In this study, the biological activity of water and ethanol extracts from Chrysanthemum incidicum Linne by ultrafine grinding for functional food source are examined. The content of phenolic compounds from Chrysanthemum incidicum Linne were the highest when extracted for 6 hr with 70% ethanol. The extraction yield of water and ethanol extracts were $7.12{\pm}1.61$ mg/g and $7.51{\pm}2.14$ mg/g, respectively. With ultrafine grinding, water and ethanol extracts were $8.63{\pm}1.15$ mg/g and $9.33{\pm}1.35$ mg/g, respectively. In determining anti-oxidative activity of Chrysanthemum incidicum Linne extracts, DPPH of normal grinding extracts was 83.52% and ultrafine grinding was 92.37%. In ABTS radical cation decolorization, normal grinding, fine grinding, and ultrafine grinding extracts were 90% or higher. In antioxidant protection factor (PF), water and ethanol extracts of ultrafine grinding showed relatively high anti-oxidative activities of each 1.82 PF and 2.16 PF, respectively. The TBARS value of ultrafine grinding extracts were lower than normal grinding and fine grinding extracts. The inhibition activity on xanthin oxidase of Chrysanthemum incidicum Linne extracts was 67.53% in ultrafine grinded water extracts and 83.45% in ultrafine grinded ethanol extracts. Inhibition on xanthin oxidase of ethanol extracts showed a higher inhibition effect than water extracts, and ultrafine grinding was higher than normal grinding. In angiotensin converting enzyme inhibition activity, ultrafine grinding water extract was 24% or higher, and ethanol extract was 34% or higher. The elastase inhibition activity of ultrafine grinding extract was 25.56%, which was higher than 20.34% of fine grinding extracts. Water extracts did not show hyaluronidase inhibition activity but ethanol extracts showed 35% of hyaluronidase inhibition activity. The determining expression inhibition of iNOS and COX-2 protein in macrophage by Chrysanthemum incidicum Linne extracts with a Western blot analysis, iNOS and COX-2 protein expression inhibition by Chrysanthemum incidicum Linne ethanol extracts were 40% and 15%, respectively at 100 ${\mu}g/mL$ concentration. The inhibitory patterns of iNOS and COX-2 protein expression was concentration dependent. The result suggests that Chrysanthemum incidicum Linne extracts by ultrafine grinding may be more useful than normal grinding as potential sources due to anti-oxidation, angiotensin converting enzyme and xanthine oxidase inhibition, anti-inflammation effect.
To investigate the effect of ethanol extract of Saxifraga stolonifera MEERBURGH on skin care, we measured anti-oxidant and anti-aging activity. S. stolonifera ethanol extract itself had anti-oxidant activity in a dose-dependent manner in DPPH radical scavenging. Silica dose-dependently increased the intracellular ROS generation in RAW 264.7 cells. S. stolonifera ethanol extract inhibited silica-induced intracellular superoxide anion generation, $H_2O_2$ and hydroperoxide generation in RAW 264.7 cells. S. stolonifera ethanol extract significantly inhibited both hyaluronidase and elastase activity, also significantly inhibited MMP-1(collagenase) activity as well. In NIH 3T3 fibroblast cells, S. stolonifera ethanol extract significantly increased collagen-like polymer synthesis, which suggesting the S. stolonifera ethanol extract might be used as hydration and anti-wrinkle agents. From the above results, it is suggested that the main ingredients of S. stolonifera ethanol extract play an important role in anti-oxidant and anti-aging activity.
The extracted phenolic compounds from Pinus densiflora root were examined biological activities for beauty food. The tyrosinase inhibitory activity which was related to skin-whitening was observed. The tyrosinase inhibitory activity was confirmed to be 92% in ethanol extract at $50{\mu}g/mL$ phenolic. The elastase and collagenase inhibitory activity as anti-wrinkle effect were showed 61 and 78% in ethanol extract at $200{\mu}g/mL$ phenolic, respectively. Astringent effect of ethanol extract was showed to be 82% at $50{\mu}g/mL$ phenolic. Hyaluronidase inhibitory activity of ethanol extract as anti-inflammation effect was confirmed to be 94% of inhibition at $200{\mu}g/mL$ phenolic. These results demonstrated that isolated phenolic compounds from P. densiflora root could be expected to use as a functional cosmetic materials.
The marine microalga Spirulina maxima was extracted using water or ethanol at 100 or $80^{\circ}C$ and by ultrasonification in water at $60^{\circ}C$. The ultrasonification technique generated the highest yield (19.8%). To be therapeutically useful, the extraction should yield a product with low cytotoxicity and high immunity against skin infections. The cytotoxicity of all extracts (1.0 mg/mL) was below 25%. Moreover, the cytotoxicity of the extract generated by ultrasonification was 5%. Extracts prepared in the described manners could inhibit hyaluronidase activity by up to 40% compared to the control. Increased growth of human B, T and NK cells and an increase in cytokine secretion were observed, confirming the interrelationship between both human immune and skin immune activity. The extract prepared by ultrasonification increased the growth of human B, T and NK cells up to $10.3{\times}10^4$ cells/mL, $11.3{\times}10^4$ cells/mL and $19.1{\times}10^4$ cells/mL, respectively. The extract prepared by ultrasonification also greatly increased the secretion of both IL-6 and $TNF-{\alpha}$. Moreover, it was estimated that protein, Na and leucine occupy a high ratio. Accordingly, this study has confirmed that extracts prepared as described have the potential to effectively increase skin immunity.
This study was designed to determine the biological activities of Chionanthus retusus flower extracts. Water and 90% ethanol extracts of C. retusus flower were prepared. The inhibitory activities of water and ethanol extracts with a phenolic content of $200{\mu}g/mL$ against xanthine oxidase were 25.60% and 15.92%, respectively. Further, the water extract did not show any inhibitory activity against ${\alpha}$-glucosidase whereas the ethanol extract showed 100.00% inhibition of ${\alpha}$-glucosidase. The inhibitory activities of the extracts against tyrosinase were 17.27% (water extract) and 36.13% (ethanol extract), which suggest that the extracts may have a whitening effect. The water extract did not inhibit elastase activity but showed a collagenase-inhibitory activity of 20.21%. On the contrary, the ethanol extract showed 96.26% and 35.93% inhibition of collagenase and elastase, respectively. These findings suggest that the extracts may have an anti-wrinkle effect. Lastly, the extracts showed a hyaluronidase inhibitory activity of 36.96% (water extract) and 88.70% (ethanol extract), suggesting that they may have an anti-inflammatory effect. The results indicate that C. retusus flower extracts containing phenolic compounds can be used as functional resources because they have anti-gout, carbohydrate degradation-inhibitory, whitening, anti-wrinkle, and anti-inflammatory effects.
Inflammation that is considered to be mainly related to pathogenesis of atopic dermatitis (AD) is the biological response of a host to stimuli, such as cellular injury or infection. In this study, we investigated the anti-inflammatory and anti-oxidative activities of white ginseng roots by ultra high pressure extraction (Gin-UHP), fermentation followed by ultra high pressure extraction (Gin-UHPF), and polyol extraction (Gin-POL). As a result, ginseng extracts were able to decrease the secretion of pro-inflammatory cytokines (interleukin-8 and tumor necrosis factor-alpha) and immunoglobulin E. Also, Gin-POL had the highest DPPH radical scavenging activity and when we compared the SOD-like activity, Gin-UHP had the highest. Moreover, we looked into the effect of these ginseng extracts on anti-aging to show the possible usefulness as a raw material of cosmetics. As a result, ginseng extracts were able to reduce the production of melanin, and inhibit the tyrosinase and elastase activities in a dose-dependent manner. The extracts also decreased the expression of MMP-1 and had a significant hyaluronidase inhibitory activity. Taken together, these results demonstrate that ginseng extracts may have an improvement effect on AD by using its anti-inflammatory and antioxidant properties.
Purpose : This study was undertaken to evaluate the effects of the different administration duration of Ginseng Radix Alba extract solution on the spermatogenic abilities. Methods : We used the 2-month-old mice and administered Ginseng Radix Alba extract solution 0.3mg/g/day for 30, 60, 90 and 120 days. The control group was administered the normal saline in the same way. We examined the number of total, motile and normal sperm from the cauda epididymis, the activities of sperm hyaluronidase. Also we observed changes of isolated testis before and after administration of Ginseng Radix Alba extract solutions in the mice. And we compared to the testicular tissue especially seminiferous tubules between control and treated group by histochemical methods. Results : The significant differences were observed in the concentration of total sperm and normality of spermatozoa of the Ginseng Radix Alba extract solution administered groups compared to the control group in 60, 90 and 120days groups. The significant differences were observed the motility of the Ginseng Radix Alba extract solution administered groups compared to the control group in 90 days group respectively. In the histological analysis of the testicular tissues, the enlargement of testicular lobe diameter and apparent vasculogenesis between testicular lobes were observed in the Ginseng Radix Alba extract solution administered groups compared to the control group, respectively. but the activity of hyaluronidase was not significantly increased in the Ginseng Radix Alba extract solution administered groups Conclusion : This study shows that Ginseng Radix Alba has the beneficial effect on the concentration, morphology and motility of sperm especially in 90 days administration group. We can suggest that Ginseng Radix Alba extract solution be useful for the treatment of male sexual dysfunctions and infertility.
The root extract of Scilla scilloides (which has been used as a traditional folk medicine in Korea) was evaluated with regard to antimicrobial, anti-inflammatory, and anti-oxidative activities. The roots of S. scilloides were minced and extracted with 95% ethanol (root:ethanol=25:75, w/v). The inhibitory effects of S. scilloides root extract on the growth of Staphylococcus aureus ATCC 35556, Salmonella enteritidis ATCC 12021, Escherichia coli O157:H7, and Candida parapsilosis KCCM 35428 were tested. The results indicate that the antimicrobial effects of both 0.1 and 1.0% extract of S. scilloides were greater against the growth of S. aureus ATCC 35556 and C. parapsilosis KCCM 35428 than the growth of S. enteritidis ATCC 12021 and E. coli O157:H7. The anti-inflammatory effects were evaluated by measurement of the inhibition of hyaluronidase activity in vitro. It appears that both 0.1 and 1.0% concentrations of extract have inhibitory effects on hyaluronidase relative to the control. Finally, the anti-oxidative effect of 1.0 and 10% extract solutions were measured according to the thiocyanate method and were compared with 1.0% BHT. The results indicate that the anti-oxidative effect of 10% S. scilloides root extract (anti-oxidative index (AOI); $72.3{\pm}4.2$) is not significantly different from that of 1.0% BHA (AOI; $76.8{\pm}3.5$) (p<0.05). However, it appears that the anti-oxidative effect of S. scilloides root extract is at least three-fold greater than that of BHA when accounting for the amount of dissolved solids in each.

In this study, we investigated the anti-oxidant, anti-aging, and skin whitening effects of Korean mistletoe (Viscum album var. coloratum). The mistletoe fraction was composed of four types: hexane (HX), ethyl acetate (EA), butanol (BU), and methylene chloride (MC). In total phenol content assay, HX showed the highest phenol content among four fractions. In addition, EA significantly increased 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and catalase-like activities, and MC significantly increased superoxide dismutase (SOD)-like activity. When we compared IC50 value in the hyaluronidase and elastase inhibition assay, MC had the lowest IC50. In addition, we also performed tyrosinase inhibition assay to demonstrate the possibility of Korean mistletoe as a cosmetic component. HX showed the highest tyrosinase inhibition rate among the fractions.
Objectives: Myrrh have been used as a traditional remedy to treat infectious and inflammatory diseases. However, it is largely unknown whether myrrh ethanol extract could exhibit the inhibitory activities against particulate matter (PM)-induced skin injury on human keratinocytes, HaCaT cells. Therefore, this study was aimed to investigate the inhibitory activity of myrrh ethanol extract on PM-induced skin injury in HaCaT cells. Methods: To investigate the inhibitory effects of myrrh ethanol extract in HaCaT cells, the skin injury model of HaCaT cells was established under PM treatment. HaCaT keratinocyte cells were pre-treated with myrrh ethanol extract for 1 h, and then stimulated with PM. Then, the cells were harvested to measure the cell viability, reactive oxygen species (ROS), pro-inflammatory cytokines including interleukin (IL) 1-beta, IL-6, and tumor necrosis factor (TNF)-𝛼, hyaluronidase, collagen, MMPs. In addition, we examined the mitogen activated protein kinases (MAPKs) and inhibitory kappa B alpha (I𝜅-B𝛼) as inhibitory mechanisms of myrrh ethanol extract. Results: The treatment of myrrh ethanol extract inhibited the PM-induced cell death and ROS production in HaCaT cells. In addition, myrrh ethanol extract treatment inhibited the PM-induced elevation of IL-1beta, IL-6, and TNF-𝛼. Also, myrrh ethanol extract treatment inhibited the increase of hyaluronidase, MMP and decrease of collagen. Furthermore, myrrh ethanol extract treatment inhibited the activation of MAPKs and the degradation of I𝜅-B𝛼. Conclusions: Our result suggest that treatment of myrrh ethanol extract could inhibit the PM-induced skin injury via deactivation of MAPKs and nuclear factor (NF)-𝜅B in HaCaT cells. This study could suggest that myrrh ethanol extract could be a beneficial agent to prevent skin damage or inflammation.
