KINETICS OF AUTOTROPHIC DENITRIFICATION FOR THE BIOFILM FORMED ON SULFUR PARTICLES : Evaluation of Molecular Technique on Monitoring Biomass Growth

  • Characteristics of sulfur-based autotrophic denitrification in a semi-continuous type reactor and the kinetic parameters were studied. Enriched autotrophic denitrifying culture was used for the reactor operation. Biomass growth on sulfur particles and in the liquid medium was monitored using the DAPI staining method. From the result of ion concentration changes and the biomass growth, maximum specific growth rate, ${\mu}_{max}$, and the half velocity constant, $K_M$, were estimated as $0.61\;d^{-1}$ and 3.66 mg/L, respectively. Growth yield coefficient, Y values for electron acceptor and donor were found as 0.49 gVSS/g N and 0.16 gVSS/g S. The biomass showed specific denitrification rate, ranging 0.86-1.13 gN/g VSS-d. A half-order equation was found to best simulate the denitrification process in the packed bed reactor operated in the semi-continuous mode.

Effects of the Combined-Preparation of Crude Drugs on Pentobarbital-induced Sleeping Time (복합한약제제가 Pentobarbital에 의해 유도된 수면시간에 미치는 영향)

  • This experiment was performed to investigate whether 50% ethanol extracts of the combined-preparation of Longanae Arilus, Chrysanthemi Flos, Zizyphi Fructus and Ginseng Radix alba (CPE) has hypnotic effects and/or enhances pentobarbital-induced sleeping time. Locomotor activity was evaluated using a ambulometer of tilting-type. The sedative-hypnotic effects were evaluated by measuring the sleeping onset time and sleeping time in pentobarbital-treated mice 30 min. after oral administration of CPE and muscimol. The intracellular $Cl^-$ concentration of cerebellar granule cells was estimated using $Cl^-$ sensitive fluorescence probe N-(ethoxycarbonylmethyl)-6-methoxyquinolinium (MQAE). CPE (150 mg/kg) decreased the locomotor activity, but CPE itself did not induce sleep. However, CPE reduced sleeping onset and prolonged sleeping time induced by pentobarbital (42 mg/kg). In addition, CPE (2 ${\mu}g/ml$) and pentobarbital (2.5 ${\mu}M$) itself did not affect on the chloride influx in primary cultured cerebellar granule cells, but the combination of CPE and pentobarbital (2.5 ${\mu}M$) increased the chloride influx onto the cells. In conclusion, it is suggested that CPE might augment pentobarbital-induced sleep through the increase of chloride influx.

Determination of agrochemical residues in tobacco using matrix solid-phase dispersion and GC/MS

  • A matrix solid-phase dispersion (MSPD) method was developed for extracting and cleaning-up the selected agrochemicals in tobacco using gas chromatography-mass spectrometry with selected ion monitoring (GC/MS-SIM). Different parameters of the method were investigated and optimized, such as the type of solid-phase (alumina, $C_{18}$ and Florisil) and eluent (acetone, acetonitrile, ethylacetate and n-hexane). The best results were obtained using 0.5 g of tobacco sample, 1.0 g of $C_{18}$ as dispersant sorbent, 1.0 g of Florisil as clean-up sorbent and acetonitile saturated with n-hexane as eluting solvent. The method was validated using tobacco samples fortified with agrochemicals at their different concentration levels. This method gave good linearity for the selected agrochemicals of ranging from $0.01{\mu}g/mL$ to $0.1{\mu}g/mL$. Recoveries of the selected agrochemicals in tobacco were more than 80 % and reproducibilities were found to be better than 10 % RSD. Those results suggested that the analytical procedure including MSPD method in combining with GC/MS could be applicable to the rapid determination often the selected agrochemicals in tobacco.

Biochemical and Immunological Characterization of the DNA Polymerase and RNase H in Feline Leukemia Virus (고양이 백혈병 바이러스의 DNA Porymerase와 RNase H의 생화학적 및 면역학적 연구)

  • Feline leukemia virus DNA polymerase was purified by ion-exchange and nucleic acid affinity chromatographies. The enzyme consists of a single polypeptide chain of approximately 72, 000 molecular weight as determined by both of a glycerol density gradient centrifugation and SDS-polyacrylamide gel electrophoresis. The preferred divalent cation for DNA synthesis is $Mn^2+$ on a variety of template-primers, and its optimum concentration appears to be significantly lower than reported results of other mammalian type-C viral enzymes. The divalent cation requirement for maximum activity of RNase H is similar to those of DNA polymerase. Both DNA polymerase and RNase H activities appear to reside on the same molecule as demonstrated by the copurification of both activities through various purification steps. An additional RNase H without detectible polymerase activity was generated by a limited chymotrypsin digestion. This RNase H activity was inhibited equally effectively as RNase H in the intact reverse transcriptase by antisera prepared against reverse transcriptase of feline leukemia virus. Neutralization and binding test showed that antibody binding to reverse transcriptase molecule did not completely inhibit the polymerase activity.

Isolation and Determination of Alkaloids in Evodiae Fructus by forming Complex Compound (착 화합물 생성에 의한 오수유 중 알칼로이드의 분리와 정량)

  • The fruit of Evodia officinalis (Rutaceae) is known for containing a number of indoloquinazoline and quinoline type alkaloids. Evodiamine, evocarpine and rutaecarpine are the major constituents of Evodiae Fructus. These alkaloids were isolated and determined by forming complex compounds from Evodiea Fructus. For the determination of these alkaloids, a new spectrophotometric method was developed with a simple and selective sample clean-up using thiocyanatocobaltate [II] complex ion. The absorbance of alkaloidal complex in 1.2-dichloroethane solution was measured at 625 nm. A calibration curve for the alkaloids isolated from Evodia Fruit was linear over the concentration range of $1.0{\sim}6.0\;mg/ml$. The method proved to be rapid, simple and reliable for the isolation and the determination of the alkaloids in Evodiae Fructus.

Interaction of NpO+2 with Cl- in Na-Ca-Cl-type solutions at ionic strength of 6M: Effect of presence of Ca ion on interaction

  • The interaction of $NpO^+_2$ with $Cl^-$ was studied using visible-near-infrared spectroscopy in $NaCl-Ca-Cl_2-NaClO_4$, $NaCl-NaClO_4$, and $CaCl_2-NaClO_4$ solutions with ionic strength (I) of 6M. The spectra of $NpO^+_2$ around 980 nm varied with $Cl^-$ concentration in the $NaCl-CaCl_2-NaClO_4$ and $NaCl-NaClO_4$ solutions at [$Cl^-$] ${\geq}3.5M$, but not in the $CaCl_2-NaClO_4$ solution. Assuming the 1:1 interaction between $NpO^+_2$ and $Cl^-$, the apparent equilibrium constants at I = 6M were evaluated. The presence of $Ca^{2+}$ was found to destabilize overall interaction between $NpO^+_2$ and $Cl^-$. The observations were consistent with the density functional theory calculation.

A small-scale membrane electro-dialyser for domestic use

  • A small-scale electro-dialysis system was constructed for domestic use. It is composed of six compartments in which five special polystyrene ionic membranes are housed. A series of experiments on the transport of sodium and chloride ions through polystyrene membranes was performed and the effects of electric current and voltage on the pH of water were investigated. This electrodialyser could reduce the NaCl content to an acceptable level (5307 mg/L) when water containing 9945 mg/L of sodium chloride is fed to the electrodialyser. The reduction was by the action of direct current 60 mA/100 mA when a 15 V / 20 V potential is maintained across the membrane. The results showed that the pH of the treated water attained a value in the range of 7-8, with the chloride concentration of 5307 mg/L when the voltage was in the range of 20 volts. This was achieved when two of the small-scale electro-dialysers were placed in series and the solutions from the respective compartments were mixed. This is considered useful because this complies to the requirement of drinking water standard both in terms of chloride and pH. Therefore, this type electrodialyserhas the potential for domestic uses in isolated houses where potable water supply is not available.

  • The effects of additives on the Zn electrodeposition in chloride-based electrolyte were investigated using circulation cell with three electrodes system. The cathodic polarization increased with the addition of polyethylenglycol (hereafter PEG) in electrolyte. This was attributed to the adsorption of the additives on the electrode and the inhibition of migration of metal ion. The PEG, however, did not have any noticeable effect on the properties of plating solutions at the concentration used. The effect of PEG on the electrocrystallization was related to its molecular weight. With the increase of molecular weight, the cathodic polarization increased, while the surface roughness was improved with the decrease of brightness. Especially, the PEG mixed with different molecular weights was the most effective. The orientation and the type of the deposited grains were changed and refined by PEG, which resulted in the modification of deposited surface roughness and brightness.

  • We have investigated the effect of $H_2$ plasma treatment on the BZO (ZnO:B, Boron doped ZnO) thin films. The BZO thin films are prepared by LP-MOCVD (Low Pressure Metal Organic Chemical Vapor Deposition) technique and the samples of BZO thin film are performed with $H_2$ plasma treatment by plasma treatment system with 13.56 MHz as RIE (Reactive Ion Etching) type. After exposing $H_2$ plasma treatment, measurement of transmittance, reflectance and haze spectra in 300~1100 nm, electrical properties as resistivity, mobility and carrier concentration and work function was analysed. Regarding the results of the $H_2$ plasma treatment on the BZO thin films are application to the TCO for solar cells, such as the a-Si thin films solar cell.

Application of Organized Media for Rapid Spectrofluorimetric Determination of Trace Amounts of Cr(VI) in the Presence of Cr(III)

  • A simple, selective and sensitive fluorescence quenching method was developed to the determination Cr(VI). The method is based on the oxidation of $I^-\;to\;{{I_3}^-}$ by Cr(VI) in sulfuric acid solution followed by immediate formation of ion association compound between I3 − and rhodamine 6G in Tween-80 micellar media at room temperature. The influence of several surfactants on rhodamine 6G fluorescence signal was studied; particular attention was paid in the aggregation behavior of rhodamine 6G–Tween-80 system. The experimental parameters (e.g., type of surfactant, reagent concentration) were studied and the optimal conditions were established. The linear calibration graph was obtained in the range 2.0 - 100.0 ng m$L^{-1}$ Cr(VI). The detection limit of the method was 0.37 ng m$L^{-1}$. The relative standard deviation (R.S.D.) is less than 5% (n = 5). The efficiency of the method for the determination of Cr(VI) in the presence of Cr(III) in the sample was investigated. The method was applied successfully to the determination of Cr(VI) and total Cr in water, and liver tissue samples.
