Cytotoxic Effect of Triglycerides via Apoptotic Caspase Pathway in Immune and Non-immune Cell Lines

  • Hyperlipidemia is defined as conditions of the accumulation of lipids such as free fatty acids (FFA), triglyceride (TG), cholesterol and/or phospholipid in the bloodstream. Hyperlipidemia can cause lipid accumulation in non-adipose tissue, which is lipid-cytotoxic effects in many tissues and mediates cell dysfunction, inflammation or programmed cell death (PCD). TG is considered to be a major cause of atherosclerosis through inflammatory necrosis of vascular endothelial cells. Recently, TG have also been shown to exhibit lipid-cytotoxicity and induce PCD. Therefore, we investigated the effect of TG on the cytotoxic effect of various cell types. When exposed to TG, the cell viability of U937 monocytes and Jurkat T lymphocytes, as well as the cell viability of MCF-7, a non-immune cell, decreased in time- and dose-dependent manner. In U937 cells and Jurkat cells, caspase-9, an intrinsic apoptotic caspase, and caspase-8, an extrinsic apoptotic caspase, were increased by exposure to TG. However, in TG-treated MCF-7 cells, caspase-8 activity increased only without caspase-9 activity. In addition, the reduction of cell viability by TG was recovered when all three cell lines were treated with pan-caspase inhibitor. These results suggest that activation of apoptotic caspases by TG causes lipotoxic effect and decreases cell viability.

  • 시판 막걸리에서 분리한 균주(Lactobacillus paracasei ssp. paracasei M13-65-3)로 타락을 제조하였다. M13-65-3균주를 이용하여 제조한 타락 추출물에 의하여 T lymphocyte인 Jurkat 세포의 증식이 촉진되었고 피부세포의 멜라닌 생성이 억제되었다. 본 연구의 결과는 타락이 면역 증강 기능성과 피부색소 조절 기능성을 보유할 가능성을 제시하고 있다.

오가피의 면역조절작용 (Immunoregulatory Action of OGAPI)

  • The purpose of this research was to investigate the immunoregulatory effect and the leukemia cell apoptosis of EtOH extract of OGAPI(OGP). The proliferation of cultured splenocytes, thymocytes and mesenteric lymph node cells were enhanced by the addition of OGP. Splenic and thymic T lymphocytes, especially TH and Tc cells were significantly increased in OGP-administered mice. OGP markedly increased the production of γ-interferon in mice serum and accelerated the phagocytic activity in peritoneal macrophages. OGP treatment enhanced the apoptosis of L1210 mouse leukemia and Jurkat, Molt4 human leukemia cells, and increased the expression of apoptosis-related ICE, c-myc, p53 gene in Jurkat cell. These results suggest that OGP have an immunoregulatory action and anti-cancer activity.

Construction of tat-and nef-defective HIV-1 and screening of natural extracts with anti-HIV-1 activity

  • Human immunodeficiency virus type 1 (HIV-1) contains several nonstructural genes which are required for the viral replication and disease pathogenesis. Among them, tat and nef genes encode an essential transactivator of HIV-1 LTR and a pluripotent protein which seems to be essential for the in vivo but not in vitro viral replication, respectively. We constructed two tat and n of defective HIV-1 and tested for their ability to replicate in several T cells. The defective viruses did not replicate in CD4$\^$+/ T cells, but rescued in the recombinant Jurkat-tat cell which also contains tat gene. The replication of tat and nef defective HIV-1 which expresses chloramphenicol acetyltransferase(CAT) gene was easily detected by a sensitive CAT assay. No revertant was identified during the passages of the mutant viruses for more than two months in Jurkat-tat cells. tat and n of defective HIV-1 could be used instead of wild type viruse for several purposes such as inhibitor screening and development of attenuated AIDS vaccine.

Alteration of Inflammatory Cytokines by Volatile Organic Compounds in T Lymphocytes

  • In the present study, we investigated whether volatile organic compounds induce inflammatory response in human T lymphocytes by evaluating the alteration of inflammatory cytokines. Volatile organic compounds such as formaldehyde, o-xylene, benzene, and hydroquinone have no cytotoxic effects on Jurkat T cells at a high concentration of 50 ${\mu}M$ for 48 h. IL-2, IL-4, IL-13, TNF-${\alpha}$ and IFN-${\gamma}$ were increased after the treatment with volatile organic compounds, although alteration of cytokines is different among volatile organic compounds. LPS as a positive control increased the secretion of IL-2, IL-4, IL-13, TNF-${\alpha}$ and IFN-${\gamma}$. MCP-1 and CCL17 (thymus and activation-regulated chemokine, TARC) were weakly increased after the treatment with volatile organic compounds but the amount of the increased cytokine was below 20 pg/ml. These results suggest that the measurement of cytokine in Jurkat T cells may be used as a useful method for evaluating the toxicity of volatile organic compounds in immune response.

Twist2 Regulates CD7 Expression and Galectin-1-Induced Apoptosis in Mature T-Cells

  • In the periphery, a galectin-1 receptor, CD7, plays crucial roles in galectin-1-mediated apoptosis of activated T-cells as well as progression of T-lymphoma. Previously, we demonstrated that $NF-{\kappa}B$ downregulated CD7 gene expression through the p38 MAPK pathway in developing immature thymocytes. However, its regulatory pathway is not well understood in functional mature T-cells. Here, we show that CD7 expression was downregulated by Twist2 in Jurkat cells, a human acute T-cell lymphoma cell line, and in EL4 cells, a mature murine T-cell lymphoma cell line. Furthermore, ectopic expression of Twist2 in Jurkat cells reduced galectin-1-induced apoptosis. While full-length Twist2 decreased CD7 promoter activity, a C-terminal deletion form of Twist2 reversed its inhibition, suggesting an important role of the C-terminus in CD7 regulation. In addition, CD7 expression was enhanced by histone deacetylase inhibitors such as trichostatin A and sodium butyrate, which indicates that Twist2 might be one of candidate factors involved in histone deacetylation. Based on these results, we conclude that upregulation of Twist2 increases the resistance to galectin-1-mediated-apoptosis, which may have significant implications for the progression of some T-cells into tumors such as Sezary cells.

The Cytotoxic and Anti-proliferative Effects of 3-Hydrogenkwadaphnin in K562 and Jurkat Cells Is Reduced by Guanosine

  • 3-hydrogenwadaphnin (3-HK) is a new daphnane-type diterpene ester isolated from Dendrostellera lessertii with strong anti-tumoral activity in animal models and in cultures. Here, prolonged effects of this new agent on proliferation and viability of several different cancerous cell lines were evaluated. Using [$^3H$]thymidine incorporation, it was found that the drug inhibited cell proliferation and induced G1/S cell cycle arrest in leukemic cells 24 h after a single dose treatment. The cell viability of Jurkat cells was also decreased by almost 10%, 31% and 40% after a single dose treatment (7.5 nM) at 24, 48 and 72 h, respectively. The drug-treated cells were stained with acridine orange/ethidium bromide to document the chromatin condensation and DNA fragmentation. These observations were further confirmed by detection of DNA laddering pattern in the agarose gel electrophoresis of the extracted DNA from the treated cells. Treatment of K562 cells with the drug at 7.5, 15 and 30 nM caused apoptosis in 25%, 45% and 65% of the cells, respectively. Exogenous addition of $25-50\;{\mu}M$ guanosine and/or deoxyguanosine to the cell culture of the drug-treated cells restored DNA synthesis, released cell arrest at G1/S checkpoint and decreased the apoptotic cell death caused by the drug. These observations were not made using adenosine. However, the drug effects on K562 cells were potentiated by hypoxanthine. Based on these observations, perturbation of GTP metabolism is considered as one of the main reasons for apoptotic cell death by 3-HK.

인간의 Jurkat T세포에서 프로스타글란딘 PGE2) (PGE2)의 cAMP 경로를 통한 인터페론 감마(INF--γ ) 유전자의 methylation (PGE2 Mediated INF-γ Gene Methylation Through cAMP Signaling Pathway in Human Jurkat T Cells)

  • 본 연구에서 인간의 백혈병세포주인 Jurkat T 세포에서 인터페론 감마(INF-${\gamma}$ 유전자의 methylation에 대한 S-nitroso-N-acetylpenicillamine (SNAP), 프로스타글란딘 $E_2$ (PG $E_2$) 그리고 dibutric cyclic AMP (dbcAMP)의 효과를 조사하였다. 인터페론 감마 유전자의 프로모터기능에 아주 중요한 디뉴클레오티드인 CpG는 SNAP, PG $E_2$, 그리고 dbcAMP를 각각 처리하였을 때 methylation되었다. PG $E_2$에 의해서 유도된 그 methylation은 아데닐산 사이클라제의 저해제의 하나인 2',5'-dideoxyadenosine (DDA)에 의해서 억제되었지만, SNAP에 의해서 유도된 methylation은 DDA에 의해서 억제되지 않았다. PG $E_2$나 dbcAMP를 처리한 세포에서 일산화질소(NO)의 생성의 증가는 나타나지 않았으며, PG $E_2$나 dbcAMP에 의해 유도된 인터페론 감마유전자의 methylation도 일산화질소합 성효소의 저해제인 $N^{G}$ -methyl-L-arginine (L-NMMA)에 의해서 억제되지 않았다. 따라서 인간의 Jurkat T 세포에서 PG $E_2$에 의한 인터페론 감마 유전자의 발현 억제는 세포내의 cAMP생성경로를 통한 인터페론 감마 유전자의 methylation과 연관되어있으나 일산화질소의 생성경로와는 무관한 것으로 보인다.화질소의 생성경로와는 무관한 것으로 보인다.

  • 1. Extracts of the leaves of Eurya emarginata markedly inhibited the growth of leukemia cells such as HL-60, KG-1, U937, K562, and Jurkat. 2. When HL-60 cells were treated with the extract, DNA fragmentation, morphological changes and sub-G1 hypodiploid cells were observed. The expressions of c-myc mRNA were also dramatically decreased. 3. Cornoside and Eutigoside were isolated from the leaves of Eurya emarginata. 4. Eutigoside induced apoptosis through down-regulation of Bcl-2 expression and activation of caspases.

Identification of Immune Responsive Genes on Benzene, Toluene and o-Xylene in Jurkat Cells Using 35 k Human Oligomicroarray

  • Volatile organic compounds (VOCs) are a major component of urban air pollution. It is documented that low exposure levels of VOCs induce alterations in immune reactivity resulting in a subsequent higher risk for the development of allergic reactivity and asthma. Despite these facts, there are few reports on the affected primary target and the underlying effective causal mechanisms. So in this study, to better understand the risk of BTX (benzene, toluene and o-xylene) which are the major VOCs and to identify novel biomarkers on immune response to these VOCs exposure in human T lymphocytes, we performed the toxicogenomic study by analyzing of gene expression profiles using 35 k human oligo-microarray. BTX generated specific gene expression patterns in Jurkat cell line. By clustering analysis, we identified some genes as potential markers on immuno-modulating effects of BTX. Four genes of these, HLA-DOA, ITGB2, HMGA2 and 5TAT4 were the most significantly affected by BTX exposure. Thus, this study suggests that these differentially expressed immune genes may play an important role in the pathogenesis on BTX exposure and have significant potential as novel biomarkers of exposure, susceptibility and response to BTC.
