for wastewater treatment based on photoautotrophic nutrient utilization and heterotrophic organic removal while producing renewable biomass. There remains a need for a control strategy to enrich them in a photobioreactor. This study performed a series of batch experiments using a mixotroph, Chlorella sorokiniana, to characterize optimal guidelines of mixotrophic growth based on a statistical design of the experiment. Using a central composite design, this study evaluated how temperature and light irradiance are associated with $CO_2$ capture and organic carbon respiration through biomass production and ammonia removal kinetics. By conducting regressions on the experimental data, response surfaces were created to suggest proper ranges of temperature and light irradiance that mixotrophs can beneficially use as two types of energy sources. The results identified that efficient mixotrophic metabolism of Chlorella sorokiniana for organics and inorganics occurs at the temperature of $30-40^{\circ}C$ and diurnal light condition of $150-200{\mu}mol\;E{\cdot}m^{-2}{\cdot}s^{-1}$. The optimal specific growth rate and ammonia removal rate were recorded as 0.51/d and 0.56/h on average, respectively, and the confirmation test verified that the organic removal rate was $105mg\;COD{\cdot}l^{-1}{\cdot}d^{-1}$. These results support the development of a viable option for sustainable treatment and effluent quality management of problematic livestock wastewater.

기내 배양환경이 거베라 유묘의 기외이식 후 생육에 미치는 영향 (Effects of Several Culture Conditions on in vivo Growth and Development in Gerbera hybrida)

  • 배양환경을 달리하여 생산한 배양묘 (타가영양묘, 혼합영양묘, 자가영양묘)를 기외 이식한 후에 생존율은 타가영양묘에 비해 혼합영양묘와 자가영양묘가 높았고 특히 자가영양묘는 97.6%로 가장 높았으며 정식 일수가 경과함에 따라 엽병장, 엽수, 엽면적은 자가영양묘가 타가영양묘와 혼합영양묘에 비하여 다소 좋았으나 유의성은 없었다. 정식 30일 후 유묘의 총광합성량은 자가영양묘가 154 mg$CO_2$.mgDW$^{-1}$h$^{-1}$로서 혼합영양묘 (148 mg$CO_2$.mgDW$^{-1}$h$^{-1}$)와 타가영양묘 (102 mg$CO_2$.mgDW$^{-1}$h$^{-1}$)보다 높았다. 그리고 개화시기는 자가 영양묘가 타가영양묘와 혼합영양묘에 비해 7~10일 정도가 빨랐고 엽병장, 엽수, 엽면적, 엽록소 함량 등도 양호하였다.

Enhanced Biomass and ${\gamma}$-Linolenic Acid Production of Mutant Strain Arthrospira platensis

  • A mutant of Arthrospira platensis PCC 9108, strain M9108, obtained by mutagenesis with UV treatment, was able to mixotrophically grow in an SOT medium containing 40 g of glucose/l. The biomass and specific growth rate of strain M9108 (4.10 g/l and 0.70/d) were 1.9-fold and 1.4-fold higher, respectively, than those of the wild type (2.21 g/l and 0.58/d) under mixotrophic culture condition. In addition, when compared with the wild type, the content of ${\gamma}$-linolenic acid (GLA) in the mutant was increased when glucose concentration was increased. Compared with the wild type, the GLA content of the mutant was 2-fold higher in autotrophic culture and about 3-fold higher in mixotrophic culture. Thus, the mutant appears to possess more efficient facility to assimilate and metabolize glucose and to produce more GLA than its wild-type strain.

글리세롤을 이용한 복합영양소에서 Chlorella sp., Nannochloris sp.와 Botryococcus braunii 의 바이오매스 생산량과 오일 함유량 비교 (Comparison of Biomass and Oil Content of Chlorella sp., Nannochloris sp., and Botryococcus braunii in the Mixotrophic Conditions using Glycerol)

  • The focus of this study was to observe the growth of Chlorella sp., Nannochloris sp., and Botryococcus braunii under mixotrophic conditions (i.e., added glycerol) with the aim of increasing the growth of biomass and algae oil content. A significant growth of biomass was obtained when grown in glycerol rich environment comparing to autotrophic conditions. 5 g/L glycerol yielded the highest biomass concentration for these strains. Mixotrophic conditions improved both the growth of the microalgae and the accumulation of triacylglycerols (TAGs). The maximum amount of TAG in Botryococcus braunii was reached in the growth medium with 10 g/L glycerol and Chlorella sp., Nannochloris sp. with 2 g/L glycerol. The content of saturated fatty acids of Chlorella sp., Nannochloris sp., and Botryococcus braunii was found to be 34.94, 14.23 and 13.39%, and the amount of unsaturated fatty acids was 65.06, 85.78 and 86.61% of total fatty acids, respectively. The fatty acid profiles of the oil for the culture possibility met the necessary requirements and are, therefore, promising resource for biofuel production.

Effects of temperature on the growth and ingestion rates of the newly described mixotrophic dinoflagellate Yihiella yeosuensis and its two optimal prey species

  • Water temperature is known to affect the growth and feeding of marine dinoflagellates. Each dinoflagellate species grows well at a certain optimal temperature but dies at very cold and hot temperatures. Thus, changes in water temperatures driven by global warming and extremely high or low temperatures can affect the distribution of dinoflagellates. Yihiella yeosuensis is a mixotrophic dinoflagellate that can feed on only the cryptophyte Teleaulax amphioxeia and the chlorophyte Pyramimonas sp. Furthermore, it grows fast mixotrophically but rarely grows photosynthetically. We explored the direct and indirect effects of water temperature on the growth and ingestion rates of Y. yeosuensis feeding on T. amphioxeia and the growth rates of T. amphioxeia and Pyramimonas sp. under 7 different water temperatures (5-35℃). Both the autotrophic and mixotrophic growth rates of Y. yeosuensis on T. amphioxeia were significantly affected by temperature. Under the mixotrophic and autotrophic conditions, Y. yeosuensis survived at 10-25℃, but died at 5℃ and ≥30℃. The maximum mixotrophic growth rate of Y. yeosuensis on T. amphioxeia (1.16 d-1) was achieved at 25℃, whereas the maximum autotrophic growth rate (0.16 d-1) was achieved at 15℃. The maximum ingestion rate of Y. yeosuensis on T. amphioxeia (0.24 ng C predator-1 d-1) was achieved at 25℃. The cells of T. amphioxeia survived at 10-25℃, but died at 5 and ≥30℃. The cells of Pyramimonas sp. survived at 5-25℃, but died at 30℃. The maximum growth rate of T. amphioxeia (0.72 d-1) and Pyramimonas sp. (0.75 d-1) was achieved at 25℃. The abundance of Y. yeosuensis is expected to be high at 25℃, at which its two prey species have their highest growth rates, whereas Y. yeosuensis is expected to be rare or absent at 5℃ or ≥30℃ at which its two prey species do not survive or grow. Therefore, temperature can directly or indirectly affect the population dynamics and distribution of Y. yeosuensis.
