A Study on the Consumption Patterns, the Importance of Product Properties, and the Satisfaction Levels of Blackberry and Mulberry Fruit Beverage Products in the Jeonbuk Area (전북지역 복분자와 오디 음료 제품의 구매실태 및 제품 속성의 중요도와 만족도 비교)

  • The principal objective of this study was to evaluate the consumption patterns, the importance of product properties, and the satisfaction levels of blackberry and mulberry fruit beverage products(BMBPs) in the Jeonbuk area, Korea. Self-administered questionnaires were collected from 294 customers(male 40.8%, female 59.2%) residing at Jeonju in Jeonbuk area. Descriptive and comparative statistical analyses were completed using SPSS PC program(version 19.0). Approximately, 61.6% of participants consumed BMBPs 1-2times a year. Only 1.7% of participants consumed BMBPs everyday. The main information source about BMBPs was 'families/friends'. About 58.8% of participants responded that the main reason for purchasing BMBPs was 'for health'. The result indicated the overall importance level(3.94) was higher than the satisfaction level(3.41)(p < .001). The importance and satisfaction levels of the older group (50's) were significantly higher than the levels of younger group(20-30's)(p < .01 ; p < .05). According to the IPA(Importance-Performance Anslysis), 'price' and 'credibility' of companies were the most important properties that had to be changed as soon as possible to increase satisfaction with BMBPs. Therefore, to increase BMBPs'-consumption, quality improvements on these products through cooperation between BMBPscompanies, the regional government, and research institutions are necessary.

Effect of Fermented Blackberry Drinks Formed from Radiation-induced Mutant on Liver Repair Capacity in Rats (방사선 유도 돌연변이체 블랙베리로 제조한 발효음료의 간 손상 회복 효과)

  • This study was conducted to investigate the effect of fermented blackberry drinks (BD) on carbon tetrachloride ($CCl_4$)-induced liver injury in rats. Male Sprague-Dawley rats were randomly divided into four groups with 6 rats per group: control, $CCl_4$, $CCl_4$ plus BD $3ml\;kg^{-1}$, and $CCl_4$ plus BD $6ml\;kg^{-1}$. We found that the levels of serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were significantly increased and the activity of antioxidant enzyme glutathione peroxidase (GPx) in the liver was decreased in rats treated with $CCl_4$ alone when compared with the control group. However, the administration of BD attenuated the levels of serum AST and ALT in $CCl_4$-treated rats. Moreover, the administration of BD significantly increased the activity of GPx in $CCl_4$-treated rat livers. Taken together, these results suggest that BD could protect the liver from $CCl_4$-induced hepatic damage.

Protective efficacies of Aronia melanocarpa (Blackberry) on the Allyl Alcohol-damaged Hepatocyte of Mice (쥐의 알코올성 간세포 손상에 대한 Aronia melanocapa(Blackberry)의 보호효과)

  • The results of the alcohol decomposition ability of Aronia melanocarpa are as follows. Plasma alcohol concentration of the Aronia group was ca. 48.9% lower than the control group, but half an hour faster in that of Aronia before alcohol group, ca. 54.9% higher and half an hour later than that of the control group. ALDH(acetaldehyde dehydrogenase) of the Aronia group was ca. 243% higher than that of the control group. But maximal ALDH of the group taking Aronia before alcohol administration showed 0.5h faster and ca. 267% higher than that of the control group. This result shows that the activity of ALDH was increased by the Aronia. Aronia group's AST and ALT are increasing with similar patterns and their levels continually under the control's, but ca. 12.6% lower at AST and ca. 19.0% lower at ALT than those of control group. Ca. 21.7% lower at AST and ca. 40.5% lower at ALT in the Aronia group before alcohol administration than those of the control group. This result shows that Aronia has a role of suppressor against the liver damage. Therefore, this study proved lucidly the protective effects of Aronia on the hepatocyte.

Protective Effects of Radiation-induced Blackberry Mutant Extract on Carbon Tetrachloride (CCl4)-induced Liver Injury in Sprague-Dawley Rats (방사선 돌연변이 블랙베리 추출물의 사염화탄소(Carbon Tetrachloride, CCl4) 투여에 의한 흰쥐의 간 손상에 대한 보호 효과)

  • The objective of the present study was to investigate the protective effects of anthocyanin-enriched extract from radiation-induced blackberry (Rubus fruticosus L.) mutant (${\gamma}$-B201) against carbon tetrachloride ($CCl_4$)-induced liver injury in Spargue-Dawley (SD) rats. The in vivo results show that ${\gamma}$-B201 attenuated the levels of serum aspartate aminotransferase, alanine aminotransferase, and liver lipid peroxidation in $CCl_4$-treated SD rats. Histopathological examination of rat livers showed that ${\gamma}$-B201 reduced the incidence of liver lesions induced by $CCl_4$. Moreover, ${\gamma}$-B201 prevented DNA damage in $CCl_4$-treated SD rats. Furthermore, administration of ${\gamma}$-B201 significantly increased the activity of antioxidant enzymes, such as superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), and glutathione reductase (GR), in $CCl_4$-treated rat livers. In conclusion, the present study suggests that ${\gamma}$-B201 blackberry extract protects the liver from $CCl_4$-induced hepatic damage through an antioxidant mechanism. Therefore, ${\gamma}$-B201 blackberry may be functional food material for human health.

Rust of Belamcanda chinensis Caused by Puccinia belamcandae (Puccinia belamcandae에 의한 범부체 녹병)

  • Rust disease was occurred on blackberry lily (Belamcanda chinensis) in several farmer's fields located in Heohwa-myon, Goseong-gun, Gyeongnam province in Korea. The typical symptoms of the disease appeared as small, yellowish spot on leaves at first. The spots then turned brown, their edges rised slightly, and powdery mass of yellow or yellowish green spores appeared on the lesions. Severely infected leaves were blighted and eventually died. Urediniospores were yellow or yellowish brown in color, globoid to ovoid in shape and $21{\sim}46{\times}18{\sim}38\;{\mu}m$ in size. Teliospores were brown in color, oblong or clavate in shape and $32{\sim}64{\times}12{\sim}26\;{\mu}m$ in size. The causal fungus was identified as Puccinia belamcandae based on morphological characteristics and host specificity. This is the first report on the rust of B. chinensis caused by P. belamcandae in Korea.

Enhancement of Antioxidative Activities of Berry or Vegetable Juices through Fermentation by Lactic Acid Bacteria (젖산균 발효를 통한 베리류 또는 과채류 주스의 항산화 활성 증진)

  • Berry or vegetable juices contain a diverse range of antioxidants. Through oxygen radical absorbance capacity assays, acai berry, aronia, wild grape, blackberry, cranberry, and spinach juices were verified to possess high antioxidant activities. Lactic acid bacteria fermentation was applied to each juice as the sole medium to improve antioxidant activity. After fermentation by Lactobacillus plantarum, the antioxidant activities of acai berry, blackberry, and spinach juices increased by 20–30% from 943.2 to 1239.2, from 110.87 to 128.04, and from 77.92 to 107.20 µmol TE/g, respectively. In this study, we found that the antioxidant activities of a number of juices were enhanced through lactic acid bacteria fermentation.

Blueberry, blackberry, and blackcurrant differentially affect plasma lipids and pro-inflammatory markers in diet-induced obesity mice

  • BACKGROUND/OBJECTIVES: Evidence indicates that berry anthocyanins are anti-atherogenic, antioxidant, and anti-inflammatory. However, berries differ vastly in their anthocyanin composition and thus potentially in their biological and metabolic effects. The present study compared hypolipidemic, antioxidant, and anti-inflammatory properties of blueberry (BB), blackberry (BK), and blackcurrant (BC) in a diet-induced obesity (DIO) mouse model. MATERIALS/METHODS: Male C57BL/6J mice were fed a high fat (HF; 35% fat, w/w) control diet or a HF diet supplemented with freeze-dried 5% BB, 6.3% BK or 5.7% BC for 12 weeks (10 mice/group) to achieve the same total anthocyanin content in each diet. Plasma lipids, antioxidant status and pro-inflammatory cytokines were measured. The expression of genes involved in antioxidant defense, inflammation, and lipid metabolism was determined in the liver, epididymal adipose tissue, proximal intestine, and skeletal muscle. Histological analysis was performed to identify crown-like structure (CLS) in epididymal fat pads to determine macrophage infiltration. RESULTS: No differences were noted between the control and any berry-fed groups in plasma levels of liver enzymes, insulin, glucose, ferric reducing antioxidant power, superoxide dismutase, and tumor necrosis factor ${\alpha}$. However, BK significantly lowered plasma triglyceride compared with the HF control and other berries, whereas BC significantly reduced F4/80 mRNA and the number of CLS in the epididymal fat pad, indicative of less macrophage infiltration. CONCLUSIONS: The present study provides evidence that BB, BK and BC with varying anthocyanin composition differentially affect plasma lipids and adipose macrophage infiltration in DIO mice, but with no differences in their antioxidant capacity and anti-inflammatory potential.

Chemical Characterization and Utilization of $19{\alpha}-Hydroxyursane-type$ Triterpenoids in Rubus species (Rubus속 식물에 존재하는 $19{\alpha}-Hydroxyursane-type$ Triterpenoid의 특성과 이용)

  • The plant Rubus species (Rosaceae) mainly contains $19{\alpha}-hydroxyurane-type$ triterpenoids $(19{\alpha}-HUT)$ as bioactive components. Available functional food includes blackberry (the fruit of thornless Rubus sp.), red raspberry (R. idaeus) and black raspberry (R. occidentalis). However, the fruit of R. coreanus, which is used in Korea as a functional food, substitutes black raspberry. Rubi Fructus, which has been traditionally used as an oriental medicinal drug, designates only unripe fruit of R. coreanus but not its ripe fruit which indicates that it needs high content of $19{\alpha}-HUT$ as a crude drug. Throughout our experiment, we found that ripe fruits contain very little amount of $19{\alpha}-HUT$ when compared to unripe fruits. In addition, various and rich $19{\alpha}-HUT$ has been reported from Rubus species. The most common structure of $19{\alpha}-HUT$ of Rubus species, euscaphic acid or tormentic acid with $3{\alpha}-OH$ or $3{\beta}-OH$, respectively, can be glycosylated or oxidized to produce a number of $19{\alpha}-HUTs $as euscaphic acid and tormentic acid derivatives and even esterified to form dimeric triterpenoids. In this review, the bioactivity and biosynthetic pathway and chemical characterization of $19{\alpha}-HUTs$ found in Rubus species are discussed.

Protective Effect of Radiation-induced New Blackberry Mutant γ-B201 on H2O2-induced Oxidative Damage in HepG2 Cells (H2O2 에 의해 유도된 HepG2 세포의 산화적 스트레스에 대한 신품종 방사선 돌연변이 블랙베리 γ-B201의 세포 보호 효과)

  • The objective of the present study was to investigate the chemical composition of anthocyanin-enriched extract of radiation-induced blackberry (Rubus fruticosus L.) mutant (${\gamma}$-B201) as well as the protective effect of ${\gamma}$-B201 against oxidative stress in vitro. The cytotoxicity, reactive oxygen species (ROS) scavenging capacity, and DNA damage were assessed by WST-1 assay, flow cytometry, and comet assay, respectively. Lactate dehydrogenase, superoxide dismutase, and catalase activities were determined by using a commercial kit. The in vitro results showed that ${\gamma}$-B201 increased the cell viability, reduction of lactate dehydrogenase release, and intracellular ROS scavenging capacity in hydrogen peroxide ($H_2O_2$)-treated HepG2 cells. Furthermore, treatment with ${\gamma}$-B201 attenuated DNA damage in $H_2O_2$-treated HepG2 cells and treatment with ${\gamma}$-B201 restored the activity of superoxide dismutase and catalase in $H_2O_2$-treated HepG2 cells. In conclusion, the present study suggests that ${\gamma}$-B201 blackberry extract can exert a significant cytoprotective effect against $H_2O_2$-induced cell damage.

A mixture of blackberry leaf and fruit extracts decreases fat deposition in HepG2 cells, modifying the gut microbiome

  • More effective treatments are needed for non-alcoholic fatty liver disease (NAFLD). We hypothesized that water extracts of blackberry fruits (BF) and leaves (BL) and their combinations (BFL) reduce fat deposition in HepG2 cells and modulate shor-tchain fatty acids (SCFA) and fecal bacteria in vitro. HepG2 cells were treated with BF, BL, BFL1:2, and BFL1:3 for 1 h, and 0.5 mM palmitate was added to the cells. Moreover, low ($30{\mu}g/mL$) and high doses ($90{\mu}g/mL$) of BL and BF were applied to fecal bacteria in vitro, and SCFA was measured by GC. BL, BF, BFL1:2, and BFL1:3 reduced triglyceride deposition in the cells in a dose-dependent manner, and BFL1:2 and BFL1:3 had a stronger effect than BF. The content of malondialdehyde, an index of oxidative stress, was also reduced in BL, BF, and BFL1:2 with increasing superoxide dismutase and glutathione peroxidase activities. The mRNA expression of acetyl CoA carboxylase, fatty acid synthase, and sterol regulatory element-binding protein-1c was reduced in BL, BF, BFL1:2, and BFL1:3 compared to the control, and BFL1:2 had the strongest effect. By contrast, the carnitine palmitolytransferase-1expression, a regulator of fatty acid oxidation, increased mostly in BFL1:2 and BFL1:3. Tumor necrosis factor-${\alpha}$ and interleukin-$1{\beta}$ expression was reduced in BL compared to that in BF and BFL1:2 in HepG2 cells. Interestingly, BL increased propionate production, and BF increased butyrate and propionate production and increased total SCFA content in fecal incubation. BF increased the contents of Bifidobacteriales and Lactobacillales and decreased those of Clostridiales, whereas BL elevated the contents of Bacteroidales and decreased those of Enterobacteriales. In conclusion, BFL1:2 and BFL1:3 may be potential therapeutic candidates for NAFLD.
