Immune Cells Are Differentially Affected by SARS-CoV-2 Viral Loads in K18-hACE2 Mice

  • Viral load and the duration of viral shedding of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) are important determinants of the transmission of coronavirus disease 2019. In this study, we examined the effects of viral doses on the lung and spleen of K18-hACE2 transgenic mice by temporal histological and transcriptional analyses. Approximately, 1×105 plaque-forming units (PFU) of SARS-CoV-2 induced strong host responses in the lungs from 2 days post inoculation (dpi) which did not recover until the mice died, whereas responses to the virus were obvious at 5 days, recovering to the basal state by 14 dpi at 1×102 PFU. Further, flow cytometry showed that number of CD8+ T cells continuously increased in 1×102 PFU-virus-infected lungs from 2 dpi, but not in 1×105 PFU-virus-infected lungs. In spleens, responses to the virus were prominent from 2 dpi, and number of B cells was significantly decreased at 1×105 PFU; however, 1×12 PFU of virus induced very weak responses from 2 dpi which recovered by 10 dpi. Although the defense responses returned to normal and the mice survived, lung histology showed evidence of fibrosis, suggesting sequelae of SARS-CoV-2 infection. Our findings indicate that specific effectors of the immune response in the lung and spleen were either increased or depleted in response to doses of SARS-CoV-2. This study demonstrated that the response of local and systemic immune effectors to a viral infection varies with viral dose, which either exacerbates the severity of the infection or accelerates its elimination.

Development of Digital Literacy Curriculum Framework Connected Computational Thinking in the information Education (정보과 교육과정에서 컴퓨팅사고력과 연계한 디지털소양 교육과정 프레임워크 개발)

  • This study provided digital literacy concept, core area and class achievement connected computational thinking in order to reestablish digital literacy keeping pace with trend change. Main study's contents analyzed digital literacy concept and changing trend of digital technologies, provided characters of information subject matter education. Implications from these analyses is that necessary digital literacy is to support software education, contents being connected computational thinking and contents being necessary. And we committed Delphi investigation to 16 information education expert on total 67 achievements based on these previous studies. Survey results totally surpassed CVR criterion of digital literacy called information life and then readjusting parts of achievements through panel discussions because of partially being lower than Validity criterion. Finally achievements of Information Life came into existence 5 items for 1st, 2nd of elementary school level(ESL), 13 items for 3rd, 4th of ESL, 23 items for 5th, 6th of ESL, 16 items for middle school Level.

  • Computational thinking is complexity and independent subject matter being capable to learn concept of computer science and providing methodology of problem solving. Also many experts have said that computational thinking will be grow essential tool in the further developing information society. Thus our country has been trying to introduce it in the K12 informatics subject matter education. Therefore we proposed a introducing method of computational thinking being appropriated of a character of it in the informatics curriculum. To do this, we analyzed character and worthy of it, advanced model cases introducing it into the curriculum. And we proposed that introduced case of it into curriculum is divided 3 cases archiving computational thinking itself, being connected aim of general subject matter with it and computer science education. According to this advanced cases, this study selected permeative style of computational thinking with the informatics subject matted curriculum. This method is divided achievement criterion into contents and means. also we proposed that contents area of informatics subject matter achievement criterion is composed Computing System, Information Life, Software and means area can be filled with subset of computational thinking. This introducing method can make informatics subject matter education settle subject matter helping problem solving through computer system beyond character of technology oriented subject matter.

  • Objectives : This study was designed to evaluate comparative effects of body acupuncture and 8 constitution acupuncture on acute stage lumbago. Methods : 50 patients checked up the Pancreotonia with acute stage lumbago were randomly divided into 2 groups. One group(Control group) took body acupuncture therapy, and the other group(Test group) took 8 constitution acupuncture therapy. Visual analogue scale(VAS), lumbar flexion and extension were checked before and after acupuncture therapy for evaluating the improvement of lumbago. Wilcoxon signed rank test and Mann-Whitney U test of SPSS 15.0 for windows program were used for a statistical analysis. Results : Both body acupuncture therapy and 8 constitution acupuncture therapy showed good effect on acute stage lumbago. And test group showed better effect on decreasing of VAS and improving of lumbar flexion and extension than control group. There was only statistically significant difference between two groups in improving of lumbar flexion, while, decreasing of VAS and improving of lumbar extension were not significant statistically. Conclusions : 8 constitution acupuncture therapy can be recommended as a useful therapy to treat patients checked up the Pancreotonia with acute stage lumbago. Futher study is needed to confirm the effectiveness of 8 constitution acupuncture.

  • Cystic lesions or progressive cystic changes in adenocarcinoma of the lung have rarely been reported. We report a case of lung adenocarcinoma that progressed from ground-glass opacities (GGOs) and consolidations or nodules to extensive cystic lesions during 12 months in a young adult patient. A 29-year-old male was initially diagnosed with primary lung adenocarcinoma by transbronchial lung biopsy of the right lower lobe and lung to lung metastasis in both lungs according to imaging findings. The initial chest computed tomography (CT) scans showed multifocal GGOs, consolidations, and nodules in both lungs. Despite treatment with palliative chemotherapy, the patient's follow-up CT scans showed multiple, cystic changes in both lungs and that the lesions had progressed more extensively. He died of hypoxic respiratory failure one year after his diagnosis.

Comparative Clinical Study between Oriental Medical Treatment and Oriental-Western Combination Treatment on Herniated Lumbar Disc (요추추간판탈출증의 한방치료와 한.양방치료 병행에 대한 비교 연구)

  • Objectives : The purpose of this study is to investigate the difference of treatment effect between Oriental Medical Treatment and Oriental-Western Combination Treatment on Herniated Lumbar Disc. Methods : From 1st November, 2008 to 31th August, 2009, 18 Herniated Lumbar Disc patients who admitted the Department of Acupuncture & Moxibustion, college of Oriental medicine, Dae-gu Hanny University were divided into two groups. Group A was treated with oriental medical theraphy (Acupuncture, Moxibustion, Herb, Cupping, Physical theraphy) and Group B with both oriental medical theraphy and western medical theraphy(Acupuncture, Moxibustion, Herb, Cupping, Physical theraphy, Intramuscular injection of Dexamethasone and Dicknol 2ml). We evaluated the treatment effect of each group with the Visual Analog Scale(VAS) and Pain Rating Scale(PRS), deviding two period(From first day to fourth day after Admission and From first day to ninth day after Admission). Results : Improvement Degree of Group A and Group B was significantly improvement (p<0.05) according to the VAS & PRS. Group B showed better performance compared with Group A. Conclusions : These results proved that combination treatment of oriental and western medicine was more effective than just oriental medical treatment of Herniated Lumbar Disc. And the difference between the two groups were significant according to PRS.

Risk Factors Associated with Subdural Hygroma after Decompressive Craniectomy in Patients with Traumatic Brain Injury : A Comparative Study

  • Objective : Subdural hygroma (SDG) is a complication occurring after head trauma that may occur secondary to decompressive craniectomy (DC). However, the mechanism underlying SDG formation is not fully understood. Also, the relationship between the operative technique of DC or the decompressive effect and the occurrence and pathophysiology of SDG has not been clarified. Purpose of this study was to investigate the risk factors of SDG after DC in our series. Methods : From January 2004 to December 2008, DC was performed in 85 patients who suffered from traumatic brain injury. We retrospectively reviewed the clinical and radiological features. For comparative analysis, we divided the patients into 2 groups : one group with SDG after craniectomy (19 patients; 28.4% of the total sample), the other group without SDG (48 patients; 71.6%). The risk factors for developing SDG were then analyzed. Results : The mean Glasgow Outcome Scale (GOS) scores at discharge of the groups with and without SDG were 2.8 and 3.1, respectively (p<0.0001). Analysis of radiological factors showed that a midline shift in excess of 5 mm on CT scans was present in 19 patients (100%) in the group with SDG and in 32 patients (66.7%) in the group without SDG (p<0.05). An accompanying subarachnoid hemorrhage (SAH) was seen in 17 patients (89.5%) in the group with SDG and in 29 patients (60.4%) in the group without SDG (p<0.05). Delayed hydrocephalus accompanied these findings in 10 patients (52.6%) in the group with SDG, versus 5 patients (10.4%) in the group without SDG (p<0.05). On CT, compression of basal cisterns was observed in 14 members (73.7%) in the group with SDG and in 18 members of the group without SDG (37.5%) (p<0.007). Furthermore, tearing of the arachnoid membrane, as observed on CT, was more common in all patients in the group with SDG (100%) than in the group without SDG (31 patients; 64.6%) (p<0.05). Conclusion : GOS showed statistically significant difference in the clinical risk factors for SDG between the group with SDG and the group without SDG. Analysis of radiological factors indicated that a midline shifting exceeding 5 mm, SAH, delayed hydrocephalus, compression of basal cisterns, and tearing of the arachnoid membrane were significantly more common in patients with SDG.

Analysis of Molecular Expression in Adipose Tissue-Derived Mesenchymal Stem Cells : Prospects for Use in the Treatment of Intervertebral Disc Degeneration

  • Objective : Recent studies have shown encouraging progress toward the use of autogenic and allogenic mesenchymal stem cells (MSCs) to arrest, or even lead to partial regeneration in, intervertebral disc (IVD) degeneration. However, this technology is still in its infancy, and further development is required. The aim of this study was to analyze whether rat adipose-derived mesenchymal stem cells (ADMSC) can differentiate towards IVD-like cells after treatment with transforming growth factor ${\beta}3$ (TGF-${\beta}3$) in vitro. We also performed quantitative analysis of gene expression for ADMSC only, ADMSCs treated with TGF-${\beta}3$, and co-cultured ADMSCs treated with TGF-${\beta}3$. Methods : ADMSCs were sub-cultured to homogeneity and used in fluorocytometry assays for CD11, CD45, and CD90/Thy1. ADMSCs were differentiated in spheroid culture towards the chondrogenic lineage by the presence of TGF-${\beta}3$, dexamethasone, and ascorbate. We also co-cultured pure ADMSCs and nucleus pulposus cells in 24-well plates, and performed immunohistochemical staining, western blotting, and RT-PCR for quantitative analysis of gene expression. Results : Results of fluorocytometry were positive for CD90/Thy1 and negative for CD11 and CD45. TGF-${\beta}3$-mediated induction of ADMSCs led to the expression of the differentiation markers of intervertebral disc-like cells, such as aggrecan, collagen II, and sox-9. Co-cultured ADMSCs treated with TGF-${\beta}3$ showed higher expression of differentiation markers and greater extracellular matrix production compared with ADMSCs treated with TGF-${\beta}3$ alone. Conclusion : ADMSC treated with TGF-${\beta}3$ may be an attractive source for regeneration therapy in degenerative IVD. These findings may also help elucidate the pathologic mechanism of MSC therapy in the degeneration of IVD in vivo.

Accuracy of several implant bite registration techniques: an in-vitro pilot study

  • PURPOSE. This study evaluated the accuracies of different bite registration techniques for implant-fixed prostheses using three dimensional file analysis. MATERIALS AND METHODS. Implant fixtures were placed on the mandibular right second premolar, and the first and second molar in a polyurethane model. Aluwax (A), Pattern Resin (P), and Blu-Mousse (B) were used as the bite registration materials on the healing abutments (H) or temporary abutments (T). The groups were classified into HA, HP, HB, TA, TP, and TB according to each combination. The group using the bite impression coping was the BC group; impression taking and bite registration were performed simultaneously. After impression and bite taking, the scan bodies were connected to the lab analogs of the casts. These casts were scanned using a model scanner. The distances between two reference points in three-dimensional files were measured in each group. One-way ANOVA and Duncan's test were used at the 5% significance level. RESULTS. The smallest distance discrepancy was observed in the TB group using the temporary abutments. The Blu-Mousse and HP groups showed the largest distance discrepancy. The TB and BC groups showed a lower distance discrepancy than the HP group (P=.001), and there was no significant difference between the groups using the temporary abutments and healing abutments (P>.05). CONCLUSION. Although this study has limitations as an in-vitro investigation, the groups using the temporary abutments to hold the Blu-Mousse record and bite impression coping showed greater accuracy than the group using the healing abutments to hold the pattern resin record.

Literature Review of Tool-based Manipulation for Musculoskeletal Diseases-with Focus on Guasha and IASTM (근골격계 질환에서 도구를 이용한 수기요법의 연구 동향 고찰: 괄사요법과 IASTM을 중심으로)

  • Objectives Our goal was to review literatures on tool-based manipulation for musculoskeletal diseases with emphasis on guasha and IASTM (Instrument Assisted Soft Tissue Manipulation) by searching foreign and domestic controlled trials. Methods We found literatures published up to August 2016 in electronic databases (OASIS, KoreaMed, KMbase, NDSL, RISS, KISS, MEDLINE/PubMed, EMBASE, Cochrane Central Register of Controlled Trials) without any limitations on language. Results 17 studies were included in the review. 13 out of 17 articles were published within this decade. VAS was the main evaluation tool for most of the articles (14 out of 17). 10 studies showed statistically significant difference between the experimental group and the control group. In terms of materials used for treatment, 6 out of 17 studies used stainless steel. Considering anatomical locations for treatment, trunk area was treated the most, accounting for 7 out of 17 studies. Conclusions Our results show that recently, research on tool-based manipulation treatments for musculoskeletal diseases are actively being performed. Diseases of the vertebral column were the main target for tool-based manipulation treatments, and stainless steel was the most popular and safe material used. Further research should be performed for more accurate data.
