Effects of Ethanol-extract of Allium wageki on the Renal Function of Fructose-induced Hypertensive Rats (쪽파 에탄올 추출물이 과당 유도 고혈압 백서의 신장 기능에 미치는 영향)

  • The present study was aimed to investigate whether ethanol-extract of Allium wageki has an ameliorative effect on the renal function in high fructose-diet induced hypertensive rats .. The urine osmolality (Uosmol) was decreased in rats with high fructose-diet (60%) during the whole experiment period without change of the urine volume (UV). The urinary excretion of sodium (UNaV) and chloride (UCIV) were decrease significantly in rats with fructose-induced hypertensive rats, whereas urinary excretion of potassium (UKV) was Increased. The creatinine clearance (Ccr) and solute-free water reabsorption were also decreased by treatment of fructose-rich diet. Among these renal functional parameters, Ccr was partially restored by the administration of ethanol-extract of Allium wageki. The Uosmol was also partially restored by the administration ethanol-extract of Allium wageki at the end of the experimental period. Taken together, ethanol-extract of Allium wageki has the ameliorative effect on glomerular filtration rate in rats with high fructose-diet induced hypertension.

A Study on the Regulation of Endometrial and Placental Cell Function by Water Extract of 3 Types of Herbal Medicines and Ethanol Extract on Scutellariae Radix (3종 한약 처방 물 추출물과 황금 에탄올 추출물에 의한 자궁내막과 태반세포 기능조절 연구)

  • Methods: We investigated the ability to induce decidualization of human endometrial stromal cells and invasive ability of human trophoblast cells by water extract of three types herbal medicines (Dangguijakyak-san, Siryung-tang, Antae-eum) and ethanol extract on Scutellariae Radix. Results: In the process of decidualization of endometrial stromal cells, three herbal medicines including Dangguijakyak-san, Siryung-tang, and Scutellariae Radix increased the production of decidual markers such as prolactin (PRL) and insulin-like growth factor-binding protein 1 (IGFBP1). However, Antae-eum increased mRNA levels of PRL and IGFBP and secretion of IGFBP in decidual stromal cells, but not PRL. Four herbal medicines inhibited the invasion of trophoblast cells. Conclusions: Four herbal medicines may play a role in implantation in women with reproductive failures. However, further studies are warranted to elucidate whether these medications are helpful in the maintenance of pregnancy.

  • Objectives : This study was carried out to investigate the acute toxicity and safety of Palmultang and Fermented Palmultang Extract in Mice. Methods : To evaluate their acute toxicity and safety, 0 (control group), 2000 mg/kg of Palmultang and Fermented Palmultang Extracts were orally administered to 15 male and 15 female ICR mice. After a single administration, we observed survival rates, behavioral pattern, clinical sign, body weight. The results of biochemical analysis and hematological analysis were no any significant change. Results : Compared with the control group, we could not find any toxic alteration in all treated mice. Conclusions : Overall, the results suggest that, the oral administration of Palmultang and Fermented Palmultang extracts did not produce significant toxic effect in mice. Hence, the fermented extract can be utilized for herbal therapy.

Acute Toxicity Study on Insampaedok-san Extracts in Mice (ICR 마우스를 이용한 인삼패독산(人蔘敗毒散)의 급성독성 연구)

  • Objective : This study was carried out to investigate the acute toxicity and safety of Insampaedok-san extract in ICR Mice. Methods : SPF ICR male and female mice were administered orally with Insampaedok-san extract of 0 (control group), 1250, 2500 and 5000 mg/kg. After single administration, we daily examined number of deaths, clinical signs, gross findings and changes of body weight for 14 days. Hematological parameters and isolated organ weights were determined after 14 days of administration. Results : No dead animal and no significant changes of body weights were found during experimental period. In addition, no differences were found between control and all of treated groups in clinical signs, organ weights and hematology, and other findings. Conclusions : Insampaedok-san extract did not show any toxic effects and oral LD50 values of the extracts was over 5000 mg/kg in ICR mice.

Study on the Anti-microbacterial Activity, Anti-inflammatory and Anti-allergic Effects of Several Herb-Extract (수종의 한약 복합 추출물의 항균, 항염 및 항알레르기 효과에 대한 실험적 연구)

  • Herbal mixture water extract of (Chrysanthemum morifolium, Portulaca oleracea, Sanguisorba officinalis, Sophora flavescens, Phellodendron amurense, Cnidium monnieri) which exhibit several beneficial effects including acne and skin diseases, was tested for anti-microbial activity and anti-inflammation effects. The herbal mixture extract showed antimicrobial activity against Stapylococcus epidermis, and Propionibacterium acne. The growth of Stapylococcus epidermis, and Propionibacterium acne was inhibited completely by addition of 1.0% of the extract. Also in the present study we examined the mixture extract on compound 48/80 induced allergy and LPS induced cyclooxygenase-2(COX-2) gene expression in RAW 264.7 macrophage. The results indicated the ear swelling and histamine release induced by compound 48/80 were dose-dependently reduced, ranging 28-60%, and 48-72% , respectively. Furthermore the extract inhibited the expression of LPS-induced COX-2 proteins and mRNAs without an appreciable cytotoxic effects on RAW 264.7 cells. The LPS-induced cytokine gene expression including IL-$1{\beta}$, TNF-$\alpha$, and IL-6 were dose-dependently suppressed by the mixture extract. Based on these results, it is concluded that the herbal mixture water extract can be applied to the acne and skin diseases therapy.

Antioxidative Activity and Nitrite Scavenging Ability of Ethyl Acetate Extract from Acanthopanax sessiliflorus (오가피 메탄올 추출물의 항산화 효과 및 아질산염 소거작용)

  • Efficacy of antioxidative activity and nitrite scavenging ability of methanol extract from Acanthopanax sessiliflorus was investigated. Electron-donating ability of extract at $RC_{50}$ was 29.93 ${\mu}g/mL$. After addition of 0.96 mg/mL extract, autooxidation of pyrogallol decreased to 22.85% by superoxide dismutase-like activity. In antioxidative activity of extract against linoleic acid during incubation times of 24, 48, 96 hours at $40^{\circ}C$, lipid peroxidation values significantly decreased by 48.89%, 45.0%, 46.34% with addition of 0.2 mg/mL, respectively. Total phenolic content was determined as gallic acid equivalents (GAE) and values revealed $410.25{\pm}4.74$ GAE ${\mu}g/mg$ of extract. Nitrite scavenging ability showed the most remarkable effect at pH 1.2, exhibited to 88.3% by addition of 0.2 mg/mL. These results suggest that methanol extract from A. sessiliflorus can be used as bioactive and functional material.

Review of the Studies on the Influence of Experimental Arthritis Model of Herbal Medicine Extract (한약 추출물이 실험적 관절염 모델에 미치는 영향에 대한 연구 분석)

  • Objectives The aim of this study was to analyze the herbal prescription and composition of the leading experimental studies of herbal medicine extract in arthritis model and to analyze the way of setting model and the way of validating effect of the leading experimental studies of herbal medicine extract in arthritis model. Methods The Subjects were selected by select criteria and exclusion criteria from January 1, 2000 to September 30, 2017. The herbal prescription and composition of the selected subjects were analyzed. The effects and statistical significance of selected studies were verified by comparing the control and treatment groups of frequently used cytokines (tumor necrosis factor $[TNF]-{\alpha}$, interleukin $[IL]-1{\beta}$, IL-6). Results 52 kinds of herbal prescriptions and 118 kinds of individual herbs were used in 64 studies. Among the 52 kinds of herbal prescriptions, Daeganghwal-tang (4 times) was used most frequently. Among the 118 kinds of individual herbs, Glycyrrhizae Radix (49 times), Angelicae Giantis Radix (34 times), Osterici Radix (24 times) were used most frequently in order. The prescription that has the highest $TNF-{\alpha}$ decline is Changchooldoin-tang. The prescription that has the highest $IL-1{\beta}$ decline is Banggibokryeong-tang. The prescription that has the highest IL-6 decline is Gwanjeol8ho-bang. Conclusions Daeganghwal-tang (herbal prescription) and Glycyrrhizae Radix, Angelicae Giantis Radix, Osterici Radix (individual herbs) were used most frequently. The prescriptions that have the highest cytokines decline were Changchooldoin-tang, Banggibokryeong-tang and Gwanjeol8ho-bang.

  • Objectives : To get to know the effects of placental extract and laser theraphy for Temporomandibular Joint Disorder, we have tried a injection of placental extract and laser theraphy into 聽宮(SI19), 聽會(GB02) for patient with Temporomandibular Joint Disorder. Methods : In this case clinical assessment of temporomandibular joint dysfunction is assessed by the VAS score and the Korean Version of Research Diagnostic Criteria for Temporomandibular Disorders(RDC/TMD). Results : The VAS score was significantly decreased after Placental extract herbal acupuncture and laser theraphy Conclusion : Placental extract herbal acupuncture and laser theraphy effectively resolved pain for Temporomandibular Disorders.

Study of Skin Depigmenting Mechanism of the Ethanol Extract of Fagopyrum esculentum (교맥 에탄올 추출물의 피부 미백기전 연구)

  • The aim of this study was to investigate the effect of ethanol extract of Fagopyrum esculentum on the melanogenesis. To determine whether ethanol extract of Fagopyrum esculentum suppress melanin synthesis in cellular level, B16F10 melanoma cells were cultured in the presence of different concentrations of Fagopyrum esculentum ethanol extract. In the present study, we examined the effects of Fagopyrum esculentum ethanol extract on cell proliferation, melanin contents, tyrosinase activity, expression of melanogenic enzyme proteins including tyrosinase, tyrosinase-related protein 1 (TRP-1) and tyrosinase-related protein 2 (TRP-2). Cell proliferation was slightly increased by treatment with ethanol extract of Fagopyrum esculentum $(25-200 {\mu}g/m{\ell}).$ The ethanol extract of Fagopyrum esculentum effectively suppressed melanin contents at a dose of $100 {\mu}g/m{\ell}).$ It was observed that the color of cell pellets was totally whitened compared with the control. The ethanol extract of Fagopyrum esculentum inhibited tyrosinase activity, regulate melanin biosynthesis as the key enzyme in melanogenesis. Using western blot analysis, the ethanol extract of Fagopyrum esculentum dose-dependently decreased tyrosinase and TRP-1 protein levels, and tyrosinase and TRP-1 were detected in similar manner. ${\alpha}-MSH$ leads to a stimulation of melanin synthesis through increase of tyrosinase activity, melanin contents and cytoplasmic dendricity. In this study, ethanol extract of Fagopyrum esculentum down-regulated the ${\alpha}-MSH$-induced tyrosinase activity, melanin contents and cytoplasmic dendricity. Regarding protein levels of the melanogenic enzymes, the amounts of tyrosinase and TRP-1 was increased after incubation with a-MSH. The treatment of ethanol extract of Fagopyrum esculentum decreased the ${\alpha}-MSH$-induced expression levels of tyrosinase and TRP-1. These results suggest that the ethanol extract of Fagopyrum esculentum exerts its depigmenting effects through the suppression of tyrosinase, TRP-1 and cytoplasmic dendricity. And it may be a potent depigmetation agent in hyperpigmentation condition.

The Efficacy of Ampule Containing Herbal Extract for Improving Skin Wrinkles in Women (한약재 추출물 함유 앰플의 주름개선 효능에 관한 기초 임상연구)

  • Objectives : This study was performed to evaluate the efficacy of ampule containing herbal extract(Ulmus davidiana, Ginkgo biloba, Perilla ocymoides, Morus alba, Glycyrrhiza uralensis (licorice), and Angelica gigas) on skin wrinkles in adult women with facial wrinkles. Methods : A total of 12 women, 30 to 46 years of age, with wrinkles in the corner of eyes, applied ampule containing herbal extract twice daily for up to 12 weeks. Silicon replicas of right crow's feet area and antecubital fossa were taken before use of test product and at 4, 8 and 12 weeks. The replicas were analyzed by optical profilometry with Skin Visiometer SV600. The wrinkle and roughness parameters, R1, R2, R3, R4 and R5 were calculated and statistically analyzed. In addition, a subjective evaluation of product efficacy was conducted by patient's assessment. Results : The mean values of all the skin roughness parameters were decreased at 4-week, and decreased significantly at 8 and 12-week except R4 at 12-week after test product use. The subjective evaluation of wrinkle by patient's assessment was also improved. However, one patient noted stiffness of face after use of test product. Conclusions : These study suggested that the application of ampule containing herbal extract twice daily for 12 weeks may effectively improve the facial wrinkles without severe side effect.
