Recent publications showed that metal nanoparticles which are made from $TiO_2,\;CeO_2,\;Al_2O_3,\;CuCl_2,\;AgNO_3$ and $ZnO_2$ induced oxidative stress and pro-inflammatory effects in cultured cells and the responses seemed to be common toxic pathway of metal nanoparticles to the ultimate toxicity in animals as well as cellular level. In this study, we compared the gene expression induced by two different types of metal oxide nanoparticles, titanium dioxide nanoparticles (TNP) and cerium dioxide nanoparticles (CNP) using microarray analysis. About 50 genes including interleukin 6, interleukin 1, platelet-derived growth factor $\beta$, and leukemia inhibitory factor were induced in cultured BEAS2B cells treated with TNP 40 ppm. When we compared the induction levels of genes in TNP-treated cells to those in CNP-treated cells, the induction levels were very correlated in various gene categories (r=0.645). This may suggest a possible common toxic mechanism of metal oxide nanoparticles.

Arginine deiminase (ADI, E.C. 3.5.3.6), one of the arginine deprivation enzymes, exhibits anticarcinogenic activities. The present study investigated the anti-inflammatory activities of the purified recombinant ADI originating from Lactococcus lactis ssp. lactis ATCC7962 (LADI). LADI dose-dependently inhibited lipopolysaccharide (LPS)-induced upregulation of inducible nitric oxide synthase and the production of nitric oxide in RAW 264.7 murine macrophages. The induction of cyclooxygenase-2 expression and subsequent production of prostaglandin $E_2$ by LPS was also attenuated by LADI treatment. Moreover, LADI inhibited the production of interleukin-6 in LPS-stimulated RAW 264.7 macrophages. These results indicate that LADI exerts anti-inflammatory effects, which may in part explain its chemopreventive potential.

Purpose : Interleukin-4(IL-4) is a critical component of the Th2 cytokine pathway and contributes to severity of respiratory syncytial virus(RSV) bronchiolitis. Previous studies observed an association between severe RSV bronchiolitis in Korean children with a common haplotype of the IL4 promoter. This study was performed to investigate functional differences of the variant IL4 promoter haplotypes. Methods : Genomic DNA was obtained from 20 children from 6 to 48 months of age in the Department of Pediatrics, Seoul National University Bundang Hospital. The IL4 promoter spanning an 1.2 kb region was amplified and haplotype was determined by cloning and the PHASE reconstruction. Transcriptional activity of Jurkat T cells which were transfected with each IL4 haplotype were analyzed by use of luciferase assay. Results : Three haplotypes of the IL4 promoter have been identified with the frequency of GCC(7 percent), TCC(17 percent), and TTT(76 percent). The TTT haplotype demonstrated the highest luciferase values in both unstimulated and PMA-stimulated Jurkat T cells. Increases in transcriptional activity compared to GCC have been shown in TTT(5.3 fold higher) followed by TCC(4.2 fold higher) in unstimulated Jurkat T cells. Conclusion : We provided evidence that increased transcriptional activity of the TTT haplotype of the IL4 promoter, which has previously been over-represented in Korean children with severe RSV bronchiolitis. Therefore, IL-4 could play a potential role in the pathogenesis of RSV infection, possibly via an altered transcriptional activity of the different IL4 haplotypes.
세균성 치태는 치은의 염증과 치주 조직 파괴를 동반하는 치주염의 주요한 인자로서 치주 조직 건강을 유지하기 위하여 적절한 치태 조절이 필요하다. 본 논문의 목적은 12주 동안 만성 초기 및 중등도 치주염 환자에서 치은염에 대한 임상 지수의 감소, interleukin-6 (IL-6) 농도와 치주질환 원인균인 Prevotella intermedia (P. intermedia), Actinobacillus actinomycetemcomitans (A. actinomycetemcomitans)에 대한 소니케어 전동 칫솔의 효과를 일반 칫솔과 비교해 보고자 하는 데 있다. 총 82명의 환자를 선택하였으며, 30명은 일반 칫솔, 52명은 소니케어 전동 칫솔 군으로 분류하여 칫솔질 교육을 실시하였다. 전악을 전치부와 구치부로 나누어 초진, 1, 4, 12주에서의 치태, 치은 지수 및 탐침 시 출혈 여부를 조사하였으며, 가장 깊은 치주낭 탐침을 보이는 치아 3개를 선택하여 탐침 깊이와 부착 정도를 측정하였고, 선택된 치아에서 초진, 1, 12주에 채취된 샘플을 통해 치은열구액 내의 IL-6 농도와 P. intermedia, A. actinomycetemcomitans의 CT값을 추가적으로 조사하여 다음과 같은 결과를 얻었다. 1 소니케어 전동 칫솔과 일반 칫솔군 모두 치은 염증을 나타내는 임상 지수 (치태지수, 치은지수, 탐침 시 출혈)는 12주 기간 동안 유의한 감소 (p<0.05)를 보였으나, 전동 칫솔 군에서 통계학적으로 더욱 유의하게 (p<0.05) 나타났다. 2. 전치부를 제외한 구치부 치아에서 소니케어 전동 칫솔군은 12주 기간 동안 탐침 시 출혈의 감소가 통계학적으로 유의하게 (p<0.05) 나타났다. 3. 가장 깊은 치주낭 탐침 깊이를 보이는 3개의 선택된 치아에서 치주낭 탐침 깊이와 부착 정도는 두 군 모두 초진에 비해 유의한 감소 (p<0.05)를 보였다. 퍼센트 변화 비교에서 치주낭 탐침 깊이는 소니케어 전동 칫솔군이 $18.47{\pm}10.05%$, 일반 칫솔군이 $14.19{\pm}8.16%$로, 부착 정도는 소니케어 전동 칫솔군이 $24.26{\pm}12.51%$, 일반 칫솔군이 $15.65{\pm}9.92%$로 각각 나타났으나, 군 간 통계적 유의차는 보이지 않았다. 4. 치은열구액의 IL-6 농도는 두 군 모두 12주 기간 동안 통계적으로 유의한 감소 (p<0.05)를 나타내었다. 퍼센트 변화 비교에서 전동 칫솔군은 51%, 일반 칫솔군은 37%로 각각 나타났으나, 군 간 통계적 유의차는 보이지 않았다. 5. Prevotella intermedia, Actinobacillus actinomycetemcomitans의 관찰에서 두 군 모두 유의한 차이는 없었다. 위 결과를 통해 본 연구에서는 소니케어 전동 칫솔의 사용이 일반 칫솔에 비하여 만성 초기 및 중등도 치주염 환자에서 치태의 제거, 치은 염증 및 임상 지수의 감소에 유의한 효과가 있으며 IL-6의 감소 경향에도 효과가 있음을 관찰하였다.
Biorenovation is a method for converting materials using the enzyme properties of microorganisms. Natural products converted by that method increase physiological activity or reduce cytotoxicity. In this study, we investigated the anti-inflammatory activity of crypsinus hastatus prothallium (CH) and biorenovated CH prothallium (CHB) using RAW 264.7 cells stimulated with lipopolysaccharide (LPS). CHB inhibited the production of nitric oxide, prostaglandin E2 and cytokines (interleukin-6, interleukin-1β, tumor necrosis factor-α) compared to CH at a concentration of 50-200 ㎍/mL. In addition, CHB concentration of 200 ㎍/mL inhibited the expression of inducible nitric oxide synthase and cyclooxygenase-2 protein by LPS stimulation to the level of the untreated control group. These results indicate that CHB could be a novel anti-inflammatory agent for cosmetic and pharmaceutical ingredients.It also suggests that the application of biorenovation has potential usefulness in developing anti-inflammatory materials. Brain insults, including neurotrauma, infection, and perinatal injuries such as hypoxic ischemic encephalopathy, generate inflammation in the brain. These inflammatory cascades induce a wide spectrum of cytokines, which can cause neuron degeneration, have neurotoxic effects on brain tissue, and lead to the development of seizures, even if they are subclinical and occur at birth. Cytokines are secreted by the glial cells of the central nervous system and they function as immune system mediators. Cytokines can be proinflammatory or anti-inflammatory. Interleukin (IL)-$1{\beta}$ and IL-8 are proinflammatory cytokines that activate additional cytokine cascades and increase seizure susceptibility and organ damage, whereas IL-1 receptor antagonist and IL-10 act as anti-inflammatory cytokines that have protective and anticonvulsant effects. Therefore, the immune system and its associated inflammatory reactions appear to play an important role in brain damage. Whether cytokine release is relevant for the processes of epileptogenesis and antiepileptogenesis, and whether epileptogenesis could be prevented by immunomodulatory treatment should be addressed in future clinical studies. Furthermore, early detection of brain damage and early intervention are essential for the prevention of disease progression and further neurological complications. Therefore, cytokines might be useful as biomarkers for earlier detection of brain damage in high-risk infants.
The hawthorn fruit (Crataegus pinnatifida Bunge var. typica Schneider) is used as a traditional medicine in Korea. The objective of this study was to understand the mechanisms of the anti-inflammatory effects of the water fractionated portion of hawthorn fruit on a lipopolysaccharide (LPS)-stimulated RAW 264.7 cellular model. The level of nitric oxide (NO) production in the water fraction and LPS-treated RAW 264.7 cells were determined with an ELISA. The cytotoxicity of the water fraction and LPS was measured with an MTT assay. Expression of nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), tumor necrosis factor (TNF)-${\alpha}$, interleukin 6 (IL-6), and interleukin $1{\beta}$ (IL-$1{\beta}$) mRNA were analyzed with a reverse transcription polymerase chain reaction (RT-PCR). The water fraction of hawthorn fruit was determined to be safe and significantly inhibited NO production in LPS-stimulated RAW 264.7 cells and suppressed COX-2, (TNF)-${\alpha}$, IL-$1{\beta}$, and IL-6 expression. The observed anti-inflammatory effects of the water fraction of hawthorn fruit might be attributed to the down-regulation of COX-2, (TNF)-${\alpha}$, IL-$1{\beta}$, and IL-6 expression in LPS-stimulated RAW 264.7 cells.

Previously, wogonin (5,7-dihydroxy-8-methoxyflavone) was found to suppress proinflammatory enzyme expression including cyclooxygenase-2 (COX-2), contributing to in vivo anti-inflammatory activity against skin inflammation. However, the detailed effect on each skin cell type has not been understood. Therefore, present investigation was carried out to find the effect of wogonin on inflammation-associated gene expression from skin fibroblasts in culture using reverse transcriptase-polymerase chain reaction. As a result, it was found that wogonin (10 - 100 ${\mu}$M) clearly down-regulated COX -2 expression from NIH/3T3 cells treated with 12-O-tetradecanoylphorbol 13-acetate, interleukin-1${\beta}$ or tumor necrosis factor-a. But, the expression levels of COX-1, interleukin-1${\beta}$ and fibronectin were not significantly affected. This finding was well correlated with significant reduction of prostaglandin E$_2$(PGE$_2$) production by wogonin. As a comparison, NS-398 (selective cyclooxygenase-2 inhibitor) did not suppress COX -2 expression and other gene levels, while PGE$_2$production was potently reduced at 0.1 - 10 ${\mu}$M. All these results suggest that COX -2 down-regulation of skin fibroblasts may be, at least in part, one of anti-inflammatory mechanisms of wogonin against skin inflammation.

Newcastle disease vaccine (NDV)-loaded chitosan microspheres (NDV-CM) were prepared. Stimulatory effects of these NDV-CM on antibody response compared to free NDV were examined in vitro and in vivo. In vitro stimulation of macrophages with virus vaccine resulted in higher number of cells compared to saline-treated control. Both NDV and NDV-CM induced secretion of interleukin-1 (IL-1) in dose dependent manner and the secretion of IL-1 by NDV-CM was delayed compared to free NDV. Irrespective of vaccine formulation, NDV subunit antigen was not effective in preventing mortality of the birds after challenge. However, CM loaded with NDV made of whole viron had antibody responses and protection similar to those shown by ND-K, a commercial inactivated oilemulsion vaccine.
