Thioacetamide 유발 흰 쥐 간독성에 대한 인삼 사포닌 및 에타놀 추출물의 효과 (Effects of Saponin and Ethanol Extract of Panax Ginseng against Thioacetamide - Induced Hepatotoxicity in Rats)

  • Panax ginseng has been used for various diseases including hepatic disorders. The aim of the present study was to investigate the hepatoprotective effects of ethanol extract and saponin of Panax ginseng in thioacetamide-intoxicated rats and to compare with silymarin, a known hepatoprotective agent. Male Sprague-Dawley rats were given single intragastric administration of thioacetamide. Aqueous solutions of ethanol extract and saponin of Panax ginseng with or without silymarin were administered intragastrically daily for six days from four days before until one day after thioacetamide administration. At the end of the treatment, the rats were fasted overnight and sacrificed. As a result, thioacetamide caused significant increase in serum levels of AST, ALT, 5'-nucleotidase and bilirubin. Thioacetamide increased $Ca^++$ content but decreased protein content in liver tissue. These thioacetamide-induced biochemical changes were prevented both by ethanol extract of ginseng and silymarin, but not by ginseng saponin. Silymarin did not potentlate the effect of either ethanol extract or saponin of ginseng on these parameters. Thioacetamide-induced confluent necrosis was not protected by the test drugs. In conclusion, ethanol extract of ginseng protects the liver possibly by stabilizing the cell membrane and by inhibiting thioacetamide-induced $Ca^++$ increase in the hepatocytes, which was comparable to that of silymarin.

Hepatoprotective Effects of Allium monanthum MAX. Extract on Ethanol-Induced Liver Damage in Rat

  • This study investigated the effects of an ethanol extract of Allium monanthum MAX. (AME) on ethanol-induced hepatotoxicity in rat liver. Sprague-Dawley rats weighing 100~150 g, were divided into 5 groups; normal group (NOR), AME 200 mg/kg treated group (S1), ethanol (35%, 10 mL/kg) treated group (S2), AME 200 mg/kg and ethanol (35%, 10 mL/kg) treated group (S3) and AME 400 mg/kg and alcohol (35%, 10 mL/kg) treated group (S4). AME was fractionated by the following solvents: n-hexane, chloroform, EtOAC and n-BuOH. Antioxidant index of the n-BuOH fraction was 600 ppm, highest among fractions. The growth rate and feed efficiency ratio were decreased by ethanol, but gradually increased to the corresponding level of the normal group by administering AME. The serum ALT activities that were elevated by ethanol were significantly decreased by AME administration. It was also observed that the hepatic activities of SOD, catalase, xanthine oxidase and GSH-Px that were increased by ethanol were also markedly decreased in the AME treated group with compared to ETB. These results suggest that ethanol extracts of Allium monanthum MAX. may have a protective effect on ethanol-induced hepatotoxicity in rat liver.

Antihepatotoxic and Antioxidant Activities of Polysaccharide Obtained from Cultured Mycelia of Ganoderma lucidum

  • The purpose of this study was to observe the effects of the polysaccharide (GLP) obtained from the liquid cultured Ganoderma lucidum on the lipidperoxidation in a rat liver microsome and hepatotoxicity in the primary cultured rat hepatocytes. It is well known that the polysaccharide of Ganoderma lucidum exhibits hepatoprotective activity, antitumor activity etc., which many suggest a relationship to lipidperoxidation. The effect of GLP on $CCl_4-$ and galactosamineintoxicated cytotoxicity in the primary cultured rat hepatocytes were reduced the GPT value. In order to the estimate the effects of anti-lipidperoxidation of the polysaccharide, enzymatic and nonenzymatic reaction assays were performed, in vitro, in the rat liver microsome. An enzymatic lipidperoxidation reaction by $ADP+FeCl_3+NADPH$ and $CCl_4+NADPH$, GLP (1 mg/mL) inhibited 77.4% and 39.4%, respectively, and the nonenzymatic reaction displayed a 97.4% strongly inhibition. In the enzymatic and nonenzymatic inducers treated with GLP, the formation of malondialdehyde (MDA) progressively decreased by raising the GLP concentration. These results suggest that the anti-lipidperoxidation and radical scavenging activity of GLP may play an important part in the liver protection action.

Hepatoprotective Effect of Grifola frondosa Water Extract on Carbon Tetrachloride-induced Liver Injury in Rats

  • The present study aimed at assessing the protective effect of water extract from fruit body of the Grifola frondosa (GFW) on carbon tetrachloride ($CCl_4$)-induced hepatotoxicity. Rats orally administered with GFW 0.5, 1.0, 2.0 g/kg for 14 days were treated with $CCl_4$ to induce hepatotoxicity. Pretreatment with GFW remarkably prevented the elevation of serum AST, ALT, ALP, LDH, $\gamma$-GTP, and liver lipid peroxides in $CCl_4$-treated rat and GFW administration in liver injured rats by $CCl_4$ showed significant (p<0.05) protection of liver as evidenced from normal serum enzymes and malondialdehyde (MDA) levels. In the ultrastructural changes, administration of $CCl_4$-induced damage of hepatocytes with vacuolation, a highly damaged endoplasmic reticulum, and degenerating nuclei. However, pre-administration with GFW preserved normal ultrastructure of hepatocytes. These results suggest that GFW had an effect to inhibit $CCl_4$-induced liver injury in rat, and that it could be used as an effective hepatoprotective agent against chemical-induced liver damage.

애엽 에탄올 추출물이 사염화탄소로 유발된 흰쥐의 간 손상 보호효과 (Protective Effect of Ethanol Extract of Artemisiae vulgaris L. on hepatic injury Induced by Carbon tetrachloride In Rat.)

  • Dawley 계 숫컷 흰쥐(200~210g)를 정상군, CCl4-대조군, CCl4-실험군으로 나누어 실험군은 애엽 에탄올 추출물을 1,000mg/kg,b.w의 용량으로 1일1회7일간 경구 투여 후 사염화탄소를 0.6mg/kg,b.w의 용량으로 복강내 주사 후 다음날 개복하여 혈청내의 Alanine aminotransferase(ALT), Aspartate aminotransferase(AST),Alkaline phosphatase(ALP), Glutamyltranspeptidase(γ-GT), Lactate dehydrogenase(LDH)의 활성도와 중성지방, 콜레스테롤 함량을 측정한 결과 애엽 추출물 투여군에서 CCl4-대조군과 비교하여 유의적인 감소를 나타내었으며, HDL-콜레스테롤은 유의적인 증가를 나타내었다. 간조직중의 지질과산화 함량은 감소를 glutathione함량은 유의적인 증가를 나타내었다 이 실험 결과 애엽 에탄올 추출물은 사염화탄소 투여에 의한 간 손상을 억제하는 보호물질을 함유하고 있음을 알 수 있었다.

구기자 복합물 약침액이 간기능 개선에 미치는 영향 (Protective Effect of Herbal Mixture Including Lycii Fructus on Hepatotoxicity Induced by Thioacetamide in Mice)

  • Objectives : This study investigated the hepatoprotective effect of herbal mixture including Lycii fructus (HML) in thioacetamide (TAA)-induced hepatotoxicity in mice. Methods : To confirm the liver protective effect, induced by TAA for 3 days injection at 100 mg/kg mice, HML were treated for 8 weeks at 300 mg/kg/day, 1000 mg/kg/day. Positive control was treated silymarin 50 mg/kg/day after TAA injection. The changes of mortality rate, clinical signs, organ weight, relative liver, blood chemistry and histopathological findings were analyzed after experiment. Results : Body weight gain was observed in all groups, but TAA treated group at 4th week and all treated groups decreased weight compared to the untreated group. As a result of organ weight measurement, organ weight gain due to hepatic injury was observed statistically significantly in TAA-treated group and TAA+Silymarin treated group, and the herbal mixture-treated group showed a tendency to decrease compared to the TAA treated group. Blood biochemistry showed that total cholesterol and very low density lipoprotein cholesterol decreased statistically in TAA+low-dose and high dose herbal mixture treated group compared to the TAA-treated group. Histopathological examination showed that liver abnormalities were not observed in untreated group, liver fibrosis was observed in liver injury with TAA treated and herbal mixture treated group. And, TAA+high dose herbal mixture group showed relaxation tendency on liver calcification compared to the TAA treated group. Conclusions : According to the above results, HML provided hepatoprotective effects on the hepatic injury by reduction of inflammatory responses.

Enzyme hydrolysate of silk protein suppresses tert-butyl hydroperoxide-induced hepatotoxicity by enhancing antioxidant activity in rats

  • The purpose of current study is to investigate the beneficial effect of enzyme (Alcalase) hydrolysates of silk protein in rat. Alcalase-treated silk protein hydrolysate (ATSH) itself did not show any cytotoxicity on the hepatic tissues and blood biochemistry, similar to the normal condition. ATSH played a protective role in tert-butyl hydroperoxide (t-BHP)-induced hepatotoxicity and liver damage. The values of AST (aspartate aminotransferase) and ALT (alanine aminotransferase), which are the indicators of the liver function, were effectively alleviated with the ATSH treatment in a dose dependent manner. The level of Lactate dehydrogenase (LDH) and Malondialdehyde (MDA), which were increased with t-BHP treatment, were significantly reduced by ATSH. High dose of ATSH (2 g/kg) reduced the t-BHP-induced LDH release by 48%. Antioxidant and antioxidant enzymes in liver cells were significantly increased by ATSH treatment in their level and activities. ATSH (2 g/kg) increased glutathione (GSH), an intracelluar antioxidant, by 2.5-fold compared with the t-BHP treated group. The activities of glutathione-s-transferase (GST), superoxide dismutase (SOD), and catalase were also elevated by 38%, 60%, and 45%, respectively, with ATSH (2 g/kg) treatment. The antioxidative effect of ATSH was recapitulated to the protection from t-BHP induced liver damages in hematoxylin and eosin (H&E) staining. Thus, ATSH might be used as a hepatoprotective agent.

산수유의 유리자유기에 의한 간손상 보호효과 (Study on the Protective Effect of Corni Fructus against Free Radical Mediated Liver Damage)

  • Carbon tetrachloride ($CCl_4$)-induced liver injury depends on a toxic agent that has to be metabolized by the liver NAPDH-cytochrome P450 enzyme system to a highly reactive intermediate. Alternations in the activity of cytochrome P450 enzymes affect the susceptibility to hepatic injury from $CCl_4$. In this study, we evaluated the potential protective activity of the traditional Korean medicinal herb, Corni fructus (CF), against an experimental model of hepatotoxicity induced by $CCl_4$. The CF exhibited a hepatoprotective activity against $CCl_4-induced$ liver damage in Sprague-Dawley (SD) rats, as measured by GOT, GPT, ALP and histological observation. The CF also showed significant decrease of malodialdehyde (MDA) and increase of glutathion (GSH), catalase activity in rat liver homogenate. In addition, the expression of CYP2E1, as measured by reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blot analysis, was significantly decreased in the liver of CF treated SD rats. But $CCl_4$ and CF has no significant effect on 1A1 and 3A1 isoform of cytochrome P450. Based on these findings, it is suggested that hepatoprotective effects of CF possibly related to antioxidative effects and regulation of CYP2E1 expression.

랫드 간세포 일차배양에서 셀레늄이 카드뮴에 의해 유도된 독성 및 지질과산화에 미치는 영향 (The Effects of Selenium on Cadmium-Induced Toxicity and Lipid Peroxidation in Rat Hepatocyte Primary Culture)

  • 본 연구의 목적은 랫드 간세포 일차배양에서 카드뮴에 의해 유발된 세포독성 및 지질과산화에 대한 셀레늄의 항산화 및 간보호 효과를 조사하기 위함이다. 이를 위해 2개의 실험을 수행하였다. 실험 1에서는, 1, 10, 50, 100 및 $50\;{\mu}M$의 다양한 농도의 카드뮴으로 6시간 동안 간세포를 일차 배양하였다. 간세포 생존율과 지질과산화는 각각 MTT 방법과 TBARS 방법으로 측정하였다. 항산화 효과와 간보호 효과는 각각 GOT와 GSH-Px 활성을 측정함으로써 결정하였다. 카드뮴은 농도 의존적으로 세포 생존율을 감소시켰으며 지질과산화는 증가시켰다. 세포 생존율과 지질과산화 간에 유의적인 음의 상관관계(r=-0.943, p<0.01)가 관찰되었다. 카드뮴은 $50\;{\mu}M$ 농도에서 GOT의 활성을 증가시켰으나 GSH-Px의 활성은 감소시켰다. 실험 2에서는 셀레늄이 카드뮴에 의해 유발된 독성 및 지질과산화에 대한 효과를 연구하기 위해 $100\;{\mu}M$의 카드뮴과 0.01, 0.1 및 1 ppm의 다양한 농도의 셀레늄으로 6시간 동안 간세포 일차 배양하였다. 세포 생존율과 GSH-Px의 활성은 1 ppm의 셀레늄에 의해 증가되었다. 반면에, 지질과산화와 GOT의 활성은 0.1 ppm의 셀레늄에 의해 감소되었다. 이러한 연구 결과는 셀레늄이 카드뮴에 대한 항산화 및 보호 효과를 나타내 보이고 있다.

감국 발효 열수 추출물의 에탄올에 의해 손상된 HepG2 세포의 간보호 효과 (Hepatoprotective effect of fermented Chrysanthemum indicum L. water extract on ethanol-induced liver injury in HepG2 cells)

  • 본 연구에서는 발효한 감국 발효 열수 추출물의 luteolin 및 luteolin-7-glucoside 함량, 항산화 활성 및 간세포 보호효과를 비교 분석함으로써 발효에 의한 활성차이를 분석하였다. 감국 내 luteolin 및 luteolin-7-glucoside 함량은 발효 전과 비교하여 유의하게 증가하였고 총 폴리페놀 함량 역시 발효 전보다 증가하는 것으로 나타났다. 유산균 발효에 의해 ABTS radical 소거능은 추출물의 농도가 50, 100, 200 및 400 ㎍/mL의 범위에서 7.30, 15.55, 28.30, 44.48%로 감국 열수 추출물과 비교하여 증가하는 것으로 나타났다. DPPH radical 소거능 역시 ABTS radical 소거능을 비교한 결과와 유사하게 같은 농도의 범위에서 감국 발효 열수 추출물의 경우 각각 6.03, 12.25, 26.18, 41.05%로 감국 열수 추출물과 비교하여 증가하는 것으로 나타났다. HepG2 세포에 감국 열수 추출물과 감국 발효 열수 추출물을 처리한 후 48시간 배양한 결과 모든 처리구에서 세포독성이 없는 것으로 나타났다. HepG2 세포에 에탄올을 처리하여 간독성을 유발한 결과 에탄올 처리한 모든 처리구에서 GGT, AST, ALT 및 LDH 활성이 증가하였고, 감국 발효 열수 추출물을 100 ㎍/mL을 처리하였을 때 대조구과 비교하여 GGT, AST 및 ALT 활성이 유의적으로 감소하는 것으로 나타났으며, LDH 활성은 감국 발효 열수 추출물을 200 ㎍/mL을 처리하였을 때 대조구과 비교하여 감소하는 것으로 나타났다. 이상의 결과로 볼 때 감국 발효 열수 추출물은 luteolin 및 luteolin-7-glucoside 함량 증가, 항산화 활성 증가 및 GGT, AST, ALT, LDH의 활성을 감소시킴으로써 에탄올에 의한 간 손상을 예방하는 것으로 사료되며, 동물실험을 통한 추가적인 간 기능개선 검증이 필요하다고 판단된다.
