Ginseng seed oil ameliorates hepatic lipid accumulation in vitro and in vivo

  • Background: Despite the large number of studies on ginseng, pharmacological activities of ginseng seed oil (GSO) have not been established. GSO is rich in unsaturated fatty acids, mostly oleic and linoleic acids. Unsaturated fatty acids are known to exert a therapeutic effect in nonalcoholic fatty liver disease (NAFLD). In this study, we investigated the protective effect and underlying mechanisms of GSO against NAFLD using in vitro and in vivo models. Methods: In vitro lipid accumulation was induced by free fatty acid mixture in HepG2 cells and by 3 wk of high fat diet (HFD)-feeding in Sprague-Dawley rats prior to hepatocyte isolation. The effects of GSO against diet-induced hepatic steatosis were further examined in C57BL/6J mice fed a HFD for 12 wk. Results: Oil Red O staining and intracellular triglyceride levels showed marked accumulation of lipid droplets in both HepG2 cells and rat hepatocytes, and these were attenuated by GSO treatment. In HFD-fed mice, GSO improved HFD-induced dyslipidemia and hepatic insulin resistance. Increased hepatic lipid contents were observed in HFD-fed mice and it was lowered in GSO (500 mg/kg)-treated mice by 26.4% which was evident in histological analysis. Pathway analysis of hepatic global gene expression indicated that GSO increased the expression of genes associated with ${\beta}$-oxidation (Ppara, Ppargc1a, Sirt1, and Cpt1a) and decreased the expression of lipogenic genes (Srebf1 and Mlxipl), and these were confirmed with reverse transcription and quantitative polymerase-chain reaction. Conclusion: These findings suggest that GSO has a beneficial effect on NAFLD through the suppression of lipogenesis and stimulation of fatty acid degradation pathway.

Cellulose Digestibility Increased with CelD Transgenic Pigs

  • This study was performed to test the cellulose digestibility using the transgenic pigs harboring cellulose degradation gene D (CelD). After delivered offsprings between normal pig and transgenic swine, DNA was isolated from piglets tail for PCR analysis. In first generation, five out of 65 piglets showed CelD positive. Unfortunately, four CelD-positive pigs were died during growing, but one survived pig was used as a transgenic founder to produce F$_1$ descendents. Among 3 F$_1$ transgenic pigs produced, one died and the remaining two pigs were used to test the fiber digest efficiency. An assorted feed was composite of 5% fiber with other ingredients. The feed of 3 kg per day was provided to the pigs including transgenic founders and littermate controls. The manure quantity was measured daily for a month, and all manures were dried for three days to analysis nitrogen, phosphate and fiber concentrations. The fiber digestion efficiencies of the transgenic F$_1$ pigs showed approximately 10% higher than those of control pigs. Fiber digestion was not greatly improved in transgenic pigs as it had been expected approximately 30%. Nitrogen concentration of transgenic pig's manure was slowly decreased compare to the control pigs. Because there were only two transgenic pigs tested, a large number of transgenic pigs may be necessary to obtain more reliable data. Breeding of animals to obtain sufficient transgenic pigs subjected for a further study is on progress. Taken together, this study demonstrated successful production of transgenic pigs with increase of cellulose digestibility in the porcine feed.

  • 본 연구에서는 반복적인 온도변화가 목분(50 wt.%와 70 wt.%)이 첨가된 폴리프로필렌 WPC(Wood Plastic Composites)의 기계적 특성에 미치는 영향을 조사하였다. WPC의 휨탄성계수(flexural modulus)와 휨강도 (flexural strength)는 반복 회수에 상관없이 동결 융해 시험에서 계면접착력의 약화 때문에 감소하는 경향을 보였다. 목분의 함량이 높을 때, 휨탄성계수의 감소가 비교적 높았다. WPC의 휨탄성계수와 휨강도는 고온($60^{\circ}C$) 저온 ($-20^{\circ}C$) 반복시험 후 고온에서 감소하고 저온에서 증가되었다. 폴리프로필렌(polypropylene, PP)의 유리전이점 (glass transition temperature: $-10^{\circ}C$) 보다 낮은 저온($-20^{\circ}C$에서 WPC는 높은 강성(stiffness)과 강도 (strength)를 유발시키는 유리상태(glassy state)로 존재한다. 고온에서 목분의 함량이 낮은 WPC가 연성의 증가 때문에 낮은 휨탄성계수와 휨강도를 보였다.

Sulfonylurea계 제초제가 처리된 다년생 잡초의 재생양상과 저장전분량의 변화 (Regrowth Patterns of the Perennial Weeds Treated with Sulfonylureas and Starch Degradation in the Rhizome)

  • Sulfonylurea계 제초제 pyrazosulfuron-ethyl과 bensulfuron-methyl을 대상으로 다년생 잡초의 재생특성과 저장전분의 변화 양상을 조사하였다. Pyrazosulfuron-ethyl을 처리하였을 때 너도방동사니가 24일 후 재생되어 5가지 다년생 잡초 중 가장 빠르게 재생하였으며, 올방개, 올미, 벗풀 등이 각각 $30{\sim}39$일 이후부터 재생하였고, 가래는 55일 이후 재생되었다. 너도방동사니의 마디의 수와 엽기별 처리에 따른 재생력의 차이에서는 bensulfuron-methyl에 대하여 4마디 식물체가 2마디 식물체보다 약 2배의 재생력을 보였고, 처리시기가 2엽기, 3엽기, 4엽기로 늦어질수록 재생력이 증가하였다. 크기에 따른 올방개의 재생력은 $0.3{\sim}0.5g$ 크기의 올방개보다 $1{\sim}1.5g$ 크기의 올방개가 bensulfuron-methyl에 대하여 재생력이 강한 것으로 나타났다. 처리시기에 따른 올방개의 재생력은 2엽기 처리보다 3엽기 처리에서 재생율이 감소되었다. 모경에 대한 의존기간은 벗풀의 경우 맹아된 후 12일이며, 올방개는 18일로 나타났다. 정상적으로 생장할 경우 영양번식체 내의 전분이 고갈되는 시기는 벗풀의 경우 맹아 후 18일, 올방개의 경우 27일로 나타났으며, 유리당의 최고치는 벗풀의 경우 맹아 후 9일, 올방개의 경우 18일로 나타났다. 올방개 괴경은 벗풀 괴경보다 1.5배 많은 전분을 함유하고있었다.

오미자 색소 추출물의 가열 변색에 대한 속도론적 연구 (Kinetic Study on the Color Deterioration of Crude Anthocyanin Extract from Schizandra Fruit (Schizandra chinensis fructus))

  • 건강기능성 식품소재로써 각광을 받고 있는 오미자의 특징적인 붉은 색소인 anthocyanin의 가열 변색에 대한 안정성을 연구하고자 오미자 조추출물(crude pigment extracts from Schizandra fructus)을 색소원으로 하여 가열 변색에 대한 지표로서 browning index(BI)를 이용하여 $80{\sim}100^{\circ}C$에서 kinetic parameter를 조사하였다. 그 결과 pH 2.0, 3.0, 4.0, 5.0 범위에서 pH가 높을수록 가열 변색 인자인 BI값이 감소하여 높은 pH에서 가열에 의한 변색이 활발하게 이루어졌음을 시사하였다. 지표인자인 BI와 가열시간의 관계를 분석한 결과 가열변색 반응은 2차 반응을 따랐으며 qinoidal base가 우세해지는 pH 5.0 구간을 제외하고는 pH가 높아짐에 따라 반응 속도상수(k)는 증가하여 높은 pH에서 가열 변색이 빠르게 이루어지고 있음을 나타내었다. Arrhenius식에 의해 계산된 오미자 anthocyanin 색소의 활성화 에너지(Ea)는 $24.87{\sim}42.54\;kJ/mol$이었으며, pH가 높아짐에 따라 활성화 에너지(Ea)값이 증가하여 온도 의존성이 커지는 것을 알 수 있었다. 또한 각 pH조건에서의 지수앞인자($k_0$)와 활성화 에너지(Ea)가 직선 상관관계를 보여 시험한 pH 범위에서 오미자 anthocyanin 색소의 가열 변색 기작은 모두 같은 것으로 나타났다.

Effects of Addition Level and Chemical Type of Propionate Precursors in Dicarboxylic Acid Pathway on Fermentation Characteristics and Methane Production by Rumen Microbes In vitro

  • Two in vitro experiments were conducted to examine the effects of propionate precursors in the dicarboxylic acid pathway on ruminal fermentatation characteristics, $CH_4$ production and degradation of feed by rumen microbes. Fumarate or malate as sodium salts (Exp. 1) or acid type (Exp. 2) were added to the culture solution (150 ml, 50% strained rumen fluid and 50% artificial saliva) to achieve final concentrations of 0, 8, 16 and 24 mM, and incubated anaerobically for 0, 1, 3, 6, 9 and 12 h at $39^{\circ}C$. For both experiments, two grams of feed consisting of 70% concentrate and 30% ground alfalfa (DM basis) were prepared in a nylon bag, and were placed in a bottle containing the culture solution. Addition of fumarate or malate in both sodium salt and acid form increased (p<0.0001) pH of culture solution at 3, 6, 9 and 12 h incubations. The pH (p<0.0001) and total volatile fatty acids (VFA, p<0.05) were enhanced by these precursors as sodium salt at 3, 6 and 9 h incubations, and pH (p<0.001) and total VFA (p<0.01) from fumarate or malate in acid form were enhanced at a late stage of fermentation (9 h and 12 h) as the addition level increased. pH was higher (p<0.001) for fumarate than for malate as sodium salt at 3 h and 6 h incubations. Propionate ($C_3$) proportion was increased (p<0.0001) but those of $C_2$ (p<0.05) and $C_4$ (p<0.01 - p<0.001) were reduced by the addition of sodium salt precursors from 3 h to 12 incubation times while both precursors in acid form enhanced (p<0.011 - p<0.0001) proportion of $C_3$ from 6h but reduced (p<0.018 - p<0.0005) $C_4$ proportion at incubation times of 1, 3, 9 and 12 h. Proportion of $C_3$ was increased (p<0.05 - p<0.0001) at all incubation times by both precursors as sodium salt while that of $C_3$ was increased (p<0.001) from 6h but $C_4$ proportion was decreased by both precursors in acid form as the addition level increased. Proportion of $C_3$ was higher (p<0.01 - p<0.001) for fumarate than malate as sodium salt from 6 h incubation but was higher for malate than fumarate in acid form at 9 h (p<0.05) and 12 h (p<0.01) incubation times. Increased levels (16 and 24 mM) of fumarate or malate as sodium salt (p<0.017) and both precursors in acid form (p<0.028) increased the total gas production, but no differences were found between precursors in both chemical types. Propionate precursors in both chemical types clearly reduced (p<0.0001 - p<0.0002) $CH_4$ production, and the reduction (p<0.001 - p<0.0001) was dose dependent as the addition level of precursors increased. The $CH_4$ generated was smaller (p<0.01 - p<0.0001) for fumarate than for malate in both chemical types. Addition of fumarate or malate as sodium type reduced (p<0.004) dry matter degradation while both precursors in both chemical types slightly increased neutral detergent fiber degradability of feed in the nylon bag.

  • Photovoltaic (PV) module can generate electricity using sunlight without causing any environmental degradation. Due to higher fossil fuel prices and environmental awareness, PV applications are becoming more popular as clean source of electricity generation. PV output is sensitive to the operating temperature and can be drastically affected in Building Integrated PV (BIPV) systems. PV Solarwall (SWPV) combination and PV systems have been evaluated in this study for improvement in electrical output and system costs. PV modules under forced ventilation. A 75W polycrystalline silicon PV module was fixed on SW in front of the ventilation fan as it was indicated to be the coolest position on the SW in phoenix simulations. The effectiveness of cooling by means of the forced ventilating air stream has been studied experimentally. The results indicate that there appears to be significant difference in temperature as well as electricity output comparing the SWPV and BIPV options. Electrical output power recovered is about $4\%$ during the typical day of the month of February. RETScreen(R) analysis of a 3kW PV system hypothetically located at Taegu has shown that with typical temperature reduction of $15^{{\circ}C$, it is possible to reduce the simple payback periods by one year. The work described in this paper may be viewed as an appraisal of a SWPV system based on its improved electrical and financial performances due to its ability to operate at relatively lower temperatures.

Monitoring the Bacterial Community Dynamics in a Petroleum Refinery Wastewater Membrane Bioreactor Fed with a High Phenolic Load

  • The phenolic compounds are a major contaminant class often found in industrial wastewaters and the biological treatment is an alternative tool commonly employed for their removal. In this sense, monitoring microbial community dynamics is crucial for a successful wastewater treatment. This work aimed to monitor the structure and activity of the bacterial community during the operation of a laboratory-scale continuous submerged membrane bioreactor (SMBR), using PCR and RT-PCR followed by denaturing gradient gel electrophoresis (DGGE) and 16S rRNA libraries. Multivariate analyses carried out using DGGE profiles showed significant changes in the total and metabolically active dominant community members during the 4-week treatment period, explained mainly by phenol and ammonium input. Gene libraries were assembled using 16S rDNA and 16S rRNA PCR products from the fourth week of treatment. Sequencing and phylogenetic analyses of clones from the 16S rDNA library revealed a high diversity of taxa for the total bacterial community, with predominance of Thauera genus (ca. 50%). On the other hand, a lower diversity was found for metabolically active bacteria, which were mostly represented by members of Betaproteobacteria (Thauera and Comamonas), suggesting that these groups have a relevant role in the phenol degradation during the final phase of the SMBR operation.

칼날형 마모시험기를 이용한 C/B충전 NR 배합고무의 마모거동 (Wear Behavior of C/B filled NR Compounds using a Blade-type Abrader)

  • 칼날형 마모 마찰 시험기를 이용하여 NR배합고무의 마모 거동을 조사하였다. 온도, 하중, 그리고 회전속도를 변화시켜 마모속도에 미치는 영향과 열화된 배합고무의 마모 현상에 대해 평가하였다. 시편의 회전속도와 수직 하중이 증가할수록 마모속도가 증가하였다. 실험온도가 증가할수록 마찰계수가 감소하였고, 마모속도도 감소하였다. 마찰에너지와 마모속도 사이에 Power-Law 관계를 나타냈으며 마모속도는 열화에 의해 급격히 증가하였다. 시편의 마모패턴은 마모조건에 의해 영향을 받았으며 특히 수직 하중을 변화시켰을 때 마모융기(ridge)간격에 큰 변화를 보였다. 배합고무의 마모속도를 결정하는데 간헐적 마모손실 무게측정 대신 칼날형 마모시험기를 이용한 연속적인 마모이동거리측정으로 대체할 수 있음을 확인하였다.

$Co^{60}-{\gamma}$ ray을 조사시킨 MOS 구조에서의 I-V특성의 방사선 조사 효과 (Radiation effects of I-V characteristics in MOS structure irradiated under $Co^{60}-{\gamma}$ ray)

  • MOS 커패시터가 이온화 방사선에 노출되었을 경우, MOS 커패시터의 방사선 조사 효과는 소자의 전기적 특성 및 동작 수명에 심각한 영향을 일으킬 수 있다. MOS 커패시터는 (100) 방향의 P-type Si wafer 위에 산화막층을 $O^2$+T.C.E. 분위기에서 성장하였으며, 그 두께는 40~80 nm로 만들었다. MOS 커패시터에 대한 방사선 조사는 $Co^{60}-{\gamma}$선을 사용하였고, 조사선량은 $10^4{\sim}10^8$으로 조사하였다. MOS 커패시터에서 전기적 전도 특성의 방사선 조사효과는 산화막 두께와 조사선량을 변화하면서 측정된 P-type MOS 커패시터는 조사선량에 의해서 강하게 영향을 받는다는 것과 저전계 영역에서의 Ohmic 전류가 전체 선량에 의존한다는 것을 알았다. 이 결과는 방사선 조사에 의해 산화막 트랩전하와 산화막-반도체($SiO_2$-Si)계면 트랩전하에 의해서 설명 할 수 있다.
