Embryonic Development of Haploid Olive Flounder Paralichthys olivaceus (반수체 넙치(Paralichthys olivaceus)의 발생학적 특성 분석)

  • We investigated the characteristics of embryonic and abnormal organ development in haploid olive flounder, Paralichthys olivaceus, by comparing egg development and histological sections in haploid and diploid individuals. After the mid-blastula transition, abnormal development was observed in haploid fish, including delayed epiboly and malformation of the germ ring and embryonic body. In haploid flounder, Kupffer’s vesicles are irregularly shaped and of variable size compared to diploids. The embryonic body of haploids was shorter and broader than that of diploids and the tail length and size were variable. Most haploid embryos failed to hatch and the few larvae that did, did not survive for more than 6 h. The histological analysis of haploid larvae revealed deformed development in diverse organs, including the eye, otic vesicles, notochord, and neural tube. These results may be related to an abnormality in the axial system of haploid larvae. This study confirmed that the abnormalities of haploid olive flounder were similar to the reported characteristics of haploid syndrome. The abnormalities are caused by delayed epiboly and involution and deformity of Kupffer’s vesicle during egg development.

Effects of Water Temperature and Salinity on the Growth and Survival of Larvae and Juvenile of Platycephalus indicus (수온과 염분이 양태 자치어의 성장과 생존에 미치는 영향)

  • The water temperature and salinity have an important effect on the growth and survival of rearing fish. This study investigates the effect of water temperature and salinity on larvae and juveniles of Platycephalus indicus. The experimental water temperature was set to 13, 16, 19, 22, and 25℃, respectively, and the salinity was set to 7, 14, 21, 28, and 32 psu, respectively. Ten individuals were randomly collected daily and measured the total length using a stereo microscope. The growth rate was the highest at 25℃ (21.62±0.14 mm), 28 psu (15.02±0.05 mm) and the lowest at 13℃ (7.04±0.05 mm), 7 psu. The survival rate was the highest at 22℃ (69.2%), 32 psu (84.1%) and the lowest at 13℃ (15.1%), 7 psu. This study demonstrates that the water temperature and salinity affected the survival and growth of Platycephalus indicus larvae and the juvenile.

  • From November 1992 to January 1993, the rockfish, Sebastes inermis was reared in the laboratory, and observed the developmental stage of the eggs and larvae squeezed from the parent fish. On December 4 1992, developing eggs at morula stage were obtained from female parent fishes($17.4{\sim}20.3mm$ in total lenght(TL)). The diameter of eggs ranged from 1.20 to 1.35mm(n=50), and oil globules in white color were found in the eggs. Hatching began about 55 hours after morula stage at the water temperature of $9.2{\sim}12.3^{\circ}C$. The newly hatched larvae measured from 2.90 to 3.35mm in TL with $8+12{\sim}13=20{\sim}21$ myomeres. Total lengths of the larvae will be from 5.75 to 6.15mm in 27 and 30 days after the hatching, respectively. Myomere number was $8+17{\sim}18=26$.

The Egg Morphology and Larval Development of the Pipefish, Syngnathus schelegeli Kaup (실고기, Syngnathus schlegeli 난(卵)의 형태(形態) 및 산출(産出) 자치어(仔稚魚)의 형태(形態) 발달(發達))

  • The pipefish Syngnathus schelegeli was reared in the laboratory from May to June 1991 and observed the morphology of eggs and larvae squeezed from the parent fish(male). The diameter of inseminated eggs ranged from 0.72 to 1.01 mm (n=50), and yolk in yellow color were found in the eggs. The newly beared larvae were 10.9 mm in average standard length and had 59~60 myomeres. In 6 days after bearing, the post larvae attained 13.8 mm in average standard length and the low jaw was developed. The larvae of 14.1 mm in average standard length (8 days after bearing) had 40~42 fin rays in dorsal fin, 9 in caudal fin and 13~14 in pectoral fin. The juvenile of 14.7 mm in average standard length (10 days after bearing) had the well elongated snout along with the opercular and caudal fin similar to adult stage's.

Studies on parasitic trematodes in bivalves -Variation of infection rate of Cercaria tapidis and Cercaria harengulae- (조개류에 기생하는 흡충류에 관한 연구 - Cercaria tapidis, Cercaria harengulae 의 기생율 변동)

  • We had reported the infection rates of trematode larvae, Cercaria tapidis and C. harengulae in the gonad tissues of Tapes philippinarum and Solen strictus in the estuary of Kum river, the west coast in Korea in 1980~1982. At the same sites from January 1994 to January 1995, we investigated the variation of the infection rates of these trematode larvae in the two clams. The results are follows. The infection rate of C. tapidis parasitized in T. philippinarum increased in 2.4 times (14.0%) higher than that (5.7%) in 1981. Monthly maxium infection rate was 32.0% in October 1994. Comparing the rate (23.3%) in December 1981, the rate in 1994 showed a higher trend than that in 1981. The mean infection rate of C. harengulae in T. philippinarum showed 2.3% in 1994. Comparing that (5%) in 1980, it showed a decreased rate than that in 1980. And monthly maximum infection infection rate of this clam was 6.7% in May 1994, compared with the rate (19.0%) in March 1980. It appeared furthermore decreased rate in 1994. The mean infection rate of the year of C. harengulae in S. strictus was 6.9% in 1994, while it was 10.2% between 1981 and 1982. It showed a decreased trend in 1994. Monthly maximum infection rate was 23.4% in August 1981, while it was 13.3% in April 1994. It showed a decreased trend in 1994 also. In case of C. tapidis the more large sizes of shell length the more infection rates were higher as same in the investigation in 1980, while in C. harengulae the smaller sizes, the more infection rates showed a higher trend.

Development of Eggs, Larvae and Juvenile of the Korean Bullhead, Pseudobagrus fulvidraco (Richardson) reared in the laboratory (동자개의 형태발달과 성장특성)

  • The early developmental stages, growth and morphological changes of the Korean bullhead, Pseudobagrus fulvidraco, were studied from a series of reared specimens. Details of the early developmental stages are illustrated with special reference to morphological transformations. Egg and sperm of Korean bullhead were obtained from mature adults under hormonal treatment, fertilized artificially, and incubated in the aquarium. The incubation period of fertilized eggs was 55 to 66 hours at a temperature of 24.9${\pm}$0.34$^{\circ}$. Larvae were fed successively with Artemia salina and Daphnia magna for 2 to 15 days and artificial food after 20 days. Fertilized eggs were adhesive and spherical with a diameter of 2.04mm(n = 100). The mean total length of newly hatched larvae was about 4.92${\pm}$0.33 mm. Mouth opening occurred on one-day-old yolk-sac larvae, and initial feeding was observed on the third day after hatching. The morphological transitions from larvae to juvenile and juvenile to young stages occurred when the fish reached about 17 mm in total length (about 13days after hatching) and about 32 mm in total length (about 30 days after hatching), respectively. Many changes in proportion of body parts to total length were observed at about 7~8 mm and 30~32 mm, corresponding to the transformations from larvae to juvenile and from juvenile to young, respectively. In comparing relative growth of each body part against total length, those characteristics related to head parts showed positive growth in the pre-larval stages, while those concerning mobile abilities showed positive growth in the post-larval stage.

Nutritional Value and Digestibility of Tenebrio molitor As a Feed Ingredient for Rockfish (Sebastes schlegeli) (조피볼락의 사료원으로 갈색거저리의 영양적 가치 및 소화율 평가)

  • Tenebrio molitor larvae, also known as yellow mealworms (MW), are rich in protein and lipid and can serve as a potential alternative protein and energy source in commercial aquafeeds. Therefore, this study attempts to evaluate the effects of different drying methods on the nutritional value of MW meal. For this, live MW were cold-anaesthetized before being subjected to three different types of drying methods, including freeze-drying, oven-drying at $60^{\circ}C$ and air-drying at room temperature for three days, and compared for proximate composition and energy content. An in-vivo digestibility test was then conducted to evaluate the nutrient digestibility of MW meal in diets for rockfish, Sebastes schlegeli. A test diet was prepared by mixing the MW meal with a reference diet (Ref) in a 30:70 ratio with chromium oxide as an inert marker at the inclusion level of 0.5%. Rockfish with mean body weight of 150 g were stocked into a fecal collection system equipped with fiberglass tanks of 400 L capacity. Each group of fish was fed one of the experimental diets to apparent satiation for 4 weeks. The results of the proximate analysis showed that drying methods had no significant effect on crude protein, crude lipid, ash and energy contents of MW. Despite being a rich source of protein and lipid, MW meal was deficient in certain amino acids, particularly methionine, and highly unsaturated fatty acids, particularly 22:6n-3 (DHA) and 20:5n-3 (EPA). MW meal showed high digestibility values for protein (93%), lipid (97%) and energy (88%). These results may indicate that MW meal is a nutritious and acceptable feed ingredient, with comparable digestibility values to conventional animal and plant feedstuffs such as fish meal and soybean meal, in practical diet for rockfish at grower stage.

Egg Development and Morphology of Larval Blenniid Fish, Istiblennius stellifer (Jordan et Snyder) Reared in the Laboratory (실험실(實驗室)에서 사육(飼育)한 저울베도라치의 난발생(卵發生) 및 부화자어(孵化仔魚))

  • Blenniid fish, Istiblennius stellifer(jordan et Snyder) is distributed in the coastal waters of south-eastern Korea and Japan. Matured adults of blenniid fish were collected from the rocky shore of Namchun-dong, Nam-gu, Pusan, Korea on May 15, 1988. The fertilized eggs were incubated and the larvae were reared in laboratory. The eggs of this species were demersal and adhesive, and their diameters varied from 0.84 to 0.88 mm(mean 0.86 mm, n=30). They have a number of small oil globules. The water temperature throughout incubation ranged from 18.5 to $23.3^{\circ}C$ and salinity was maintained at $28.2-29.5\;^{\circ}/_{\circ\circ}$. The hatching took place in 130 hours after fertilization. The newly hatched larvae were 2.70 mm in total length with 11 (abdominal)+22~25 (caudal)=33-36 myomeres. The larvae absorbed the yolk material and oil globule completely in 10 days after hatching and became postlarvae. Total lengths of the larvae reached 4.65 and 5.75 mm in 10 and 13 days after the hatching, respectively.

Biochemical Composition of Rotifer, Brachionus plicatilis Enriched with Different Commercial Enrichments (영양 강화제 종류에 따른 rotifer, Brachionus plicatilis의 생화학적 조성)

  • This study was carried out to investigate changes in biochemical composition of rotifer, Brachionus plicatilis, enriched with the commercial enrichments (Enhance, Advantage, Algamac-2000, DHA-Selco and Advantage + Chlorella) at various durations of enrichment (0, 6, 12 and 24 hr) to improve the growth and survival of marine fish larvae. Total lipid content of rotifers enriched with various enrichments tended to increase with an increase in durations of enrichment up to 6 hr, but after that, was not significantly affected by enrichment materials. However, total protein content of rotifers enriched groups except for Advantage+Chlorella decreased with the increase in duration of enrichment. The highest protein/lipid ratio showed 2.7 in rotifer enriched with the Advantage +Chlorella. The phospholipid/lipid ratio of rotifer enriched with the Enhance, Advantage and Advantage+Chlorella groups was significantly higher than that of enriched rotifer with the Algamac-2000 and DHA-Selco groups. The highest DHA level, 2.5%, of rotifer enriched for 24 hr was obtained in the Advantage, but was not significantly different among other groups, except for Algamac-2000. No significant difference in DHA level of rotifer enriched with the DHA-Selco, Algamac-2000 and Advantage+Chlorella groups was observed between l2h and 24hr of enrichment. The DHA/EPA ratio in the enriched rotifers varied among enrichment material groups, ranged from a high level of 11.1:1 in the Advantage+Chlorella group to a low level of 4.1:1 in DHA-Selco group. The results from this study indicate that rotifers enriched with Enhance, Advantage and Advantage+Chlorella seemed to be effective to improve nutritional value of rotifer for marine fish larvae because phospholipid, DHAJEPA and protein/lipid ratios of rotifer enriched with Enhance, Advantage+Chlorella were higher than those of rotifer enriched with either DHA-Selco or Algamac-2000. Especially, supplementation of the Chlorella to these enrichments would appear to be effective for improvement of fish larval performance because of no reduction of protein level in rotifer, which is critical for growth of fish larvae.

Early Life History of Tanakia latimarginata (낙동납자루(Tanakia latimarginata)의 초기생활사)

  • This study is to observe species identification and early life history of Korean endemic species of Tanakia latimarginata and to use it as a basis for taxonomic studies. As a result of morphological identification, a dark band appeared at the margin of the anal fin, and the ovipositor color of the female was light orange. The shape of the egg was fusiform and sticky. The egg size (long${\times}$short diameter) averaged $4.41{\times}1.44mm$. The incubation time was 126 hours after the fertilization at an average water temperature of $21.0^{\circ}C$. Immediately after hatching, the larvae had egg yolk at an average total length of $5.91{\pm}0.18mm$ (n=5). At 18 days after hatching, the trunk fur was developed in the caudal fin with an average total length of $8.02{\pm}0.08mm$ (n=5). At 41 days after hatching, the larvae absorbed egg yolk at an average total length of $8.70{\pm}0.23mm$ (n=5). At 80 days after hatching, the average length of the fins was $12.6{\pm}0.28mm$ (n=5). The number of fin of the dorsal fin was iii.8, the anal fin iii.9~10, the caudal fin 19, lateral line scales 32~35 were similar to their brood stork.
