Comparative Study of Nitric Oxide Scavenging Effect in Several Herbal Extracts

  • In this study, we investigated in vitro nitric oxide (NO) scavenging effect on nine herbal extracts using an NO donor S-nitroso-N-acetylpenicillamine (SNAP) compared with vitamin C. All herbal extracts effectively reduced the generation of NO radicals in a dose-dependant manner and over 65% in 10h. Especially, Ephedrae herba, Carthami flos and Lonicerae flos declined over 75% of NO scavenging effect, suggested that the herbal extracts are the powerful free radical scavengers and may be effective in clinical applications.

항 종양성 생약의 Cytotoxicity에 관한 연구 ( I ) (A Study on the Cytotoxicities of Domestic Antitumour Crude Drugs)

  • The study on cytotoxicities of domestic antitumour crude drugs were carried out in order to evaluate the antitumour activity. The eleven crude drugs were studied in this paper. No cytotoxicities were observed both at 0.1ml of water extracts and alcohol extracts deuted against monkey kidney cell and HeLa-cell after 3 days cultivation at $37^{\circ}C$. The sample shown the heavy cytotoxicities against monkey kidney cell at 0.3ml alcohol extracts diluted sample solution are Lonicerae Flos and Puchrestae Radix.

금은화가 LPS로 유발된 급성 폐 손상에 미치는 영향 (Effects of Lonicerae Flos Extracts on LPS-induced Acute Lung Injury)

  • Objective : The object of this study was to observe the effects of Lonicerae Flos (LF) aqueous extracts on lipopolysaccharide (LPS)-induced rat acute lung injury. Method : Five different dosages of LF extracts were orally administered once a day for 28 days before LPS treatments, and then all rats were sacrificed after 5 hour-treatment of LPS. Eight groups of 16 rats each were used in the present study. The following parameters caused by LPS treatment were observed ; body weights, lung weights, pulmonary transcapillary albumin transit, arterial gas parameters (pH, $PaO_2$ and $PaCO_2$) bronchoalveolar lavage fluid (BALF) protein lactate dehydrogenase (LDH), and proinflammatory cytokines tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin-$1{\beta}$ (IL-$1{\beta}$) contents, total cell numbers, neutrophil and alveolar macrophage ratios, lung malondialdehyde (MDA), myeloperoxidase (MPO), proinflammatory cytokines TNF-${\alpha}$ and IL-$1{\beta}$ contents. In addition, the histopathologic changes were observed in the lung in terms of luminal surface of alveolus, thickness of alveolar septum, number of polymorphonuclear neutrophils. Result : As results of LPS-injection, dramatical increases in lung weights, pulmonary transcapillary albumin transit increases, increases in $PaCO_2$, decreases in pH of arterial blood and $PaO_2$, increases of BALF protein, LDH, TNF-${\alpha}$ and IL-$1{\beta}$ contents, total cells, neutrophil and alveolar macrophage ratios, TNF-${\alpha}$ and IL-$1{\beta}$ contents increases were detected with decreases in LSA and increases of alveolar septum and PMNs numbers, respectively as compared with intact control. These are means that acute lung injuries (resembling acute respiratory distress syndrome) are induced by treatment of LPS mediated by inflammatory responses, oxidative stress and related lipid peroxidation in the present study. However, these LPS-induced acute lung injuries were inhibited by 28 days continuous pretreatment of 250 and 500mg/kg of LF extracts. Because of lower three dosages of LF treated groups, 31.25 and 62.5 and 125mg/kg did not showed any favorable effects as compared with LPS control, the effective dosages of LF in LPS-induced acute lung injuries in the present study, is considered as about 125mg/kg. The effects of 250mg/kg of LF extracts showed almost similar effects with ${\alpha}$-lipoic acid 60mg/kg in preventing LPS-induced acute lung injuries. Conclusion : It seems that LF play a role in protecting the acute respiratory distress syndrome caused by LPS.

Screening of Hepatoprotective Activity of Medicinal Plant Extracts on Carbon Tetrachloride-induced Hepatotoxicity in Rats

  • The purpose of this study was to find the effect of 36 traditional medicinal plant species on hepatoprotective activity screening. For this study, carbon tetrachloride $(CCl_{4})$ intoxificated rats were used. Test extracts were made with the traditional medicinal plants refluxed in 95% MeOH and orally administered to the rats. Sixteen species, such as Mentha arvensis, Sophora japonica, Benincasa hispida, Lonicera japonica (Lonicerae Flos), Agaricus blazei, Epimedium koreanum, Aralia continentalis, Lithospermum erythrorhizon, Cimicifuga foetida, Gastrodia elata, Sanguisorba officinalis, Cephalonoplos segetum, Bupleurum falcatum, Alisma plantago-aquatica var. orientale, Lonicera japonica (Lonicerae Folium) and Sinomenium acutum showed protective effect against increased serum alanine aminotransferase (ALT) and/or serum asparteate aminotransferase (AST) activities.

Protective Effect of Lonicerae Flos Aqueous Extracts on a Pressure Overload-induced Heart Failure Model

  • Objectives: Lonicerae flos (LF), a dried flower part of Lonicera japonica Thunb., has been widely used in Korean medicine as anti-inflammatory and antioxidative agent. The purpose of this study was to determine the cardioprotective effects of LF, through potential antioxidant effects, on the pressure overload (PO)-induced heart failure (HF) in C57BL/6 mice after transverse aortic constriction (TAC) surgery. Methods: Resveratrol (10 mg/kg body weight) or LF (125, 250 or 500 mg/kg body weight) was orally administered, once daily for 14 days, starting 14 days after TAC surgery. Changes in the mortality, body weights, heart weights, histopathology of the heart, and antioxidant defense systems of the heart were analyzed. Results: Marked and noticeable increases of heart weights, mortalities, and hypertrophic, focal, and lytic fibrotic histological changes in the LVs were observed, with destruction of heart antioxidant defense systems after surgery. However, HF signs, induced by TAC surgery through PO, and destruction of heart antioxidant defense systems were significantly and dose-dependently inhibited by 14 days of maintained oral treatment with LF 500, 250 or 125 mg/kg. Treatment with 250 mg/kg LF was comparable to treatment with 10 mg/kg resveratrol. Conclusions: The results in this study suggest that oral administration of LF favorably relieves PO-induced HF following TAC, through increase of heart antioxidant defense systems. The overall effects of 250 mg/kg LF were similar to those of 10 mg/kg resveratrol. More detailed mechanistic studies should be conducted in the future, with screening of the biologically active compounds in LF.

Evaluation of Antibacterial Effects of a Combination of Coptidis Rhizoma, Lonicerae Flos, Paeonia Japonica Extracts, and Dioctahedral Smectite Against Salmonella Typhimurium in Murine Salmonellosis

  • 본 연구는 황련, 금은화, 그리고 백작약 혼합분말(1:1:1)의 메탄올 추출물(CLP1000)과, 이에 dioctahedral smectite를 혼합한 합제 (CLPS1000)의 살모넬라에 대한 항균효과를 평가하기 위해 수행되었다. CLP1000와 CLPS1000는, 0. 5mg/ml 농도에서 S. typhimurium에 대한 항균효과를 보이지 않았으나, 1.0 mg/ml의 농도에서는 S. typhimurium에 대한 항균활성이 관찰되었다. BALB/c 마우스를 이용한 살모넬라 감염시험에서 Smectite, CLP1000 그리고 CLPS1000을 각각 10 mg/ml 농도로 12일 동안 투여한 결과, 사망률이 각각 90%, 90% 그리고 70%을 보여, CLPS1000이 마우스 살모넬라증에 강력한 효과를 갖고 있는 것으로 나타났다.

인동 추출물의 성장호르몬 유발 효과 (Induction of Growth Hormone Release by the Extracts of Lonicera japonica $T_{HUNB.}$)

  • Lonicerae Flos (LF) has been used as an anticancer, anti-viral, and anti-inflammatory agent in traditional herbal medicine. In this study, induction of rat growth hormone (rGH) by addition of methanol (MeOH) extract of LF of Lonicerae (L.) Folium or several constituents of L. Folium were carried out in the pituitary cell culture system. Induced rGH level by addition of 70% MeOH extract of LF was increased to $732.65{\pm}105.64%$ of control (n=18, p<0.01), however, the other sequential fractions were not significantly different from the control. Ochnaflavone, a constituent of L. Folium, induced rGH level in the cell culture to $329.73{\pm}160.00%$ of control (n=6, p<0.01). An I..v. injection of the MeOH extract of LF did not increase plasma rGH level in anesthetized rats. Unfortunately, the MeOH extract of LF induced prolactin and LH release about 7 and 5 fold of the control, respectively (p<0.05, each). In conclusions, 70% MeOH extract of LF exerted induction of rGH release in rat pituitary cell culture. Further studies to investigate mechanisms of the inducded rGH by LF are in progress.

仙方活命飮 및 구성약물의 세포독성에 관한 실험적 연구 (Experimental Study on Cytotoxity of Sunbanhwalmyungeum and Its Composition Oriental Medicines)

  • The purpose of this research was to investigate cytotoxity of Sunbanhwalmyungeum extract. Cytotoxity was determined by MTT assay method. After tumor cell lines(G361, BI6F10, MDA, A549) transplantation, the extracts of SunBangHwalMyungEum and its composition oriental medicines were administered, cytotoxity was measured by absorbance. The results were obtained as follows. 1. Sunbanhwalmyungeum extract and its composition oriental medicines extracts showed the concentration was higher, the more cytotoxity increased. 2. Both water and ethanol extracts of Sunbanhwalmyungeum showed excellent cytotoxity against G361, B16F10, MDA, A549 and high cytotoxity over 80$\%$ against G361, B16F10, MDA except A549 at the concentration of 1000ppm. 3. In water extract, Rhei Radix et Rhizoma, Gleditsiae Spina, Trichosanthis Radix, Glycyrrhizae Radix, Ledebouriellae Radix showed excellent cytotoxity. In ethanol extract, Gleditsiae Spina, Citri Pericarpium, Trichosanthis Radix, Paeoniae Radix Rubra, Myrrha showed excellent cytotoxity. 4. Rhei Radix et Rhizoma, Gleditsiae Spina, Trichosanthis Radix, Paeoniae Radix Rubra showed high cytotoxity in both water and ethanol extrats. 5. In water extract, Glycyrrhizae Radix, Ledebouriellae Radix, Myrrha showed high cytotoxity against A361, Lonicerae Flos, Olibanum, Fritillariae cirrhosae Bulbus, Paeoniae Radix Rubra, Manitis Squama against B16F10, Paeoniae Radix Rubra, Manitis Squama against MDA, Rhei Radix et Rhizoma, Angelicae gigantis Radix against A549. 6. In ethanol extract, Lonicerae Flos, Trichosanthis Radix showed high cytotoxity against G361, Rhei Radix et Rhizoma, Angelicae gigantis Radix, Gleditsiae Spina, Olibanum, Angelicae dahuricae Radix, Fritillariae cirrhosae Bulbus, Paeoniae Radix Rubra, Glycyrrhizae Radix, Ledebouriellae Radix, Myrrha against B16F10, Rhei Radix et Rhizoma, Manitis Squama against MDA, Citri Pericarpium, Manitis Squama against A549.

금은화 물추출물의 항산화 효과와 Ultraviolet(UV)B로 유도된 사람 각질형성세포 손상에 대한 보호효과 (The antioxidative and cytoprotective effect of Lonicerae japonicae Flos water extracts on the ultraviolet(UV)B-induced human HaCaT keratinocytes)

  • Objectives : Lonicerae japonicae Flos(LJF) has been reported to exhibit anti-oxidant, anti-inflammatory, anti-viral, anti-rheumatoid properties. However, it is still largely unknown whether LJF inhibits the ultraviolet(UV)B-induced oxidative damage in human HaCaT keratinocytes. Therefore in this paper, we investigated the anti-oxidative capacity and protective effect of LJF against UVB-induced oxidative demage in human HaCaT keratinocytes. Methods : To evaluate the anti-oxidative activity of LJF extracts, we measured total phenolic contents, total flavonoid contents, antioxidant capacity, and superoxide scavenging activity. To give an oxidative stress to HaCaT cells, UVB was irradiated with $200mJ/cm^2$ to HaCaT cells. To detect the protective effect of LJF against UVB, we measured cell viability, DNA fragmentation and reactive oxygen species (ROS) production. In addition, we performed high-performance liquid chromatography (HPLC) analysis to find a major component of LJF. Results : LJF contained phenolic and flavonoid contents, and showed the anti-oxidant and superoxide scavenging activity. The UVB-induced oxidative conditions led to the cell death, DNA fragmentation and reactive oxygen species (ROS) production. However, pretreatment with LJF reduced oxidative conditions, including inhibition of cell death, DNA fragmentation and ROS production. In addition, we found out chlorogenic acid as major component of LJF. Conclusions : These results could suggest that LJF contained anti-oxidative contents and exhibited protective effects against UVB on human HaCaT keratinocytes. And the effective compound of LJF which could show protective activities against UVB is chlorogenic acid. Thus, LJF and chlorogenic acid would be useful for the development of drug or cosmetics treating skin troubles.

MTT Assay에 의한 천연물질의 항 HIV-1 활성 검색 (Screening of Anti-HIV-1 Activity of Natural Products by MTT Assay)

  • Methanol and/or boiling water extraction of 201 natural products and subsequent MTT assay using MT-4 cell line was carried out to screen the anti-HIV-1 activity. Among 97 methanol extracts, 7 extracts from Chrysanthemi Indicium Flos, Magnoliae Cortex, Machili Cortex, Reynoutriae Rhizoma, Lithospermi Radix, Agastachis Herba, and Chaenomelis Fructus showed anti-HIV-1 activity and their SI value were 2.25 to 5.77. In addition, among 119 boiling water extracts, 10 extracts from Lonicerae Caulis et Foloium, Elsholtziae Herba, Leonuri Herba, Portulacae Herba, Schizonepetae Herba, Curcumae Rhizoma, Amomi Cardamomi Fructus, Cirsii Radix et Herba, Carpesii Herba, and Siegesbeckiae Herba showed anti-HIV-1 activity and their SI value were 1.30 to 7.64. Methanol extracts of above seven natural products were fractionated and the anti-HIV-1 activity of each fraction was examined. Extraction was carried out with hexane, chloroform, butanol, and water to trace active anti-HIV-1 componets. As a result, the water fraction of Magnoliae Cortex, Machili Cortex, Reynoutriae Rhizoma, Agastachis Herba, Chaenomelis Fructus and the butanol fraction of Chrysanthemi Indicium Flos, Reynoutriae Rhizoma showed anti-HIV-1 activity and their SI value were 1.40 to 8.02. We could reach a conclusion that studies to trace the anti-HIV-1 active component of each natural products in further fractionation and to identify its structure by Infrared spectroscopy, NMR spectroscopy and gel permeation chromatography were needed.
