하수 처리 과정에서 염분이 세균 군집에 미치는 영향 (Effect of Salinity on the Bacterial Community in the Sewage Treatment System)

  • 하수종말처리장의 처리효율에 미치는 염분의 영향을 알기 위하여 염분의 농도 변화에 따른 종속영양세균, 총세균 그리고 FISH 방법으로 세균 군집 구조를 파악하였다. 총세균수는 염분과 관계없이 큰 변화가 없었으나, 종속영양세균수와 군집구조는 염분에 따라 다르게 변화하였다. 염분이 1% 인 경우에는 대조구에 큰 변화가 없었으나, 염분이 2% 이상이 되면 종속영양세균과 Eubacteria의 비율이 줄어들고, 세균 군집도 큰 변화가 있었다. 특히 Proteobacteria $\alpha$-group, $\gamma$-group 및 Cytophaga-Flavobacterium group의 비율이 감소하였다. 염분이 증가하면, 종속영양세균수와 세균 군집이 변화하므로, 하수처리과정에서 염분이 유입될 경우에는 염분을 1% 이하로 유지하여야 한다.

소양호 세균 군집의 계절적 분석 (Seasonal analysis of heterotrophic bacterial community in lake Soyang)

  • The numerical taxonomy of heterotrophic bacterial community in Lake Soyang was analysed. 95, 115, 88 and 75 strains which were isolated at each season from spring in 1987 to winter in 1988 were clustered by single matching coefficient. The diversity indices (H') were in the range of 0.511-1.684, and the community was most diverse in spring. THe seasonal variation of generic composition was significant. Of the domonant genera, Acinetobacter, Pseudomonas, and Flavobacterium were representative.

낙동강하구에서의 미생물 다양성과 환경변화에 따른 내성한계 (Variations of diversity and tolerance indicies of heterotrophic bacterial communities in Naktong estuary)

  • To determine the characteristics of heterotrophic bacterial community in estuarine ecosystem, water and sediment samples were taden from Naktong estuary. All isolates were compared with 73 characters and described by cluster analysis. With same characters, 30 reference strains were able to divide into approximate species level at 80% similarity (S value). Diversity indices ($H^{1}$) of sediment column isolates were higher than water column isolates. The bacterial community commonly appeared in water and sediment column was reduced with going to downstream. Tolerance indices for temperature (Pt) and salinity (Ps) were also higher in sediment isolates than in water isolates. The bacterial community in sediment column is believed to be composed with diverse populations compared to water column and maintains its stability against various environmental changes with high physiological tolerances.

소양호에서의 종속영양세균의 종구성 및 Alkaline phosphatase 분비 세균에 관한 연구 (Heterotrophic Bacterial Community and Alkaline Phosphatase Releasing Bacteria in Lake Soyang)

  • 1987년 9월부터 1988년 8월까지 소양호 4개 정점에서 총 세균수, 종속영양세균수의 분포와 각 종의 alkaline phosphatase의 분비 비율과 활성도를 측정하였다. 총 세균은 $3.2{\times}10^{5}$ - $3.2{\times}10^{6}$ cells/${\mu}\ell$의 범위였으며, 종속 영양세균의 총 세균수는 0.07-2.63%이였다. 종속영양세균의 종 당양성은 여롬과 양어장 인근, Peridinium이 발생하는 곳에서 낮았다. Pseudomonas. Flavobacterium은 모든 정점에서 우점종으로 나타났다. Alkaline phosphatase를 분비하는 비율과 활성도는 Flavobacterium에서 높게 나타나, 유기인산염 분해에 우점세균 중 가장 큰 기여를 하는 것으로 밝혀졌다.

입상 활성탄 여과지에서 세균의 재성장과 생물막 형성 세균의 분리 및 동정 (Bacterial regrowth in biofilms formed in granular activated carbon filter adsorbers and the bacterial isolation and identification)

  • This study aimed to investigate the biofilm formation, bacterial regrowth, and bacterial community structure in the granular-activated carbon (GAC) filter adsorbers (FAs) used in water treatment plants. In 2005 and 2006, raw water, settled water, GAC FA by depth, and filtered water were collected twice a year from water treatment plants (WTPs) B and S. The number of heterotrophic bacteria, including mesophilic and psychrophilic bacteria, in such collected waters was investigated along with the total number of coliforms therein. Heterotrophic bacteria were detected in most samples, mainly at the surface layers of the GAC FAs, and fewer such bacteria were found in the lower and bottom layers. An increase in the bacterial number, however, was observed in the samples from various depths of the GAC FAs in WTPs B and S compared with the surface layers. An increase in the bacterial number was also detected in the filtered water. This may indicate that there is a regrowth of the bacteria in the GAC FA. Considering, however, that heterotrophic bacteria were not found in the filtered water, it can be deduced that most bacteria are removed in the chlorination process. Coliforms were detected at the surface layer of the GAC FAs, but their regrowth was not observed. MicroLog systems were used to identify the bacteria community distribution. Eight genera and 14 species, including Pseudomonas spp., were detected in WTP B, and 8 genera and 9 species, including Aeromonas spp., in WTP S. Further studies are required to elucidate their role in the biofilms in water treatment processes.

Abundance and Structure of Microbial Loop Components (Bacteria and Protists) in Lakes of Different Trophic Status

  • The abundance, biomass, size distribution, and taxonomic composition of bacterial and protistan (heterotrophic and autotrophic nanoflagellates and ciliates) communities were investigated in six lakes of Masurian Lake District (north-eastern Poland) differing in trophic state. Samples were taken from the trophogenic water layer during summer stratification periods. Image analysis techniques with fluorescent in situ hybridization (FISH) as well as [$^3H$]-methyl-thymidine incorporation methods were applied to analyze differences in the composition and activity of bacterial communities. The greatest differences in trophic parameters were found between the humic lake and remaining non-humic ones. The same bacterial and heterotrophic nanoflagellate (HNF) cell size classes dominated in all the studied lakes. However, distinct increases in the contributions of large bacterial (>$1.0{\mu}m$) and HNF (>$10{\mu}m$) cells were observed in eutrophic lakes. The bacterial community was dominated by the ${\beta}$-Proteohacteria group, which accounted for 27% of total DAPI counts. Ciliate communities were largely composed of Oligotrichida. Positive correlations between bacteria and protists, as well as between nanoflagellates (both heterotrophic and autotrophic) and ciliates, suggest that concentrations of food sources may be important in determining the abundance of protists in the studied lakes.

Bacterial Community and Biological Nitrate Removal: Comparisons of Autotrophic and Heterotrophic Reactors for Denitrification with Raw Sewage

  • An autotrophic denitrification reactor (ADR-l) and a heterotrophic denitrification reactor (HDR-2) were operated to remove nitrate and nitrite in an anoxic environment in raw sewage. The $NO_3$-N removal rate of ADR-l was shown to range from 52.8% to 78.7%, which was higher than the $NO_3$-N removal rate of HDR-2. Specific denitrification rates (SDNR) of ADR-l and HDR-2 were 3.0 to 4.0 and 1.1 to $1.2\;mgNO_3$-N/gVSS/h, respectively. From results of restriction fragment length polymorphism (RFLP) of the 16S rRNA gene, Aquaspirillum metamorphum, Alcaligenes defragrans, and Azoarcus sp. were $\beta$-Proteobacteria that are affiliated with denitritying bacteria in the ADR-l. Specifically, Thiobacillus denitrificans was detected as an autotrophic denitrification bacteria. In HDR-2, the $\beta$-Proteobacteria such as Denitritying-Fe-oxidizing bacteria, Alcaligenes defragrans, Acidovorax sp., Azoarcus denitrificans, and Aquaspirillum metamorphum were the main bacteria related to denitrifying bacteria. The $\beta$-and $\alpha$-Proteobacteria were the important bacterial groups in ADR-l, whereas the $\beta$-Proteobacteria were the main bacterial group in HDR-2 based on results of fluorescent in situ hybridization (FISH). The number of Thiobacillus denitrificans increased in ADR-l during the operation period but not in HRD-2. Overall, the data presented here demonstrate that many heterotrophic denitritying bacteria coexisted with autotrophic denitrifying bacteria such as Thiobacillus denitrificans for nitrate removal in ADR-l. On the other hand, only heterotrophic denitritying bacteria were identified as dominant bacterial groups in HDR-2. Our research may provide a foundation for the complete nitrate removal in raw sewage of low-COD concentration under anoxic condition without any external organic carbon or the requirement of post-treatment.

Bacterial Community Variations in Hot Pepper-Sown Soil Using FAME Analysis as an Indicator of Soil Quality

  • The bacterial compositions of seven hot-pepper sown soil were compared in this study. From the 624 isolates, 95 species and 49 genera were identified by fatty acid methyl ester analysis (FAME). The FAME results of seven soil showed two distinct clusters for aerobic and Gram-negative bacteria in the high productivity and monoculture soil samples. While Arthrobacter ($17\%$), Kocuria ($11\%$), Pseudomonas ($8\%$), and Bacillus ($8\%$) were predominant among bacteria which were cultured on heterotrophic (YG) agar medium, Pseudomonas ($56\%$), Stenotrophomonas ($16\%$), and Burkholderia ($8\%$) were predominant on crystal violet agar medium. Shannon Weaver indices (H) indicated that colonies obtained from heterotrophic agar medium (3.1) were found to be more diverse than those obtained from the crystal violet media (1.9). The results suggest that FAME analysis may be a potential indicator for of soil quality.

통영 바다목장 해역의 종속영양세균 군집의 연차적 분포 (Annual Distribution of Heterotrophic Bacterial Community in the Marine Ranching Ground of Tongyeong Coastal Waters)

  • [ $2003{\sim}2007$ ]년 통영 바다목장 해역 5개 정점을 대상으로 표층수와 저층수의 종속영양세균의 개체수를 측정하고, 종속영양세균 군집의 종조성 및 우점종을 분석하였다. 종속영양세균은 겨울이나 봄 및 가을에 비하여 해수의 온도가 높은 여름에 더 많이 검출되었으며, 정점별 변화는 특별한 변화 경향이 없었다. 2003년 10월 및 2004년 9월에는 태풍과 집중호우의 영향으로 표층수와 저층수의 개체수가 큰 차이를 나타내지 않거나 저층수의 개체수가 표층수보다 오히려 더 많은 경우도 있었다. 통영 바다목장 해역의 종속영양세균 개체수는 해를 거듭할수록 감소하는 추세를 나타내었다. 종속영양세균 군집의 종조성은 Pseudomonas fluorescens TY1, Pseudomonas stutzeri TY2, Acinetobacter lwoffii TY3, Sphingomonas paucimobilis TY4, Burkholderia mallei TY5, Pasteurella haemolytica TY6, Pasteurella multocida TY7, Comamonas acidovorans TY8, Actinobacillus ureae TY9 및 Chryseobacterium indologenes TY10으로 조사되었고, P. fluorescens TY1과 A lowffii TY3이 우점종으로 분석되었다.

  • 낙동강 하구생태계에 분포한 종속영양세균의 환경 변화에 따른 군집구조의 변화를 파악하기 위해 총 858 균주를 분리하여 수리학적 방법에 의해 분석하였다. 종의 다양성(H') 은 1.54-3.49 로, 하구언 건설 전의 수치와 큰 차이가 없었으나, 균등성지수(evenness index)(J') 는 0.31-0.80 으로 감소하였다. Physiological tolerance index 중 수온에 대한 지수(P') 는 수심이 얕은 정점에서, pH 와 염분에 대한 $P_{h}$$P_{s}$ 는 담수와 해수가 교차하는 정점에서 높았다. 우점의 cluster 를 속 (genus) 수준에서 동정한 결과 Aeromonas, Vibrio, Pseudomonas, Acinetobacter-Morexella, Alcaligenes, Flavobacterium, Micrococcaceae, Enterobacteriaceae 등으로 분류되어 하구언 축조 전과 동일한 종류의 세균뷴포를 나타냈으나 그 출현비율은 달랐다. 이와 같은 결과로 미루어 하구언 축조 등에 따른 낙동강 하구역의 환경변화는 분포세균의 종류보다는 생리적 특셩에 영양을 미쳐 특정종의 우점을 나타나게 한 것으로 사료된다.
