Antioxidative Effect of Ethanol Extract for 5 Kinds of Spice (5종의 향신료 에탄올 추출물의 항산화 효과)

  • This study was performed to investigate the antioxidative effect of ethanol extracts of 5 spices. They were separately extracted in ethanol from dried samples at room temperature, and freeze-dried. In vitro testing were conducted by DPPH (1,1-diphenyl-2-picryl hydrazyl) radical scavenging activity, inhibition of iron-induced linoleate peroxidation and the inhibition of malondialdehyde (MDA) and bovine serum albumin (BSA) conjugation reaction. The ethanol extracts of clove (92.9%) and cinnamon (89.9%) showed the most effective results among five spices in the DPPH radical scavenging capacities. The inhibition rate of ethanol extract of clove on the lipid peroxidation was 55.8%. The ethanol extracts of mustard, wasabi and black pepper were effective in the inhibition of MDA and BSA conjugation reaction showing 73.2%, 72.2% and 61.6%, respectively. These results suggest that five spices tested in this study may enhance the antioxidative capacity, although the results were different according to the assay method and sample.

Effect of Hizikia fusiforme Extracts on Antioxidant Enzyme Activity and Vitamin E Concentration in Rats (톳 추출물의 경구투여가 흰쥐의 항산화효소 활성과 비타민 E 농도에 미치는 영향)

  • The purpose of this study was to investigate antioxidant enzyme activity and vitamin E concentrationin in Sprague-Dawley rat after being fed various extracts of Hizikia fusiforme. There were six experimental groups: control group (C), H. fusiforme ethanol extract group (EtOH), H. fusiforme dichloromethane fraction group ($CH_2Cl_2$), H. fusiforme ethylacetate fraction group (EtOAc), H. fusiforme butanol fraction group (n-BuOH), H. fusiforme water fraction group ($H_2O$). H. fusiforme extracts (400 mg/kg B.W) were orally administrated to the rats every day for 4 weeks. The activities of superoxide dismutase (SOD), catalase, glutathione peroxidase (GSH-Px), and concentrations of malondialdehyde (MDA) and vitamin E in the liver and blood were measured. The activity of SOD in the liver was significantly higher in the $CH_2Cl_2$ and $H_2O$ groups (p<0.05) than in the control and other extract groups. The SOD activity in serum increased significantly in all H. fusiforme groups (p<0.05) compared to the control group and it was also significantly higher in the EtOH and $H_2O$ groups (p<0.05) than in other extract groups. The serum catalase activity increased significantly in the n-BuOH group (p<0.05) compared to the control and other extract groups. The plasma MDA concentration decreased significantly in the n-BuOH and $H_2O$ group (p<0.05) compared to the control group. Serum concentration of ${\alpha}$-tocopherol showed no significant differences in most of the experimental groups, but it was significantly higher in the EtOAc group (p<0.05). The ${\alpha}$-tocopherol concentrations in the liver showed a significant increase in the $CH_2Cl_2$ and $H_2O$ groups (p<0.05) compared to the control and other extract groups. The liver ${\gamma}$-tocopherol concentrations in H. fusiforme extract groups showed a tendency to increase compared to the control group and it was significantly higher in the $H_2O$ group (p<0.05) than in other extract groups. These results suggest that supplementation of water extracts of H. fusiforme extract could be effective in improving the antioxidant system.

영지버섯(Ganoderma lucidum)추출물의 항산화 및 항돌연변이 효과

  • 버섯류는 항산화, 항암, 항변이원성, 항콜레스테롤성 효과 등의 여러 가지 생리활성 기능을 나타내기 때문에 암 및 성인병에 대한 예방 및 개선효과가 기대되는 좋은 소재라 할 수 있다. 그러므로 본 연구에서는 영지버섯의 생리 기능에 대한 활성을 탐색하고자 영지버섯을 에탄올로 추출하여 항산화효과와 항돌연변이 효과를 측정하였다. MDA에 대한 단백질의 보호효과를 알아본 결과 영지버섯추출물은 MDA의 교차 결합 형성을 31.68%저해하였고 처리농도가 증가할수록 저해율은 증가하였다. (중략)

Antioxidant activities from the Different Parts of Artemisia argyi H. using an in vitro System (In vitro 시스템에서 남해 약쑥(Artemisia argyi H.)의 부위별 항산화 활성)

  • The antioxidant activities of 60% ethanol extracts from different parts (leaf, stem, and root) of Artemisia argyi using an in vitro system were examined to find a possibility as natural antioxidant substances. The highest total phenolics (23.08 mg/g) was obtained by 60% ethanol extract of leaf. The scavenging activity of 60% ethanol extract from different parts of A. argyi on the DPPH and ABTS radical was raised with increasing amount of extracts, and 60% ethanol extract from leaf showed the highest radical scavenging activities. The leaf extract presented the highest reducing power and strong inhibitory effect on autooxidation of linoleic acid. In MDA formation using mouse brain homogenates, the 60% ethanol extracts of A. argyi also exhibited antioxidant activity as inhibition of MDA (71.38% at $200{\mu}g/mL$). Therefore, these results suggest that the 60% ethanol extract of A. argyi leaf possess excellent antioxidant activities and thus it has great potential as a source for natural antioxidant.

A Study on the Possibility of Malondialdehyde(MDA) as Indicator of Forest Therapy Effectiveness (산림치유 효과 측정 지표로써 과산화지질의 활용가능성 분석)

  • This study aimed to analyze and compare results from psychological relaxations by using profile of mood state (POMS) and Malondialdehyde (MDA) level as a scale of oxidative damage before and after walking in the forest and on the street. 29 participations (15 men and 14 women) walked in the forest and on the street for 30 minutes of each for two days. The participations filled a questionnaire for POMS and conducted an heart rate variability test, oxidative damage test (MDA as biological marker through a urine test) before and after walking. To compare the psychological states after walking in the two difference places, walking in the forest showed statistically more meaningful results in the section of tension, depression, anger, fatigue, confuse and total mood disturbance (p<0.01) than walking on the street. According to the MDA results, a concentration of MDA showed significant increase after walking on the street (p=0.014). On the contrary to this, a concentration of MDA tended to decrease after walking in the forest (p=0.076). The study showed that the psychological states were more stable and oxidative damages were more decreasing after walking in the forest than walking on the street. In the conclusion, MDA would be biological indicator for assessing the effect of alleviation on the oxidative stress after walking in the forest.

Effect of Eupatorium japonicum Extract on the Metastasis, Invasion and Adhesion of MDA-MB-231 Human Breast Cancer Cells (등골나물 추출물이 인간의 유방암세포인 MDA-MB-231 세포의 이동, 침윤 및 부착에 미치는 영향)

  • The metastatic effect of Eupatorium japonicum extract (EJE) on MDA-MB-231 human breast cancer cells was investigated. MDA-MB-231 cells were treated with various concentrations of EJE (0, 5, 10 and $20{\mu}g/mL$). EJE inhibited cell migration, invasion and adhesion of MDA-MB-231 cells in dose-dependent manners. Gelatin zymography exhibited that EJE significantly down regulated secretion of matrix metalloproteinase (MMP)-9 and MMP-2. EJE decreased the protein levels of tissue inhibitor of metalloproteinase (TIMP)-1 but increased TIMP-2 levels. Additionally, EJE reduced the protein and mRNA levels of urokinase-type plasminogen activator (uPA), vascular endothelial growth factor (VEGF) and intercellular adhesion molecule (ICAM). In several solvent fractions of EJE, the hexane fraction markedly decreased MDAMB-231 cell migration. Thus, these finding suggest that EJE may be a potential antimetastatic agent, which can considerably inhibit the metastatic and invasive capacity of breast cancer cells.

In vitro Anti-proliferative Characteristics of Flavonoids and Diazepam on MDA-MB-231 Breast Cancer Cells (Flavonoid류와 diazepam의 시험관 내 MDA-MB-231 유방암세포 증식 억제 효과)

  • The beneficial use of sedatives is often required for medically ill patients. This study examined the effect of plant flavonoids and diazepam peripheral-type benzodiazepine receptor (PBR) activation and glucose utilization in breast cancer cells, along with their interactions. In estrogen receptor negative MDA-MB-231 cells, the anti-proliferative activity of fisetin (3,7,3',4'-tetrahydroxyflavone) and diazepam was more prominent than in estrogen receptor positive MCF-7 cells. Unlike PBR ligands, treatment with $10^{-6}$ M concentration of diazepam for 3 days exhibited anti-proliferative effects, while similar to apigenin (4',5,7-Trihydroxyflavone) and fisetin, diazepam hardly affected the PBR mRNA expression by MDA-MB-231 cells. Treatment with $10^{-6}$ M concentration of flavonoids and diazepam for 3 days inhibited the glucose utilization of MDA-MB-231 cells. Treatment with $10^{-6}$ M concentration of flavonoids and diazepam for 6 days showed increased cytotoxicity and reduced the PBR mRNA expression of the MDA-MB-231 cells. Apigenin enhanced diazepam-induced anti-proliferative effects on the MDA-MB-231 cells as well. All together, this study showed the in vitro anti-proliferative activity of flavonoids and diazepam on MDA-MB-231 breast cancer cells, plus additive enhancements. In conclusion, this study provides experimental basis for advanced trials in the future.

Inhibition of cellular proliferation and apoptosis by Scutellaria Baicalensis in MDA-MB-231 breast cancer cells (황금의 유방암세포주에 대한 항암작용)

  • Objective : 황금의 유방암세포주에 대한 항암효과 및 기전에 대한 연구는 아직 미흡하며, 특히 에스트로젠리셉터를 가지지않은 유방암세포주인 MDA-MB-231에 대한 효과 및 기전에 대한 연구는 아직 발표된바 없어, 이에 대한 연구가 진행되었다. Methods : 인간 유방암세포주 MDA-MB-231 MTT assay를 이용 성장방해비율을 조사하였으며. FACS analysis를 이용 cell cycle analysis를 시행하였고, Western Blot Analysis 및 Annexin V analysis를 시행하였다. Results : MDA-MB-231에 대한 황금의 IC50는 180 ug/ml 이었으며 최대 세포성장억제효과는 $500{\mu}g/ml$로 한약재중 비교적 강한 세포독성을 보여 주었다. 유세포분석 에서 황금 $500{\mu}g/ml$의 농도를 72시간 투여한 경우 세포사멸(Sub Gl) 분율이 대조군의 1.7%에 비해 21%로 높아 현저한 용량의존적인 세포사멸현상을 보여주었으며, 세포사멸을 보다 명확히 규명할 수 있는 Annexin V analysis에서도 황금 $200{\mu}g/ml$농도일때 48시간에서 17%의 뚜렷한 세포사멸효과를 나타내었다. 한편 세포사멸촉진인자인 Bax, 세포사멸실행단백질인자인 caspase 3의 활성과 PARP의 분할은 세포사멸이 세포주기정지와 더불어 세포사멸의 과정에 p53이 관여함을 알 수 있다. 앞으로의 연구는 p53발현이 다른 세포주와 각 단백질의 억제제를 통해 인과적인 관련성을 즘 더 명확히 할 필요가 있어야 할 것으로 생각되어진다. Conclusion : 유방암의 예후에 있어 호르몬치료에 부적절함으로 인해 예후가 나쁜 에스트로젠리셉터 발현이 없는 유방암에 대해서도 황금이 탁월한 항암효과를 보여주고 있으며, 임상적으로 황금단독, 다른 항암약재와의 배합, 그리고 기존의 항암화학요법이나 방사선요법과의 병용투여를 통한 초기 및 진행된 유방암의 치료에 대한 새로운 접근의 실마리를 제공할 것으로 생각된다.

Induction of Apoptosis by Immature Prunus salicina Lindl. cv. Soldam (피자두(Prunus salicina Lindl. cv. Soldam) 미숙과의 apoptosis 유도 효과)

  • Apoptosis of Prunus salicina Lindl. cv. Soldam, which possesses hematopoiesis, osteoporosis prevention, and antimutagenic effects, at different growth stages was evaluated. Cytotoxic effect of acetone extracts of immature fruits against various tumor cell lines was higher than that of mature fruits, particularly in hormone-independent human breast cancer, MDA-MB-231 cell line. Immature fruit extract increased expression level of pro-apoptotic protein Bax and reduced that of anti-apoptotic protein Bcl-2, and stimulated caspase-3 activity in MDA-MB-231 cells. Results suggest immature fruit of P. salicina Lindl. cv. soldam to be natural source for development of functional food and medical agents to prevent human breast cancer.
