Studies on the Interspecific and Intergeneric Hybridization in Herbage Grasses I. Effects of hybrid embryo age on callus formation and plant regeneration (화본과목초의 종.속간 잡종에 관한 연구 I. 교잡* 일령에 따른 Callus형성과 식물체재분화)

  • A breeding program in progress at Suweon Livestock Experiment Station, RDA, involves the hybridization of italian ryegrass (Lm) and several leading forage grasses in an effort to combine the nutritive, productivity and palatability qualities of Lm with the adaptive and cold resistance qual; .les of several leading grasses. In order to study the fate of the hybridization between remotely related species, immatured hybrid embryos were cultured on media. The emasculated plants of Lm were Sikem and Tetrone. Reweille, 2n=14, of perennial ryegrass (Lp.), Forager, 2n=42, of tall fescue(Fa), First, 2n=14,of meadow fescue(Fp), Potomac, 2n=28, of orchardgrass(Dg), and Richmond, 2n=42, of timothy(Ph.p) were used as pollinators. Embryos were isolated on 4, 8, 12, 16, and 20 days after pollination and cultured them on modified Murashige and Skoog media. Calluses and plantlets have been obtained after 8 days old embryos crossing between $Lm{\times}Fa$, after 12 days embryos crossing between $Lm{\times}Lp$, Dg, and Ph.p, and after 16 days embryos crossing between $Lm{\times}Fp$. Both callus and shoot formation occurred on 6 % or less of the plated embryos of $Lm{\times}Fp$, Dg, and Ph.p. Embryoderived callus forming shoots have been obtained from 4 days old embryo crossing between $Lm{\times}Dg$ which has not successfully been done anywhere as far as we know. It means that hybrid plants of species without crossability can be obtained through the use of immatured embryo culture. Some of plated embryos developed directly shoots from embryos and the others shoot-forming callus. Cross between related species showed a high frequency of directly shoot formation from plated embryos and cross between remotely related species a high frequency of callus formation.

Serum Carbohydrate Antigen 19-9 as an Indicator of Liver Metastasis in Colorectal Carcinoma Cases

  • Purpose: The liver is the organ to which colorectal carcinomas (CRCs) most commonly metastasize, and surgical resection has been established as the most effective and potentially curative treatment for CRC with liver metastasis (LM). Therefore, surveillance of LM is vital for improvement of prognosis of CRC patients. In this study, we aimed to explore the potential value of carbohydrate antigen 19-9 (CA 19-9), carcinoembryonic antigen (CEA), and marker enzymes in indicating LM with CRC. Methods: Three groups of eligible patients with metastatic cancers were retrospectively included: CRC patients with LM (CRC-LM) or without LM (CRC-NLM), and non-CRC patients with LM (NCRC-LM). All metastatic lesions were identified by CT or MRI. Data on characteristics of the patients, the primary site, the locations of metastasis, CA 19-9, CEA, and biochemical parameters were collected for analysis. Results: A total of 493 patients were retrospectively included. More alcohol consumption was found in CRC-LM than CRC-NLM. Some biochemical enzymes were found to be significantly higher in groups with LM than without (CRC-LM or NCRC-LM v.s CRC-NLM). Both CEA and CA 19-9 were much higher in CRC-LM than CRC-NLM or NCRC-LM. For CRC patients, CA 19-9, ${\gamma}$-glutamyl transpeptidase, CEA and alcohol consumption were identified as independent factors associated with LM. Conclusion: Our analysis suggested the CA 19-9 might be a potential valuable indicator for LM of CRC in the clinic.

Multiplex PCR method for environmental monitoring of approved LM cotton events in Korea (국내 승인 LM면화의 자연환경 모니터링을 위한 multiplex PCR 개발)

  • The growth area of living modified (LM) cotton has steadily increased every year, since its first commercialization in 1996. Development of environmental risk assessment tools and techniques for LM cotton is required for ecosystem safety. We therefore developed multiplex PCR assays for simultaneous detection of two (MON15985, MON531) and four (GHB614, LLCOTTON25, MON88913 and MON1445) LM cotton events approved in Korea, with event specific primer pairs. The PCR reactions were optimized by using event specific primers of six LM cottons at various concentrations. The reactions allows amplification of estimated amplicons of MON15985 (214 bp), MON531 (270 bp), GHB614 (119 bp), LLCOTTON25 (164 bp), MON88913 (276 bp), and MON1445 (389 bp) from multiplex PCR reactions. The multiplex PCR assay developed allowed that two annealing steps (15 cycles at $55^{\circ}C$ and 25 cycles at $60^{\circ}C$) were performed for amplification of distinguished two LM cottons, and only one annealing step (50 cycles at $60^{\circ}C$) was necessary for tetraplex PCR. Primer extension step of all PCR reactions was skipped for time-effective amplification. Our methods suggest that two multiplex PCR assays can be cost-effective and a rapid diagnostic tool for environmental LMO monitoring of six LM cottons.

A Data Fitting Technique for Rational Function Models Using the LM Optimization Algorithm (LM 최적화 알고리즘을 이용한 유리함수 모델의 데이터 피팅)

  • This paper considers a data fitting problem for rational function models using the LM (Levenberg-Marquardt) optimization method. Rational function models have various merits on representing a wide range of shapes and modeling complicated structures by polynomials of low degrees in both the numerator and denominator. However, rational functions are nonlinear in the parameter vector, thereby requiring nonlinear optimization methods to solve the fitting problem. In this paper, we propose a data fitting method for rational function models based on the LM algorithm which is renowned as an effective nonlinear optimization technique. Simulations show that the fitting results are robust against the measurement noises and uncertainties. The effectiveness of the proposed method is further demonstrated by the real application to a 3D depth camera calibration problem.

  • To position precision machines accurately, linear motion (LM) guides having rolling elements can be used. For ultra-accurate positioning control of the precision machines, the understanding of the dynamic behavior of the LM guide at the macro and/or micro scales is most critical, but the research on this subject is rare. The objective of the present research is to observe the vibration phenomena of the LM guide. Bails are used as the rolling elements in this work. Several experiments show the nonlinear characteristics of the LM guide such as hysteresis behavior and force-dependent natural frequencies phenomena.

  • This paper consider the testing problem of variance component for the unbalanced two-way error component model with nuisance parameter. We derive the one-sided LM test statistic for testing zero individual(time) effects assuming that the other time-specific(individual) effects are present. Using the Monte Carlo experiments, the computational more demanding LR test slightly underestimates the nominal size and has the low powers relative to LM test statistic.
