폴리페놀 및 항산화 관점에서의 쌍화탕 및 단미혼합쌍화탕의 동등성 (Equivalence of Traditional and Individual Preparation of Ssanghwa-tang in terms of Polyphenol Contents and Radical Scavenging Activity)

  • Traditional medicines usually use combinational formula that is prepared by mixing different varieties of medicinal herbs and boiling them in water to yield a decoction. In recent years a modified method has been proposed and practiced wherein the individual herbs are boiled with water separately and later these extracts are mixed together for use. This practice has lead to a constant controversy concerning the chemical and biological equivalence between the preparations arriving out of these two methodologies. In this research, we attempted to study the effects of traditional and individual preparation of Ssanghwa-tang (ST) to evaluate their equivalence in terms of its antioxidant activity and polyphenol content. The study results indicate that the polyphenol contents as well as the extraction yields of these two preparations were very similar to each other.

민간 생약재의 아플라톡신 $B_1$에 대한 항돌연변이 효과 (Antimutagenic Effects of Traditional Herbal Drugs on the Aflatoxin $B_1$)

  • The antimutagenic effects of 46 kinds of medicinal plants that have been used as traditional folk antitumor agents in Korea were studied by using Ames mutagenicity test. Most of the methanolic extracts from the plants which were used in this experiment showed strong antimutagenic activity toward aflatoxin B1(AFB1) in Salmonella typhimrium TA100 and TA98. However, N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) induced mutagenicity was not blocked by adding the methanolic extracts of the plants except persimmon leaves (Diospyros kaki Thunberg)and Elaeagnus umbellata.

  • Herbal plants or traditional Oriental medicine have been considered as a potential resource of new drug development worldwide. However, traditional Korean medicine has given little effort to the field of new drug development. This study reports on a plant-derived hepatotherapeutic drug, silymarin, which has been popularly used in many countries. It was discovered as an active compound from Silybum marianum (milk thistle) which has been known as a medicinal plant having hepatoprotective properties in both European and Asian countries. While it has been used as an herbal prescription in Asia, its active compounds or scientific mechanisms were intensively studied in Europe. Currently, silymarin is one of the most powerful herbal extracts in the world, and its usage is being expanded to many other medical purposes. This report would be helpful for providing an informative example of herbal-derived drug development to Oriental doctors or scientists in the Oriental medicinal field.

Inhibitory Effects of Herbal Medicines on Hyaluronidase Activity

  • Inhibitory effects of 130 medicinal plants on hyaluronidase activity were analyzed. The medicinal plants are clinically used as herbal medicines for korean traditional prescriptions. Six out of the 130 herbal medicines exhibited more than 50% of inhibition on hyaluronidase activity by their total methanol extracts with 5mg/ml as a final concentration. The active total methanol extracts were prepared from cortex of Acantbopanax gracilistylus, lignum of Caesalpinia sappan, radix of Glycyrrhiza uralensis, radicis cortex of Morus alba, herba of Prunella vulgaris, and radix of Sanguisorba officinalis. These active total methanol extracts were sequentially fractionated with dichloromethane, ethyl acetate, n-butanol, and then water. Among the solvent-fractionated extracts, the butanol fractions of Acantbopanax gracilistylus and Glycyrrhiza uralensis with 1 mg/ml as the final concentration exhibited more than 50% of inhibition on hyaluronidase activity, and the other fractions with the same concentration did less than 20% of inhibition.

한약재 추출물이 지방세포주 3T3-L1 분화에 미치는 영향 (The Effect of seven Korean Traditional Herbal Medicines on 3T3-L1 Adipocyte Differentiation)

  • Objectives : The aim of the present study is to evaluate the effects Rehmannia glutinosa, Radix puerariae, Semen coicis, Fructus hordei, Cortex acanthopanacis, Fructus chaenomelis, or Radix glycyrrhizae extracton 3T3-L1 preadipocytes differentiation. Methods : We adopted Oil Red O staining methods to observe the formed lipid droplets. And the amount of lipids in adipocytes was measured using absorptiometric analysis. Results : The extracts of Rehmannia glutinosa, Radix puerariae, Semen coicis, Fructus hordei, Cortex acanthopanacis, Fructus chaenomelis, or Radix glycyrrhizae stimulated the preadipocytes differentiation and lipid droplet formation. And the complex extract of the traditional herbal medicines stimulated there actions more than single extracts. Conclusions : The extract of Rehmannia glutinosa, Radix puerariae, Semen coicis, Fructus hordei, Cortex acanthopanacis, Fructus chaenomelis, or Radix glycyrrhizae has stimulatory effects. on adipogenesis. Moreover, the complex extract of the traditional herbal medicines has more effect than single extracts.

한약, 한약재, 생약과 천연물의 법규상 개념 및 정의의 문제점과 개선안 (A study on the Problems and Improvement Proposals on Legal Definitions in Regards to Herbs, Herbal Drugs, Crude Drugs and Natural Products)

  • Objectives : This study was to analyze the definitions of herbs, herbal drugs, crude drugs and natural products in the relevant laws and regulations, understand the related problems, and propose directions for improvement. Methods : I analyzed the legal definitions in respect of herbs, herbal drugs, crude drugs and natural products in relevant laws and regulations since 1945, explained the problems, and suggested the solution-considering the academic stance of Traditional Korean Medicine and the dualistic medical and pharmaceutical system. Results : Herbs are defined as "refined things that are cut and dried in their most original state". The definition of crude drugs includes herbs and the "cell contents, secretion, extracts, minerals and other parts of animals and plants that are used medicinally". The concept of natural products is expanded to adding tissue cultures to the definition of crude drugs. Conclusions : The definition of herbs should at least include all products that are "processed, extracted and prepared" as well as contents that consist of various forms of hospital-prepared herbs. The term "herbal drug" corresponds to a traditional term of "drug", and this should be established as a concept to explain "drugs in raw materials that are used to prepare herbs and/or manufacture herbal medicine". The legal definition of herbs should include the concept of crude drugs. Herbal drug preparations and crude drugs should be included in the definition of herbal drugs.

항톡소포자충 효과를 가진 천연물 유래 추출물에 관한 연구 (Anti-Toxoplasmosis effects of herbal extracts against Toxoplasma Gondii)

  • Toxoplasmosis is a parasitic disease caused by Toxoplasma gondii, with very few therapeutic treatment options. The choices for treatment are pyrimethamine and sulfadiazine, however their utility is limited because of drug toxicity and serious side effects. In this study, ethanol extracts of 13 traditional medicines used to treat Toxoplasma gondii were tested in vitro for their anti-Toxoplasma gondii cytotoxicity. The median effective concentration ($EC_{50}$) values for the herbal extracts ranged from 173 mg/mL to 1995.35 mg/mL. In HeLa cell, the selectivity of Alpinia oxyphylla (2.75), Mucunae Caulis (2.96), Dictamnus dasycarpus (7.52) which was higher than sulfadiazine (2.08). This indicates that Alpinia oxyphylla, Mucunae Caulis, Dictamnus dasycarpus extracts may be sources of new anti-Toxoplasma gondii compounds.

한약재 추출물에 대한 전혈응집억제 효능 검색 (Effect of Extracts for Herbal Medicines on the Inhibition of Whole Blood Aggregation)

  • 고미의 성질을 강하게 나타내는 한약재를 대상으로 맛과 효능과의 상관관계를 실험적으로 알아보기 위한 연구를 시도하였다. 본 연구에서는 콜라겐으로 유도된 인간전혈응집을 이용하여 고미약재 20품목에 대한 혈소판응집 억제능을 측정하였다. 이 중에 3종의 열수추출물과 2종의 70% 에탄올추출물이 p<0.001 수준에서 유의하게 전혈응집 억제 활성을 나타내었다. 특히, 전혈응집 억제능이 가장 뛰어난 후보소재로 황련이 선별되었으며 관능요원에 의한 관능평가에서 쓴맛이 가장 강한 한약재로 평가되었다. 이상의 실험결과는 전통의학이론을 바탕으로 한약재의 사용에 따른 효능을 실험적 근거로 제시할 수 있고 쓴맛이 약한 한약재보다 쓴맛이 강한 한약재가 혈액순환이 원활하게 하는 가능성의 경향이 있는 것으로 기대되었다.

한방 추출물의 역류성 식도염 점막보호 효과에 대한 스크리닝 (Screening for Mucosal Protective Effects of Various Korean Herbal Medicine Extracts in Gastroesophageal Reflux Disease)

  • Objectives : This study evaluates how various traditional Korean herbal medicines assess MUC5AC expression for esophageal mucosal defense and analyzes the associated mechanisms involved in inflammation. Methods : Forty types of traditional Korean herbal medicines were assessed for in vitro antioxidant activities, and the real-time PCR method was employed to analyze MUC5AC expression under pH 4.5 conditions in human esophageal epithelial cells (HET-1A). Eight types of Korean herbal medicines were evaluated for in vitro antioxidant activities, and Reactive oxygen specise (ROS) expression was analyzed under bile salt (480 𝜇M) and pH 5.5 conditions in human esophageal epithelial cells (HET-1A). Simulation experiments involving bile salts and acidity were conducted for one hour to assess the efficacy of four drugs, and the activities of Mitogen-activated Protein Kinase (MEK), Nuclear Factor Kappa B (NF-𝜅B), and Cyclooxygenase-2 (COX-2) were detected through Western blot analysis. Results : Compared to the Normal group, the Control group exhibited higher ROS generation. Such increased ROS levels were significantly reduced by four extracts: Citrus Unshius Pericarpium (CUP), Cnidium officinale Rhizoma (CR), Ginseng Radix (GR), and Linderae Radix (LR). The protein expression of COX-2 decreased with the treatment of LR, CUP, and CR. Particularly, CUP and CR exhibited superior effects compared to other groups in inhibiting the phosphorylation of NF-𝜅B. Conclusion : Based on the results obtained, we have identified drugs that inhibit oxidative stress and inflammation caused by bile acid in esophageal epithelial cells. Our future plans involve comparing and analyzing the efficacy of these herbal medicines through animal experiments.

배양된 흰쥐 뇌간 신경세포에서 5-Hydroxytryptamine 흡수에 대한 각종 전통 생약 추출물의 억제 효과 (Inhibitory Effects of Extracts from Traditional Herbal Drugs on 5-Hydroxytryptamine Uptake in Primary Cultured Rat Brainstem Neurons)

  • Crude methanolic(80%) extracts from 109 kinds of traditional herbal drugs were randomly screened for inhibitory effects on 5-hydroxytryptamine(5-HT) uptake in primary cultured rat brainstem neurons. Rat brainstem neurons were cultured from embryonic day 14, and maintained for 7-9 days in vitro. Clomipramine (500 nM), a reference drug, decreased 5-HT uptake to 16% of control values. Of the 109 herbal drugs screened, Citri immaturi Pericarpium(靑皮), Coptidis Rhizoma(黃蓮), Cnidii Rhizoma(土川芎) showed the most potent 5-HT uptake inhibiting activities. These herbal drugs, at the concentration of $10{\;}{\mu}g/ml$, inhibited 5-HT uptake 69, 69, and 57% respectively, when inhibition(%) was expressed as a relative value compared to the 500 nM clomipramine-induced inhibition.
