  • Salmon calcitonin (sCT) is a therapeutic polypeptide hormone consisting of 32 amino acids (3432 Da). As with other bioactive peptide therapeutics, however, therapeutic use of sCT has been limited due to the problems of short circulating half-life and rapid proteolytic degradation. To get over this problem, the three positional isomers of mono-PEGylated sCT were prepared and among these, the best drug candiate for nasal application was chosen. (omitted)

  • Low velocity impact test and compressive residual strength test after impact were performed by using Hercules AS4/3501-6[45/0/-45/90]$_{2s}$ laminated plate to investigate the low velocity impact damage behavior and the post-impact strength degradation on orthotropic composite laminate plate. Due to the lateral impact losd, the load path showed "" shape according to the laminate central deflection. Damage in a laminate occurs by inclined matrix crack at the damage initiation load stage and vertical matrix crack, occurs on the outer surface. Evaluating the compressive residual strength after the low velocty impact test, it could be found that there is a transient range where the compressive residual strength drop suddenly in the initial damage which is in the matrix crack range and the initial delamination area. is in the matrix crack range and the initial delamination area.

Process Characteristics for $YB_{2}Cu_{3}O_{7-d}$ Films Fabricated by Single Target Sputter and Surface Modification Technique

  • Thin films of $YB_{2}Cu_{3}O_{7-d}$ were prepared on various substrated of MgO(100), $SrTiO_{3}$, and $LaAlO_{3}$ by using off-axis magentron sputtering methods and annealing in-situ. The prarameters of film fabrication processes had been optimized through a "follow the lcoal maxima" strategy to yield good quality films in therms of the critical temperature $T_{c}$ and the critical current density $J_{c}$. Optimizedproecsses employing a plane magndtron and an cylindrical magnetron yielded $T_{c}$>90K along with $J_{c}$$10^{6}$A/$\textrm{cm}^2$ at 77K and > 2${\times}$$10^{7}$A/$\textrm{cm}^2$ at 5K. The sampels, however, showed degradationinthe properties, after chemical etching for fabrication of microbridges with the line width of 2-10 mocrons. In particular, the value of $T_{c}$ for the microbridges of 2microns was as small as 80%. The degradation was strongly dependent on the line width through a formula : $T_{c}$(e)=$T_{c}$)b) [1-a exp(-1000 bL)} where $T_{c}$(e) and $T_{c}$ (b) are the values of $T_{c}$ in the absolute scale measured after and before chemical etching, respectively and L is the line width in mm. By utilizing a best fitting technique, the proper constant values of a and to b were found as exp(-1.2) and 0.22, respectively. This formula was very useful in estimatiing the upper limit of the device operationtemperature.

  • 22.9[kV]/50[MVA]/30[m] HTS transmission power cable has been developed and tested at Korea Electrotechnology Research Institute and LG Cable Company by 21 century center for applied superconductivity technology. It is necessary to measure of critical current degradations, AC loss, insulation test and etc at the HTS cable development. This paper is analyzed characteristics that critical current of HTS cable bending condition according to this paper. We will be able to decide the diameter of drum which HTS cable is wound around and minimum curvature radius of HTS cable from results of this research.

20(S)-ginsenoside Rh2 induces caspase-dependent promyelocytic leukemia-retinoic acid receptor A degradation in NB4 cells via Akt/Bax/caspase9 and TNF-α/caspase8 signaling cascades

  • Background: Acute promyelocytic leukemia (APL) is a hematopoietic malignancy driven by promyelocytic leukemia-retinoic acid receptor A (PML-RARA) fusion gene. The therapeutic drugs currently used to treat APL have adverse effects. 20(S)-ginsenoside Rh2 (GRh2) is an anticancer medicine with high effectiveness and low toxicity. However, the underlying anticancer mechanisms of GRh2-induced PML-RARA degradation and apoptosis in human APL cell line (NB4 cells) remain unclear. Methods: Apoptosis-related indicators and PML-RARA expression were determined to investigate the effect of GRh2 on NB4 cells. Z-VAD-FMK, LY294002, and C 87, as inhibitors of caspase, and the phosphatidylinositol 3-kinase (PI3K) and tumor necrosis factor-α (TNF-α) pathways were used to clarify the relationship between GRh2-induced apoptosis and PML-RARA degradation. Results: GRh2 dose- and time-dependently decreased NB4 cell viability. GRh2-induced apoptosis, cell cycle arrest, and caspase3, caspase8, and caspase9 activation in NB4 cells after a 12-hour treatment. GRh2-induced apoptosis in NB4 cells was accompanied by massive production of reactive oxygen species, mitochondrial damage and upregulated Bax/Bcl-2 expression. GRh2 also induced PML/PML-RARA degradation, PML nuclear bodies formation, and activation of the downstream p53 pathway in NB4 cells. Z-VAD-FMK inhibited caspase activation and significantly reversed GRh2-induced apoptosis and PML-RARA degradation. GRh2 also upregulated TNF-α expression and inhibited Akt phosphorylation. LY294002, an inhibitor of the PI3K pathway, enhanced the antitumor effects of GRh2, and C 87, an inhibitor of the TNF-α pathway, reversed NB4 cell viability, and GRh2-mediated apoptosis in a caspase-8-dependent manner. Conclusion: GRh2 induced caspase-dependent PML-RARA degradation and apoptosis in NB4 cells via the Akt/Bax/caspase9 and TNF-α/caspase8 pathways.

Effects of steaming on saponin compositions and antiproliferative activity of Vietnamese ginseng

  • Background: Steaming of ginseng is known to change its chemical composition and biological activity. This study was carried out to investigate the effect of different steaming time-scales on chemical constituents and antiproliferative activity of Vietnamese ginseng (VG). Methods: VG was steamed at $105^{\circ}C$ for 2-20 h. Its saponin constituents and antiproliferative activity were studied. The similarity of chemical compositions between steamed samples at $105^{\circ}C$ and $120^{\circ}C$ were compared. Results: Most protopanaxadiol and protopanaxatriol ginsenosides lost the sugar moiety at the C-20 position with 10-14 h steaming at $105^{\circ}C$ and changed to their less polar analogues. However, ocotillol (OCT) ginsenosides were reasonably stable to steaming process. Antiproliferative activity against A549 lung cancer cells was increased on steaming and reached its plateau after 12 h steaming. Conclusion: Steaming VG at $105^{\circ}C$ showed a similar tendency of chemical degradation to the steaming VG at $120^{\circ}C$ except the slower rate of reaction. Its rate was about one-third of the steaming at $120^{\circ}C$.

주사슬에 곁사슬기를 갖는 폴리히드록시아미드의 물성 및 난연특성 (Physical Properties and Flame Retardency of Polyhydroxyamides (PHAs) Having Pendant Groups in the Main Chain)

  • 폴리(에틸렌글리콜)메틸에테르[poly (ethyleneglycol)nlethyl ether, MPEG] 곁사슬기와 짧고 강직한 디메틸페녹시(dimethylphenoxy) 곁사슬기를 갖는 폴리히드록시아미드(poly (hydroxyamide)s, PHAs)의 물성 및 난연특성을 DSC, TGA, FTIR, pyrolysis combustion flow calorimeter(PCFC), X-ray diffractometer를 사용하여 조사하였다. 중합체들의 최대분해온도는 공기 분위기하에서 $276{\sim}396^{\circ}C$의 범위를 보였다. PHAs의 heat release (HR) capacity와 total heat release (total HR) 값들은 MPEG의 분자량 증가에 따라 증가됨을 보였다. $290^{\circ}C$에서 열처리된 M-PHA 2의 경우 열처리 시간에 따라서 HR capacity 값들은 253 J/gK에서 42 J/gK로 감소하였고, 60% 중량 감소 온도는 $408^{\circ}C$에서 $856^{\circ}C$로 증가하였다. PHAs의 분해 활성화 에너지는 $129.3{\sim}235.1kJ/mol$의 범위를 보이고, 전환율에 따라 증가하였다. PHAs의 인장 모듈러스는 MPEG의 사슬길이가 증가함에 따라 감소하였으며, PBO로 전환된 후에는 초기 모듈러스보다 더 상승하였다.

Isolation, Screening and Identification of Swine Gut Microbiota with Ochratoxin A Biodegradation Ability

  • The potential for ochratoxin A (OTA) degradation by swine intestinal microbiota was assessed in the current study. Intestinal content that was collected aseptically from swine was spiked with 100 ppb OTA and incubated for 6 and 12 h at $39^{\circ}C$. An OTA assay was conducted using the incubated samples, and it was found that 20% of the OTA toxin was detoxified, indicating the presence of microbes capable of OTA degradation. Twenty-eight bacterial species were isolated anaerobically in M 98-5 media and 45 bacterial species were isolated using nutrient broth aerobically. Screening results showed that one anaerobic bacterial isolate, named MM11, detoxified more than 75% of OTA in liquid media. Furthermore, 1.0 ppm OTA was degraded completely after 24 h incubation on a solid 'corn' substrate. The bacterium was identified by 16S rDNA sequencing as having 97% sequence similarity with Eubacterium biforme. The isolation of an OTA-degrading bacterium from the swine natural flora is of great importance for OTA biodegradation and may be a valuable potential source for OTA-degradation enzymes in industrial applications.

  • Modified 9Cr-1Mo steels possess excellent high-temperature mechanical properties and are widely used in energy conversion industries. However, in-service materials degradation, such as softening, carbide-induced embrittlement, temper embrittlement, etc., can take place during long-term operation. Evolution of microstructure due to service exposure to high temperature has a strong effect on the performance of heat resistant steels. In case of modified 9Cr-1Mo steels, precipitation of $Fe_2Mo$-type laves phases and coarsening of $M_{23}C_6$-type carbides are the primary cause of degradation of mechanical properties such as toughness, hardness, tensile strength and creep resistance. This study was aimed at finding reliable parameter for assessing the integrity of modified 9Cr-1Mo steels. Characteristic parameters were attained between mechanical and ultrasonic properties.

High energy swift heavy ion irradiation and annealing effects on DC electrical characteristics of 200 GHz SiGe HBTs

  • The total ionizing dose (TID) and non ionizing energy loss (NIEL) effects of 100 MeV phosphorous ($P^{7+}$) and 80 MeV nitrogen ($N^{6+}$) ions on 200 GHz silicon-germanium heterojunction bipolar transistors (SiGe HBTs) were examined in the total dose range from 1 to 100 Mrad(Si). The in-situ I-V characteristics like Gummel characteristics, excess base current (${\Delta}I_B$), net oxide trapped charge ($N_{OX}$), current gain ($h_{FE}$), avalanche multiplication (M-1), neutral base recombination (NBR) and output characteristics ($I_C-V_{CE}$) were analysed before and after irradiation. The significant degradation in device parameters was observed after $100MeV\;P^{7+}$ and $80MeV\;N^{6+}$ ion irradiation. The $100MeV\;P^{7+}$ ions create more damage in the SiGe HBT structure and in turn degrade the electrical characteristics of SiGe HBTs more when compared to $80MeV\;N^{6+}$. The SiGe HBTs irradiated up to 100 Mrad of total dose were annealed from $50^{\circ}C$ to $400^{\circ}C$ in different steps for 30 min duration in order to study the recovery of electrical characteristics. The recovery factors (RFs) are employed to analyse the contribution of room temperature and isochronal annealing in total recovery.
