Polymorphism of Carp $\beta$-globin Gene on Chromosome (잉어 $\beta$-globin 유전자의 염색체상에서의 다형해석)

  • Common DNA fragments of the ${\beta}$-globin gene were observed from six races of the adult common carp: Hybrid-Yamato, Japanese wild type, Mirror, Suwa-Yamato, Scale German, and Saku-Yamato. Chromosomal DNAs isolated from the above six races were digested with restriction endonucleased EcoRI and PstI. The digested fragments were transferred onto nitrocellulose filter and hybridized with a probe of carp ${\beta}$-globin cDNA. Molecular sizes of the hybridized DNA fragments digested with EcoRI were 3.6Kb(Kilo base), 4.3Kb and 15Kb in Hybrid-Yamato, Japanese wild type, Mirror, Scale German and Saku-Yamato carp DNAs. In Scale German and Saku-Yamato carp DNAs, two and one more hybridized DNA fragments were observed, respectively. Molecular sizes of the hybridized DNA fragments digested with PstI were 2.2Kb, 6.5Kb, 7.8Kb and 9.2Kb in Hybrid-Yamato, 2.2Kb, 6.5Kb and 9.2Kb in Japanese wild type, 2.2Kb, 6.5Kb, 7.8Kb, and 13Kb in Mirror, 2,2Kb, 5,5Kb, 6.5Kb, 7.8Kb, 9.2Kb and13Kb in Scale German, and 2.2Kb, 5.5Kb, 6.5Kb, 9.2Kb and Saku-Yamato carp DNA. Therefore, depending on carps, three to six DNA fragments were hybridized with ${\beta}$-globin gene probe. Thus it indicated polymorphysm in the globin gene family of carp.

Effects on Lethal Concentration 50%, Hematological Parameters and Plasma Components of Mirror carp, Cyprinus carpio nudus Exposed to Waterborne Nitrate (수인성 질산 급성 노출에 의한 향어, Cyprinus carpio nudus의 반수치사농도, 혈액학적 성상 및 혈장성분에 미치는 독성 영향)

  • To determine the toxic effects of the mirror carp, Cyprinus carpio nudus on waterborne nitric acid exposure, they were exposed to waterborne nitric acid at 0, 4, 20, 100, 500, and 2,500 mg NO3-/L for 96 h. The semi-lethal concentration (LC50) of the scented fish by 96 hours of acute exposure to nitric acid was 1,433.54 mg NO3-/L. The physiological changes of waterborne nitric acid exposure on mirror carp were evaluated through hematologic properties, and RBC count showed a significantly decreased (p < 0.05). The change in the ion control ability of the mirror carp according to exposure to waterborne nitric acid was evaluated through the plasma inorganic component, and calcium and magnesium were significantly decreased (p < 0.05) in the plasma inorganic component. The health and stress status of the mirror carp due to exposure to waterborne nitric acid were evaluated through plasma organic ingredients, and the plasma organic component glucose showed a significantly increase (p < 0.05). Changes in liver damage and enzyme activity due to waterborne nitric acid exposure were evaluated through plasma enzyme components, the enzymatic activities of plasma AST, ALT, and ALP were significantly decreased (p < 0.05) in high levels of waterborne nitric acid exposure. The results of the present study suggest that waterborne nitric acid exposure to C. carpio nudus can toxicly affect survival, hematologic properties, and plasma components.

  • Mirror carp, Cyprinus carpio nudus (weight 26.2±3.1 g, length 11.8±0.5 cm) were exposed to waterborne total ammonia nitrogen (TAN) at 0, 2, 4, 8, 16, 32, 64 and 128 mg TAN/L. The lethal concentration 50 (LC50) of mirror carp, C. carpio nudus exposed to waterborne ammonia was 60.38 mg TAN/L. The red blood cell (RBC) count was significantly reduced by ammonia exposure, whereas there was no significant changes in the hemoglobin concentration and hematocrit value. In the plasma components, glucose, total protein, aspartate aminotransferase (AST) and alkaline phosphatase (ALP) were significantly increased by ammonia exposure. The results of this study suggest that the ammonia exposure to C. carpio nudus affects the survival rates, hematological parameters and plasma components as toxicity.

Growth Comparison of Israeli Carp (Cyprinus carpio) to Different Breeding Combination

  • The purpose of this study was to investigate the improvement of growth in Israeli carp (Cyprinus carpio), and the cross experiment was carried out with two strains of Israeli carp. Four combinations of Israeli carp from Jeonbuk fisheries farm and Songpu mirror carp from Heilong Jiang, China (KK; Jeonbuk ♀ ${\times}$ Jeonbuk ♂, KC; Jeonbuk ♀ ${\times}$ China ♂, CC; China ♀ ${\times}$ China ♂ and CK; China ♀ ${\times}$ Jeonbuk ♂) were developed and reared. Body length, body weight and condition factor were determined at 20, 40, 60 and 170 days post-hatch (DPH). The results showed that there were differences in growth rate of the four groups. Body length of four groups were CK > CC > KC > KK and body weight were CC > CK > KC > KK at 170 DPH. The growth perfomance of four groups were statistically significant difference (P<0.05). During the rearing, CC group had longer length and higher weight at 170 DPH compared to other three groups and also condition factor was highest in the CC group, but there was no significant difference in a survival rate. These results indicated that the growth performance mainly depended upon brooder combination but survival rate could not significantly affect brooder.

Toxic Effects of Waterborne Copper Exposure on the Hematological Parameters and Plasma Components of Mirror Carp Cyprinus carpio nudus (수인성 구리 급성노출에 의한 향어(Cyprinus carpio nudus)의 혈액학적 성상 및 혈장성분의 독성영향)

  • Mirror carp Cyprinus carpio nudus (weight 42.0±3.8 g, length 14.3±0.4 cm) were exposed to different concentrations of waterborne copper (0, 100, 200, 400, 800, and 1,600 ㎍ Cu2+·L-1) at 20.3℃ for 96 h. The lethal concentration 50 of waterborne copper was 1,176.45 ㎍ Cu2+·L-1. Among hematological parameters, red blood cell count was significantly decreased, whereas there were no significant changes in the hemoglobin concentration and hematocrit value. Among the inorganic plasma components, calcium was significantly decreased following copper exposure. Conversely, organic plasma components such as glucose and total protein were significantly increased. Similarly, enzymatic components, such as aspartate transaminase, alanine transaminase, and alkaline phosphatase, were also significantly increased. These findings suggest that the copper exposure is detrimental to the survival rates and physiology of C. carpio nudus.

  • Jinyangho is a natural water supply source of tap water in west Gyeongnam area, but mass mortality of crusian carp occurred during the time of temperature rise in spring. Examinations on diseased fishes were able to isolate four bacteria isolates and then identified the bacteria as a member of Aeromonas sp. Challenge experiment with mirror carp (Gyrinus carpiospecilaris) was proved the virulence, the isolates were in turn believed as the causative agent of mass mortality in Jinyangho.

Assessment Selective Breeding Effect of Israeli carp (Cyprinus carpio) from Korea (국내 이스라엘 잉어의 선발육종효과 평가)

  • Since the introduction of Israeli carp into Korea for farming in 1973, there are no breeding studies on developing Korea Israeli carp (domestic) so far. This study performed gene-based cross-breeding studies to restore genetic diversity of lowered Israeli carp through continuous inbreeding, and for rapid growth and better scales. This study produced four cross-breeding groups (F1) using Koean Israeli carp and Chinese Songpu mirror carp for the improvement of growth and scale of Israeli carp in Korea. And mating scheme for breeding groups was set in consideration of the morphological analysis and genetic distance of broodstock. In addition, this study used microsatellite markers and genotype data to analyze genetic diversity and parentage analysis. As a result, the average NA and HE values of Korean select broodstock are 8.3 and 0.743, and F1 is 13.0 and 0.764. This study shows that the genetic diversity of F1 has been recovered over Korean Israeli carp through breeding between Korean Israeli carp and Chinese Songpu mirror carp. Common Israeli carp in Korea reached 1.7 kg in 17 months, and improved Israeli carp reached to 2.2 kg. The KC (Korea×China, KC) group was 2.52 and broodstock group was 3.15. F1 showed lower scale score (0.63) than broodstock. The improved carp (F1; CK, KC) had 20% better scales than the parent group (F0), which improved 27% in weight and 25% in scales compared to common Israeli carp. The Israeli carp developed by the genetics-based breeding grew quicker and had improved genetic diversity and fewer scales, which will be of great value for Korean Israeli aquaculture industry due to good marketability.

Antibacterial efficacy and safety of copper sulfate pentahydrate to cultured fish (양식 어류에 있어서 황산동(copper sulfate pentahydrate)의 안전성과 살균효과에 대한 연구)

  • Antibacterial efficacy and safety of copper sulfate pentahydrate (5% solution) was evaluated in laboratory and field studies for disinfection of some fish bacterial pathogens in cultured fish, flounder(Paralichthys olivaceus), mirror carp(Cyprinus carpio), gold fish(Carrassius auratus) and eel(Anguilla japonica). Dipping of fishes for acute toxicity was performed for a period of 48h, and the TLm value(median tolerance limit) were 4.0ppm in mirror carp and flounder, 4.4ppm in rainbow trout, and 10.0ppm in gold fish. After exposure below than TLm value for 1 month, no side effects and histopathological changes were observed. The test drug were shown high antibacterial activities against most bacterial pathogens by exposure in the laboratory and field trials. However, Aeromonas salmonicida showed a resistance to the dosage. We can prevent outbreaks of bacterial diseases of fish by the dosage of 1ppm for 6hr and the test drug is thought to be used effectively and widely as a disinfectant against most bacterial fish pathogens.

Comparisons of amino acid sequences of ${\beta}$-globin gene between carp and other vertebrates (잉어와 척추동물들의 ${\beta}$-globin 아미노산배열의 비교)

  • The purpose of this study was to understand the evolutionary relationships between fish and other vertebrates which had DNA with the genetic defects in homoglobin expression, with comparison to the nucleotide homologies of the ${\beta}$-globin genes. The predicted amino acid sequence from carp ${\beta}$-globin gene was compared with those of other vertebrates from the published data. The nucleotide homologies of the predicted amino acid sequence from the carp ${\beta}$-globin gene with those of goldfish and mirror carp were high, and the rates were 97.3% and 93.9%, respectively. On the other hand, with the previously reported ${\beta}$-globins of goat, frog, human, rat, goose, chicken, and duck, it showed low homology ranging from 45.9 to 58.1%. The carp ${\beta}$-globin has one inserted amino acid residue, which was also found in other fish ${\beta}$globin, but not in other vertebrate ${\beta}$-globins.

  • A novel wet release process ($\mu$ CARP - Micro Channel Assisted Release Process) for releasing an extreme large-area plate structure without etching hole is proposed and experimented. Etching holes in conventional process reduce a effective area and degrade an optical characteristics by a diffraction. In addition, as the area of a released structure increases, the stietion becomes more serious. The proposed process resolves these problems by the introduction of a micro fluidic channel beneath the structure which will be released. In this paper, a 5 mm${\times}$5mm-single crystal silicon plate structure was released by the proposed $\mu$CARP without etch holes on the structure. The variation in etching time with respect to the of the introduced micro channel is also examined. This process is expected to be beneficial for the actuator of a nano-scale data storage and the scanning mirror.
