Comparison of survival prediction models for pancreatic cancer: Cox model versus machine learning models

  • A survival prediction model has recently been developed to evaluate the prognosis of resected nonmetastatic pancreatic ductal adenocarcinoma based on a Cox model using two nationwide databases: Surveillance, Epidemiology and End Results (SEER) and Korea Tumor Registry System-Biliary Pancreas (KOTUS-BP). In this study, we applied two machine learning methods-random survival forests (RSF) and support vector machines (SVM)-for survival analysis and compared their prediction performance using the SEER and KOTUS-BP datasets. Three schemes were used for model development and evaluation. First, we utilized data from SEER for model development and used data from KOTUS-BP for external evaluation. Second, these two datasets were swapped by taking data from KOTUS-BP for model development and data from SEER for external evaluation. Finally, we mixed these two datasets half and half and utilized the mixed datasets for model development and validation. We used 9,624 patients from SEER and 3,281 patients from KOTUS-BP to construct a prediction model with seven covariates: age, sex, histologic differentiation, adjuvant treatment, resection margin status, and the American Joint Committee on Cancer 8th edition T-stage and N-stage. Comparing the three schemes, the performance of the Cox model, RSF, and SVM was better when using the mixed datasets than when using the unmixed datasets. When using the mixed datasets, the C-index, 1-year, 2-year, and 3-year time-dependent areas under the curve for the Cox model were 0.644, 0.698, 0.680, and 0.687, respectively. The Cox model performed slightly better than RSF and SVM.

Nitrification of low concentration ammonia nitrogen using zeolite biological aerated filter (ZBAF)

  • This study focuses on nitrification through a biological aerated filter (BAF) that is filled with a zeolite medium at low concentrations of ammonia. The zeolite medium consists of natural zeolite powder. The BAF is operated under two types of media, which are a ball-type zeolite medium and expanded poly propylene (EPP) medium. Nitrification occurred in the zeolite BAF (ZBAF) when the influent concentration of ammonia nitrogen was 3 mg L-1, but the BAF that was filled with an EPP medium did not experience nitrification. The ammonia nitrogen removal efficiency of ZBAF was 63.38% and the average nitrate nitrogen concentration was 1.746 mg/L. The ZBAF was tested again after a comparison experiment to treat pond water, and municipal wastewater mixed pond water. The ZBAF showed remarkable ammonia-nitrogen treatment at low concentration and low temperature. During this period, the average ammonia nitrogen removal efficiency was 64.56%. Especially, when water temperature decreased to 4.7℃, ammonia nitrogen removal efficiency remained 79%. On the other hand, the chemical-oxygen demand (COD) and phosphorus-removal trends were different. The COD and phosphorus did not show as efficient treatment as the ammonia-nitrogen treatment.

Analysis of Wear Properties for $Ni_{3}Al$ Layer coated on Ferrous Materials by Diffusion Treatment after Combustion Synthesis at low Temperature (저온 연소합성 후 확산 열처리한 $Ni_{3}Al$ 금속간화합물 코팅층의 미끄럼 마모거동)

  • Coating brittle intermetallic compounds on metal can enlarge the range of their use. It is found that intermetallic compound coating layers made by only combustion synthesis in an electric furnace have porous multi-phase structures containing several intermediate phases, even though the coating layers show good wear resistance. In this study, dense $Ni_{3}Al$ single phase layer corresponding to the initial composition of the mixed powder is coated on two different ferrous materials by the diffusing treatment after combustion synthesis. After- ward, sliding wear behaviors of the coating layer are evaluated in comparison with that of the coating layer with porous multi-phase structure made by only combustion synthesis. As a result, the wear properties of the coating layer composed of dense $Ni_{3}Al$ single phase are considerably improved at the range of low sliding speed com- pared with that of the coating layer with porous multi-phase structure, particularly in the running-in wear region. This is attributed to the fact that wear of the coating layer is progressed by shearing as a sequence of adhesion, not by occurring of pitting on the worn surface due to having dense structure without pores.

A Study on the Dyeing of PTT(polytrimethylene terephthalate)/Silk Mixture Fabrics with Disperse Dyes/Acid Dyes (분산염료/산성염료에 의한 PTT(polytrimethylene terephthalate)/견 교직물의 염색에 관한 연구)

  • The dyeing of poly(trimethylene terephthalate)(PTT)/silk mixture fabrics can be accomplished by a two bath dyeing method with separate application of the disperse dyes on the PTT, reduction cleaning of the stained silk and then dyeing the silk with the acid dyes, or by one bath dyeing method with mixed dye ranges, possibly followed by a cleaning treatment. The two bath dyeing method has the advantage of better results with respect to dry cleaning fastness properties thanks to the possibility of an intermediate reduction clear. On the other hand, as compared with the two bath dyeing method, one bath dyeing method with a mixed dye range permits rapid and more reproducible dyeing, without the risk of great difference with respect to the shade of the strike on both substrates as well as savings of time, energy and water usage. This study was carried out to investigate dyeing characteristics of PTT/silk mixture fabrics with disperse dyes/acid dyes by one bath dyeing method in comparison with two bath dyeing method in the interests for rationalization of the dyeing process.

Synthesis and characterization of polyamide thin-film nanocomposite membrane containing ZnO nanoparticles

  • We report in this study the synthesis of mixed matrix reverse osmosis membranes by interfacial polymerization (IP) of thin film nanocomposite (TFNC) on porous polysulfone supports (PS). This paper investigates the synthesis of ZnO nanoparticles (NPs) using the sol-gel processing technique and evaluates the performance of mixed matrix membranes reached by these aerogel NPs. Aqueous m-phenyl diamine (MPD) and organic trimesoyl chloride (TMC)-NPs mixture solutions were used in the IP process. The reaction of MPD and TMC at the interface of PS substrates resulted in the formation of the thin film composite (TFC). NPs of ZnO with a size of about 25 nm were used for the fabrication of the TFNC membranes. These membranes were characterized and evaluated in comparison with neat TFC ones. Their performances were evaluated based on the water permeability and salt rejection. Experimental results indicated that the NPs improved membrane performance under optimal concentration of NPs. By changing the content of the filler, better hydrophilicity was obtained; the contact angle was decreased from $74^{\circ}$ to $32^{\circ}$. Also, the permeate water flux was increased from 26 to 49 L/m2.h when the content of NPs is 0.1 (wt.%) with the maintaining of lower salt passage of 1%.

Effects of Sayeok-tang on Papain-Induced Osteoarthritis in Mice (Papain으로 유도된 골관절염 생쥐 모델에서 사역탕(四逆湯)의 항골관절염 효능에 관한 연구)

  • This study intends to clarify how Sayeok-tang(here in after reffered to SYT) affect C57BL/10 mice whose osteoarthritis was induced by papain. Osteoarthritis was induced by injecting papain in the knee joint of 3 groups(n=6) of mice. Normal group was non-treatment group and was not injected papain, whereas control mice were orally administered with $200{\mu}{\ell}$ of physiological saline. Positive comparison group was medicated with 100 mg/kg of Joins$^{(R)}$ mixed with $200{\mu}{\ell}$ of physiological saline. Experimental group was medicated with 400 mg/kg of SYT mixed with $200{\mu}{\ell}$ of physiological saline. Both Positive and experimental comparison groups were orally medicated once per day for 4 weeks. After the experiment, the functions of liver and kidney, inflammation cytokine values within serum, degree of revelation for inflammation cytokine genes, immune cells within blood, metabolism of arachidonic acid and amount of cartilage were measured and histopathological changes in the knee joint structures were observed. As results, SYT had no significant effect on the liver and kidney functions. Interleukin-$1{\beta}$(IL-$1{\beta}$), interleukin-6(IL-6), monocyte chemo attractant protein-1(MCP-1) and tumor necrosis factor-${\alpha}$(TNF-${\alpha}$) were significantly decreased. Inflammation cytokines in joints were all significantly decreased. Prostaglandin $E_2(PGE_2)$, thromboxane $B_2(TXB_2)$ were significantly decreased. Destruction of cartilage on micro computed tomography(CT)-arthrography was meaningfully decreased. In terms of histopathology, infiltration of inflammation, proliferation of synovial membrane, subsidence of cartilage and bone due to penetration of excessive formation of synovial cell and destruction of cartilage were small. Based on all results mentioned above, Sayeok-tang(SYT) is believed to be meaningful for suppressing the progress of osteoarthritis and its treatments because of its anti-inflammatory effects and alleviation of pain with histopathological effective efficacy.

The Assessment of Cattle Slurry Application on Productivity and Feed Values of Barley and Hairy Vetch Influenced by Sowing Methods (보리와 헤어리 베치의 단, 혼파 재배 시 우분뇨의 시용에 따른 생산성과 사료가치의 평가)

  • This trial was conducted to assess effects of cattle slurry application on productivity and feed values of barley and hairy vetch when they were influenced by single or mixed-sowed cultivation in paddy fields, and to obtain good quality of organic forage resources. The results summarized are as follows. For barley and hairy vetch, single-sowed cultivation was lower in annual dry matter (DM) and total digestible nutrients (TDN) yields than mixed-sowed (P<0.05). Although crude protein (CP) contents for the barley within single-sowed treatments were lowest as 6.5%, those of hairy vetch within the same sowed method were highest as 16.7%. However, mixed-sowed cultivation with barley and hairy vetch, showing 13.8% CP content, was significantly (P<0.05) higher than CP and relative feed value (RFV) of barley alone treatment. For barley alone treatment, cattle slurry application significantly increased annual DM and TDN yields in comparison with treatments of P+K fertilization as chemical fertilizers and no fertilizer as controls. Results from cattle slurry application showed 84% and 82% in contrast with chemical fertilizer for annual DM and TDN yields, respectively. For mixed-sowed cultivation with barley and hairy vetch, cattle slurry application showed 90% and 94% annual DM and TDN yields, respectively as compared with N+P+K fertilization as chemical fertilizers. Crude protein contents ($14.2{\sim}15.9%$) for cattle slurry application treatments were significantly (P<0.05) higher than those of other treatments. Moreover, cattle slurry application treatment had the highest TDN and RFV among treatments, showing $60.7{\sim}61.8%$ and $112.2{\sim}118.1$, respectively. For hairy vetch alone treatments, annual DM and TDN yields of cattle slurry alone application treatment were highest among fertilization treatments. Furthermore, CP, TDN and RFV of cattle slurry alone application treatments were significantly (P<0.05) higher than those of other treatments. The results showed that mixed-sowed cultivation rather than single-sowed for barley or hairy vetch improved their nutritive value and quality, and also within mixed-sowed cultivation, cattle slurry application increased production yield per ha and CP contents. In the application of above system to organic livestock farming, it would be expected that forages produced by cattle slurry application under mixed-sowed method might become a substitute for foreign organic grain as protein sources.

An Efficient Method for the Release of Recombinant Penicillin G Amidase from the Escherichia coli Periplasm (대장균의 periplasm으로부터 재조합 PGA 단백질의 효율적이고 간단한 방출 방법)

  • In this study, we report on a simple, efficient method for obtaining penicillin G amidase (PGA) from recombinant Escherichia coli using a formulation mixed with detergent and lysozyme. Research was conducted on the extraction efficiency of PGA from the periplasmic space in cells in terms of the type of detergent, detergent concentration, pH, reaction time, and temperature of permeabilization. The extraction yield of PGA in the formulated surfactant/lysozyme treatment was increased by approximately (55-65 U/ml) in comparison with that in the single surfactant treatment. The released PGA solution was concentrated and exchanged with buffer using an ultrafiltration (U/F) system. The yields of diatomite filtration, membrane filtration (M/F), and U/F were 69.7%, 93.8%, and 77.3%, respectively. A total of 212 KU of PGA was recovered. At the 25-L culture scale, the overall yield of extraction using the mixed surfactant/lysozyme method was 49.2%. The specific activity of extracted PGA was 11 U/mg in protein. The concentrated PGA solution was immobilized on microporous silica beads without further purification of PGA. The total immobilization yield of PGA on the resin was 48.7%, while the enzyme activity was 101 U/g. The immobilized PGA was successfully used to produce 6-APA from penicillin G. Our results indicated that a simple extraction method from periplasmic space in E. coli may be used for the commercial scale production of ${\beta}-lactam$ antibiotics using immobilized PGA.

  • This study was performed to assess the possibility of application of sewage sludge which is the side-product from domestic sewage treatment plant to the materials for biodiesels by investigating the yields and composition of the lipids and fatty acid methyl esters(FAMEs) from soxhlet extraction and in-situ transesterification. As the results, yields of in-situ transesterification were higher than soxhlet extraction. In comparison by sewage sludge type, yields of sewage sludge mixed nightsoil or livestock were higher than a single sewage sludge. And maximum yield showed up to 14 wt%. Fatty acid composition of extracted lipids and synthesized FAMEs consists of palmitic acid(C16:0), palmitoleic acid(C16:1), stearic acid(C18:0), oleic acid(C18:1), and linoleic acid(C18:2).

Inhibitory effects of calcium against intestinal cancer in human colon cancer cells and $Apc^{Min/+}$ mice

  • The aim of the study was to investigate the inhibitory effects of calcium against intestinal cancer in vitro and in vivo. We first investigated the effects of calcium treatment in HCT116 and HT29 human colon cancer cells. At the concentration range of 0.8-2.4 mM, calcium significantly inhibited cell growth (by 9-29%), attachment (by 12-26%), invasion (by 15-31%), and migration (by 19-61%). An immunofluorescence microscope analysis showed that the treatment with calcium (1.6 mM) for 24 h increased plasma membrane ${\beta}$-catenin but decreased nuclear ${\beta}$-catenin levels in HT29 cells. We then investigated the effect of dietary calcium on intestinal tumorigenesis in $Apc^{Min/+}$ mice. Mice received dietary treatment starting at 6 weeks of age for the consecutive 8 weeks. The basal control diet contained high-fat (20% mixed lipids by weight) and low-calcium (1.4 mg/g diet) to mimic the average Western diet, while the treatment diet contained an enriched level of calcium (5.2 mg calcium/g diet). The dietary calcium treatment decreased the total number of small intestinal tumors (by 31.4%; P < 0.05). The largest decrease was in tumors which were ${\geq}$ 2 mm in diameter, showing a 75.6% inhibition in the small intestinal tumor multiplicity (P < 0.001). Immunohistochemical analysis showed significantly reduced nuclear staining of ${\beta}$-catenin (expressed as nuclear positivity), but increased plasma membrane staining of ${\beta}$-catenin, in the adenomas from the calcium-treated groups in comparison to those from the control group (P < 0.001). These results demonstrate intestinal cancer inhibitory effects of calcium both in human colon cancer cells and $Apc^{Min/+}$ mice. The decreased ${\beta}$-catenin nuclear localization caused by the calcium treatment may contribute to the inhibitory action.
