Phytotoxic Effect of 5-Aminolevulinic Acid, a Biodegradable Photodynamic Biomaterial, on Rice and Barnyardgrass

  • ALA (5-aminolevulinic acid) has been proposed as a tetrapyrrole-dependent photodynamic herbicide by the action of the protoporphyrinogen IX oxidase (Protox IX). A study was conducted to determine photodynamic herbicidal effect of ALA on seedling growth of rice (Oryza sativa L.) and barnyard grass (Echinochloa crus-galli Beauv. var. oryzicola Ohwi) under dry and wet conditions. ALA effect on early plant growth of rice and barnyardgrass was greatly concentration dependant, suggesting that it promotes plant growth at very low concentration and inhibits at high concentration. No significant difference in herbicidal activity of biologically and synthetically produced ALAs on plant lengths of test plants was observed ALA exhibited significant photodynamic activity regardless of PSDIP and its duration. Significant shoot growth inhibition by ALA soaking treatment exhibited apparently, indicating that ALA absorbed through root system was translocated into shoot part of plants. ALA reduced plant heights of rice and barnyardgrass seedlings by 6% and 27%, respectively, showing more tolerant to ALA in rice under wet condition. Leaf thickness was reduced markedly by ALA with increasing of ALA concentration, due to mainly membrane destruction and severe loss of turgidity in mesophyll cells, although the epidermal was little affected. It was observed that photodynamic herbicidal activity of ALA applied by pre-and post-emergence application exhibited differently on plant species, and that the activity of ALA against susceptible plants was highly correlated with growing condition.

Effects of Ditching on Seedling Stand in Wet Direct Seeding Rice Culture

  • In order to develop more stable seedling stand method in wet direct rice seeding culture, the effect of making the drainage ditches was studied in both methods of broadcast seeding on floody paddy surface and puddled-soil drill seeding. In a broadcast seeding on floody paddy surface, the ditching after seeding using a tractor or small ditch maker improved the seedling stand, and reduced the floating seedling and herbicide injury by accelerating the drainage. Suitable ditching time was at 2 days after seeding (DAS) for the tractor and at 3 DAS for the ditch maker. In the puddled-soil drill rice seeding culture, the ditching before seeding with a ditch maker at 3 days after draining effectively improved the seedling stand due to quick draining through well formed the seeding furrows. In the puddled-soil drill seeding, seedling stand number showed higher in both ditching plots synchronized with seeding compared with both only draining treatment at 1 DAS without ditching and the flooding plot condition for 6 DAS. And the suitable ditching depth was 6 cm, as considered the ditching status and drained status. These results suggest that the ditching in wet direct rice seeding is an resonable practice for improving the seedling stand through the accelerating drainage of field.

A Survey on the Pesticide Use and Perceptions about the Hazards of Pesticiedes among the Farmers in Kyoungju Area, Korea (일부 경주지역 농민의 농약사용실태 및 농약의 위험성에 대한 인식 조사)

  • This study was done for the purpose of investigating the status of the use and perceptions of pesticides among the farmers who spray them in person. And it was also done to provide the basic statistics for the Agricultural Health Policy and study. Over the part of agricultural area in Kyungju and Ulsan City, Korea, from July to September 1999, 1032 questionnaires were distributed and 561 of them were collected and 447 were analysed. Major results of this study are as follows. The proportion of those who have special location for pesticides storage was only 5.3%, and that of those who lock hte storage location was only 13.6%. As to the treatment of bottles after use, most common response was 'burn in home'(47.7%). Most farmers purchase pesticides through 'pesticide store' or 'Agricultural Cooperatives'. The knowledge and Usage of pesticides depends in large part on experiences. The farmers who cultivate orchard spray pesticides more frequently than those cultivate paddy rice mainly. Most common pesticides in use are oganophosphates and carbamates. And also the pesticides that contains so-called 'Environmental Hormone' are used. The perception about the hazards of pesticides is rather superficial and insufficient. 'Herbicide(Gramoxon·Paraco)'(38%) is accepted as the most hazardous by farmers.

Weeding Hypothesis on Direct Seeding Rice Field as Applied by the Old Firing and Water Dressing Method (고대 화경수누법(火耕水?法)을 응용한 직파 논[直播畓] 제초법)

  • It was a method of weed removing, called as whagyeongsoonoobeob. Namely, the Hwanoobeob was the weeding method came from "Jeminyosool" in old China, and had been descended to "Wibinmyeongnonggi", "Nonggajibseong" and "Gwanongsocho" in the $16\sim18^{th}$ century of our country. The method was applied to rice paddy after applying direct-seeding, burning by unit before watering to remove weeds and to save rice plants. It would be applied with a method of using non-selective herbicide such as paraquat and oxyfluorfen, or radiation treatment, as a simple and safe weed removing method.

Difference in Physiological Responses to Environmental Stress in Protox Inhibitor Herbicide-Resistant Transgenic Rice and Non-transgenic Rice (Protox 저해형 제초제 저항성 형질전환벼와 비형질전환벼의 환경스트레스에 대한 생리적 반응 차이)

  • The objective of this research was to confirm the difference in physiological responses to environmental stresses such as chilling, high temperature, NaCl, and chemical stress (paraquat) in Protox inhibitor resistant-transgenic rice (MX, PX, and AP37) and its non-transgenic counterpart (WT). Transgenic and non-transgenic rice plants were exposed to a chilling temperature of $5^{\circ}C$ for 1 day or a high temperature of $45^{\circ}C$ for 4 days and allowed to recover at $25^{\circ}C$ for 6 days after the chilling treatment or 8 days after the high temperature treatment. Leaf injury, shoot fresh weight, porphyrin biosynthesis substances, and chlorophyll content were investigated in transgenic and non-transgenic rice at 6 days after 0.5% and 1% NaCl treatments or at 5 days after 0~300 ${\mu}M$ paraquat treatments. No significant difference in leaf injury and shoot fresh weight were observed between transgenic and non-transgenic rice during chilling and recovery. Plant height and shoot fresh weight were also similar between transgenic and non-transgenic rice during the high temperature and recovery period (0~5 days). However, plant height and shoot fresh weight in transgenic rice line MX and PX were lower than in non-transgenic rice at 6 days for recovery. Leaf injury, chlorophyll, and Mg-Proto IX ME contents had no significant difference between transgenic rice and non-transgenic rice after NaCl treatment, but Proto IX content for AP37 and shoot fresh weight for PX and AP37 in 0.5% NaCl treatment were significantly reduced compared with non-transgenic rice. There was no difference in leaf injury and shoot fresh weight when comparing transgenic rice and non-transgenic rice after paraquat treatment. Although transgenic rice and non-transgenic rice showed a little difference at a particular measurement period in certain environmental stresses, there was generally no difference in physiological responses between transgenic rice and non-transgenic rice.

Different Physiological Activity of Selected Rice Cultivars to Diphenylether Herbicide, Oxyfluorfen - III. Differential Protoporphyrinogen Oxidase(Protox) Activity and Protoporphyrinogen IX(PPIX) Accumulation (Oxyfluorfen에 대한 내성(耐性) 및 감수성(感受性) 벼품종(品種)의 생리활성(生理活性) 기구 (機構) - III.   • This study was conducted to investigate the inhibition of protox activity and the PPIX accumulation of the oxyfluorfen-tolerant and-susceptible rice cultivars with barnyardgrass, a typical susceptible weed in accordance by oxyfluorfen treatment. The susceptible rice cultivars and barnyardgrass showed more inhibition of protox activity due to the treatment of oxyfluorfen than the tolerant rice cultivars. Especially in the concentration at $10^{-6}$M treatment, protox activity of the susceptible rice cultivars and barnyardgrass were the completely inhibited but the tolerant rice cultivars kept 32~59% of activity compared to the control. As the treatment concentration increased, the content of PPIX accumulation increased and it increased untill four hours of light exposure but it tended to decrease these after. The content of PPIX accumulation by the treatment of oxyfluorfen was more pronounced in the light condition than in the dark. Under the light and dark conditions, the susceptible rice cultivars and barnyardgrass showed more PPIX accumulation than the tolerant rice cultivate. Especialiy the susceptible barnyardgrass had more than the rice. With the treatment of GC and DA, tetrapyrrole biosynthesis inhibitor, the herbicidal activity by oxyfluorfen was inhibited, and the susceptible rice cultivars and barnyardgrass tended to have less effective than the tolerant rice cultivars and the content of chlorophyll or PPS accumulation tended to be similar.

Effects of Hot Water and Specific Gravity Treatment on Germination of Legumes and Green Manure Crops (열탕처리와 염수선 처리가 콩과 녹비작물 발아에 미치는 영향)

  • Green manure crops have a variety of benefits, including improving soil fertility, removing soil salinity, enhancing soil aeration and moisture regulation, reducing the use of chemical fertilizers, reducing nitrous oxide emissions, reducing herbicide use through weed-suppressive effects, promoting agricultural ecosystem protection and carbon dioxide absorption capacity. To find ways to increase the germination rate of legumes and green manure crop seeds, which are imported in large quantities every year. It was carried out that two seed priming methods, osmotic priming and heat treatment, and compared their effects. Heat treatment was treated for 10 minutes at 40℃ or 60℃, followed by rinsing with water. Osmotic priming was applied for 30 seconds in a saline solution with a specific gravity of 1.13. Overall, there was no significant difference in the final germination rate, but it was found that osmotic priming and heat treatment affected the germination speed. However, applying heat treatment and osmotic priming simultaneously did not affect the germination characteristics. Therefore, it is suggested that heat treatment and osmotic priming can increase the germination speed of soybean and green manure crops.

Differential herbicide response of sulfonylurea-resistant Monochoria vagnalis accessions to sulfonylurea herbicides (서로 다른 지역에서 채집된 Sulfonylurea계 제초제 저항성 물달개비의 제초제 반응 차이)

  • Four sulfonylurea(SU)-resistant Monochoria vaginalis(M. vaginalis) accessions were tested for levels of resistance to four SU herbicides which have been widely using in paddy fields of Korea, based on whole plant response and sensitivity of the target enzyme, acetolactate synthase(ALS). The resistant Naju, Nonsan and Gimje accessions were not affected to the survival by treatment with recommended dose of all SU herbicides tested. The $GR_{50}$ values for the Naju, Nonsan and Gimje accessions were 8- to 33-fold, 8- to 30-fold and 7- to 32-fold higher to recommended doses of all SU herbicides tested than the susceptible Cheongdo accession, respectively. However, the $GR_{50}$ values for Kimhae accession displayed an intermediate response and was only 4-to 13-fold more resistant than the susceptible accession. The ALS $I_{50}$ values for the Naju, Nonsan and Gimje accessions were 25- to 66-fold, 9- to 26-fold and 10- to 24-fold higher to recommended doses of all SU herbicides tested than the susceptible Cheongdo accession, respectively. However, the $I_{50}$ value for Kimhae accession was 4- to 9-fold more resistant than the susceptible accession, as determined by $I_{50}$ values of ALS.

Effect of Herbicide 'Kerb' on Growth of Perennial Ryegrass (Lolium perenne L. 'Accent') and Zoysiagrass (Zoysia japonca 'Zenith') under Shading Treatments (차광조절하에서 제초제 Kerb가 퍼레니얼 라이그래스와 한국잔디의 생육에 미치는 영향)

  • In order to investigate an artificially transition method of perennial ryegrass(Lolium perenne L. 'Accent') overseeded on zoysiagrass (Zoysia japonca Zenith') with Kerb (3,5-jichloro-N (1,1-dimethyl-2-propynyl). We observed turfgrass growth and physiological responses of the two turfgrasses under the shading treatments. The experiment was conducted in greenhouse during April- August. Shading was generally using a black shade net product whose the shade level was done into 0, 35 and 75%, actual light intensities were 589, 378 and $135\;{\mu}mol{\cdot}m^{-2}{\cdot}s$ PAR, respectively. The four levels of Kerb treatment were 0, 0.1 0.2, and $0.4\;g{\cdot}m^{-2}$. As shading was getting increased, growth of perennial ryegrass was increased slowly, but zoysiagrass was reduced and almost all of experiments showed significant differences statistically. Perennial ryegrass exhibited inclination that growth was reduced in more than Kerb $0.4\;g{\cdot}m^{-2}$ by shades. On the other hand, it was shown that the growth of zoysigrass was reduced by shades largely and it was not effected by Kerb. Through this study, it was determined that Kerb is effective for elimination of overseeded perennial ryegrass on zoysiagrass. Moreover, when excluding perennial ryegrass under the condition like shade, it is desirable for treating high concentration more than the basic processing amount.

Analytical Method of Bentazone Residue in Agricultural Commodities Using HPLC-UVD/MS (HPLC-UVD/MS를 이용한 농산물 중 bentazone의 분석법 확립)

  • Bentazone is benzothiadiazole group herbicide, and used to foliage treatment. This herbicide have already been widely used for cereals and vegetables planting in worldwide. This experiment was conducted to establish a determination method for bentazone residue in crops using HPLC-UVD/MS. Bentazone residue was extracted with acetone (adjusted pH 1 with phosphoric acid) from representative samples of five raw products which comprised hulled rice, soybean, apple, green pepper, and Chinese cabbage. The extract was diluted with saline water, and dichloromethane partition was followed to recover bentazone from the aqueous phase. Florisil column chromatography was additionally employed for final clean up of the extract. The bentazone was quantitated by HPLC with UVD, using a YMC ODS AM 303 ($4.6{\times}250$ mm) column. The crops were fortified with bentazone at 3 levels per crop. Mean recovery ratio were ranged from 82.0% for a 0.2 mg/kg in apple to 97.9% for a 0.02 mg/kg in Chinese cabbage. The coefficients of variation were ranged from 0.5% for a 0.02 mg/kg in soybean to 9.7% for a 0.02 mg/kg in Chinese cabbage. Quantitative limit of bentazone was 0.02 mg/kg in representative five crop samples. A LC/MS with selected-ion monitoring was also provided to confirm the suspected residue. Therefore, this analytical method was reproducible and sensitive enough to determine the residue of bentazone in agricultural commodities.
