Resonance May Elucidate New Bone Formation Induced by Low amplitude and High frequency Mechanical Stimuli (고주파 미세자극에 의한 뼈의 생성에 관한 모델링)

  • Bone is a self-assembly material. It is known that the low amplitude and high frequency mechanical stimulus, which is much less amplitude but higher frequency than those induced by the normal activity, can induce new bone formation. The vibrating resonance is employed to elucidate why new bone is formed by this kind of mechanical stimulus. For example, as 30 Hz and $5{\mu}{\epsilon}$ mechanical stimulus is applied at the wall of canaliculus (the tiny tube type pathway of bone fluid flow and the diameter of canaliculus is less than 200nm), the osteocytic cell membrane experiences $1,000{\mu}{\epsilon}$ strain due to the vibrating resonance. Two experiments will follow after this pilot study; (1) observing the MAPK pathway of osteocytes by using in-vitro cell culture and (2) visualizing the actin filament network in the osteocytes by using the imaging technique, such as confocal laser scanning microscope.

Development of Measurement Equipment of Membrane Stress Using White Noise Sound Wave (화이트 노이즈 음파를 이용한 막구조물의 장력 측정장치 개발)

  • One of the most important matters in keeping membrane structures in healthy condition is to maintain the proper tension distribution over the membrane. However, it is not easy to know the real stress level in the membrane quantitatively after completion of the structures. Authors suggested measurement method that can measure membrane stress using sound wave, and have been holding experimental tests of membrane stress measurement that used the sound external excitation with sine wave and white noise. The concept of the method is the fact that measurement of resonance frequency by vibrating membrane having rectangular boundary by audible frequency can measure membrane stress indirectly. In this paper, through the experimental tests it is proved that the equipment can be used for not only the membrane material of type A but also for types B and C. In addition, it is proved that the developed measurement equipment is available to stably measure the membrane stress which exists in the membrane material of the actual membrane structures.

Localization of the Membrane Interaction Sites of Pal-like Protein, HI0381 of Haemophilus influenzae

  • HI0381 of Haemophilus influenzae was investigated by circular dichroism (CD) and nuclear magnetic resonance (NMR) spectroscopy. HI0381 is a 153-residue peptidoglycan-associated outer membrane lipoprotein, and a part of the larger Tol/Pal network. Here, we report its backbone $^1H$, $^{15}N$, and $^{13}C$ resonance assignments, and secondary structure predictions. About 97% of all of the $^1HN$, $^{15}N$, $^{13}CO$, $^{13}C{\alpha}$, and $^{13}C{\beta}$ resonances covering 131 non-proline residues of the 134 residue, mature protein, were clarified by sequential and specific assignments. CSI and TALOS analyses revealed that HI0381 contains five ${\alpha}$-helices and five ${\beta}$-strands. To characterize the structure of HI0381, the effects of pH and salt concentration were investigated by CD. In addition, the structural changes occurring when HI0381 was in a membranous environment were investigated by comparing its HSQC spectra and CD data in buffer and in DPC micelles; the results showed that helix ${\alpha}4$ and strand ${\beta}4$ became aligned with the membrane. We conclude that the conformation of HI0381 is affected by the membrane environment, implying that its folded state is directly related to its function.

Preparation of Spherical TiO2 Nanoparticles Using Amphiphilic PCZ-r-PEG Random Copolymer Template Membrane (양친성 PCZ-r-PEG 랜덤 공중합체 분리막을 이용한 구형 이산화티타늄 나노입자의 제조)

  • Amphiphilic PCZ-r-PEG random copolymer assisted solvothermal process is used to prepare mesoporous $TiO_2$ microspheres generated from nanoparticles by self-assembly method. Synthesized PCZ-r-PEG is characterized by Fourier transform infrared spectroscopy (FT-IR), nuclear magnetic resonance (NMR), gel permeation chromatography (GPC) and transmission electron microscopy (TEM). The mesoporous $TiO_2$ are prepared by PCZ-r-PEG, glucose, water in tertrahydrofuran solution at $150^{\circ}C$ for 12 h and the $TiO_2$ microspheres are calcined at $550^{\circ}C$ for 30 min to further crystallize and organic residue are removed. Morphology and crystallization phase is characterized by scanning electron microscopy (SEM) and X-ray diffraction (XRD) respectively. The mesoporous $TiO_2$ crystallized in pure anatase phase with diameter of $300{\pm}20nm$.

Parametric resonance of composite skew plate under non-uniform in-plane loading

  • Parametric resonance of shear deformable composite skew plates subjected to non-uniform (parabolic) and linearly varying periodic edge loading is studied for different boundary conditions. The skew plate structural model is based on higher order shear deformation theory (HSDT), which accurately predicts the numerical results for thick skew plate. The total energy functional is derived for the skew plates from total potential energy and kinetic energy of the plate. The strain energy which is the part of total potential energy contains membrane energy, bending energy, additional bending energy due to additional change in curvature and shear energy due to shear deformation, respectively. The total energy functional is solved using Rayleigh-Ritz method in conjunction with boundary characteristics orthonormal polynomials (BCOPs) functions. The orthonormal polynomials are generated for unit square domain using Gram-Schmidt orthogonalization process. Bolotin method is followed to obtain the boundaries of parametric resonance region with higher order approximation. These boundaries are traced by the periodic solution of Mathieu-Hill equations with period T and 2T. Effect of various parameters like skew angle, span-to-thickness ratio, aspect ratio, boundary conditions, static load factor on parametric resonance of skew plate have been investigated. The investigation also includes influence of different types of linearly varying loading and parabolically varying bi-axial loading.

Application of Hyaluronic Acid Membrane Cross-linked with 1,3-Butadiene Diepoxide (1,3-Butadiene diepoxide로 가교된 히아루론산 막의 응용)

  • The biodegradable hyaluronic acid membranes cross-linked with lactide using the crosslinking agent, 1,3-butadiene diepoxide (BD), were prepared as a potential biocompatible material for tissue engineering. The degree of lactide and BD reaction of the crosslinked membrane was determined by the analysis of nuclear magnetic resonance spectroscopy 6% of growth inhibition was observed in case of high BD concentration but the value is low enough not to affect cell growth. As the crosslinking reaction temperature increased, elongation increased and swelling ratio decreased. The rate of degradation was found to increase with the crosslinking temperature. The drug release experiment showed that the transport of drug through the membrane decreased with the crosslinking temperature.

Structural and Thermal Analysis and Membrane Characteristics of Phosphoric Acid-doped Polybenzimidazole/Strontium Titanate Composite Membranes for HT-PEMFC Applications

  • A series of novel PBI/SrTiO3 nanocomposite membranes composed of polybenzimidazole (PBI) and strontium titanate (SrTiO3) with a perovskite structure were fabricated with various concentrations of SrTiO3 through a solution casting method. Various characterization techniques such as proton nuclear magnetic resonance, thermogravimetric analysis, atomic force microscopy (AFM) and AC impedance spectroscopy were used to investigate the chemical structure, thermal, phosphate absorption and morphological properties, and proton conductivity of the fabricated nanocomposite membranes. The optimized PBI/SrTiO3-8 polymer nanocomposite membrane containing 8wt% of SrTiO3 showed a higher proton conductivity of 7.95 × 10-2 S/cm at 160℃ compared to other nanocomposite membranes. The PBI/SrTiO3-8 composite membrane also showed higher thermal stability compared to pristine PBI. In addition, the roughness change of the polymer composite membrane was also investigated by AFM. Based on these results, nanocomposite membranes based on perovskite structures are expected to be considered as potential candidates for high-temperature PEM fuel cell applications.

  • $^1H-^{15}N$ heteronuclear dipolar coupling solid-state NMR experiments on lipid bilayer or bicelle samples are very useful for the structural studies of membrane proteins. However, to study these biological samples using solid-state NMR, a specific probe with high efficiency and high capability is required. In this paper, we describe the optimized design, construction, and efficiency of a 400 MHz wide-bore $^1H-^{15}N$ solid-state NMR probe with 5-mm solenoidal rf coil for high power, multi-pulse sequence experiments, such as 2D PISEMA or 2D SAMMY.

  • Film Bulk Acoustic Resonator (FBAR) using thin piezoelectric films can be made as monolithic integrated devices with compatibility to semiconductor process, leading to small size, low cost and high Q RF circuit elements with wide applications in communications area. This paper presents an MMIC compatible suspended FBAR using SOI micromachining. It is possible to make a single crystal silicon membrane using a SOI wafer In fabricating active devices, SOI wafer offers advantage which removes the substrate loss. FBAR was made on the 12㎛ silicon membrane. Electrode and Piezoelectric materials were deposited by RF magnetron sputter. The maximum resonance frequency of FBAR was shown at 2.5GHz range. The reflection loss, K$^2$$\_$eff/, Q$\_$serise/ and Q$\_$parallel/ in that frequency were 1.5dB, 2.29%, 220 and 160, respectively.

Home-built Solid-state NMR Probe for Membrane Protein Studies

  • Proteins in highly oriented lipid bilayer samples are useful to study membrane protein structure determination. Planar lipid bilayers aligned and supported on glass slide were prepared. These stack of glass slide with planar lipid bilayers are not well fit for commercial solid-state NMR probe with round coil. Therefore, homebuilt solid-state NMR probe was built and used for a stack of thin glass plates and RF coil is wrapping directly around the flat square sample. The overall filling factor of the coil is much better and the large surface area enhances the extent to orientation by providing uniform environments for the phospholipids and the high ratio of circumference to area reduces edge effects. $^1H\;and\;^{15}N$ double resonance probe for 400 MHz NMR (9.4T) with a flat coil (coil size: 11 mm ${\times}$ 20 mm ${\times}$ 4 mm) is constructed and tested.
