Selection of Lactococcus lactis HY7803 for Glutamic Acid Production Based on Comparative Genomic Analysis

  • Comparative genomic analysis was performed on eight species of lactic acid bacteria (LAB)-Lactococcus (L.) lactis, Lactobacillus (Lb.) plantarum, Lb. casei, Lb. brevis, Leuconostoc (Leu.) mesenteroides, Lb. fermentum, Lb. buchneri, and Lb. curvatus-to assess their glutamic acid production pathways. Glutamic acid is important for umami taste in foods. The only genes for glutamic acid production identified in the eight LAB were for conversion from glutamine in L. lactis and Leu. mesenteroides, and from glucose via citrate in L. lactis. Thus, L. lactis was considered to be potentially the best of the species for glutamic acid production. By biochemical analyses, L. lactis HY7803 was selected for glutamic acid production from among 17 L. lactis strains. Strain HY7803 produced 83.16 pmol/μl glutamic acid from glucose, and exogenous supplementation of citrate increased this to 108.42 pmol/μl. Including glutamic acid, strain HY7803 produced more of 10 free amino acids than L. lactis reference strains IL1403 and ATCC 7962 in the presence of exogenous citrate. The differences in the amino acid profiles of the strains were illuminated by principal component analysis. Our results indicate that L. lactis HY7803 may be a good starter strain for glutamic acid production.

Effects of Supplementation of Probiotics on the Performance, Nutrient Digestibility and Faecal Microflora in Growing-finishing Pigs

  • Two experiments were conducted to investigate the effect of dietary supplementation of Bacillus, Saccharomyces and lactic acid bacteria (LAB) on performance and nutrient digestibility in grower and finisher pigs. In Exp. 1, 80 pigs (32 females and 48 males), $28.7{\pm}0.9\;kg$ body weight (BW), were randomly divided into 4 treatment groups balanced for sex and weight (5 pigs per pen, 4 pens per treatment). They were fed one of four diets: a basal grower (20-50 kg BW) and finisher (>50 kg BW) diet without any addition of probiotic or antibiotic (diet C), the basal diet supplemented with Bacillus subtilis H4 (diet B), diet B supplemented with Saccharomyces boulardi Sb (diet BS) and diet BS supplemented with a LAB complex (diet BSL). The LAB complex consisted of Enterococcus faecium 6H2, Lactobacillus acidophilus C3, Pediococcus pentosaceus D7, and Lactobacillus fermentum NC1. In Exp. 2, 16 male pigs, $29.2{\pm}0.8\;kg$ BW, were kept in individual pens and divided into 4 groups (4 pigs in each group). All 4 groups were given exactly the same growing-period diets (diet C, B, BS and BSL) as in Exp 1. The total faeces and urine were collected during 5 days (day 20-24) to determine nitrogen retention and total tract digestibility. In the growing period, average daily feed intake (ADFI), average daily gain (ADG) and feed conversion ratio (FCR) were not affected by diet B and BS (p>0.05), but ADG increased (+5.9%) (p<0.05) and FCR improved (+5.9%) (p<0.05) on diet BSL compared with the control, although ADFI was not different (p>0.05). Digestibility of crude protein and organic matter was higher (p<0.05) in diet BSL and digestibility of crude fibre was higher (p<0.05) in diet BS and BSL than in diet C. Nitrogen retention was not affected by diet (p>0.05). The faecal LAB counts were increased in grower pigs fed diet BSL (p<0.05) and faecal E. coli counts were decreased in pigs fed diets BS and BSL (p<0.05). In the finishing period, no effects of diet were found in ADFI, ADG, FCR, nutrient digestibility, and nitrogen retention (p>0.05). Faecal LAB and E. coli counts in the finisher pigs were not affected by diet (p>0.05). In conclusion, the current study demonstrates that a mixture of bacteria and yeast has the potential to be used as a probiotic dietary supplement in grower pigs.

Effects of Lactobacilli on the Performance, Diarrhea Incidence, VFA Concentration and Gastrointestinal Microbial Flora of Weaning Pigs

  • Two experiments were conducted to evaluate the effects of a complex Lactobacilli preparation on performance, resistance to E. coli infection and gut microbial flora of weaning pigs. In exp. 1, twelve pigs (7.65$\pm$1.10 kg BW), weaned at 28 d, were randomly allotted into 2 groups and placed in individual metabolic cages. During the first 7 d, one group of pigs was provided ad libitum access to water containing $10^5$ colony forming units (CFU) Lactobacilli per ml and the control group was provided tap water. The Lactobacilli preparation included Lactobacillus gasseri, L. reuteri, L. acidophilus and L. fermentum, which were isolated from the gastrointestinal (GI) tract mucosa of weaning pigs. On d 8, 20 ml of $10^8$ CFU/ml E. coli solution (serovars K99, K88 and 987P at the ratio of 1:1:1) was orally administered to each pig. Diarrhea scores and diarrhea incidence were recorded from d 7 to 14. On d 14, pigs were euthanized and digesta and mucosa from the stomach, duodenum, jejunum, ileum, cecum and colon were sampled using aseptic technique to determine microflora by culturing bacteria in selective medium. The results showed that Lactobacilli treatment significantly decreased E. coli and aerobe counts (p<0.01) but increased Lactobacilli and anaerobe counts (p<0.01) in digesta and mucosa of most sections of the GI tract. A 66 and 69.1% decrease in diarrhea index and diarrhea incidence, respectively, was observed in the Lactobacilli treated group. In exp. 2, Thirty-six crossbred Duroc$\times$Landrace$\times$Yorkshire piglets, weaned at 28$\pm$2 days, were selected and randomly allocated into 2 groups. There were 18 piglets in each group, 3 piglets in one pen and 6 replicates in each treatment with 3 pens of barrow and 3 pens of female piglet in each treatment. Piglets had ad libitum access to feed and water. The initial body weight of piglet was 7.65$\pm$1.09 kg. Dietary treatments included a non-medicated basal diet with Lactobacilli ($10^5$ CFU/g diet) or carbadox (60 mg/kg) as control. On d 21, six pigs per group (one pig per pen) were euthanized. Ileal digesta was collected to determine apparent amino acid digestibility. Microflora content was determined similarly to exp.1. The results showed that Lactobacilli treatment significantly improved average daily feed intake (ADFI) of pigs compared to carbadox (p<0.05) during the first 2 wks after weaning and average daily gain (ADG) and ADFI increased significantly (p<0.05) from d 8 to 14. Nitrogen and total phosphorus digestibility also increased (p<0.05). Bacterial counts were similar to exp. 1. The results indicate that the complex Lactobacilli preparation improved performance for 2 wks after weaning, enhanced resistance to E. coli infection, and improved microbial balance in the GI tract.

전통막걸리에서 분리한 효모균주를 이용한 막걸리 발효과정 중의 물성 및 미생물 군집의 변화 (Variations of Properties and Microbial Community during Fermentation of Makgeollies by Isolated Yeasts from Traditional Makgeollies)

  • 전통막걸리들에서 순수분리한 효모균주 5종을 이용하여 각각 막걸리를 제조하면서 발효 시일에 따른 막걸리의 물성 변화와 PCR-DGGE를 이용하여 미생물 군집구조 변화를 조사하였다. 막걸리의 pH는 0일째 pH 6에서 발효 2일째 pH 3 정도의 큰 감소를 보였고 이후 큰 변화가 없었다. 발효일별로는 차이를 보였지만(ANOVA, p<0.001) 균주별로는 차이가 없었다(p=0.60). 산도는 0.19~1.04% 범위였으며 발효 2일째부터 증가하였고 발효일자별(p<0.001) 및 균주별(p=0.006) 모두 차이가 있었다. C 균주의 산도 증가가 가장 컸으며 S 균주의 산도 증가가 가장 적었다. 아미노태 질소는 S 균주가 8일째 0.442%로 높은 값을 보였지만 다른 균주들은 모두 0.150% 이하를 나타내었다. 발효일자별(p=0.4558) 및 균주별(p=0.3513) 모두 큰 차이를 보이지 않았다. 당도는 빵효모균주인 C가 발효 4일 째부터 다른 균주들에 비해 높았고 발효일자별(p<0.0001) 및 균주별(p=0.007) 모두 유의미한 차이를 보였다. 알코올 함량은 모든 효모균주에서 발효 2일째 10%의 급격한 증가를 보였으며 그 이후에 큰 차이가 없었다. 발효일자별(p<0.0001)로는 차이를 보였지만 균주별로는 차이는 없는 것으로 나타났다(p=0.1464). DGGE 밴드로 검출된 우점세균은 산성물질을 생산하며 기능성을 보이는 Lactobacillus fermentum와 Pediococcus pentosaceus였고, 우점진균은 Saccharomyces cerevisiae였는데 이는 효모균주 첨가에 의한 결과로 판단된다.

시판 막걸리로 제조한 전통발효유 타락의 발효 특성 (Fermentation and Microbial Characteristics of Korean Traditional Fermented Milk, Tarak)

  • 한국 전통 발효유인 타락을 제조하기 위하여 4종의 시판 막걸리와 시판 우유를 사용한 '타락'을 제조하여 발효 특성을 분석하였으며 관여 미생물을 분석하였다. 제조된 타락에서의 pH는 발효시간에 따라 유의적으로(P<0.001) 감소되어 pH 5.56~6.49를 나타내고 산도는 유의적으로(P<0.001) 증가하여 0.17~0.40%의 값을 나타내었다. 점도는 유청이 분리되기 전까지 유의적으로(P<0.001) 증가하였다. 당도는 유의적으로(P<0.001) 감소하였다. 에탄올 함량은 시간에 따라 증가하여 발효 24시간에 0.51~0.71 mg/mL를 나타내었다. 발효산물로는 주된 유기산이 lactic acid로 발효시간에 따라 점차 증가하여 발효 24시간에서는 모든 시료에서 전체 유기산 생성량의 80% 이상을 차지하였으며 이외에 미량의 acetic acid, citric acid, succinic acid도 검출되었다. 발효와 더불어 검출된 주된 유리당은 lactose였고 소량의 glucose가 나타났다. 타락의 젖산균 수는 발효시간에 따라 증가하여 발효 24시간에는 9.87~10.41 log CFU/mL로 나타났으며 효모 수는 6.99~7.73 log CFU/mL였다. 타락에서의 분리된 균주는 효모인 Saccharomyces cerevisiae와 다양한 Pediococcus acidilactici, Lactobacillus fermentum, Leuconostoc mesenteroides 등의 다양한 젖산균이 나타났다. 따라서 타락은 효모와 젖산균이 공존하는 효모-젖산균 발효유임을 알 수 있었다.

유산균을 이용한 발효인삼 제조 및 품질 특성 (Preparation and Quality Characteristics of the Fermentation product of Ginseng by Lactic Acid Bacteria (FGL))

  • 인삼과 유산균은 각각 면역 활성, 생리기능조절 등 생체 효용성을 인정받고 있는 가능성 식품소재이다. 따라서 유산균을 이용한 인삼 발효물은 두 소재의 장점을 모두 이용할 수 있는 가능성이 있으므로 본 연구는 여러 가지 유산균을 이용하여 인삼 발효물을 제조하고 생균수, pH 및 산도 등을 조사하여 인삼 발효물 제조에 적합한 최적의 유산균주를 선정하고 인삼의 발효공정을 확립하였다. 27종의 유산균 중 인삼농도가 증가할수록 생육이 촉진되는 유산균은 Lac.plantarum MG208, Lac.casei Mg311, Lac.rhamnosus, MG315, Lac.acidophilus MG501, Lac.acidophilus MG501C, Lac.reuteri MG505, Lac.fermentum MG590, Bif.longum MG723 등 8종이었다. 특히, Lac.plantarum MG208이 5% 홍삼배지에 배양했을 때 가장 생육이 촉진되는 균주였다. 발효 전 배지 pH를 6.8로 동일하게 보정하였을 때 최종 인삼발효물의 pH는 유의하게 낮아졌고($pH3.45{\sim}4.83$), 특히, 홍삼농도가 높을수록 pH는 더 낮았다($pH3.42{\sim}4.30$). 이와 유사하게 백삼 및 홍삼 발효제조물의 적정산도는 약 10배정도 증가했고, 적정산도 증가율이 가장 높은 균종은 Lac.plantarum MG208, Lac.acidophilus MG501, 및 Bif.longum MG723이었으며, 발효 후 적정산도는 백삼보다 홍삼을 첨가한 경우 좀 더 높았다. 이와같이 여러 가지 유산균주를 이용하여 인삼 발효물을 제조한 결과 생균수와 유산균 발효제품에 적용할 수 있는 pH 및 적정산도 유도면에서 발효인삼 제조에 가장 적합한 유산균주는 Lac.plantarum MG208이며 인삼첨가농도는 1-5%, 배양시간은 48시간이 적합한 것으로 생각된다. 인삼의 유산균 발효물의 생리활성 효능은 앞으로 추가연구를 통하여 검증되어야 하며 그 유용성에 대한 고찰 및 검토가 필요하다.

PCR-DGGE를 이용한 구기자-맥문동 막걸리의 발효 과정과 저장 기간 중 효모와 세균 균총의 변화 (Changes in Yeast and Bacterial Flora during Fermentation and Storage of Gugija-Liriope tuber Makgeolli using PCR-DGGE)

  • 구기자-맥문동 막걸리의 미생물 분포와 최적 저장 기간을 확립하기 위하여 먼저 구기자-맥문동 막걸리를 제조한 후 $4^{\circ}C$$20^{\circ}C$에 저장하면서 효모, 총 세균 그리고 젖산균의 생균 수 변화를 조사하였다. 발효가 완료되어 $4^{\circ}C$에서 15일간 저장하는 동안 효모 수는 큰 변화가 없었다. 하지만 30일간 저장한 후에는 약 62%가 감소되었다. $20^{\circ}C$에서 수행한 저장 실험에서는 15일간 저장한 후 초기 효모균수의 87%가 감소하였으며 30일간 저장한 후에는 약 95.2%가 감소되었다. 구기자-맥문동 막걸리의 DGGE 분석에 의해 S. cerevisiae와 미동정의 Saccharomyces sp.가 동정되었는데 $20^{\circ}C$에서 저장하였을 때Saccharomyces sp.가 더 빨리 사멸하였다. 저장 기간 중 총 세균 수와 젖산균의 변화 양상은 유사하였다. $4^{\circ}C$에서 저장 하였을 때 저장 15일 시점에서 총 세균 수는 약 64%가 감소하였으나 그 이후 저장 30일까지 총 세균수의 변화는 크게 없었다. $20^{\circ}C$에서 저장하였을 경우에는 초기 세균 수가 저장 15일 시점에서 총 세균수의 약 95%가 감소하였으며 그 이후 저장 30일 시점까지 총 세균수는 크게 변화하지 않았다. DGGE분석을 통해 발효 개시 시점에서는 W. cibaria가 우점균이었다가 점차 L. fermentum과 P. acidilactici가 증가하여 저장 기간 동안 이들이 우점균으로 존재 함을 확인하였다.

Effects of Feeding Solid-state Fermented Rapeseed Meal on Performance, Nutrient Digestibility, Intestinal Ecology and Intestinal Morphology of Broiler Chickens

  • This trial was conducted to determine the effects of feeding a diet containing solid-state fermented rapeseed meal on performance, nutrient digestibility, intestinal ecology and intestinal morphology of broiler chickens. A mixed liquid culture, containing approximately 5 log cfu/ml Lactobacillus fermentum, Enterococcus faecium, Saccharomyces cerevisae and Bacillus subtilis was prepared in a 1:1:1:1 ratio. A basal substrate (BS) containing 75% rapeseed, 24% wheat bran and 1% brown sugar was mixed with the liquid culture in a ratio of 10:3. Over the 30-day fermentation, isothiocyanates were reduced from 119.6 to 14.7 mmol/kg. A total of 168, day-old male Arbor Acres broiler chicks were assigned to one of three dietary treatments including a corn-soybean meal based control diet as well as two experimental diets in which the control diet was supplemented with 10% of the BS containing unfermented rapeseed meal or 10% of the BS containing rapeseed meal subjected to solid state fermentation. There were 8 pens per treatment and 7 birds per pen. From days 19-21 and days 40-42, uncontaminated excreta were collected from each pen for digestibility determinations. In addition, digesta from the colon and ceca were collected to determine the number of lactobacilli, enterobacteria and total aerobes. The middle sections of the duodenum, jejunum, and ileum were collected for intestinal morphology. Over the entire experimental period (d 1-42), the weight gain and feed conversion of birds fed fermented rapeseed meal were superior (p<0.05) to that of birds fed nonfermented rapeseed meal and did not differ from the soybean control. On day 42, birds fed fermented rapeseed meal had higher (p<0.05) total tract apparent digestibility coefficients for dry matter, energy, and calcium than birds fed non-fermented rapeseed meal. Colon and ceca digesta from broilers fed the fermented feed had higher (p<0.05) lactobacilli counts than birds fed the control and non-fermented rapeseed meal diets on day 21 and 42. Fermentation also improved (p<0.05) villus height and the villus height:crypt depth ratio in the ileum and jejunum on day 21 and 42. The results indicate that solid-state fermentation of rapeseed meal enhanced performance and improved the intestinal morphology of broilers and may allow greater quantities of rapeseed meal to be fed to broilers potentially reducing the cost of broiler production.

Psychobiotic Effects of Multi-Strain Probiotics Originated from Thai Fermented Foods in a Rat Model

  • This work aimed to investigate the psychobiotic effects of six bacterial strains on the mind and behavior of male Wistar rats. The probiotic (PRO) group (n=7) were rats pre-treated with antibiotics for 7 days followed by 14-day probiotic administration, antibiotics (ANT) group (n=7) were rats treated with antibiotics for 21 days without probiotics. The control (CON) group (n=7) were rats that received sham treatment for 21 days. The six bacterial strains with probiotic properties were mostly isolated from Thai fermented foods; Pedicoccus pentosaceus WS11, Lactobacillus plantarum SK321, L. fermentum SK324, L. brevis TRBC 3003, Bifidobacterium adolescentis TBRC 7154 and Lactococcus lactis subsp. lactis TBRC 375. The probiotics were freeze-dried into powder (6×109 CFU/5 g) and administered to the PRO group via oral gavage. Behavioral tests were performed. The PRO group displayed significantly reduced anxiety level and increased locomotor function using a marble burying test and open field test, respectively and significantly improved short-term memory performance using a novel object recognition test. Antibiotics significantly reduced microbial counts in rat feces in the ANT group by 100 fold compared to the PRO group. Probiotics significantly enhanced antioxidant enzymatic and non-enzymatic defenses in rat brains as assessed using catalase activity and ferric reducing antioxidant power assay, respectively. Probiotics also showed neuroprotective effects with less pyknotic cells and lower frequency of vacuolization in cerebral cortex. This multi-strain probiotic formulation from Thai fermented foods may offer a potential to develop psychobiotic-rich functional foods to modulate human mind and behaviors.

신생아 분변 및 동치미에서 분리한 젖산균 대사산물의 항균특성 (Antimicrobial Characteristics of Metabolites of Lactic Acid Bacteria Isolated from Feces of Newborn Baby and from Dongchimi)

  • 식중독 미생물의 증식을 억제하며, probiotic으로 작용할 수 있는 균주로서 가능성이 있는 젖산균을 신생아 분변과 동치미서 젖산균을 분리(0.15%의 담즙산염과 pH 3.0인 산성조건)하였고, 항균활성이 우수한 젖산균을 선발하였다. D2와 F35-2 균주는 Lactobacillus plantarum, F20-3 균주는 L. fermentum으로 동정되었다. 젖산균 대사산물의 특성과 항균 기작을 알아보기 위해 몇 종의 식중독 미생물(Listeria monocytogenes, Bacillus cereus, Staphylococcus aureus, Salmonlla Typhimurium, Escherichia, coli O157:H7, Salmonella Enteritdis)을 사용하였으며, 항균활성 Bioscreen C로 측정하였다. 젖산균의 대사산물의 특성을 확인한 결과 pH가 중성으로 갈수록 항균효과가 감소하는 경향을 나타내었다. Catalase 효소처리 후에는 F20-3 균주만 항균활성이 소실되어 항균활성의 원인으로 $H_2O_2$의 영향이 있을 것으로 추정되었다. 단백질 분해 효소(trypsin, pepsin)처리에는 항균활성에 영향이 없었고, $121^{\circ}C$ 15분간 열처리에도 안정한 물질로 추정되었다. 분리균주 대사산물은 유사한 pH와 젖산 농도보다 식중독 미생물의 증식 억제가 우수하여 분리된 균주의 증식억제 물질은 유기산 및 대사산물과 관계가 있을 것으로 추정되었다. 분리균주가 생성하는 유기산을 HPLC로 정량 한 결과, D2와 F35-2는 발효 24시간에 각각 1.84, 1.85%의 젖산만이 생성되었고, F20-3은 발효 24시간에 0.91%의 젖산과 0.22%의 초산을 생성하였다. 이것으로 볼 때 젖산발효형태는 D2와 F35-2는 homo형이고, F20-3은 hetero형의 발효를 하는 것으로 확인되었다.
