Assessment on Antioxidant Properties of Oplopanax elatus Nakai in vitro

  • Effects of Oplopanax elatus have known to various pharmaceutical therapies. However, chemical prosperities in the plant are rarely investigated. In order to detect biological activity, we evaluated the antioxidant activity of five fractions from methanolic extracts in each part of O. elatus. Also, contents of polyphenols and polysaccharides were measured. Five fractions were of sub-fractions using n-hexane, ethyl-acetate, n-butanol, and water from methanolic extracts. Ethylacetate and n-butanol fractions from stem and root exhibited strong antioxidant activity and high total phenolics content. On the HPLC analysis, ten free phenolics, including p-hydroxybenzoic acid, chlorogenic acid, caffeic acid, syringic acid, p-cou maric acid, ferulic acid, naringin, hesperidin, quercetin and trans-cinnamic acid, were identified from the fractions and were shown to different quantitative proportions. Furthermore, ethylacetate and n-butanol fraction had the highest amount of chlorogenic acids, one of the cinnamic acid derivatives possessing pharmacological properties. These results indicated that the fractions of O. elatus, as well as methanolic extracts, could be used as natural antioxidative ingredients.

Antioxidant and Antiproliferative Activities of Methanolic Extracts from Thirty Korean Medicinal Plants

  • To study the health promoting effects of medicinal plants, 30 medicinal plants commonly available in Korea have been evaluated for their antioxidant compounds and antioxidant and antiproliferative activities. Total polyphenolics and flavonoids in the methanolic extracts were measured by spectrophotometric methods and 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging activities and chelating effects have been determined for antioxidant activities. Moreover, the effects of medicinal plants on cell proliferation of intestinal (Caco-2) and pituitary (GH3) tumor cells were investigated using thiazolyl blue terazolium bromide (MIT) assay. The methanolic extracts of Pueraria thunbergiana and Artemisiae asiatria contained the highest total polyphenolic and flavonoid contents, respectively. P. thunbergiana exhibited the highest antioxidant activities. A. asiatria showed the strongest antiproliferative activity against Caco-2 and Ponciruc trifoliata Rafin and Lophathrum gracile Bronghiart exhibited the highest activities against GH3. Although there was positive correlation between ABTS radical scavenging activity and polyphenolic contents ($R^2=8189$), no relationship was found between antiproliferative and antioxidant activities.

Potential use of ultrasound in antioxidant extraction from Ecklonia cava

  • Water and methanolic extracts of Ecklonia cava, a marine brown alga, were prepared by ultrasonic extraction (UE) and conventional extraction (CE) methods. The radical-scavenging activity and the inhibitory effects against hydrogen peroxide ($H_2O_2$)-induced DNA damage of the extracts were investigated. All extracts prepared by CE exhibited higher total polyphenolic content than that in the extracts prepared by UE. Extraction yield and total phenolic content increased as the UE time increased. The radical-scavenging activities increased as the UE time increased. All extracts prepared by CE exhibited higher 1,1-diphenyl-2-pricrylhydrazyl (DPPH) and hydroxyl radical-scavenging activities than did those prepared by UE. Extracts prepared by UE showed stronger scavenging activities on alkyl radical and $H_2O_2$ than those prepared by CE did. Methanolic extract with UE 12 h (100MEU-12h) and methanolic extract with CE 24 h (100MEC-24h) were selected and evaluated by comet assay for their inhibitory effect against $H_2O_2$-induced DNA damage. 100MEU-12h showed slightly greater protective effect against $H_2O_2$-induced DNA damage than 100MEC-24h. Thus, UE can be effectively used as a seaweed extraction technique, and there is potential for scale-up of the extraction process.

포도씨 추출물과 분획물의 Tyrosinase 저해활성 (Inhibitory Effect of Methanol Extract and Its Fractions from Grape Seeds on Mushroom Tyrosinase)

  • 본 연구에서는 포도 가공 부산물인 포도씨를 이용하여 피부노화 및 색소침착에 관련된 tyrosinase 저해활성을 알아보고자 하였으며 폴리페놀 화합물과 총항산화력과의 상관관계를 비교·분석하고자 하였다. 포도씨 80% 메탄올 추출물은 hexane, chloroform, ethyl acetate, water 층의 순서로 분획 되었다. Tyrosinase 저해활성은 각 분획물보다 crude한 형태인 methanol 추출물의 효과가 더 높았으며 분획물 중 ethyl acetate 층이 가장 높은 tyrosinase 저해활성을 나타내었고 추출물과 분획물 모두 농도 의존적인 경향을 나타내었다. 포도씨 분획물 중 ethyl acetate층이 polyphenol과 flavonoid 함량이 가장 높았고 ABTS 라디칼 소거능 또한 가장 높은 결과를 나타내었으며 water 분획층도 비교적 높은 활성을 나타내었다. 본 연구 결과 tyrosinase 활성과 항산화 성분,항산화 활성 경향이 일치하는 것을 볼 수 있었으며 포도씨는 미백, 항노화 효과로 화장품 산업에서 천연 물질로 잠재적인 기능성을 가지고 있을 것으로 사료된다.

활성산소 소거활성에 따른 꾸지뽕잎 추출물의 신경세포 보호 효과 (Neuroprotective Effect according to Reactive Oxygen Species Scavenging Activity from Extracts of Cudrania tricuspidata Leaves)

  • In an attempt to identify the neuroprotective effect of Cudrania tricuspidata (CT) leaves against ROS (reactive oxygen species)-induced oxidative stress in neuronal cells, the extracts from CT leaves were investigated using PC12 cells and N18-RE-105 cells. The methanolic and ethanolic extracts from CT were denoted as CTM (Cudrania tricuspidata Leaves methanolic extracts) and CTE (Cudrania tricuspidata Leaves ethanolic extracts), respectively. The neuroprotective effects of the extracts were measured by DCF-DA assay, MTT reduction assay, and LDH release assay. The PC12 cells exposed to $H_2O_2$-induced oxidative stress and the N18-RE-105 cells exposed to glutamate-induced oxidative stress were treated with various concentrations of CTM and CTE. The results, CTM treatments resulted in the induction of a dose-dependent protective effect in PC12 cells and N18-RE-105 cells. Interestingly, CTE also showed neuroprotective effect in PC12 cells and N18-RE-105 cells. Therefore, these results suggest that CTM and CTE could be a new potential candidate as neuroprotective agents against ROS-induced oxidative stress in neuronal cells.

Methanolic Extract of Asterina pectinifera inhibits LPS-Induced Inflammatory Mediators in Murine Macrophage

  • This study aimed to elucidate anti-inflammatory activities from extracts of Asterina pectinifera on nitric oxide (NO) production, TNF-${\alpha}$ and IL-6 release in lipopolysaccharide (LPS)-stimulated murine macrophage cell, RAW264.7. We prepared the methanolic extracts (60-MAP, 70-MAP, 80-MAP and 90-MAP), aqueous extract (W-AP) and functional bioactive compound fraction (He-AP and EA-AP) from Asterina pectinifera according to extract method. The 60-MAP, 70-MAP, 80-MAP, 90-MAP and W-AP were significantly suppressed LPS-induced production NO, TNF-${\alpha}$ and IL-6 secretion in a concentration-dependent manner (P < 0.05). Especially, 80-MAP by extracted 80% methanol had the strongest activity in reduction of inflammatory mediators among these extracts. Indeed, to identify active fraction, which contained potential bioactive compounds, from 80-MAP of Asterina pectinifera, we tested anti-inflammatory activity of the He-AP or the EA-AP. The He-AP was next extracted from 80-MAP and the EA-AP were extracted from the other methanol layer except the He-AP. The EA-AP demonstrated a strong anti-inflammatory effect through its ability to reduce NO production and it also inhibited the production of proinflammatory cytokines such as IL-6 and TNF-${\alpha}$ at low concentration. These results suggested that the methanolic extract from Asterina pectinifera had the potential inhibitory effects on the production of these inflammatory mediators.

Carbohydrase inhibition and anti-cancerous and free radical scavenging properties along with DNA and protein protection ability of methanolic root extracts of Rumex crispus

  • The study elucidated carbohydrase inhibition, anti-cancerous, free radical scavenging properties and also investigated the DNA and protein protection abilities of methanolic root extract of Rumex crispus (RERC). For this purpose, pulverized roots of Rumex crispus was extracted in methanol (80% and absolute conc.) for 3 hrs for $60^{\circ}C$ and filtered and evaporated with vacuum rotary evaporator. RERC showed high phenolic content ($211{\mu}g$/GAE equivalent) and strong 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging ($IC_{50}$ = 42.86 (absolute methanol) and $36.91{\mu}g/mL$ (80% methanolic extract)) and reduced power ability. Furthermore, RERC exhibited significant protective ability in $H_2O_2/Fe^{3+}$/ascorbic acid-induced protein or DNA damage and percentage inhibition of the HT-29 cell growth rate following 80% methanolic RERC exposure at $400{\mu}g/mL$ was observed to be highest ($10.2%{\pm}1.03$). Moreover, methanolic RERC inhibited ${\alpha}$-glucosidase and amylase effectively and significantly (P < 0.05). Conclusively, RERC could be considered as potent carbohydrase inhibitor, anti-cancerous and anti-oxidant.

Antifilarial and Antibiotic Activities of Methanolic Extracts of Melaleuca cajuputi Flowers

  • We evaluated the activity of methanolic extracts of Melaleuca cajuputi flowers against the filarial worm Brugia pahangi and its bacterial endosymbiont Wolbachia. Anti-Wolbachia activity was measured in worms and in Aedes albopictus Aa23 cells by PCR, electron microscopy, and other biological assays. In particular, microfilarial release, worm motility, and viability were determined. M. cajuputi flower extracts were found to significantly reduce Wolbachia endosymbionts in Aa23 cells, Wolbachia surface protein, and microfilarial release, as well as the viability and motility of adult worms. Anti-Wolbachia activity was further confirmed by observation of degraded and phagocytized Wolbachia in worms treated with the flower extracts. The data provided in vitro and in vivo evidence that M. cajuputi flower extracts inhibit Wolbachia, an activity that may be exploited as an alternative strategy to treat human lymphatic filariasis.

수종 생약의 티로시나제 억제효과 (Tyrosinase Inhibition Activity of Some Herbal Drugs)

  • To isolate biologically active compounds which exhibit tyrosinase inhibition activity and ultimately express skin whitening effect, 14 oriental herbal drugs were screened in ter ms of tyrosinase inhibition. For this purpose, in vitro enzyme assay system for tyrosinase, so called Pomerantz method with some modifications has been established. Crude methanolic extracts from 14 herbal drugs were made and examined for their inhibitory activity against tyrosinase. Those extracts from Cnidii Rhizoma, Arecae Semen, Caryophylli Flos, and Ephedrae Herba showed strong inhibitory activities on mushroom tyrosinase. Therefore, crude methanolic extracts from those 4 herbal drugs were further fractionated using ether, butanol and water. respectively. The ether and n-butanol extracts from Arecae Semen and the n-butanol and water extracts from Caryophylli Flos, respectively, showed relatively strong tyrosinase inhibitory activity compared to arbutin.

B16 Melanoma Cell에서 티로시나아제 유전자 발현에 황금(Scutellaria baicalensis) 추출물이 미치는 효과 (Effects of Scutellaria baicalensis Extracts on Tyrosinase Gene Expression in B16 Melanoma Cells)

  • 유전자 발현 조절 수준에서 멜라닌 색소의 생합성을 조절하는 천연물질을 탐색하고자 황금으로부터 유용성 물질을 추출 및 분획하여 티로시나아제 프로모터 부위를 도입하여 형질전환된 B16 melanoma cell에 처리한 결과, 그 메탄올 추출물의 티로시나아제 유전자 발현율은 $10\;{\mu}g/m{\ell}$$100\;{\mu}g/m{\ell}$의 농도에서 대조군 세포에 비해 각각 약 231과 256%로 매우 크게 증진하는 효과를 보여 주었다. 극성도가 서로 다른 용매 분획물들을 형질전환된 세포에 처리하였을 때 methylene chloride와 ethyl acetate 용매 분획물들은 $10\;{\mu}g/m{\ell}$의 농도에서, butyl alcohol 용매 분획물은 $10\;{\mu}g/m{\ell}$$100\;{\mu}g/m{\ell}$의 농도에서, 그리고 물 용매 분획물은 $500\;{\mu}g/m{\ell}$의 고농도에서 티로시나아제 유전자의 발현을 증진시키는 효과를 보여 주었다. 그러나 그들의 티로시나아제 발현 증진 효과는 메탄올 추출물보다 낮았다. 황금 메탄올 추출물을 B16 melanoma cell에 처리한 다음 MTT assay를 실시한 결과, $10\;{\mu}g/m{\ell}$$100\;{\mu}g/m{\ell}$의 농도에서는 대조군 세포와 세포활성능이 유사할 정도로 세포독성이 매우 낮게 나타났다. 그러나 그 이상의 농도에서는 세포 활성능이 현저하게 낮았으며, 특히 $1,000\;{\mu}g/m{\ell}$의 농도에서는 세포 독성이 심하여 세포의 활성능을 측정하는 것이 불가능하였다. 본 연구결과, 황금 메탄올 추출물 내에는 티로시나아제 유전자의 발현을 증진시키는 데에 관여하는 성분들이 함유되어 있는 것으로 추측된다. 따라서 황금 추출물은 멜라닌 색소의 생합성에 관여하는 유전자의 발현하는 증진시키는 목적으로 이용함이 바람직하며, 이를 산업적으로 응용하기 위해서는 앞으로도 보다 많은 연구가 필요하다.
