Improvement of Silkworm Egg Microinjection Using 3D Printing Technology (3D 프린팅 기술을 이용한 누에 알 미세주입 기술 개선)

  • Silkworms, which have for long been used as an insect resource for industrialization, have recently attracted attention as potential bio-factories for the production of novel biomaterials. In this regard, material production is typically achieved based on transformation technology, mediated via microinjection, in which a target gene is inserted into eggs containing an embryo. However, an essential step in the microinjection procedure is egg fixation, which can be a time-consuming and laborious task. Therefore, in this study, using the 3DCADian program, we adopted a 3D printing approach to model egg liners and glue drawers, which can contribute to facilitating egg alignment and fixation, thereby enhancing transformation efficiency by reducing time consumption and fatigue. After rendering using Fusion 360, the two supplementary tools were produced by printing with nylon resin (PA12) and Sinterit Lisa Pro. Subsequent analysis of the time required to fix eggs on glass slides using the two manufactured tools, revealed that the processing time was reduced by approximately 18.6% when the two tools were used compared with when these tools were not used. These innovations not only reduced fatigue but also contributed to more effective use of the microscope and manipulator for microinjection. Consequently, we believe that with additional research and refinement, the egg liner and glue drawer developed in this study could be used to enhance silkworm transformation efficiency and study similar transformation systems in other industrial insects.

Establishing a Demand Forecast Model for Container Inventory in Liner Shipping Companies (정기선사의 컨테이너 재고 수요예측모델 구축에 대한 연구)

  • This study attempts to establish a precise forecast model for the container inventory demand of shipping companies through forecasts based on equipment type/size, ports, and weekly system dynamics. The forecast subjects were Shanghai and Yantian Ports. Only dry containers (20, 40) and high cubes (40) were used as the subject container inventory in this study due to their large demand and valid data computation. The simulation period was from 2011 to 2017 and weekly data were used, applying the actual data frequency among shipping companies. The results of the model accuracy test obtained through an application of Mean Absolute Percentage Error (MAPE) verified that the forecast model for dry 40' demand, dry 40' high cube demand, dry 20' supply, dry 40' supply, and dry 40' high cube supply in Shanghai Port provided an accurate prediction, with $0%{\leq}MAPE{\leq}10%$. The forecast model for supply and demand in Shanghai Port was otherwise verified to have relatively high prediction power, with $10%{\leq}MAPE{\leq}20%$. The forecast model for dry 40' high cube demand and dry 20' supply in Yantian Port was accurate, with $0%{\leq}MAPE{\leq}10%$. The forecast model for supply and demand in Yantian Port was generally verified to have relatively high prediction power, with $10%{\leq}MAPE{\leq}20%$. The forecast model in this study also had relatively high accuracy when compared with the actueal data managed in shipping companies.

Stability Analysis of Waste Landfill Using Multi-interface Element Numerical Method (복합 경계면요소 수치해석에 의한 매립지 안정성 해석)

  • A finite element nonlinear stress-deformation model with multi-interface element is applied to the stability analysis of waste landfill slope. Strength parameters of waste and geosynthetic materials are obtained from the triaxial test of waste and the direct shear test of geosynthetics, respectively. The landfill models used for the numerical models are fit to regulations of the Korean waste management law. The results of the strength tests showed linear behavior for the waste and nonlinear behavior for the eosynthectic materials. The stability analysis with multi-interface element for the geosynthetic materials in the liner system showed large shear stress and slippage at the boundary of the foundation and the slope of the waste fill. This analysis verified the necessity of multi-interface analysis for waste landfills with composite liners.

  • Maxwell displacement current (MDC) measurement has been employed to study the dielectric property of Langmuir-films. MDC flowing across monolayers is analyzed using a rod-like molecular model. A method for determining the dielectric relaxation time ${\tau}$ of floating 'monolayers on the water surface is presented. MDC floing across monolayers is analyzed using a rod-like molecular model. It is revealed that the dielectric relaxation time ${\tau}$ of monolayers in the isotropic polar orientational phase is determined using a liner relationship between the monolayer compression speed a and the molecular area Am. Compression speed a was about 30,40,50mm/min. LB layers of Arachidic acid deposited by LB method were deposited onto slide glass as Y-type film.The physicochemical properties of the LB films were examined by UV absorption spectrum, SEM and AFM. The structure of manufactured device is Au/Arachidic acid/Al, the number of accumulated layers are 3~9. Also, we then examined of the MIM device by means of I-V characteristic of the device is measured from -3 to +3[V]. The insulation property of a thin film is better as the distance between electrodes is larger.

A Comparison Study between Korean and Foreign Shipping Liners on Logistics Service Attributes and Capability (국적선사와 외국적선사의 물류서비스 속성과 역량에 관한 비교 연구)

  • In order to obtain and maintain competitive advantage, under rapidly changing maritime environment, liners should make efforts to reinforce their logistics service capabilities. Most of all, for this, shipping lines should accurately recognize and evaluate their logistics service attributes. In the present research, various logistics service attributes for ocean carriers were selected from previous studies and the observed data for 6 main service attributes were collected through a questionnaire survey to 31 maritime experts. Moreover, shipping lines' relative logistics service capabilities were measured by an additional survey process. The comparative gravity between core evaluation factors were measured by AHP technique and the relative capability levels of 2 Korean liners and 5 foreign shipping companies were assessed by utilizing a fuzzy model. The empirical result presents that Korean liners are superior at customer service capability but inferior at freight rate competitiveness. This research shows that Korean liners are ranked at high position for overall logistics service capabilities and focusing on the differentiation strategy but not on the cost related strategy.

  • Container terminals at Gwangyang Port are operated by three container operators: A, B and C. Ultimately, there is consensus that a single operator should operate all terminals so that economies of scale can be achieved even in the operation of the container terminal. Integration between operators has a positive effect on both operators and shipping companies. From the operator's point of view, overlapping fixed costs between operators can be unified, reducing overall costs and utilizing spare facilities. On the other hand, from the viewpoint of the shipping company, it is possible to ensure stable use of the port facilities and always allow berthing, reduce days on demurrage and ship waiting, and provide one-stop service for work. However, existing cases of operators' integration or relocation of terminals remained to estimate the expected effects of alternatives, emphasizing only the financial point of view. The port terminal is a large system, and it is important to consider that it is an aggregate of major logistics facilities and equipment. Moreover, if the estimation can be made by quantifying the expected effect, the justification of the terminals' relocation can be further emphasized. Therefore, it is very important to estimate the expected effect from the viewpoint of systemic operation. Moreover, the need for operators' integration can be further emphasized if it can be estimated through quantification of expected effects. Currently, three alternatives are considered as alternatives to the terminals' relocation, and in this study, the optimal plan was derived for the 3 alternatives by the linear planning model of the minimum shuttle transportation cost in the terminal. The optimal plan is alternative 2, which shows the most advantageous integration effect in terms of expected effects. Alternative 2 integrates the B terminal into the C terminal, and the A terminal operates independently as it is.

Model testing of leakage effect due to multi-cell inflater of rapid protection system in subsea tunnel (해저터널 급속차폐시스템의 Inflater 분할구조에 따른 터널 내 차수효율에 대한 실험적 연구)

  • This paper presents the study of leakage effect due to multi-cell inflater of rapid protection system to protect the possibilities of tunnel damages by flooding threats and unusual leakage to be occurred during and after subsea tunnel construction. Particularly, this protect system should be necessary in subsea tunnel. This research concentrates the physical model tests due to several multi-cell inflater to study protection capacity of leakage between the inflater and tunnel liner. A 27:1 small scale model are used in the model tests. The leakage rate, water pressure and axial displacement of inflater are measured during the model tests. According to the results, the minium leakage rate clearly shows in the case of two-cell inflater compared with in other cases. It is concluded that the results of this research will be very useful to understand the fundamental information of inflater structure design and development the technology of tunnel protection structures in the future.

AGAR-ALGINATE COMBINED IMPRESSION TECHNIQUE AND DIMENSIONAL CHANGE RESULTING FROM IDOPHOR DISINFECTION (아기-알지네이트 연합 인상체의 보관방법 및 idophor 침적소독에 의한 체적 변화에 관한 연구)

  • Statement of problem : Because dental impression can be a cross-infection source, disinfection, such as immersion disinfection is nessecary. However, the disinfection process may change the volume of the impression cast. Purpose : The purpose of this study is to find out the effects on dimensional change of different storage times and methods, of agar-alginate impression when immersed in a 5% idophor disinfectant. Material and Methods : An agar-alginate impression was made from a mandible model and then was disinfected and stored according the experimental conditions and a stone model was produced. Measurements were taken between reference points on the original mandible model and they were compared to measurements taken between reference points on the stone model. The study was divided into 4 groups. In group 1, the impression was stored in a 100% humidor for 30 minutes, 1 hour, 2 hours, 3 hours, 4 hours, 6 hours, 12 hours and 24 hours and stone models were made at each time. In group 2, the impression was immersed in 5% idophor disinfectant and then stored in a 100% humidor for the same length of times as group 1, and stone models were made at each time. In group 3, the impression was stored in extend-A-pourR, a special storage solution for 30 minutes, 1 hour, 2 hours, 3 hours, 4 hours, 6 hours, 12 hours, 24 hours 3 days, and 7 days and stone models were made at each time. In group 4, the impression was immersed in 5% idophor disinfectant and then stored in stored in extend-A-pourR, a special storage solution for the same length of times as group 3, and stone models were made at each time. 5 impressions and stone models were made at each time to make a total of 180. The Student-t test (P < .05) was used to do a statistical analysis of the measurements of the mandible model and stone models. The repeated-measure 2-way analysis of variance (P < .05) was used to do a statistical analysis of the difference in the 4 groups. Results : The percent liner dimensional change was from $0.25{\pm}0.03%$ (group 1, 30 minutes) to $0.34{\pm}0.06%$ (group 4, 7 days). No significant change was noticed between the 4 groups. Conclusion : According to the above study, in both methods, least dimensional change was recorded when the storage times were short and in both methods, immersion in 5% idophor disinfectant did not effect dimensional change in the agar-alginate impression.

A Study on the Determination of Reference Parameter for Aircraft Impact Induced Risk Assessment of Nuclear Power Plant (원전의 항공기 충돌 리스크 평가를 위한 대표매개변수 선정 연구)

  • In this study, we developed a methodology to determine the reference parameter for an aircraft impact induced risk assessment of nuclear power plant (NPP) using finite element impact analysis of containment building. The target structure used to develop the method of reference parameter selection is one of the typical Korean PWR type containment buildings. We composed a three-dimensional finite element model of the containment building. The concrete damaged plasticity model was used for the concrete material model. The steels in the tendon, rebar, and liner were modeled using the piecewise-linear stress-strain curves. To evaluate the correlations between structural response and each candidate parameter, we developed Riera's aircraft impact force-time history function with respect to the variation of the loading parameters, i.e., impact velocity and mass of the remaining fuel. For each force-time history, the type of aircraft is assumed to be a Boeing 767 model. The variation ranges of the impact velocity and remaining fuel percentage are 50 to 200m/s, and 30 to 90%, respectively. Four parameters, i.e., kinetic energy, total impulse, maximum impulse, and maximum force are proposed for candidates of the reference parameter. The wellness of the correlation between the reference parameter and structural responses was formulated using the coefficient of determination ($R^2$). From the results, we found that the maximum force showed the highest $R^2$ value in most responses in the materials. The simplicity and intuitiveness of the maximum force parameter are also remarkable compared to the other candidate parameters. Therefore, it can be concluded that the maximum force is the most proper candidate for the reference parameter to assess the aircraft impact induced risk of NPPs.

Non-Liner Analysis of Shear Beam Model using Mode Superposition (모드중첩법을 이용한 전단보 모델의 비선형 해석)

  • To analyze the dynamic behavior of structure, direct integration and mode superposition may be utilized in time domain analysis. As finite number of frequencies can give relatively exact solutions, mode superposition is preferable in analyzing structural behavior. In non-linear analysis, however, mode superposition is seldom used since time-varying element stiffness changes stiffness matrix, and the change of stiffness matrix leads to the change of essential constants - natural frequencies and mode shapes. In spite of these difficulties, there are some attempts to adopt mode superposition because of low cost compared to direct integration, but the result is not satisfactory. In this paper, a method using mode superposition in non-linear analysis is presented by separating local element stiffness from global stiffness matrix with the difference between linear and non-linear restoring forces to the external force vectors included. Moreover, the hysteresis model changing with the relative deformation in each floor makes it possible to analyze non-linear behavior of structure. The proposed algorithm is applied to shear beam model and the maximum displacement is compared with the result using direct integration method.
