Simultaneous Analysis of three Marker Components in Hwangryunhaedok-tang by HPLC-DAD (황련해독탕 중 3종 생리활성 물질의 HPLC-DAD 동시 정량분석법 확립)

  • In this study, a high performance liquid chromatography-diode array detector method was established, for simultaneous determination of three compounds, berberine, palmatine and geniposide in Hwangryunhaedok-tang, To develop and validate method, $C_{18}$ column (5 ${\mu}M$, 4.6 mm${\times}$250 mm) was used with gradient mobile phase, water containing 0.1% trifluoroacetic acid (TFA) and MeOH at the column temperature of $30^{\circ}C$. UV wavelength was set at 230 and 280 nm. Validation of the chromatography method was evaluated by linearity, precision and accuracy test. Calibration curve of standard components showed good linearity ($R^2$ > 0.9999). The limits of detection (LOD) and limits of quantification (LOQ) varied from 0.05 to 0.17 ${\mu}g/ml$ and 0.15 to 0.53 ${\mu}g/ml$, respectively. The relative standard deviations (RSDs) data of intra-day and inter-day test were in less than 2.99% and 1.90%, respectively. The results of the accuracy test were in the range of 98.36 to 102.52% with RSDs values 0.32 to 1.98%. The results of validation indicated that this method was a very accurate and sensitive assay.

  • Theoretical studies on the un-catalyzed and catalyzed aminations of ketene with $NH_3$ and $(NH_3)_2$, respectively, were studied using MP2 and hybrid density functional theory of B3LYP at the 6-31+G(d,p) and 6- 311+G(3df,2p) basis sets in the gas phase and in benzene and acetonitrile solvents. In the gas phase reaction, the un-catalyzed mechanism was the same as those previously reported by others. The catalyzed mechanism, however, was more complicated than expected requiring three transition states for the complete description of the C=O addition pathways. In the un-catalyzed amination, rate determining step was the breakdown of enol amide but in the catalyzed reaction, it was changed to the formation of enol amide, which was contradictory to the previous findings. Starting from the gas-phase structures, all structures were re-optimized using the CPCM method in solvent medium. In a high dielectric medium, acetonitrile, a zwitterions formed from the reaction of $CH_2$=C=O with $(NH_3)_2$, I(d), exists as a genuine minimum but other zwitterions, I(m) in acetonitrile and I(d) in benzene become unstable when ZPE corrected energies are used. Structural and energetic changes induced by solvation were considered in detail. Lowering of the activation energy by introducing additional $NH_3$ molecule amounted to ca. −20 $\sim$ −25 kcal/mol, which made catalyzed reaction more facile than un-catalyzed one.

Instability and Transition of Nonparallel Bouyancy-Induced Flows Adjacent to an Ice Surface Melting in Water (얼음 벽면의 융해율을 고려한 비평행 자연대류에서 유동의 불안정성과 천이에 관한 연구)

  • A set of stability equations is formulated for natural convection flows adjacent to a vertical isothermal surface melting in cold pure water. It takes account of the nonparallelism of the base flows. The melting rate is regarded as a blowing velocity at the ice surface. The numerical solutions of the linear stability equations which constitute a two-point boundary value problem are accurately obtained for various values of the density extremum parameter $R=(T_m-T_{\infty})/(T_0-T_{\infty})$ in the range $0.3{\leq}R{\leq}0.6$, by using a computer code COLNEW. The blowing effects on the base flow becomes more significant as ambient temperature ($T_{\infty}$) increases to $T_{\infty}=10^{\circ}C$. The maximum decrease of heat transfer rate is about 6.4 percent. The stability results show that the melting at surface causes the critical Grashof number $G^*$ and the maximum frequency of disturbances to decrease. In comparision with the results for the conventional parallel flow model, the nonparallel flow model has a higher critical Grashof number but has lower amplification rates of disturbances than does the parallel flow model. The spatial amplification contours exhibit that the selective frequency $B_0$ of the nonparallel flow model is higher than that of the parallel flow model and that the effects of melting are rather small. The present study also indicates that the selective frequency $B_0$ can be easily predicted by the value of the frequency parameter $B^*$ at $G^*$, which comes from the neutral stability results of the nonparallel flow model.

  • A 2.45GHz microwave plasma source with a linear antenna has been developed for low temperature large scale deposition processing. Microwave power is transmitted through WR340 waveguide and a copper rod, linear antenna, is located in a quartz tube. The power matching is effectively achieved by a linear antenna is located at ${\lambda}_g/4$ or $3{\lambda}_g/4$ from the end of WR340 waveguide. The Ar plasma was generated along the surface of quartz tube and a clear standing wave pattern with nearly 10cm wavelength was observed at Ar pressure of 200mTorr and 200W input power. The electron density and electron temperature were investigated by using the electrostatic probe. The electron density and electron temperature were highly measured near the surface of quartz tube. Ar plasma density along the quartz tube is mostly uniform despite standing wave set-up and antenna of long length. A uniform temperature was measured at 10~40cm distance from the end quartz tube and 5cm distance from the surface of quartz tube.

Performance evaluation of organic matter adsorption from actual graywater using GAC: OrbitrapTM MS and optimization

  • The complex combination of organic contaminants in the wastewater made water treatment challenging; hence, organic matter in water bodies is usually measured in terms of organic carbon. Since it is important to identify the types of compounds when deciding suitable treatment methods, this study implemented a quantitative and qualitative analysis of the organic matter content in an actual graywater sample from Ulsan, Republic of Korea using mass spectroscopy (MS). The graywater was treated using adsorption to remove the organic contaminants. Using orbitrap MS, the organic matter content between an untreated graywater and the treated effluent were compared which yielded a significant formula count difference for the samples. It was revealed that CHON formula has the highest removal count. Isotherm studies found that the Freundlich equation was the best fit with a coefficient of determination ($R^2$) of 0.9705 indicating a heterogenous GAC surface with a multilayer characteristic. Kinetics experiments fit the pseudo-second order equation with an $R^2$ of 0.9998 implying that chemisorption is the rate-determining step between the organic compounds and GAC at rate constant of $52.53g/mg{\cdot}h$. At low temperatures, the reaction between GAC and organic compounds were found to be spontaneous and exothermic. The conditions for optimization were set to achieve a maximum DOC and TN removal which yielded removal percentages of 94.59% and 80.75% for the DOC and TN, respectively. The optimum parameter values are the following: pH 6.3, 2.46 g of GAC for every 30 mL of graywater sample, 23.39 hrs contact time and $38.6^{\circ}C$.

Comparative Study of the Effective Dose from Panoramic Radiography in Dentistry Measured Using a Radiophotoluminescent Glass Dosimeter and an Optically Stimulated Luminescence Detector

  • Accurate measurement of the absorbed dose and the effective dose is required in dental panoramic radiography involving relatively low energy with a rotational X-ray tube system using long exposures. To determine the effectiveness of measuring the irradiation by using passive dosimetry, we compared the entrance skin doses by using a radiophotoluminescent glass dosimeter (RPL) and an optically stimulated luminescence detector (OSL) in a phantom model consisting of nine and 31 transverse sections. The parameters of the panoramic device were set to 80 kV, 4 mA, and 12 s in the standard program mode. The X-ray spectrum was applied in the same manner as the panoramic dose by using the SpekCalc Software. The results indicated a mass attenuation coefficient of $0.008226cm^2/g$, and an effective energy of 34 keV. The equivalent dose between the RPL and the OSL was calculated based on a product of the absorbed doses. The density of the aluminum attenuators was $2.699g/cm^3$. During the panoramic examination, tissue absorption doses with regard to the RPL were a surface dose of $75.33{\mu}Gy$ and a depth dose of $71.77{\mu}Gy$, those with regard to the OSL were surface dose of $9.2{\mu}Gy$ a depth dose of $70.39{\mu}Gy$ and a mean dose of $74.79{\mu}Gy$. The effective dose based on the International Commission on Radiological Protection Publication 103 tissue weighting factor for the RPL were $0.742{\mu}Sv$, $8.9{\mu}Sv$, $2.96{\mu}Sv$ and those for the OSL were $0.754{\mu}Sv$, $9.05{\mu}Sv$, and $3.018{\mu}Sv$ in the parotid and sublingual glands, orbit, and thyroid gland, respectively. The RPL was more effective than the OSL for measuring the absorbed radiation dose in low-energy systems with a rotational X-ray tube.

Serum cholinesterase activities and serum bromide concentrations in farmers (농업인에서의 체내 잔류농약 검출)

  • The usage or agricultural chemicals is on the increasing tendency. Methyl bromide and organophosphate are the most widely used toxic agricultural chemicals in Korea. We try to set up the methods to detect the accumulation of these chemicals in Korean farmers. Blood samples were collected for 121 farmers of slack's season in February 1998. And survey about arable acreage and usage of the gloves and masks was also performed. Serum cholinesterase activities and bromide concentrations were measured with gold chloride method and colorimetric method. The reference ranges of serum cholinesterase activity and bromide concentration were 1.6~15.9 U/mL and below $72.9{\mu}g/mL$. Serum bromide concentrations of farmers and normal controls showed no differences. Serum cholinesterase activities of farmers were significantly higher than those of normal controls. According to the arable acreages and usage of the gloves and masks, serum bromide concentrations and cholinesterase activities showed no differences. In conclusion, serum cholinesterase activities and bromide concentrations showed no differences between farmers of slack's season and normal controls.

  • This study deals with an analysis on the sinking resistance for the model purse seine, in the case of different netting material and sinkers. The experiment was carried out using rune simplified model seines of knotless nettings. Dimension of model seines 420cm for corkline and 85cm for seine depth, three groups of models rigged 25, 45 and 60g with the same weighted sinkers in water were used. These were named PP-25, PA-25, PES-25, PP-45, PA-45, PES-45, PP-60, PA-60 and PES-60 seine. The densitie($\rho$) of netting materials were 0.91g/cm$cm^3$, 1.14g/cm$cm^3$ and 1.38g/cm$m^3$. Experiments carried out in the observation channel in a flume tank under still water conditions. Sinking motion was recorded by the one set of TV-camera for VTR, and reading coordinate carried out by the video digitization system. Differential equations were derived from the conservation of momenta of the model purse seines and used to determine the sinking speeds of the depths of leadline and the other portions of the seines. An analysis carried out by simultaneous differential equations for numerical method by sub-routine Runge-Kutta-Gill The results obtained were as follows : 1. Average sinking speed of leadline for the model seines rigged 60g with the same weighted sinkers in water was fastest for 12.2cm/sec of PES seine, followed by 11.4cm/sec of PA and 10.7cm/sec of PP seines. 2. The coefficient of resistance for netting of seine was estimated to be $K_D=0.09(\frac{\rho}{\rho_w})^4$ 3. The coefficient of resistance for netting bundle of seine was estimated to be $C_R=0.91(\frac{\rho}{\rho_w})$ 4. In all seines, the calculated depths of leadline closely agreed with the measured ones, each 25g, 45g, 60g of weighted sinkers were put into formulas meas.=1.04cal., meas.=0.99cal. and meas.=0.98 cal.

A Studyon the Movment of Ground Water of Banayweol Formation (반야월층(半夜月層)의 지하수운동(地下水運動)에 관(關)한 연구(硏究))

  • This study deals with the flow of bed rock ground water of Banyaweol Formation, which is presently cleared up as a laminar flow. The result obtained may be summarized as the following. 1) The Banyaweol Formation consists mainly of thin-bedded, green to blackish green shale, mudstone, and marl. The marl and mudstone alternatively occur with shale. The marl and mudstone form a aquifer of Banyaweol Formation. In this study, a group of aquifer is in convenience named as a aquifer zone. The aquifer occurs in lenticular form. The aquifer seems to be a type of artesian aquifer because it is covered with aquicludes, but it actually forms a unconfined aquifer because its piezometric surface stays under the lower aquiclude. The lowering of piezometric level is formed because of leakage of the ground water to the lower aquifer undersaturated. 2) The coefficient of permeability of Banyaweol Formation's ground water body (K) is derived by using Dupuit's equation as the following ${\log}K=\frac{CK^2-dK+f}{aK-b}\;\(M=1.365(2H-s)s\\M={\log}1.956s{\sqrt{H}}r\)$ here, $$a=\sum_{1}M_iG_i$$ $$b={\frac{1}{2}log{\sum_{i}}Q_i{^2}$$ $$c=2{\sum_{i}}M_i{^2}$$ $$d=loge{\sum_{i}}M_{i}Q_{i}+2{{\sum_{i}}N_{i}Q_{i}$$ $$f=loge{\sum_{i}}Q_i{^2}N_i$$ If the measured values substituted for the above equation, the coefficient of permeability of the aquifer is 4.1m/day. The coefficient of storge of the aquifer is $2.8{\times}10^{-4}$ if the measured values substituted for Theis's equation. Using the above constants, the filtration velocity of the aquifer is $2.1{\times}1O^{-1}m/day$and the daily flow quantity of the ground water is $847.38m^{3}/day$. 3) In order to understand the time necessary for a circulation of ground water body, the contents of tritum contained in the ground water are measured as 2.3 T.U. at the Korea Atomic Energy Research Institute. Before 1952, the average concentration of tritium per year in groundwater was reported as 10T. u., taking it as the standard, the groundwater of the present study 26.25 years old. Therofore, the groundwater of the Banyaweol Formation is judged as an relatively old groundwater. It is characteristic that the ground water of Banyawol Formation is laminar flow as well as unconfined aquifer and ground water flow of relatively long time. 4) The nature, means of flow, and circulation of Banyaweol Formation's ground water body make it possible set up this ground water body as a ground water system.

Characteristics of Stress Drop and Energy Budget from Extended Slip-Weakening Model and Scaling Relationships (확장된 slip-weakening 모델의 응력 강하량과 에너지 수지 특성 및 스케일링 관계)

  • The extended slip-weakening model was investigated by using a compiled set of source-spectrum-related parameters, i.e. seismic moment Mo, S-wave velocity Vs, corner-frequency fc, and source-controlled high-cut frequency fmax, for 113 shallow crustal earthquakes (focal depth less than 25 km, MW 3.0~7.5) that occurred in Japan from 1987 to 2016. The investigation was focused on the characteristics of stress drop, radiation energy-to-seismic moment ratio, radiation efficiency, and fracture energy release rate, Gc. The scaling relationships of those source parameters were also investigated and compared with those in previous studies, which were based on generally used singular models with the dimensionless numbers corresponding to fc given by Brune and Madariaga. The results showed that the stress drop from the singular model with Madariaga's dimensionless number was equivalent to the breakdown stress drop, as well as Brune's effective stress, rather than to static stress drop as has been usually assumed. The scale dependence of stress drop showed a different tendency in accordance with the size category of the earthquakes, which may be divided into small-moderate earthquakes and moderate-large earthquakes by comparing to Mo = 1017~1018 Nm. The scale dependence was quite similar to that shown by Kanamori and Rivera. The scale dependence was not because of a poor dynamic range of recorded signals or missing data as asserted by Ide and Beroza, but rather it was because of the scale dependent Vr-induced local similarity of spectrum as shown in a previous study by the authors. The energy release rate Gc with respect to breakdown distance Dc from the extended slip-weakening model coincided with that given by Ellsworth and Beroza in a study on the rupture nucleation phase; and the empirical relationship given by Abercrombie and Rice can represent the results from the extended slip-weakening model, the results from laboratory stick-slip experiments by Ohnaka, and the results given by Ellsworth and Beroza simultaneously. Also the energy flux into the breakdown zone was well correlated with the breakdown stress drop, ${\tilde{e}}$ and peak slip velocity of the fault faces. Consequently, the investigation results indicate the appropriateness of the extended slip-weakening model.
