  • Kaneka Corporation (Kaneka) has been manufacturing CoQ10 under GMP regulation since 1977. Kaneka has a sophisticated quality control system and has been supplying high quality CoQ10 materials to the worldwide customers (Kaneka CoQ10) for about 30 years. Kaneka CoQ10 is characterized by a lot of safety data, which are derived from clinical trials with healthy volunteers (single-dose and 4-week multi-dose safety studies), animal studies (13-week sub-chronic study in dogs and 52-week chronic study in rats), three types of mutagenicity test, six type of skin irritation test (for cosmetics), and others. The risk assessment of CoQ10 was performed by Council for Responsible Nutrition (USA). They reviewed many of available clinical data including clinical trials using Kaneka Q10, and concluded that the upper level for supplements (ULS) of CoQ10 is 1,200mg/day (Hathcock and Shao. 2006, Regulatory Toxicology and Pharmacology, $\underline{45}$, 282 - 288).

경기도 내 유통 견과종실류의 잔류농약 실태 모니터링 (Monitoring Pesticide Residues in Nuts and Seeds in Gyeonggi-do)

  • 본 연구에서는 경기도 내 유통 중인 견과 종실류의 잔류농약 실태를 조사하고자 온라인 또는 유통매장을 통해 131건을 수거하여 조사하였다. 총 14품목의 견과종실류를 수거하였으며 339종의 잔류농약을 분석한 결과 131건 중 총 8건에서 검출되었으며 범위는 0.01 mg/kg-0.07 mg/kg 수준으로 나타났다. 검출된 시료 8건 중 7건이 잔류농약 기준을 초과하여 부적합으로 판정되었으며 부적합 7건 중 6건은 수입농산물에서 판정되었다. 중국산 호박씨의 경우 PLS가 적용된 피라클로스트로빈이 5건 검출되었으며 범위는 0.02 mg/kg-0.04 mg/kg 수준으로 나타났다. 인도산 참깨와 국내산 참깨에서도 펜디메탈린, 보스칼리드, 비펜트린이 각각 0.04 mg/kg, 0.05 mg/kg, 0.06 mg/kg으로 검출되어 부적합 수치를 보였다. 또한 검출된 8종의 농약에서 EDI/ADI (%) 값이 0.002%-0.372%으로 낮은 수준임을 확인하였다.

Sprague-Dawley 랫드를 이용한 선폐정천탕의 단회투여독성시험 (Single Dose Toxicity Study of Seonpyejeongcheon-tang in Sprague-Dawley Rats)

  • Objectives : This study aimed to evaluate the single oral dose toxicity of Seonpyejeongcheon-tang (SJT) in male and female Sprague-Dawley rats. Methods : In this single oral toxicity study, rats were orally administrated in a single dose of 0 or 5,000 mg/kg SJT. There were 7 rats in each group. After single administration, mortality, clinical signs, body weight changes and gross pathological findings were observed for 14 days. Organ weight, clinical chemistry and hematology were tested after 14 days. Results : There was no mortality or other clinical signs for 14 days. There were also no significant differences in body weight, organ weights, hematological and serum chemical parameters between the SJT and control groups. Conclusions : The results obtained in this study suggest that the 50% lethal dose of SJT is over 5,000 mg/kg, so this finding can be expected to provide scientific evidence for the safety of SJT.

LLC-$PK_1$을 이용한 신독성 물질들의 $\alpha$-methyl glucose uptake에 미치는 영향의 평가 (Effect of Nephrotoxicants on $\alpha$-Methylglucose Uptake in LLC-$PK_1$)

  • Many nephrotoxic agents exert their effect primarily on the cells of the proximal tubules. We used the LLC-$PK_1$, kidney epithelial cell line as a model system for studies on nephrotoxicity and investigated whether the uptake of $\alpha$-methylglucose($\alpha$-MG) could serve as a parameter to assess effects of nephrotoxicants on the functional integrity of the cells at an early time of toxicity. The enzyme leakage test which has been used to be as a conventional cytotoxic parameter in vitro, was conducted to compare with $\alpha$-MG uptake. Treatment with cisplatin for 24 and 48 hours significantly increased activities of lactate dehydrogenase and $\gamma$-glutamyltransferase in culture medium at a concentration of 50$\mu$M. However, above 100$\mu$M of concentration, activities of these enzymes in media were dramatically decreased by cisplatin. These observations indicate that cisplatin has direct inhibitory effect on the activities of these enzymes and make it doutful to use enzyme leakage test to demonstrate damage of kidney cells by chemicals such as cisplatin over the appropriate range of concentration. Cisplatin inhibited $\alpha$-MG uptake at a low concentration which enzymes were not leaked. Also cadmium chloride and mercuric chloride which are acutely nephrotoxic in vivo, significantly inhibited $\alpha$-MG uptake at a low concentration. These results indicate that the uptake of $\alpha$-methylglucose in LLC-$PK_1$cell line is a useful biomarker for the study of nephrotoxicity.

참전복, Haliotis discus hannai의 폐사 및 글리코겐 함량에 미치는 부유토사의 영향 (The Effects of Suspended Solids on the Mortality and the Glycogen Content of Abalone, Haliotis discus hannai)

  • 연안개발에 의하여 해양으로 유입되는 부유토사는 해양생태계에 많은 영향을 미칠 것으로 판단된다. 따라서 본 연구에서는 참전복, Haliotis discus hannai 치패를 이용하여 부유토사 농도 변화에 따른 폐사율 및 글리코겐 함량 변화에 대해 알아보았다. $0{\sim}2,000\;mg/L$의 부유토사 농도구에서 96시간 동안 노출을 행한 결과 전복의 폐사는 일어나지 않았으나 연체부의 글리코겐 함량에 있어서는 1,500 mg/L 이상의 농도구에서 유의하게 감소하였다. 본 연구결과로부터 부유토사가 전복의 폐사에는 직접적인 영향을 미치지는 않으나 부유토사에 의하여 건강상 장해를 일으킬 수 있을 것으로 판단되며 장기적으로 발생할 수 있는 부유토사의 영향을 고려하여 양식장으로의 토사유입을 방지 할 수 있는 대비책을 마련해야 할 것이다.

Comparative Analysis of Tagatose Productivity of Immobilized L-Arabinose Isomerase Expressed in Escherichia coli and Bacillus subtilis

  • Although arabinose isomerase (E.C. 5.3.1.4), a commercial enzyme for edible tagatose bioconversion, can be expressed in an Escherichia coli system, this expression system might leave noxious by-products in food. To develop an eligible tagatose bioconversion with food-safe system, we compared the tagatose production activity of immobilized arabinose isomerase expressed in Bacillus subtilis (a host generally recognized as safe) with that of the enzyme expressed in E. coli. A 48% increase in tagatose production (4.3 g tagatose/L at $69.4\;mg/L{\cdot}hr$) was found using the B. subtilis-expressed immobilized enzyme system, compared to the E. coli-expressed enzyme system (2.9 g tagatose/L). The increased productivity with safety of the B. subtilis-expressed arabinose isomerase suggests that it is a more eligible candidate for commercial tagatose production.

돈사 작업장 유형에 따른 암모니아와 황화수소의 실내농도 및 발생량에 관한 현장 조사 (Field Study of Emission Characteristics of Ammonia and Hydrogen Sulfide by Pig Building Types)

  • The principal aim of this field study was to determine the concentrations and emissions of gaseous contaminants such as ammonia and hydrogen sulfide in the different types of pig buildings in Korea and allow objective comparison between Korea and the other countries in terms of pig housing types. This field study was performed from May to June and from September to October in 2002. Pig buildings investigated in this research were selected in terms of three criteria; manure removal system, ventilation mode and growth stage of pig. Measurements of concentration and emission of ammonia and hydrogen sulfide in the pig buildings were done in 5 housing types and the visited farms were 15 sites per each housing type. Concentrations of ammonia and hydrogen sulfide were measured at three locations of the central alley in the pig building and emission rates of them were estimated by multiplying the average concentration($mg/m^3$) measured near the air outlet by the mean ventilation rate($m^3/h$) and expressed either per pig of liveweight 75kg(mg/h/pig) or per area($mg/h/m^2$). Concentrations of ammonia and hydrogen sulfide in the pig buildings were averaged to 7.5 ppm and 286.5 ppb and ranged from 0.8 to 21.4 ppm and from 45.8 to 1,235 ppb, respectively. The highest concentrations of ammonia and hydrogen sulfide were found in the mechanically ventilated buildings with slats; 12.1 ppm and 612.8 ppb, while the lowest concentrations of ammonia and hydrogen sulfide were found in the pig buildings with deep-litter bed system(2.2 ppm) and the naturally ventilated pig buildings with manure removal system by scraper(115.2 ppb), respectively(p<0.05). All the pig buildings were investigated not to exceed the threshold limit values(TLVs) of ammonia(25 ppm) and hydrogen sulfide(10 ppm). The mean emissions of ammonia and hydrogen sulfide per pig(75kg in terms of liveweight) and area($m^2$) from pig buildings were 250.2 mg/h/pig and 37.8 mg/h/pig and $336.3mg/h/m^2$ and $50.9mg/h/m^2$, respectively. The pig buildings with deep-litter bed system showed the lowest emissions of ammonia and hydrogen sulfide(p<0.05). However, the emissions of ammonia and hydrogen sulfide from the other pig buildings were not significantly different(p>0.05). Concentrations and emissions of ammonia and hydrogen sulfide were relatively higher in the pig buildings managed with deep-pit manure system with slats and mechanical ventilation mode than the different pig housing types. In order to prevent pig farm workers from adverse health effect caused by exposure to ammonia and hydrogen sulfide in pig buildings, they should wear the respirators during shift and be educated sustainably for the guideline related to occupational safety.

통영항의 해수 및 저질의 위생학적 특성 (Sanitary Characteristics of Seawater and Sediments in Tongyeong Harbor)

  • 이 연구는 통영항 내의 해수와 저질의 특성과 주변횟집의 수질의 품질 특성에 대하여 2008년 6월부터 2009년 5월까지 월 1회 조사를 실시하였으며 그 결과는 다음과 같다. 통영항의 해양수온은 평균 $17.4{\pm}5.77^{\circ}C$, 투명도 평균 $1.9{\pm}0.46$ m, 부유물질 $36.7{\pm}5.00$ mg/L, COD $2.36{\pm}0.80$ mg/L, DO $6.67{\pm}1.54$ mg/L, 염분농도 $32.3{\pm}0.64$‰를 각각 나타내었다. 아질산 질소는 $0.010{\pm}0.005$ mg/L, 질산 질소 $0.050{\pm}0.011$ mg/L, 암모니아 질소 $0.030{\pm}0.007$ mg/L, 인산 인 $0.050{\pm}0.006$ mg/L로 각각 나타났으며, TBT는 검출한계 이하로 검출되었다. 통영항 해수 중의 total coliform은 조사기간 중 < 1.8~22,000 MPN/100 mL 범위에 기하학적 평균값이 164.9 MPN/100 mL이었고 230 MPN/l00 mL을 초과하는 비율은 39.6%를 나타내었다. fecal coliform은 < 1.8~7,900 MPN/100 mL 범위에 기하학적평균값은 33.7 MPN/100 mL, 43 MPN/100 mL을 초과하는 비율은 47.9%로 조사되었다. total coliform에 대한 fecal coliform 검출 비율은 69.2%로 나타났다. 통영항 해수 중의 비브리오는 6월부터 11월에 240주가 검출되었으며, V. alginolyticus가 82주로 34.2%를 차지하여 가장 많이 검출되었고, 그 밖에 parahaemolyticus가 13.8%, V. culnificus 10.0%, V. cholerae non-O1이 0%, V. mimicus 12.5%가 검출되었다. 저질 특성은 함수율이 $56.63{\pm}5.51$ %, 강열감량 $9.20{\pm}1.36$%, COD $7.64{\pm}1.13$ mg/g, AVS $0.07{\pm}0.02$mg/g으로 다른 해역에 비하여 다소 높게 나타났다. 중금속의 함량은 카드뮴(Cd)은 $0.10{\pm}0.05$, 구리(Cu) $4.79{\pm}8.20$, 비소(As) $1.95{\pm}0.17$, 수은(Hg) $0.10{\pm}0.07$, 납은 불검출, 6가크롬은 ($Cr^{6+}$) $0.34{\pm}0.12$, 아연 (Zn) $125.33{\pm}16.40$, 니켈(Ni) $16.34{\pm}1.93$ mg/kg을 나타내었다. 통영항 주변 횟집의 total coliform 및 fecal coliform은 횟집에 따라서 다소 차이가 있었지만 여름철에 주로 검출되었으며, 여름철 집중관리가 요구되었다.

랫드에 투여된 규회석이 호흡기계에 미치는 영향 연구 (Toxicity study of Wollastonite to the Respiratory Systems in Sprague-Dawely Rats)

  • Occupational exposure to wollastonite can occur during its mining, processing and using. Wollastonite was analyzed for its physicochemical properties by transmission electron microscope and scanning electron microscope equipped with energy dispersive X-ray spectrometer. Wollastonite fibers were 3.3${\mu}m$ in diameter(av.) and were 30.5${\mu}m$ in length(av.). The atomic composition of wollastonite was Si 56.41% and Ca 43.59%. Respiratory toxicity of wollastonite has been studied in term sequential in Sprague-Dawely rats. UICC chrysotile (average diameter 0.03${\mu}m$, average length 2.93${\mu}m$) was applied as the positive control. The effects of 2mg wollastonite on respiratory system and pathological changes were evaluated after 1, 4, 8, 12 weeks instilled into rat lungs. Inflammation response broke out from 1 week after instilled with wollastonite and the pathological examination further showed increased legions of granulomatous inflammation after 4 weeks, but decreased granulomatous inflammation after 8 weeks, whereas chrysotile induced progressive interstitial granulomatous inflammation and fibrosis as a function of time.

경엽 및 관주 처리에 따른 딸기 중 침투성 농약 잔류패턴 비교 (Comparison of residue patterns between foliar application and drenching in export strawberry)

  • 딸기는 한국의 주요 농산물 중 하나로 매년 3,000만 달러 이상이 수출된다. 그러나 농산물 안전은 최근 농업 무역에서 중요한 요소로 작용하고 있어, 안전한 농산물을 생산하는 것은 매우 중요하다. 안전한 농산물을 생산하려면 적절하게 농약을 관리하는 것이 필수적이다. 딸기는 고설재배를 이용해 경작하는 곳이 많아지고 있는데 점적관수를 이용해 수분, 양분 및 농약을 관주 처리하여 투여하고 있다. 농약을 살포 할 때 관주 처리는 경엽처리를 사용하는 것보다 잔류농약 수준이 더 낮게 나타났다. 본 연구에서는 딸기 중 농약을 관주처리 및 경엽처리 하였을 때, 농약의 잔류량을 경시적으로 비교 하였다. 농약을 농약안전사용기준량 및 두배량으로 처리 한 후 0, 3, 6, 9, 13, 16 및 21 일에 수집한 후 $-18^{\circ}C$에서 보관 하였다. 샘플을 LLE(액체-액체 추출) 및 QuEChERS의 전처리 후 LC 및 LC-MS/MS로 분석 하였다. 결과적으로 관주 처리 한 딸기는 모든 처리구에서 1 mg/kg 미만으로 검출되었다. 농약을 경엽 처리와 관주 처리 하였을 때 딸기의 잔류량 차이는 carbendazim의 경우 5-27배 정도 경엽처리가 높았고, azoxystrobin은 관주처리 시 불검출 되어 비교할 수 없었다.
