본 연구는 쑥 추출물의 항염 활성이 prostaglandins 합성의 저해 및 pro-inflammatory cytokine의 억제기전과 관련이 있을 것으로 예상되어짐에 따라, 큰비쑥(A. fukudo)을 대상으로 80% EtOH로 추출하고 추출물을 극성에 따라 용매분획을 실시하여, 큰비쑥 에탄올 추출물 및 용매분획물들이 염증반응의 주체가 되는 대식 세포 계열인 RAW 264.7 세포에서 LPS 로 유도된 TNF-${\alpha}$, IL-$1{\beta}$ 그리고 IL-6와 같은 pro-inflammatory cytokine과 NO의 생성억제효과, 그리고 iNOS와 COX-2의 단백질 발현 억제효과 및 $PGE_{2}$ 생성 억제효과 등을 통해 알아보았다. 대식세포 계열인 RAW 264.7 세포에 LPS로 자극을 주고 큰비쑥 추출물을 처리하여 확인해본 결과, 추출물 및 분획물들이 다소 차이는 있었지만 TNF-${\alpha}$, IL-$1{\beta}$ 그리고 IL-6에서 생성억제 효과를 나타났다. 또한 헥산, 디클로로메탄 및 에틸아세테이트 분획물에서 NO의 생성억제 효과가 강하게 나타났으며, 헥산과 디클로로메탄 분획물에서는 iNOS, COX-2 및 $PGE_{2}$ 생성 억제 효과가 다른 분획물에 비해 강하게 나타났다. 이러한 결과는 큰비쑥에서 유효성분 추출을 통한 항염증 물질의 연구 또는 예방하거나 치료할 수 있는 염증 억제 성분의 분리 및 그 작용기전 연구에 중요한 기초 자료가 될 것이라 사료된다. 또한 큰비쑥 추출물로부터 염증억제 성분을 도출하고자 활성분획인 헥산과 디클로로메탄 분획물에 대하여 활성성분의 분리가 진행 중이다.
회향(Foeniculum vulgare) 열매의 메탄올 추출물과 분획물(hexane, methylene chloride, ethylacetate, n-butanol)을 cell culture model system을 이용하여 항염증 활성을 평가하였다. 이를 위해, LPS처리로 염증이 유도된 RAW 264.7 macrophage에서 시료가 세포독성을 나타나지 않는 $100{\mu}g/ml$의 농도를 처리한 후 염증매개물질인 NO, PGE2, 염증성 사이토카인($IL-1{\beta}$, IL-6 및 TNF-${\alpha}$) 생성 및 mRNA 발현을 측정하였다. 그 결과 hexane, methylene chloride 및 ethylacetate 분획물은 대조군에 비해 NO, PGE2 생성을 억제하였으며, 이는 iNOS, COX-2 mRNA 발현 저해에서 기인함을 확인하였다. Methylene chloride와 ethylacetate 분획물은 $100{\mu}g/ml$의 농도에서 $IL-1{\beta}$, IL-6 생성 및 mRNA 발현을 효과적으로 억제하였으며, 특히 ethyl acetate 분획물은 TNF-${\alpha}$ 의 생성과 mRNA발현을 유의적으로 저해하였다. 본 연구 결과를 통해 회향 및 회향 분획물의 항염증 활성을 확인하였으며, 이는 염증 매개물질인 NO, PGE2 그리고 염증성 사이토카인 mRNA 활성 및 분비 억제에 의한 것으로 나타났다.
Twelve compounds were isolated from the MeOH extract of the leaf and stem of the Korean endemic plant Weigela subsessilis L. H. Bailey. Their chemical structures were elucidated on the basis of physicochemical and spectroscopic data and by comparison with those of published literatures. These compounds were identified as three sterols, ${\beta}$-sitosterol acetate (2), ${\beta}$-sitosterol (3), daucosterol (11), eight triterpenoids, squalene (1), ursolic acid (4), ilekudinol A (5), corosolic acid (6), ilekudinol B (7), esculentic acid (8), pomolic acid (9), asiatic acid (10), and one iridoid glycoside, alboside I (12). This is the first report pertaining to the isolation of these compounds from Weigela subsessilis L. H. Bailey. In addition, three compounds 7, 9, and 12 were found to display a strong inhibitory effect on the production of IL-8 in the HT29 cells stimulated by TNF-${\alpha}$.

Opisthorchis viverrini infection induces chronic inflammation, and a minor proportion of infected individuals develop advanced periductal fibrosis (APF) and cholangiocarcinoma (CCA). Inflammatory cytokines and/or their gene polymorphisms may link to these biliary pathologies. We therefore investigated associations among cytokine gene polymorphisms and cytokine production in 510 Thai cases infected with O. viverrini who presented with APF+ or APF-, as established by abdominal ultrasonography as well as in patients diagnosed with CCA. Levels of pro-inflammatory and anti-inflammatory cytokines were determined in culture supernatants after stimulation of peripheral blood mononuclear cells (PBMCs) with O. viverrini excretory-secretory (ES) products. Pro-inflammatory cytokines, IL-$1{\beta}$, IL-6, IFN-${\gamma}$, LT-${\alpha}$, and TNF-${\alpha}$ were significantly increased in CCA patients compared with non-CCA (APF- and APF+) cases. Polymorphisms in genes encoding IL-$1{\beta}$-511C/T, IL-6-174G/C, IFN-${\gamma}$+874T/A, LT-${\alpha}$+252A/G, and TNF-${\alpha}$-308G/A were then investigated by using PCR-RFLP or allele specific-PCR (AS-PCR) analyses. In the CCA cases, LT-${\alpha}$+252A/G and TNF-${\alpha}$-308G/A heterozygous and homozygous variants showed significantly higher levels of these cytokines than the wild type. By contrast, levels of cytokines in wild type of IFN-${\gamma}$+874T/A were significantly higher than the variants in CCA cases. IFN-${\gamma}$+874T/A polymorphisms were associated with advanced periductal fibrosis, whereas IL-6-174G/C polymorphisms were associated with CCA. To our knowledge, these findings provide the first demonstration that O. viverrini infected individuals carrying several specific cytokine gene polymorphisms are susceptible to develop fibrosis and CCA.

BACKGROUND/OBJECTIVES: Nelumbo leaves have been used in traditional medicine to treat bleeding, gastritis, hemorrhoids, and halitosis. However, their mechanisms have not been elucidated. MATERIALS/METHODS: The present study prepared two Nelumbo leaf extracts (NLEs) using water or 50% ethanol. Inflammatory response was induced with LPS treatment, and expression of pro-inflammatory mediators (inducible nitric oxide synthase, cyclooxygenase-2, tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin (IL)-$1{\beta}$, and IL-6 and nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) productions were assessed. To determine the anti-inflammatory mechanism of NLEs, we measured nuclear factor-${\kappa}B$ (NF-${\kappa}B$) activity. Major metabolites of NLEs were also analyzed and quantified. RESULTS: NLEs effectively reduced the expression and productions of pro-inflammatory mediators such as IL-$1{\beta}$, IL-6, TNF-${\alpha}$, $PGE_2$, and NO. NLEs also reduced NF-${\kappa}B$ activity by inhibiting inhibitor of NF-${\kappa}B$ phosphorylation. Both extracts contained catechin and quercetin, bioactive compounds of NLEs. CONCLUSIONS: In this study, we showed that NLEs could be used to inhibit NF-${\kappa}B$-mediated inflammatory responses. In addition, our data support the idea that NLEs can ameliorate disease conditions involving chronic inflammation.
The purpose of this study was to examine the anti allergic effect in vivo and in vitro, and to observe single and four weeks repeated toxicity in mice of Bangpung-galgeun-tang (BGT). We investigated anti DNP IgE-mediated passive cutaneous anaphylaxis in rodents and compound 48/80-induced active systemic anaphylatic shock in mice after oral administration with BGT of 0.4 g/kg and 0.8 g/kg for 8 days, and also examined MTT assay, ${\beta}-hexosaminidase$ activity, IL-4 and $TNF-{\alpha}$ from RBL-2H3 and $TNF-{\alpha}$ from Raw264.7 after pre-treatment with BGT of 0.25 mg/ml, 0.5 mg/ml, 1 mg/ml and 2 mg/ml. To ascertain safety and toxicity of BGT, we divided into single and four weeks repeated administration test. In single test, three groups were administrated different dosages and routes (2 g/kg/i.p., 4 g/kg/i.p. and 15 g/kg /p.o.) of BGT, and in four weeks repeated test, 0.8 g/kg BGT was administrated. Control groups were administrated with only saline according to on Korean Food and Drug Administration, respectively. We observed attentively motality, abnormal clinical sign, body weight change, organ weight, AST and ALT of mice after BGT administration. BGT inhibited passive cutaneous anaphylaxis and active systemic anaphylatic shock by oral administration. All the concentrations of BGT from 0.25 to 2 mg/ml didn't have an effect on cell viability and cytotoxicity. In RBL-2H3, ${\beta}-hexosaminidase$ release, IL-4 and $TNF-{\alpha}$, and in Raw264.7, $TNF-{\alpha}$ were significantly reduced by treated all concentrations of BGT. During toxicity experiment period, there was no difference in body weight change, organ weight, AST and ALT among different dose groups. Death were found 3 mice from day 2 to day 3 in single test i.p. group. (2 g/kg, 4 g/kg). Several individuals of single test i.p. group were observed that decreased locomotor activity, exophthalmos, bloodshot eyes, loss of eyesight and so on in early period after administration. But there was no difference in clinical signs among p.o. group. These results indicate that BGT have inhibition effects on allergy and suggest that no observable effect level of the test orally administration was considered to be more than 2 g/kg in mice under the conditions employed in this study.
Objective: This study aimed to use a mouse model to evaluate the effects of Gwaruhaengryeon-hwan (GHH) on chronic obstructive pulmonary disease (COPD) and particulate matter induced lung injury. Materials and Methods: The study was carried out in two ways (in vitro, in vivo). In vitro RAW 264.7 cells (mouse macrophage) were used and analyzed by flow cytometry, ELISA. In vivo lipopolysaccharide (LPS) and cigarette smoke solution (CSS), or coal, fly ash, diesel exhaust particle (CFD) challenged mice were used and its BALF was analyzed by ELISA, lung tissue by real-time PCR. Results: In vitro, GHH maintained an 80-100% rate of viability. So cytotoxicity was not shown. In the ELISA analysis with RAW 264.7 cells, GHH significantly decreased NO over $30{\mu}g/ml$. In the ELISA analysis, GHH significantly decreased $TNF-{\alpha}$, IL-6 over $300{\mu}g/ml$. In the COPD model, the GHH 200 mg/kg dosage group, the application of GHH significantly decreased the increasing of neutrophils, $TNF-{\alpha}$, IL-17A, MIP2, CXCL-1 in BALF, $TNF-{\alpha}$, $IL-1{\beta}$ mRNA expression in lung tissue and histological lung injury. In the CFD induced lung injury model, the GHH 200 mg/kg dosage group, the application of GHH significantly decreased the increase of neutrophils, $TNF-{\alpha}$, IL-17A, MIP2, CXCL-1 in BALF, MUC5AC, $TGF-{\beta}$ mRNA expression in lung tissue and histological lung injury. Conclusion: This study suggests the usability of GHH for COPD patients by controlling lung tissue injury.

Inflammatory responses from activated microglia are one of major causes of Alzheimer's disease (AD). Particularly, proinflammatory cytokines (PC), such as IL-l$\beta$ and TNF-$\alpha$, and nitric oxide (NO) are correlated with AD by inducing the chronic inflammation in the brain. In the present study, we found that microglia are activated by lipopolisaccharide (LPS) and Abeta42 (A$\beta$42), and those activated microglia produced such repertoires up to 72h with a turning point at 24h. However, no dose dependecy was found during the chasing time courses (6h to 72h). (omitted)
생체 내 (ex vivo) 실험에서 혼합 (고들빼기, 돌미나리, 메밀, 톳, 생강)시료의 물 추출물을 4주간 격일로 마우스 체중 kg 당 0, 50, 500 mg/kg B.W.의 농도로 마우스에 경구 투여한 후 비장세포 증식능, LPS에 의해 활성화된 복강 대식세포가 분비하는 염증성 사이토카인 ($IL-1{\beta}$, IL-6, $TNF-{\alpha}$)의 생성량을 측정하였다. 그 결과 혼합시료의 물추출물은 ConA나 LPS로 자극하지 않은 경우 50 mg./kg B.W. 농도에서 대조군에 비해 유의적으로 높은 비장 증식능을 보였다. LPS에 의해 활성화된 복강 대식세포의 사이토카인 분비량을 측정한 결과에서도 $IL-1{\beta}$, IL-6, $TNF-{\alpha}$의 분비량은 마우스 체중 kg 당 50 mg/kg B.W. 농도의 투여군에서 높은 생성량을 보여주었으나, 500 mg/kg B.W.의 농도 투여군에서는 IL-6 분비량은 대조군에 비해 높은 반면 $TNF-{\alpha}$의 분비량은 큰 차이를 보이지 않았다. 이상의 결과에 따라 비장세포 증식능과 사이토카인 분비능을 검색한 결과 고들빼기, 도미나리, 메밀, 톳, 생강을 혼합한 혼합시료의 추출물 50 mg/kg B.W. 농도 투여가 면역 세포와 면역 기관의 주요기능을 증진시키리 가능성이 있을 것으로 사료된다. 이러한 연구결과를 토대로 앞으로 혼합식을 통한 기능성 식품 개발에 기초 연구 자료가 될 것으로 기대된다.
In the present study, immune gene expression of rainbow trout, Oncorhynchus mykiss, against bacterial endotoxin (LPS) was studied in vivo. The expression of prointflammatory cytokine genes (IL-1$\beta$ and TNF-$\alpha$) and IFN-related genes (IRF-I, Mx-3) at gill was assessed by RT-PCR at different time point of day1 and day 3 post-injection. It was shown that the proinflammatory cytokine gene expression at gill was induced 1 day after LPS injection but the expression was not sustained until day 3. Meanwhile upregulated expression of IFN-related genes was found to be only at day 3 post injection, indicating indirect effect of LPS on these genes.
