Antifibrotic Effects of Oriental Herbs Extraction on Liver Cirrhosis (한약재(자하거, 백출, 차전자) 추출물의 간경화 억제효능에 관한 연구)

  • Hepatic cirrhosis is an important feature of chronic liver disease. Liver cirrhosis is characterized by hyperaccumulation of fibrous tissue components and is commonly observed in latter or terminal states of chronic hepatic disease. The antifibrotic effects on liver cirrhosis by oriental herbs extraction material were examined in bile duct ligated rats. Oriental herbs extraction (0.99 mg/kg rat weight/day) was administrated to cirrohotic rats for 4 weeks. Liver collagen content of bile duct ligated rats was significantly increased. And liver histology showed collagen fiber deposition was increased as well as the normal architecture was lost with large zone of necrosis being observed. Herbs extraction administrated rats showed significantly decreased liver collagen content, accumulation of collagen fiber in histological analysis, and biochemical markers of hepatic diseases. Those results demonstrate the usefulness of herbs extraction materials as an antifibrotic agent for liver cirrhosis.

Optimization of fish oil extraction from Lophius litulon liver and fatty acid composition analysis

  • The Lophius litulon liver was used as raw material for the extraction of fish oil via various extraction methods. The extraction rate by water extraction, potassium hydroxide (KOH) hydrolysis and protease hydrolysis were compared and the results revealed the protease hydrolysis extraction had a higher extraction rate with good protein-lipid separation as observed by optical microscope. Furthermore, subsequent experiments determined neutrase to be the best hydrolytic enzyme in terms of extraction rate and cost. The extraction conditions of neutrase hydrolysis were optimized by single-factor experiment and response surface analysis, and the optimal extraction rate was 58.40 ± 0.25% with the following conditions: enzyme concentration 2,000 IU/g, extraction time 1.0 h, liquid-solid ratio 1.95:1, extraction temperature 40.5℃ and pH 6.5. The fatty acids composition in fish oil from optimized extraction condition was composed of 19.75% saturated fatty acids and 80.25% unsaturated fatty acids. The content of docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) were 8.06% and 1.19%, respectively, with the ratio (6.77:1) surpassed to the recommendation in current researches (5:1). The results in this study suggest protease treatment is an efficient method for high-quality fish oil extraction from Lophius litulon liver with a satisfactory ratio of DHA and EPA.

he Study of Nucleic Acid Extraction Method from Archival Paraffin Blocks (보존된 파라핀 블록에서 핵산 추출기법에 관한 연구)

  • It designed a study to examine the efficiency of DNA and RNA extraction from archival formalin-fixed, paraffin-embedded tissues using an non-heating and heating method. Archival paraffin blocks of liver, kidney, colon were randomly selected. Each paraffin block was prepared in 20 microtubes. For each paraffin blocks were tested non-heating DNA extraction to 10 microtubes and heating protocol under pH 7.0 and $100^{\circ}C$ to 10 microtubes. Evaluation of the results of DNA extraction was carried out by measuring concentration by UV spectrophotometry and then PCR amplification. DNA extraction content that non-heating method was liver $5{\pm}0.7{\mu}g/mL$, kidney $2{\pm}0.3{\mu}g/mL$, colon $6{\pm}0.4{\mu}g/mL$ and heating method was liver $12{\pm}0.6{\mu}g/mL$, kidney $7{\pm}0.5{\mu}g/mL$, colon $10.{\pm}0.3{\mu}g/mL$. Successful RNA extraction was observed, by ${\beta}$-actin amplification, in 46.7% sections for samples treated by the heating method versus 30.0% using non-heating DNA extraction. The extracted nucleic acid showed better values for samples heated at $100^{\circ}C$. Therefore heating extraction of nucleic acid is reliable, quick and efficiency.

The Effects of Protecting Liver and Improving Liver Function on Cabbage Extract (양배추 추출물의 간보호 및 간기능 개선 효과)

  • This study investigated the pharmaceutical extraction and the functional health food extraction, which have a beneficial effect on the human body and which can be used safely for a long period of time without adverse side effects and also have excellent effects of protecting liver and improving liver function. As the results, the cabbage extract does not show cytotoxicity, and thus can be used safely. In an experiment performed on an animal model with liver injury induced by a drug (APAP), it could be seen that the cabbage extract exhibited the effects of protecting liver and improving liver function by effectively reducing AST and ALT which are liver injury markers, indicating that the cabbage extract is effective as a pharmaceutical extraction for preventing or treating liver disease. In particular, the cabbage extract was effective in treating inflammation of the liver by reducing the expression of the inflammatory mediators iNOS and COX-2 and the proinflammatory cytokine $IL-1{\beta}$, which are involved in acute inflammatory reactions accompanying liver injury. In the results, an extract of cabbage heat-treated at a temperature of 100 to $150^{\circ}C$ had a better liver function-improving effect or anti-inflammatory effect than an extract of raw cabbage.

Development and Usability Testing of a User-Centered 3D Virtual Liver Surgery Planning System

  • Objective: The present study developed a user-centered 3D virtual liver surgery planning (VLSP) system called Dr. Liver to provide preoperative information for safe and rational surgery. Background: Preoperative 3D VLSP is needed for patients' safety in liver surgery. Existing systems either do not provide functions specialized for liver surgery planning or do not provide functions for cross-check of the accuracy of analysis results. Method: Use scenarios of Dr. Liver were developed through literature review, benchmarking, and interviews with surgeons. User interfaces of Dr. Liver with various user-friendly features (e.g., context-sensitive hotkey menu and 3D view navigation box) was designed. Novel image processing algorithms (e.g., hybrid semi-automatic algorithm for liver extraction and customized region growing algorithm for vessel extraction) were developed for accurate and efficient liver surgery planning. Usability problems of a preliminary version of Dr. Liver were identified by surgeons and system developers and then design changes were made to resolve the identified usability problems. Results: A usability testing showed that the revised version of Dr. Liver achieved a high level of satisfaction ($6.1{\pm}0.8$ out of 7) and an acceptable time efficiency ($26.7{\pm}0.9 min$) in liver surgery planning. Conclusion: Involvement of usability testing in system development process from the beginning is useful to identify potential usability problems to improve for shortening system development period and cost. Application: The development and evaluation process of Dr. Liver in this study can be referred in designing a user-centered system.

Effect of Various Garlic Extracts on Hyperlipidemia Rats Induced by Triton WR-1339 (추출조건을 달리한 마늘 추출물이 Triton WR-1339 유발 고지혈증 흰주의 체내 지질대사에 미치는 영향)

  • The purpose of this study was to investigate the effect of garlic(Allium sativum L.) extracts with extraction conditions on hyperlipidemia prevention. The extracts prepared for garlic by hot temperature extraction(HG), low temperature extraction(LG), UMPM extraction(UG), fermentation(FG) and black garlic hot temperature(BG) method. To evaluate of hypolipidemia effect in vivo, we examined serum and liver lipid profiles of Triton WR-1339 induced hyperlipidemia rats. Serum triglyceride and total cholesterol levels in UG group were significantly higher than control group. Liver total cholesterol content in LG group and liver triglyceride in UG group was significantly lower than control group.

Detection and Analysis of the Liver Area and Liver Tumors in CT Scans (CT 영상에서의 간 영역과 간 종양 추출 및 분석)

  • In Korea, hepatoma is the thirdly frequent cause of death from cancer occupying 17.2% among the whole deaths from cancer and the rate of death from hepatoma comes to about 21's persons per one-hundred thousand ones. This paper proposes an automatic method for the extraction of areas being suspicious as hepatoma from a CT scan and evaluates the availability as an auxiliary tool for the diagnosis of hepatoma. For detecting tumors in the internal of the liver from CT scans, first, an area of the liver is extracted from about $45{\sim}50's$ CT scans obtained by scanning in 2.5-mm intervals starting from the lower part of the chest. In the extraction of an area of the liver, after unconcerned areas outside of the ribs being removed, areas of the internal organs are separated and enlarged by using intensity information of the CT scan. The area of the liver is extracted among separated areas by using information on position and morphology of the liver. Since hepatoma is a hypervascular turner, the area corresponding to hepatoma appears more brightly than the surroundings in contrast-enhancement CT scans, and when hepatoma shows expansile growth, the area has a spherical shape. So, for the extraction of areas of hepatoma, areas being brighter than the surroundings and globe-shaped are selected as candidate ones in an area of the liver, and then, areas appearing at the same position in successive CT scans among the candidates are discriminated as hepatoma. For the performance evaluation of the proposed method, experiment results obtained by applying the proposed method to CT scans were compared with the diagnoses by radiologists. The evaluation results showed that all areas of the liver and liver tumors were extracted exactly and the proposed method has a high availability as an auxiliary diagnosis tools for the discrimination of liver tumors.

Segmentation of Liver Regions in the Abdominal CT Image by Multi-threshold and Watershed Algorithm

  • In this paper, we proposed a liver extracting procedure for computer aided liver diagnosis system. Extraction of liver region in an abdominal CT image is difficult due to interferences of other organs. For this reason, liver region is extracted in a region of interest(ROI). ROI is selected by the window which can measure the distribution of Hounsfield Unit(HU) value of liver region in an abdominal CT image. The distribution is measured by an existential probability of HU value of lever region in the window. If the probability of any window is over 50%, the center point of the window would be assigned to ROI. Actually, liver region is not clearly discerned from the adjacent organs like muscle, spleen, and pancreas in an abdominal CT image. Liver region is extracted by the watershed segmentation algorithm which is effective in this situation. Because it is very sensitive to the slight valiance of contrast, it generally produces over segmentation regions. Therefore these regions are required to merge into the significant regions for optimal segmentation. Finally, a liver region can be selected and extracted by prier information based on anatomic information.

A Comparsion of Nuclei Proteins in Chicken Liver and Erythrocyte (닭의 간과 적혈구의 핵 단백질의 비교연구)

  • Nuclei proteins were purified from chick liver to homogeneity by means of acid extraction CM Sephadex c 25 column chromatography and Bio Rex 70 column chromatography, The molecular weight of liver Nuclei proteins 1 and 2 as estimated by electrophoresis on SDS-polycrylamide gel are 29000 and 27,000 respectively. These molecular weights are identical with those of Nuclei Proteins 1 and 2 isolated from chick erythrocyte. The liver and erythrocyte Nuclei Proteins also co-migrated in acetic acid-urea gel electrophoresis. Furthermore the anti-sera raised against liver Nuclei Proteins 1 and 2 cross-reacted with erythrocyte Nuclei Proteins 1 and 2 respectively, However the amino acid compositions of liver Nuclei Prooteins 1 and 2 were found to be different from those of corresponding erythrocyte Nuclei proteins ; the contents of serine and proline in liver Nuclei proteins were higherocyte Nuclei proteins ; the contents of serine and proline in liver Nuclei protesins were higher than those in erythrocyte Nuclei proteins while the content of lycsine in liver Nuclei proteins was lower than the erythrocyte Nuclei proteins, These results suggest that in spite of similarities in many respects the liver and erythrocyte Nuclei proteins in chicks and different proteins.
