Selection of Functional Lentinula edodes Strains with High Ergothioneine Content using Mono-mono Hybridization (단핵균주간 교잡에 의한 에르고티오네인 함량이 높은 기능성 표고 균주 선발)

  • Lentinula edodes have been cultivated extensively not only in South Korea but also in other Asian countries. In terms of taste and nutrition, it is a valuable mushroom that is comparable to other mushrooms. L. edodes contains various physiologically active compounds that promote human health. One compound, ergothioneine, exerts a powerful antioxidant function that inhibits cellular senescence. As it is not biosynthesized by plants or animals, L. edodes is very important for ergothioneine supply. The L. edodes cultivars Bambithyang (NIFoS 3404) and Sansanhyang (NIFoS 3420) of the National Institute of Forest Science (NIFoS) had higher ergothioneine content than the other cultivars, and 11 strains were obtained using mono-mono hybridization between them. The strains were inoculated into sawdust media to obtain fruiting bodies, and the ergothioneine content of each hybrid strain was confirmed using high-performance liquid chromatography (HPLC) analysis. Among the 11 strains, NIFoS 5108 had a higher ergothioneine content than the parental strains, Bambithyang (NIFoS 3404) and Sansanhyang (NIFoS 3420). This study revealed that it is possible to develop cultivars with improved specific functions using mono-mono hybridization.

Biological Control of the Sciarid Fly, Lycoriella mali (Diptera: Sciaridae) Using Steinernema carpocapsae in a Button Mushroom Cultivation House (양송이버섯 재배사에서 Steinernema carpocapsae를 이용한 긴수염버섯파리 생물적 방제)

  • We used the entomopathogenic nematode, Steinernema carpocapsae, for the effective biological control of Lycoriella mali during Agaricus bisporus cultivation. The concentrations of two S. carpocapsae products distributed in Korea were $6{\times}10^4$ and $2.5{\times}10^5m^{-2}$ respectively. As a result of investigating the density of A. bisporus larvae in a rice straw medium and soil in an A. bisporus cultivation house, the mean numbers per $30cm^2$ were 0.8 and 22.2, respectively, and A. bisporus larvae were detected mainly in the soil. The control effects after once spraying S. carpocapsae at $6{\times}10^4$ and $2.5{\times}10^5m^{-2}$ was higher at the 14th day than at the 7th day. Therefore, we investigated the control effect after twice spraying of $6{\times}10^4$ and $2.5{\times}10^5m^{-2}$ at the 14th day. The control effects after this spraying were 51.9% and 96.8%, respectively, and the control effect of spraying at $2.5{\times}10^5m^{-2}$ was better than at $6{\times}10^4m^{-2}$. The control effect of diflubenzuron WP was lower than 50%.

Antiangiogenic activity of non-aqueous fraction from Sparassis crispa extract in human umbilical vein endothelial cells (혈관내피세포에서 꽃송이버섯(Sparassis crispa) 소수성 추출물의 항혈관신생 활성)

  • Sparassis crispa is an edible mushroom that is distributed in Korea, Japan, Europe, and North America. It exerts various biological activities such as immunopotentiation, anti-diabetic, anti-cancer, and anti-inflammatory effects. Recently, we separated the health functional non-aqueous fraction from the chloroform extract of S. crispa (SCF4). In this study, we evaluated the antiangiogenic activity of SCF4 in human umbilical vein endothelial cells (HUVECs). SCF4 effectively inhibited vascular endothelial growth factor (VEGF)-induced cell growth at concentrations ($5-25{\mu}g/mL$) showing no cytotoxic effects. SCF4 inhibited VEGF-induced invasiveness and tube formation ability, which are in vitro angiogenic features of HUVECs, in a dose-dependent manner. In addition, SCF4 markedly suppressed in vivo angiogenesis of chorioallantoic membrane from growing chick embryos without cytotoxicity. Furthermore, SCF4 downregulated the phosphorylation of VEGFR2, AKT, and ERK1/2, which are major angiogenic signal mediators. These results suggest that SCF4 inhibited angiogenesis by suppressing the VEGFR2 signaling pathways without cytotoxicity.

Development and Application of a Cleaved Amplified Polymorphic Sequence Marker for Discriminating A Mating Type Alleles of Lentinula edodes (표고 A 교배형 구분을 위한 CAPS 마커의 개발 및 적용)

  • Lentinula edodes is one of the most widely consumed edible mushrooms in Korea. Mating in L. edodes is regulated by a tetrapolar system, and two unlinked genetic loci, A and B, are known to be major determinants of the mating types, as reported in other heterothallic basidiomycetes. The A locus of L. edodes encodes a pair of homeodomain (HD) transcription factors. The highly variable N-termini of these HD transcription factors contribute to the diversity among the A mating types. In this study, we developed a cleaved amplified polymorphic sequence (CAPS) marker to discriminate 11 different A mating type alleles predominant among both cultivated and wild strains. Amplification of the variable region of the A locus followed by digestion with HaeIII and EcoRI restriction enzymes enabled successful discrimination among the 11 A mating type alleles. We also evaluated the applicability of this method in the identification of two A mating types of a dikaryotic strain.

Antioxidant and whitening effects of loquat (Eriobotrya japonica) fruit extracts (비파(Eriobotrya japonica) 열매 추출물의 산화방지능과 미백 효과)

  • The loquat (Eriobotrya japonica) is a fruit tree belonging to the Rosaceae family. Loquat fruit extracts from three cultivars (Tanaka, Mogi, and Jinwang) were prepared using absolute methanol and homogenization and ultrasound sonication procedures. We measured the total phenolic and flavonoid content of the fruit extracts and their antioxidant capacities. In addition, we evaluated tyrosinase activity and anti-melanogenic effects in B16F1 melanoma cells. The fruit extract from cv. Tanaka had the highest total phenolic content and showed the highest antioxidant capacity in the ABTS assay. The loquat fruit extract from cv. Jinwang had the highest total flavonoid content and showed the highest antioxidant capacity in the DPPH assay. Loquat fruit extracts from the Tanaka and Jinwang cultivars effectively inhibited mushroom tyrosinase activity. The loquat fruit extracts reduced intracellular oxidative stress in B16F1 melanoma cells. Treating B16F1 melanoma cells with loquat fruit extract from cv. Tanaka at a concentration of 125 ㎍/mL effectively inhibited melanin synthesis. However, treating the B16F1 melanoma cells with loquat fruit extracts from the Jinwang and Mogi cultivars did not. These results suggest that loquat fruit extracts from the Tanaka cultivar may serve as potential sources of antioxidants and act as a skin-whitening agent.

Protease Activity from Fruit Body of Sarcodon aspratus (능이자실체의 Protease 활성)

  • This study was performed to investigate the protease activity from fruit body of Sarcodon aspratus and its features. The specific protease activity was increased with the increasing purification steps, 2.62 times by desalting, 17 times by CMC column chromatography, 113.8 times by DEAE-Sephadex A-50 column chromatography, and 728.3 times by Sephadex G-75 column chromatography. Proteases were identified as two different enzymes having different isoelectric points at pH 4.35 (its recovery rate 8%) and pH 4.7 (its recovery rate 3.5%). Those proteases were purified by 3,025 folds and 3,257 folds in terms of specific activity. Two proteases having different isoelectric points had similar enzymatic properties. This protease was estimated to be 43,000 daltons of molecular weights by SDS-PAGE. This protease with optimum pH 4 was almost stable in the pH range of 4~7. Optimal temperature of protease activity was 40 to 50℃, and the protease activity was completely inhibited at 70℃ for 30 min.

Analysis of the Ergothioneine Content in the Fruiting Bodies of Sawdust-cultivated Lentinula edodes Cultivars in Korea (한국의 표고 톱밥재배 품종의 에르고티오네인 함량 분석)

  • Lentinula edodes is a type of mushroom widely consumed in East Asia, and many of the components contained in them are used as raw materials for pharmaceuticals or as health supplements. Ergothioneine, a sulfur-containing amino acid in L. edodes, is attracting attention due to its antioxidant and anti-inflammatory effects. L. edodes generally contain an ergothioneine content of about 200 to 800 mg per kg; there are some differences in this value depending on the cultivation conditions. In this study, 24 domestic sawdust-cultivated L. edodes cultivars were cultured under the same conditions, and the morphological characteristics and ergothioneine content of the fruiting bodies were investigated. The yield and morphological characteristics of the fruiting bodies were different in each cultivar. Taehyanggo and Sanjo 713ho had the largest and thickest pileus. The pileus of L. edodes had a higher ergothioneine content than the stipe. Sanjo 715ho had the highest ergothioneine content in the pileus and the stipe at 1,225 mg/kg and 753 mg/kg, respectively. An ergothioneine content of more than 1,000 mg/kg in the pileus was observed in Bambithyang, Sansanhyang, Sulbaekhyang, Taehyanggo, Sanjo 705ho, Sanjo 709ho, Sanjo 715ho, and Sanjo 716ho. These cultivars can be used as parental strains to develop new cultivars with enhanced ergothioneine functionalities.

Vertical Transmission of RNA Mycoviruses in Lentinula edodes (표고에서의 RNA 바이러스 수직감염)

  • Lentinula edodes is an important commercial mushroom and there have been several reports of viral infections in L. edodes. Two mycoviruses (LeV-HKB and LeNSRV1) were detected in Sanbaekhyang (NIFoS 2778) and Taehyanggo (NIFoS 4317), the sawdust-cultivated commercial strains. The vertical transmission rates of the viruses were investigated by detecting the viruses in 80 monokaryotic strains derived from basidiospores isolated from the fruiting bodies of each strain. Most of the monokaryotic strains were infected with the virus and the two viruses showed different levels of meiotic stability, with LeV-HKB showing higher meiotic stability than LeNSRV1. Therefore, it seems that the vertical transmission mechanism of mycoviruses is different depending on the virus species. We also examined the mycelial growth rate of the monokaryotic strains and compared the growth rate according to virus infection status. Although there was no statistically significant correlation between viral infection and mycelial growth rate, we found that the average growth rate was reduced by additional virus infection. We expect our data to contribute to a greater understanding of the mechanism of the vertical transmission of mycoviruses, and promoting breeding using virus-free monokaryotic strains.

  • Among all the ingredients usedin mandu, the following types were used:, 13 types of grains were used (12.38%), 30 types of vegetables, fruits, bulbs,and nuts were used (28.57%), 32 types of marine products, birds, meats, fishes, and shellfishes were used (30.48%), 10 types of functional ingredients were used (9.52%) and. For spices, 20 types of spices were used (19.05%). 2. Cooking Methods offor Mandu. The mMandu eaten at in the early Joseon era had was primarily made ofusedbuckwheat that contained boiled tofu or egg uiijuk in the kneaded dough for the most part and while kneading with buckwheat, the tofu or egg uiijuk has been boiled down to knead the dough, and and starch powder, bean powder, or rice powder, etc were mixed to make the mandu coating. Buckwheat powder was mixed toadded to the flourwer or was used by itself, while meat, vegetables, tofu, and shiitake mushroom, etc were also addedincluded. From the 18th century, the host plant, or cabbage kimchi, were prepared and combined had been sliced to be used as filling together while red pepper powder was mixed combined withto spices or vinegar soy sauce to be used together. Also, Radishes had beenwere also used as filling, but shown as not being used fromafter the start of the 1900's. For the shape of mMandu, it was madeinto different shapes such as as triangle, rectangle, date plum, gwebul, half moon, or pomegranate shapes, and then shapes to be boiled in simmering water, baked, or cooked as soup in clear broth for soup., In the 17th to 18th century, boilingthen in a steamer gradually became a cooking style, assumed the style of boiling in a steamer in $17th{\sim}18th$ century while in the 16th century,the an essay ofn fermenting flour in ‘Food Dimibang’ in 16th century had indicated it was cooked as the style ofby steaming in a rice steamer. Also, Mandu may have also contained the following: the thin-cut and boiled fish was cut out thin to put into the filling and boiled down, made by putting in added pine nuts after making bbeef jerky or boiled- down meat, fish, or shellfish itself to extractsand mold mandu only the ingredients combined withto put on starch powder, and then boiled down and put on pine nut powder finally, after it or cooled it wasdown to be eaten by dipping in vinegar soy sauce. In conclusion, many different types of mandu were made during the Joseon era using a variety ofwhile the ones using such various ingredients. are also one type of mandu.

Characteristics of Lentinula edodes Fruiting Bodies Formed by Mono-mono Hybridization Using Mating Type Analysis (교배형 분석을 활용한 표고 교배균주 육성 및 자실체 발생 특성)

  • In this study, dikaryotic strains of Lentinula edodes were generated by mono-mono hybridization using mating type analysis and their fruiting body characteristics were investigated. Approximately 100 monokaryotic strains were isolated from the basidiospores of Sanbaekhyhang, and homokaryotic strains were isolated from Chungheung 1ho using protoplast isolation and regeneration. Their mating types were evaluated and a total of 60 dikaryotic strains were hybridized. Using these strains, fruiting bodies were produced and their characteristics were examined after cultivation on sawdust media. The results indicated that the rate of hybridization was 100% and that 55 of 60 strains formed fruiting bodies. These showed normal pileus and gill structures; however,10 strains also produced fruiting bodies with abnormal pileus and gill structures. The weight and size of the fruiting bodies differed depending on the strains. Overall, further studies are needed for predicting the characteristics of hybridized strains based on their parental strains.
