Isolation and Purification of Decursin and Decursinol Angelate in Angelica gigas Nakai (참당귀(Angelica gigas Nakai) 중 Decursin 및 Decursinol Angelate의 분리 및 정제)

  • This paper is intended as an investigation of the method of extraction and the analysis by high-performance liquid chromatography mass spectroscopy of decursin and decursinol angelate in the dried root of Angelica gigas Nakai. The extracted decursin and decursinol angelate were the purity of >95% using 60% ethanol at $-20^{\circ}C$ for 12 hours by HPLC analysis. Decursin and decursinol angelate were efficiently isolated using recycling HPLC. The purity of isolated decursin and decursinol angelate was identified as 99.97 and 99.40% by HPLC analysis, respectively. The molecular weights of Decursin and decursinol angelate were also identified as m/z=329 ($[M+H]^+$) and m/z=351 ($[M+Na]^+$) by mass spectroscopy.

HPLC Analysis and Extraction Methods of Decursin and Decursinol Angelate in Angelica gigas Roots (당귀(Angelica gigas) 중 Decursin 및 Decursinol Angelate 추출 방법과 HPLC 분석)

  • This paper is intended as an investigation of the analysis of high-performance liquid chromatography and the method of extraction of decursin and decursinol angelate in Angelica gigas roots. There are three kinds of extraction methods: distilled water, 50% EtOH and 100% EtOH. The condition of HPLC was obtained on a reversed-phase column $(Polarity\;dC_{18},\;4.6{\times}250 mm,\;5\;{\mu}m)$ using a phosphate buffer-acetonitrile-sodium lauryl sulfate as the mobile phase. Under these chromatographic conditions, UV detector was 230 nm, column temperature $30^{\circ}C$ and the speed of a current 1.0 ml/min, respectively. The results of extraction with distilled water, 50% EtOH and 100% EtOH in Angelica gigas roots were as follows. The concentrations of decursin and decursinol angelate were 182 and 153 ppm (distilled water), 3,142 and 2,547 ppm (50% EtOH) and 3,341 and 2,778 ppm (100% EtOH). There were high positive correlations between the concentrations of decursin and EtOH (r=0.8928, p<0.01) and decursinol angelate and EtOH (r=0.9009, p<0.01).

Effect of High Hydrostatic Pressure on the Shelf-life and Sensory Characteristics of Angelica keiskei Juice (초고압처리 신선초 녹즙의 저온저장 안정성 및 관능적 특성 변화)

  • Microbial and sensory changes during storage at $4^{\circ}C$ of Angelica keiskei juice pressurized at $5700\;kg_f/cm^2$ for 7 min, an optimum process condition suggested from the previous study, were investigated. Pseudomonas, which is responsible for putrefaction of refrigerated food, was totally inactivated by pressurization. E. coli and coliform bacteria were also completely inactivated. According to the sensory evaluation data, high hydrostatic pressure does not change sensory characteristics of freshness, sweetness and bitterness (P<0.05). However, after 8 day storage at $4^{\circ}C$, pressurized Angelica keiskei juice showed higher freshness than untreated control. These results indicated that the high hydrostatic pressure can be used as an effective process method for preserving Angelica keiskei juice.

  • This study investigated on the nutritional components and physiological activity of four wild vegetables namely Salvia plebeia R. Br., Angelica acutiloba, Gynura procumbens and Saururus chinensis Baill for the development of representative wild vegetables in Chungbuk. Salvia plebeia possessed the highest radical scavenging activity and beta-carotene, but exhibited the lowest α-glucosidase inhibitory activity compared to the other three vegetables. Angelica acutiloba showed high crude protein content and α-glucosidase inhibitory activity, but contained low total polyphenol content, radical scavenging ability and beta-carotene compared to the other three vegetables. Gynura procumbens showed high mineral content, beta-carotene, vitamin K1 content and α-glucosidase inhibitory activity, but showed the lowest total polyphenol content and radical scavenging ability compared to the other three vegetables. Saururus chinensis showed high crude fiber content and total polyphenol content, but contained the lowest mineral and vitamin K1 content. To conclude, it is suggested to use Salvia plebeia or Saururus chinensis as antioxidant food materials and Angelica acutiloba and Gynura procumbens as food materials and sources of α-glucosidase inhibitors. In particular, it is believed that Saururus chinensis, which possessed high content of crude fiber, is suitable for low-calorie food materials such as diet products.

  • In the study of general pharmacological action, we tested effect on spontaneous momentum, phenobarbital sleeping times, secretion of gastric juice, body temperature and movement of small intestine, etc. Using Pleurospermum kamtschaticum, Angelica tenuissima, Angelica gigas and Zanthoxylum schinifolium as samples to estimate adverse effects. In the study of general pharmacological action to estimate adverse effects, we observed no side effects of sample-treated groups against the control group.

  • Thirteen compounds were isolated from the roots of Angelica genuflexa through repeated silica gel column chromatography. Nine coumarins, isoimperatorin (1), osthol (2), demethylsuberosin (3), oxypeucedanin (4), heraclenin (5), pabulenol (7), umbelliferone (8), oxypeucedanin hydrate (9) and marmesinin (11), and four chromones, hamaudol (6), cimifugin (10), sec-O-glucosylhamaudol (12) and prim-O-glucosylcimifugin (13), were identified by physicochemical and spectroscopic analysis. Among these, compounds 3, 5, 6, 8, 12, and 13 were isolated for the first time from the roots of Angelica genuflexa. These coumarins and chromones were examined for their anticomplement activity. Demethylsuberosin (3) showed a weak anticomplement activity with an $IC_{50}$ value of $390\;{\mu}M$.

  • The inhibitory effects of ethanol extract of Radex Angelica uchiyamana, on LPS- or IFN- plus LPS-induced production of NO, and $TNF-{\alpha}$, and expression of iNOS and COX-2 were investigated in the activated RAW 264.7 cells. This extract significantly inhibited the production of NO and $TNF-{\alpha}$, and suppressed the expression of iNOS and COX-2 in a dose-dependent manner. These results show that ethanol extract of Radex Angelica uchiyamana may explain some known biological activities including anti-inflammatory effect, and is of considerable benefit in the treatment for NO and pro-inflammatory cytokine overproduction related immunological diseases.

  • Essential oil of the root of Angelica koreana Max. (Umbelliferae) was investigated. Essential oil was obtained from the dried roots by steam distillation and fractionated by column chromatography. Each isolate or fraction was identified by GC, GC-MS and spectral analysis. It was found to contain nine monoterpenes such as ${\alpha}-pinene$(7.0%), camphene, ${\beta}-pinene$, myrcene, ${\alpha}-phellandrene$, ${\Delta}-3-carene$, p-cymene, limonene(2.8%), terpinolene and also found to contain m-cresol(11.6%), citronellol, citral b, methylcinnamate, eudesmol and osthol. 2-Hydroxy-5-methylacetophenone and twelve compounds were tentatively identified.

  • Essential oil of the root of Angelica dahurica Benth et Hook(Umbelliferae) was investigated. Essential oil was obtained from the dried roots by steam distillation and fractionated by column chromatography. Each isolate or fraction was identified by GC, GC-MS and spectral analysis. It was found to contain eleven monoterpenes such as ${\alpha}-pinene(4.74%),\;campben, {\beta}-pinene,\;myrcene,\;{\alpha}-phellandrene,\;{\delta}-3-carene(39.4%),\;{\alpha}-terpinene,\;{\rho}-cymene,\;{\beta}-phellandrene,\;{\alpha}-terpinene,\;terpinolene\;and\;also\;found\;to\;contain\;4-vinylguaiacol,\;iso-elemicin,\;{\beta}-elemene$, caryophyllene, ligustilide, osthol and seven tentatively identified sesquiterpenes.

Four sesquiterpenes and five sesquiterpene alcohols were tentatively identified by comparison of their mass spectra.
