Comparison of Volatile Compounds in Plant Parts of Angelica gigas Nakai and A. acutiloba Kitagawa (참당귀와 일당귀의 부위별 휘발성 정유성분 비교)

  • Volatile flavor compounds Angelica gigas Nakai and Angelica acutiloba Kitagawa were extracted by SDE (simultaneous steam distillation & extraction) using the mixture of n-pentane and diethylether (1:1, v/v) as an extract solvent and analyzed by GC-FID and GC-MS. The amount of essential oils of top part and root in Angelica gigas were obtained in 0.063% (v/w) and 0.389% (v/w) yields as a fresh weight base, respectively. The main compounds in top parts and the root were identified as nonane (7.51% and 24.49%, respectively), ${\alpha}-pinene$ (14.64% and 31.75%), limonene+${\beta}-phellandrene$ (14.01% and 9.66%), ${\gamma}-terpinene$ (7.85% and 1.20%), germacrene-d (5.85% and 0.22%), (E,E)-${\alpha}-farnesene$ (6.05% and 1.40%), ${\beta}-eudesmol$ (5.26% and 1.84%). Although these compounds were present in both parts. The results showed large differences in. the concentrations of them much varied. The amount of essential oils stem and leaf obtained (0.068% and 0.127% in A. gigas) and (0.153% and 0.243% in A. acutiloba) yields as a fresh weight base, respectively. More than 18 and 32 components in stem and leaf have been identified, which of main components in A. gigas were ${\alpha}-pinene$, myrcene, limonene, germacrene-d, eudesmol and butylphthalide, but germacrene-d and butylphthalide contents were also different in stem and leaf. And more than 21 and 32 components in A. acutiloba were ${\gamma}-terpinene$ and butylphthalide. Volatile compounds were very different in both species.

Changes in the Functionality of Cheonggukjang During Fermentation Supplemented with Angelica gigas, Rehmanniae Radix, and Red ginseng (당귀.지황.홍삼 첨가에 따른 발효 청국장의 기능성 변화 연구)

  • Cheonggukjang is one of the traditional fermented soy-based foods in Korean diets. Studies in cell cultures, humans have revealed anti-hypertension, anti-stress, anticancer, antioxidant, immune enhancing effects. Angelica gigas, Rehmanniae radix, and Red ginseng are popular medicinal plants and widely used for oriental medicine. In this study a strategy had been developed to mobilize beneficial phenolics from Angelica gigas, Rehmanniae radix, and Red ginseng combined with fermented soy by Cheonggukjang fermentation for antioxidant and Type II diabetes management. The quality and functional characteristics of Chenggukjang fermented with Angelica gigas, Rehmanniae radix and Red ginseng. Cheonggukjang (CKJ), Angelica gigas Cheonggukjang (CKJ-DD), Rehmanniae radix Cheonggukjang (CKJ-RG), Angelica gigas and Rehmanniae radix Cheonggukjang (CKJ-DD+RG) and Red ginseng Cheonggukjang (CKJ-RED) were evaluated. The mobilized phenolic profile was evaluated for antioxidant activity and the potential to inhibit ${\alpha}$-amylase linked to hyperglycaemia. This research has important implications for the development of functional soy-based-fermented foods enriched with Angelica gigas, Rehmanniae radix and Red ginseng phenolics for oxidative stress - induced diabetic complications. Furthermore, Hunter's color values of 5 types cheonggukjang, lightness (L-values), redness (a-values) and yellowness (b-values) were evaluated. Free amino acid content of CKJ-RED (0.993 mg/gd. w.) showed higher than that of CKJ (0.205 mg/g-d.w.).

  • Dried leaves of Raphanus sativus L. and Angelica keiskei L. extracts have strong antioxidant potential and in consequence profound effects on ulcerative colitis. Present study was conducted to explore the effect of diet mixtures containing dried leaves of Raphanus sativus and Angelica keiskei powder on ulcerative colitis in mice and antioxidant potential of radish green and Angelica extracts as well. Both dried leaves of Raphanus sativus and Angelica keiskei extracts exhibited higher antioxidant activity due to the presence of polyphenols, favonoids and antioxidant enzymes like superoxide dismutase (SOD). Analyzed data indicate that Angelica keiskei extract had higher polyphenol and flavonoid contents compared with radish green, while maximum SOD activity was noted in dried leaves of Raphanus sativus extract Likewise, higher antioxidant ($348.72{\pm}31.65{\mu}g/mL$) and $ABTS^+$ radical scavenging (17%) activities were noted in dried leaves of Raphanus sativus extract compared with Angelica keiskei.

Effects of Nitrogen Fertilization on Leaf Yield and Pyranocurmarine Accumulation in Angelica gigas Nakai

  • Angelica gigas Nakai is one of the most widely used herbal medicines and is known to have many pharmaceutical effects including an anti-oxidant, anti-cancer etc. This study was carried out to investigate an effect of fertilization on leaf yield, production of dry-matter and accumulation of pyranocurmarine compounds such as decursin (DE) and decursinol angelate (DA) in Angelica gigas Nakai. Effect of fertilization was determined from response surface regression equation composing of 2 by 3 factorial arrangement of urea, sodium dihydrogen phosphate and potassium chloride. Yield of leaf in Angelica gigas Nakai significantly increased until 100 days after transplanting. Production of leaf also tended to increase with increasing nitrogen fertilization. Model of regression equation showed that leaf production depended upon nitrogen ($Pr>{\mid}t{\mid}$ : 0.087, 0.256 and 0.079). Also, statistical results between nitrogen application level and production of dry-matter showed significant relationship (p<0.05) and contents of dry-matter was highest in 10 kg 10a-1 treatment on 24 Sep. Active compound isolated and purified from leaf and root of Angelica gigas Nakai was identified as DE and DA by gas chromatograph-mass spectrophotometry (GC-MS). Concentration of DA as prevalent compound in leaf was highest on 20 Aug. but decreased on 24 Sep. Amount of DE and DA accumulated in Angelica gigas Nakai significantly increased with growth stages and nitrogen level. The result of our investigation imply that nitrogen fertilization is important factor for production of leaf and accumulation of pyranocurmarine in Angelica gigas Nakai as a medicinal/food materials.

  • 'Angelica' is one of the most traditionally consumed medicinal herbs around Northeast Asia including Korea for treatments of various diseases or health care purposes like hematopoiesis, blood circulation for women, sedative, analgesic, and a tonic medicine etc. Angelica gigas Nakai, a Korean native species of Angelica, is clearly different from the others in containing a high concentration of active ingredients like pyranocoumarines including decursin, decursinol, and decursinol angelate. These compounds have various kinds of positive effects such as anti-tumor activity including the precaution of neutropenia occurred during anticancer drug administration, improvements of metabolic disorders, menstrual irregularity, impairment of renal function, respiration improvement, cognition-enhancement, anti-inflammatory effect, anti-oxidative effect, enhancing fertility and so forth. Thus it implies incredible potentialities in future development for foods and drugs. However, certain purity-related qualities and/or overdose in food products can cause side effects like toxicities; therefore, their safety profiles should also be considered. This review focuses on the positive and negative effects of three pyranocoumarines in Angelica gigas Nakai and some possibilities and considerations for future food and drug products development.

A Study on Environment Characteristics of the Angelica gigas Nakai Population (참당귀(Angelica gigas Nakai) 개체군의 환경 특성 분석)

  • This study was carried out to investigate vegetation structure and soil properties of Angelica gigas population distributed in Jeombongsan, Bangtaesan, Odaesan, Gyebangsan and Jirisan. From August 2007 until September 2008, 5m${\times}$5m quadrat was established in native area of Angelica gigas in order to record a dominants and coverage, and soil factors at 20 sites. It was found that the altitude in the distributed areas for Angelica gigas population was 710m or more. Angelica gigas population was classified into Parasencio auriculata var. kamtschatical dominant population, Pseudostellaria palibiniana dominant population, Isodon excisus dominant population and typical dominant population. In the site of study, soil pH, electrical conductivity, soil organic matter, available phosphorous, and exchangeable potassium, exchangeable calcium, exchangeable magnesium, exchangeable natrium concentration and total nitrogen were ranged from 5.1~6.2, 0.1~0.79dS/m, 2.21~22.11%, 9.56~37.97mg/kg, 1.85~23.88cmol+/kg, 0.21~1.18cmol+/kg, 1.07~5.09cmol+/kg, 0.04~0.14cmol+/kg and 0.09~1.04% respectively. The Parasencio auriculata var. kamtschatical dominant population was found in area of high altitude and had high slope degree. But Pseudostellaria palibiniana dominant population was found in area of low altitude and low slope degree. Isodon excisus dominant population was found to have higher soil pH and exchangeable potassium concentration than the other dominant population.

  • Silicagel colum chromatography of the ether extract of the fruits of Angelica decursiva $F_R$. et SAV. gave four crystalline products of coumarins and one steroid.   • Objectives : Hyaluronic acid(HA) is a mucopolysaccharide, occuring naturally in living organisms. It is one of the most hydrophilic molecules, so it has been known as being related to skin hydration and skin aging. The purpose of this study is to examine the effects of Angelica gigas(A. gigas) ethanol extract on hyaluronic acid synthesis. Methods : To determine cytotoxicity and hyaluronic acid synthase 2 gene expression, hyaluronic acid production in HaCaT cells, MTT assay and RT-PCR ELISA was used. Results : There were no cytotoxicity in $50{\mu}g/ml$ concentration A. gigas extract in MTT assay. Hyaluronic acid synthase 2(HAS2) gene expression was increased by all treated concentration A. gigas extract. Hyaluronic acid production was higher than control group in $50{\mu}g/ml$ & $100{\mu}g/ml$ concentration A. gigas extract. Conclusions : Hyaluronic acid production was increased by A. gigas extracts. Therefore, We suggest that A. gigas can make a contribution to the moisturizing effect on human skin.

Quality Characteristics of Yogurt Supplemented with Angelica gigas Nakai Leaf Extract (참당귀잎 추출물을 첨가한 요구르트의 품질 특성)

  • In this study, quality characteristics of yogurt supplemented with Angelica gigas Nakai leaf extract were examined. The pH of the yogurt ranged from 4.40 to 4.45 and the titratable acidity ranged from 0.96% to 0.98%. The viscosity tended to decrease with the addition of the Angelica gigas Nakai leaf extract, but did not affect stability during storage. In the range of 0.1% to 0.3%, lactic acid bacteria were present in the range of 1.9×109 to 3.2×109 CFU/mL. The decursin content in yogurt was quantitatively analyzed, depending on the addition of 0.1% to 0.3% of Angelica gigas Nakai leaf extract and was found to be 0.26 ㎍/g, 15.23 ㎍/g, and 23.57 ㎍/g respectively. Organic acid showed the highest generation of lactic acid. The antioxidant properties of yogurt were shown to increase with the addition of the Angelica gigas Nakai leaf extract. The sensory score of yogurt supplemented with 0.1% of the Angelica gigas Nakai leaf extract was highly valued, at a level similar to that of plain yogurt. Yogurt supplemented with 0.2% of the extract was rated above the normal score of 6.31 to 6.50. As shown by the results, the optimal concentration of Angelica gigas Nakai leaf extract for addition to yogurt was within 0.2%.
