DNA Profiles of Trichoderma spp. in Korea

  • Molecular approaches, internal transcribed spacer(ITS) sequences of ribosomal DNA, and Universal Rice Primer Polymerase Chain Reaction(URP-PCR) were used to investigate the genetic diversity, taxonomic complexity, and relationships of Trichoderma species in mushroom farms. Forty-one isolates of 13 Trichoderma spp. were used in this study and clustered into eight groups. The DNA fingerprint patterns and ITS1 region sequence alignment data showed similar results, but not in some species, such as T. virens, T. atroviride, T. harzianum, and T. aureoviride. Results of this study have proven that the morphology-based taxonomic system has some limitations in terms of classification. The data obtained in this study would be a good index for classifying indistinguishable Trichoderma strains.

Phylogenetic and Morphological Identification of the Novel Pathogen of Rheum palmatum Leaf Spot in Gansu, China

  • A new leaf spot disease was observed on leaves of Rheum palmatum (Chinese rhubarb) in Northwest China (Gansu Province) starting in 2005. A Septoria-like fungus was isolated and completion of Koch's postulates confirmed that the fungus was the casual agent of the leaf spot disease. Morphology and molecular methods were combined to identify the pathogen. The fungus produced conidiomata pycnidia and the conidia were 2~5 septate, $61.2{\sim}134.1{\mu}m$ in length and $3.53{\sim}5.3{\mu}m$ in width, which is much larger than the known Spetoria species that infects Polygonaceae species. Phylogenic analysis of the internal transcribed spacer region confirmed that this Septoria-like fungus is within the Septoria genus but distinct from known Septoria species. Together, these morphological and phylogenetic data support that the R. palmatum infecting Septoria strain is a newly-described plant pathogenic species.

Trichoderma asperellum Chi42 Genes Encode Chitinase

  • Four Trichoderma strains (CH2, SH16, PQ34, and TN42) were isolated from soil samples collected from Quang Tri and Thua Thien Hue provinces in Vietnam. The strains exhibited high chitinolytic secretion. Strain PQ34 formed the largest zone of chitinase-mediated clearance (> 4 cm in diameter) in agar containing 1% (w/v) colloidal chitin. Analysis of the internal transcribed spacer regions of these strains indicated that they were Trichoderma asperellum. The molecular weights of the chitinases were approximately 42 kDa. Chitinase genes (chi42) of T. asperellum strains TN42, CH2, SH16, and PQ34 were 98~99% homologous to the ech42 gene of T. harzianum CB-Pin-01 (accession No. DQ166036). The deduced amino acid sequences of both T. asperellum strains SH16 and TN42 shared 100% similarity.

감나무 잎에서 분리한 Phialocephala piceae에 대한 보고 (A New Report on Phialocephala piceae Isolated from Leaf of Diospyros kaki in Korea)

  • 경상북도 상주시에 재식된 감나무 잎의 표면에서 국내 미기록종인 곰팡이를 분리하여 형태학적 및 계통학적 분석을 수행하였다. 분리된 곰팡이는 Phialocephala piceae의 형태적 특징인 암갈색 균총과 굵고 짧은 분생자경 유사조직을 관찰할 수 있었다. Phialocephala속의 동정에 사용되는 internal transcribed spacer 영역과 RNA polymerase II largest subunit 유전자를 이용한 계통학적 분석을 통해 해당균이 P. piceae임이 확인되었고, 이를 통해 최초로 국내에 P. Piceae가 존재함을 보고하였다.

Ten New Recorded Species of Macrofungi on Ulleung Island, Korea

  • Ulleung Island is a biodiversity hotspot in South Korea. During a survey of indigenous fungal species from Ulleung Island conducted from 2015 to 2016, we discovered 10 unrecorded macrofungi in Korea. These macrofungi were identified to the species level using morphological features and phylogenetic analysis based on the internal transcribed spacer region: Deconica phyllogena, Mycena zephirus, Phaeomarasmius proximans, Phlebia radiata, Pluteus semibulbosus, Postia alni, Resinicium pinicola, Scytinostroma portentosum, Tricholomopsis flammula, and Tyromyces kmetii. We also provide detailed morphological descriptions for these 10 species.

단메밀과 쓴메밀의 ITS 염기서열 기반 식별마커 (Discriminability of Molecular Markers Based on Muclear Ribosomal ITS Sequences of Fagopyrum esculentum and F. tataricum)

  • We analyzed the nuclear ribosomal internal transcribed spacer (ITS) sequence of common buckwheat, Fagopyrum esculentum and tartary buckwheat, F. tataricum. The diversity of the nucleotides and haplotypes, Tajima's D, and Fu's Fs was analyzed and compared among the varieties of common buckwheat and tartary buckwheat. The diversity of nucleotides and haplotypes indicated that the buckwheat populations had undergone rapid population expansion but D and Fs did not support their expansion statistically. The phylogenetic analysis of ITS sequences did not clearly establish the phylogenetic relationships between the varieties of common buckwheat. The In/Del sequence of ITS-1 region could, therefore, be used as a DNA marker to distinguish raw or manufactured products derived from common buckwheat and tartary buckwheat.

고려인삼과 미국삼의 종간잡종으로부터 재분화된 식물체의 특성 (Characteristics of Plantlets Redifferentiated from F1 Hybrid between Panax ginseng and Panax quinquefolius)

  • 교잡 1세대의 화서조직에서 재분화된 F1 유식물체는 고려인삼 유식물체에 비하여 지상부와 지하부 생육이 모두 양호하였으며, 줄기 색도 고려인삼 재분화 식물체의 줄기에 비하여 자색을 강하게 띄었으며 잎의 색도 진한 녹색을 나타내었다. 천풍, 연풍, 선원 등의 고려인삼의 품종 내에서는 Internal Transcribed Spacer (ITS)영역의 DNA PCR 패턴간에 차이점이 나타나지 않았으나, 고려인삼과 미국인삼은 각기 다른 PCR 패턴을 보였으며, 고려인삼과 미국인삼간의 교잡 1세대는 고려인삼과 미국인삼에 나타나는 고유한 PCR패턴을 모두 나타내었다. 교잡 1세대에서 유기한 캘러스와 재분화식물체는 조직배양 모본인 교잡 1세대와 동일한 PCR 패턴을 보임에 따라 교잡 1세대의 조직배양체는 ribosomal DNA의 ITS영역에서 유전적인 안정성을 나타내는 것으로 확인되었다.

Sequence Analysis of Cochlodinium polykrikoides Isolated from Korean Coastal Waters Using Sequences of Internal Transcribed Spacers and 5.8S rDNA

  • The relativity of four isolates of C. polykrikoides was determined by comparative sequence analysis based on direct sequencing of PCR amplified ribosomal DNA (the internal transcribed spacer region and the 5.8S rDNA). Sequence comparisons indicated that four isolates had the same nucleotide sites in the ITS regions, as well as a total of 585 nucleotide length and 100% homology. The molecular data revealed that C. polykrikoides in Korean coastal waters show no genetical difference.

Identification of Entomopathogenic Fungus, Beauveria spp. F-101 Isolated from Thecodiplosis japonensis Using Internal Transcribed Spacer Sequence

  • For the development of the alternative control system against the major forest pests, Beauveria spp. F-101, isolated from a dead larva of Thecodiplosis japonensis, was selected because this isolate showed high pathogenicities against T. japonensis and Acantholyda parki. Beauveria spp. F-101 had irregular clustered conidio-phores and conidia borne on a distinctive apical zigzag extension, and it showed typical characteristic of the genus, Beauveria in morphology. For molecular based-identification, the ribosomal ITS region of Beauveria spp. F-101 was amplified with ITS1 and ITS4 primers, and cloned into pGEM- T Easy vector. The amplified PCR product was 569 bp in size and completely sequenced. The similarities of the cloned ITS sequence were 99 % and 97% to those of B. bassiana and B. brongniartii, respectively. In comparison to other species among the genus Beauveria, the ITS region of Beauveria spp. F-101 showed a similarity of 95% to B. amorpha, 95% to B. tenella, 89% to B. vermiconia and 69% to B. alba, respectively. In addition, in comparison to different genus, it had 95% similarities to Cordyceps militaris and 91% to Paecilomyces tenuipes. Accordingly, the current result suggests that Beauveria spp. F-101 was a variant of B. bassiana and it seems to be a new isolate considering sequence variation in ITS region.
