Effect of Water Uptake Rate of Chicken on Lipid Oxidation, Color of Meat, and Microbes of Chicken during Storage (닭고기 함수율이 저장기간 동안 지방산패도, 육색, 미생물 변화에 미치는 영향)

  • This study was performed to evaluate change of lipid oxidation, microbes, and color of chicken meat containing 6, 8, and 10% of water uptake rate(WUR) during cold storage. pH of chicken containing 6, 8, and 10% of WUR showed 6.00, 6.04, and 6.05%, at day 3, respectively. Lightness($L^*$) of chicken containing 6, 8, and 10% of WUR was 73.40, 75.27, and 75.31, at day 3, respectively. Redness($a^*$) of chicken with 10% of WUR was 2.52, which was the highest value, while no difference was found between chicken with 6 and 8% of WUR. Yellowness($b^*$) of chicken containing 6, 8, and 10% of WUR was 4.31, 4.33, and 4.36. Thiobarbituric acid reactive substances(TBARS) value of chicken containing 6, 8, and 10% of WUR was 0.107, 0.104, and 0.113 mg malondialdehyde/kg meat. The higher WUR, the higher TBARS value was shown. Total microbial number of chicken with 6, 8, and 10% of WUR was 4.04, 4.12, and 4.56 $log\;CFU/cm^2$ at day 3. E. coli of chicken with 6, 8, and 10% of WUR was 0.93, 1.40, and 1.28 $log\;CFU/cm^2$. From this result high WUR increases $L^*$ value, TBARS, and microbes of chicken meat during storage. Therefore, lower WUR would be beneficial to extend shelf life of chicken meat during storage.

  • In order to study the effect of aqua-acupuncture with Radix Angelicae gigantis extract solution by acuncture point. Radix Angelicae gigantis aqua-acupuncture was carried on every day for 3 days and 5 days 7 days respectively on the corresponding loci of Chondol(CV22) and non-acupuncture points (loci at the root of tail), after inducing hypothyroidism by thiourea administration and thereafter the contents of serum triiodothyronine$(T_3)$, thyroxine$(T_4)$, triiodothyronine uptake, free $T_3$, free $T_4$, total protein, albumin, creatinine, creatine, BUN, cholesterol, triglyceride, and total lipid were measured in hypothyroidism rats. The following results have been obtained : 1. $T_3$ was increased with statistical significance by Radix Angelicae gigantis aquq-acupuncture for 3 days, 5 days and 7 days Chondol(CV22) and non-acupuncture points as compared with the control group. 2. $T_3$ uptake was increased with statistical significance by Radix Angelicae gigantis aquq-acupuncture for 3 days, 5 days and 7 days to Chondol(CV22) as compared with the control group. 3. Free $T_4$ was increased with statistical significance by Radix angelicae gigantis aqua-acupuncture for 3 days, 5 days and 7 days to Chondol(CV22), and non-acupuncture points respectively as compared with the control group. 4. Albumin was decreased with statistical significance by Radix Angelicae gigantis aqua-acupuncture for 3 days, and 5 days and 7 days to Chondol(CV22), and non-acupuncture points respectively as compared with the control group. 5. Creatine was decreased with statistical significance by Radix Angelicae gigantis aqua-acupuncture for 3 days, and 5 days and 7 days to Chondol(CV22), and non-acupuncture points respectively as compared with the control group. 6. Cholesterol was decreased with statistical significance by Radix Angelicae gigantis aqua-acupuncture for 3 days, and 5 days and 7 days to Chondol(CV22) and non-acupuncture points respectively as compared with the control group. 7. Triglyceride was decreased with statistical significance by Radix Angelicae gigantis aqua-acupuncture for 3 days, and 5 days and 7 days to Chondol(CV22) and non-acupuncture points respectively as compared with the control group. 8. Total lipid was decreased with statistical significance by Radix Angelicae gigantis aqua-acupuncture for 3 days, and 5 days and 7 days to Chondol(CV22) and non-acupuncture points respectively as compared with the control group.

The Effects of Dioscorea bulbifera L. on Hyperthyroidism of Rats (황약자가 갑상선기능항진 유발 백서에 미치는 영향)

  • Objective: The aim was to study experimentally the effects of Dioscorea bulbifera L. induced by sodium levothyroxine. Methods: Sprague-Dawley rats were separated into four groups. Except for the normal group, the other three were treated with sodium levothyroxine $160{\mu}g/kg$ for 5 days. Among the three groups, two (except the control) were treated with Dioscorea bulbifera L. extract of 0.75g/kg (sample A) or 1.5g/kg (sample B). $T_3-uptake,\;T_3,\;T_4$, TSH, total cholesterol, glucose, ALP, AST, ALT, tree fatty acids, $\beta-lipoprotein$, and change in the serum of rats were measured after medication with solid extract of Radix Scrophulariae. Results: $T_4$ concentration and $T_3-uptake$ level decreased in sample A and B compared to controls. $T_3$ concentration decreased but not statistically meaningful. Considering lipid metabolism, only free fatty acid level was significantly reduced glucose level also significantly decreased in sample B. Serum AST, ALT and ALP levels were slightly elevated but there was no meaning statistically, which doesn't mean Dioscorea bulbifera L. has liver toxicity because the quantity of the extract perhaps couldn't reach the minimal dose causing liver toxicity, which according to an article was 60g. Body weight significantly increased in comparison to controls. Conclusion: These results suggest that Dioscorea bulbiftra L. lowers excess thyroid hormone and increased metabolism, resulting in improvement of hyperthyroid state.

The Mechanism of Hematoxylin on Glucose Metabolism Improvement in Sciatic Nerves from Streptozotocin-induced Diabetic Rats (Streptozotocin으로 유도한 당뇨쥐의 대퇴신경에서 Hematoxylin의 당대사 개선 메커니즘)

  • Hematoxylin is the main component of Hematoxylon campechianum which has been utilized in the southern provinces of Korea as a folk remedy for diabetic complications. In the present study, to investigate the hypoglycemic mechanism of hematoxylin, the 2-deoxyglucose uptake and phospholipid metabolism were examined in sciatic nerves from three groups of rats : normal control, diabetic control, diabetic hematoxylin-treated group. Hematoxylin significantly reduced blood glucose levels in diabetic control rats. On a wet weight basis, the nerves from diabetic rats showed a 20% decrease in total phospholipid from that of controls and a relative decrease in phosphatidylinositide. Hematoxylin treatment increased the incorporation rate of 2-[3H] myo-inositol into total phosphoinositids in diabetic rat. The effectiveness were more potent in higher dose hematoxylin-treated rats than lower dose hematoxylin-treated rats. These results suggest that hematoxylin increases glucose transport and lipid metabolism by partially normalizing concerned with myo-inositol metabolism in diabetic rat. Therefore we propose that hematoxylin can be a promising candidate for diabetes medication.

Ursodeoxycholic acid decreases age-related adiposity and inflammation in mice

  • Ursodeoxycholic acid (UDCA), a natural, hydrophilic nontoxic bile acid, is clinically effective for treating cholestatic and chronic liver diseases. We investigated the chronic effects of UDCA on age-related lipid homeostasis and underlying molecular mechanisms. Twenty-week-old C57BL/6 male and female mice were fed a diet with or without 0.3% UDCA supplementation for 25 weeks. UDCA significantly reduced weight gain, adiposity, hepatic triglyceride, and hepatic cholesterol without incidental hepatic injury. UDCA-mediated hepatic triglyceride reduction was associated with downregulated hepatic expression of peroxisome proliferator-activated receptor-γ, and of other genes involved in lipogenesis (Chrebp, Acaca, Fasn, Scd1, and Me1) and fatty acid uptake (Ldlr, Cd36). The inflammatory cytokines Tnfa, Ccl2, and Il6 were significantly decreased in liver and/or white adipose tissues of UDCA-fed mice. These data suggest that UDCA exerts beneficial effects on age-related metabolic disorders by lowering the hepatic lipid accumulation, while concurrently reducing hepatocyte and adipocyte susceptibility to inflammatory stimuli.

Current Guidelines on the Management of Dyslipidemia (이상지질혈증의 국내 및 국외 치료 가이드라인 비교)

  • Objective: Dyslipidemia is recognized as a prominent risk factor for cardiovascular and cerebrovascular diseases but it is manageable through therapeutic and lifestyle intervention. Interpreting the latest guidelines is essential for an application of recommendation from guidelines into clinical practice. Therefore, this study aimed to compare the most recent guidelines on dyslipidemia treatment recommendations in Korea and USA. Methods: This study analyzed and compared 2015 Korean guidelines for the management of dyslipidemia, 2013 American College of Cardiology/American Heart Association (ACC/AHA) guideline and 2016 supportive guidelines from ACC. Results: A comparison was made focused on the following: target patients based on cardiovascular risk assessment, target goal, and treatment strategies including statin and non-statin therapies. Four target patient groups by risk were suggested in 2015 Korean guideline and cardiovascular risk factors were also considered for initiation of lipid lowering therapy. Titrated statin regimen was recommended by Korean guideline to reach LDL cholesterol and non-HDL cholesterol target level. In 2013 ACC/AHA guideline, four statin benefit group was introduced considering ASCVD risk and high intensity statin or intermediate intensity statin use were recommended without dose titration. 2016 update was to support non-statin therapy based on updated evidence and new consideration of ezetimibe, PCSK9-inhibitor and bile acid sequestrant was brought up. Conclusion: Guidelines are continuously updating as new and important clinical data are constantly released along with the advent of newly approved drugs for lipid disorder. This article provides resources that facilitates uptake of these recommendations into clinical practice.

Effect of conjugated linoleic acid in diacylglycerol-rich oil on the lipid metabolism of C57BL/6J mice fed a high-fat high-cholesterol diet

  • The effect of conjugated linoleic acid (CLA) isomers esterified in diacylglycerol (DAG)-rich oil on lipid metabolism was investigated. Since dietary DAG has been known to induce the regression of atherosclerosis, CLA-DAG and olive-DAG oils containing similar levels of DAG (51.4~54.2%) were synthesized from olive oil. Hyperlipidemic C57BL/6J mice were then fed high-fat high-cholesterol diets supplemented with these oils (5% each) for 7 wk. The CLA-DAG diet containing 2.1% CLA isomers (0.78% c9,t11-CLA; 1.18% t10,c12-CLA) remarkably increased the levels of total plasma cholesterol and glutamic oxaloacetic transaminase (GOT) along with hepatic cholesterol and triacylglycerol (TAG) contents. Furthermore, the CLA-DAG diet inhibited fat uptake into adipose tissue whereas fat deposition (especially in the liver) was increased, resulting in the development of fatty livers. Hepatic fatty acid composition in the CLA-DAG mice was different from that of the olive-DAG mice, showing higher ratios of C16:1/C16:0 and C18:1/C18:0 in the liver. The activity of hepatic acyl-CoA:cholesterol acyltransferase (ACAT) was higher in CLA-DAG mice while plasma lecithin:cholesterol acyltransferase (LCAT) activity and the ferric reducing ability of plasma (FRAP) were lower in CLA-DAG mice compared to the olive-DAG animals. Results of the present study suggest that CLA incorporation into DAG oil could induce atherosclerosis in mice.

Nannochloris eucaryotum growth: Kinetic analysis and use of 100% CO2

  • Microalgae are receiving an increasing attention because of their potential use as $CO_2$ capture method and/or as feedstock for biofuels production. On the other hand the current microalgae-based technology is still not widespread since it is characterized by technical and economic constraints that hinder its full scale-up. In such contest the growth kinetics of Nannochloris eucaryotum (a relatively unknown marine strain) in batch and semi-batch photobioreactors is quantitatively investigated with the aim of obtaining the corresponding kinetic parameters suitable for process engineering and its optimization. In particular the maximum growth rate was evaluated to be 1.99 $10^{-3}\;h^{-1}$. Half saturation concentrations for nitrates ($K_N$) and phosphates uptake ($K_P$) were evaluated as 5.4 $10^{-4}\;g_N\;L^{-1}$ and 2.5 $10^{-5}\;g_P\;L^{-1}$, respectively. Yield factors for nitrogen ($Y_N$) and phosphorus ($Y_P$) resulted to be 5.9 $10^{-2}\;g_N\;g^{-1}$ biomass and 6.0 $10^{-3}\;g_P\;g^{-1}{_{biomass}}$, respectively. The possibility of using 100% (v/v) $CO_2$ gas as carbon source is also evaluated for the first time in the literature as far as N. eucaryotum is concerned. The strain showed a good adaptability to high concentrations of dissolved $CO_2$ as well as to low pH. The lipid content under 100% $CO_2$ is about 16.16 %wt $wt^{-1}$ and the fatty acid methyl esters composition of the extracted oil is in compliance with the European regulation for quality biodiesel.

Alpha-Lipoic Acid Inhibits Glycogen Synthesis and Modifies Glucose Metabolism and Signaling Pathways in Soleus Muscles from Healthy Rats

  • Alpha-lipoic acid is a known hypoglycemic agent that may be useful in the treatment of diabetes. The objective of this study was to investigate the fate of glucose in isolated muscles incubated with lipoic acid by determining its direct effects on specific metabolic and signaling pathways. Soleus muscles from healthy rats were incubated with lipoic acid in the absence or presence of insulin. Glucose transport, glycogen synthesis, glucose oxidation and lipid synthesis were determined and affects on major pathways associated with insulin signaling were evaluated. Glucose transport was not significantly altered by the addition of lipoic acid to the incubation medium. However, lipoic acid decreased glycogen synthesis in comparison to controls. Glucose oxidation was moderately increased while de-novo lipid synthesis from glucose was inhibited. Wortmannin repressed insulin stimulation of glucose incorporation into glycogen, an effect that was augmented by the combined treatment of wortmannin and lipoic acid. Basal and insulin-stimulated serine phosphorylation of Akt was not changed by the addition of lipoic acid to the incubation medium. These data show that in this in vitro model, lipoic acid did not significantly affect glucose uptake but dramatically modified pathways of glucose metabolism within muscle tissue.

Anti-diabetic Effect of Opuntia humifusa Stem Extract (손바닥선인장(Opuntia humifusa) 줄기 추출물의 항당뇨 효과)

  • Opuntia humifusa known as the Eastern prickly pear have been used as a treatment of burns, diarrhea, asthma, rheumatism, gonorrhea, and diabetes in alternative medicine. O. humifusa is widely cultivated in the middle and southern provinces of Korea and distributed in North America. The aim of this study is to investigate anti-diabetic effect of O. humifusa stem (OHS) water or 80% MeOH extract using 3T3-L1 adipocytes and db/db mice animal models. OHS 80% MeOH extract at a dose of $250{\mu}g/ml$ significantly increased the glucose uptake and lipid accumulation compared with the control in 3T3-L1 adipocytes. Blood glucose, plasma total cholesterol and triglyceride levels were significantly reduced by oral treatment of OHS 80% MeOH extract (200 mg/kg BW) for 6 weeks in db/db mice. Also, the oral treatment of OHS 80% MeOH extract slightly changed the plasma insulin and insulin resistance levels in db/db mice, but were no significance in comparison to control. Glucose transporter(GLUT)4 expressions of adipose tissue and muscle were significantly increased more than that in the control. Therefore, these results suggest that OHS 80% MeOH extract inhibits the blood glucose level through regulation of lipid profile, insulin resistance, and GLUT4 expression in db/db mice and its diabetic effect is effective more than water extract.
