서울 및 부여지역 여성들의 철분 영양상태에 관한 비교 연구 (An Assessment of Iron Nutritional Status of Korean Women in Seoul and Puyo Areas)

  • The iron nutritional status of 328(20-70 years old) women in Seoul and Puyo areas was evaluated using a dietary information and a measurement of hematological indices. The serum iron was measured by Red Blood Cell(RBC), Hemoglobin(Hb), Hematocrit(Hct), Serum Iron(S-Fe), Total Iron Binding Capacity (TIBC), Transferrin Saturation(TS) and Serum Ferritin(SF) and was analyzed with the information obtained by interviews which included socio-demographic variables and the dietary pattern of the subjects. The mean Hb was 12.6 $\pm$ 1.21g/dl(Seoul : 12.4 $\pm$ 1.14g/dl, Puyo : 13.2 $\pm$ 1.28g/dl), Hct was 38.7 $\pm$ 3.84%(Seoul: 37.5 $\pm$ 3.28%, Puyo: 4.19 $\pm$ 3.48%), S-Fe was 76.8 $\pm$ 31.49ug/dl(Seoul: 68.8 $\pm$ 27.3ug/dl, Puyo: 78.8 $\pm$ 32.3ug/dl) and TIBC was 277.7 $\pm$ 86.15ug/dl(Seoul: 354.1 $\pm$ 129.8ug/dl, Puyo: 259.0 $\pm$ 59.55ug/dl). The mean Ts(%) was 30.9 $\pm$ 17.9% (Seoul: 21.7 $\pm$ 10.52%, Puyo: 33.2 $\pm$ 18.68%) and the level was significantly higher in Puyo women(p<0.05). The mean SF was 45.4 $\pm$ 46.21ng/ml(Seoul: 53.6 $\pm$ 50.21ng/ml, Puyo: 36.1 $\pm$ 39.83ng/ml). The Red Blood Cell(RBC) showed a negative correlation with age and Hb, Hct showed a negative correlation with education and income level. However TIBC showed a and total energy expenditure per day. Energy, protein, carbohydrate, iron and vit.C intake showed a positive correlation with Hb and Hct. In particular, heme iron showed a positive correlation with RBC and nonheme iron of Hb, income level and iron intake affected on the level of Hct. In Seoul women, the age of menarche affected on the level of TIBC, total energy expenditure affected on the level of Hct. In Seoul women, the age of menarche affected on the level of TIBC, total energy expenditure affected on the level of S-Fe.

습부항이 혈액학적 성상에 미치는 영향에 대한 연구 (Clinical study on the effects of Bleeding Pressure Therapy in patients RBC, Hb and Hct change)

  • Objectives: We examined if their hematological status could be changed by blood loss in patients treated with bleeding pressure therapy. Methods: The patients were divided into two groups as follows: The Subject group(B) were 42 cases treated with bleeding pressure therapy. The Control group(N) were 43 cases treated with negative bleeding pressure therapy, We checked CBC & differ cell count of two groups. Results: The means of RBC, Hb count and Hct were slightly decreased after 1 week. but they were recovered after 2 weeks. Conclusions: These results indicate that Bleeding Pressure Therapy don't reduced RBC, Hb count and Hct in Patients.

Induction of Apoptosis with Kigelia africana fruits in HCT116 Human Colon Cancer Cells via MAPKs Signaling Pathway

  • Kigelia africana (Lam.) Benth. (Bignoniaceae) is a flowering plants in South, Central and West Africa and commonly known as the sausage tree (Eng.); worsboom (Afr.); umVunguta, umFongothi (Zulu); Modukguhlu (North Sotho); Muvevha (Venda). The dried, powdered fruits are used as dressing for wounds and ulcers, haemorrhoids, rheumatism, purgative, skin-firming, lactation in breast-feeding mothers. The aim of this study is to investigate the cytotoxic and apoptotic potentials of 70% ethanolic extracts of Kigelia africana fruits in HCT116 human colon cancer cells. Treatment of Kigelia africana fruits with various concentrations resulted in a sequence of characteristic of apoptosis, including loss of cell viability and morphological changes. Flow cytometry analysis showed Kigelia africana fruits increased the sub-G1 phase (apoptosis) population. Apoptosis confirmed by annexin V-fluorescein isothiocyanate and propidium iodide double staining in HCT116 human colon cancer cell lines. Moreover, analysis of the mechanism indicated that Kigelia africana fruits showed an increased Bax and Bcl-2 expressions in a dose-dependent manner, resulting in activation of hallmarks of apoptotic events, caspase-3, caspase-9 and cleaved poly-ADP-ribose polymerase. This is the first report to demonstrate the cytotoxicity of Kigelia africana fruits on HCT116 human colon cancer cells.

Induction of Apoptosis of DK-5-62, a Novel (-)-Catechin Derivative Through MAPKs Signaling Pathway in HCT116 Cells

  • The present study was designed to investigate the molecular mechanisms of DK-5-62, a novel (-)-catechin derivative on HCT116 human colorectal cancer cells. DK-5-62 inhibited the proliferation in dose- and time-dependent manner accompanied by the morphological changes. Effects of DK-5-62 appeared to be mediated by the induction of apoptosis, as manifested through DNA-binding dye Hoechst 33258 staining. Analysis of the mechanism of these events indicated that DK-5-62-treated cells exhibited an increased ratio of Bax/Bcl-2, resulting in the activation of caspase-9, caspase-3, and poly-ADP-ribose polymerase in a dose-dependent manner. Moreover, DK-5-62-induced apoptosis was accompanied by phosphorylation of the mitogen-activated protein kinase family, c-Jun N-terminal kinase, p38, and extracellular signal-regulated kinase. These results suggest that HCT116 cells are moderately sensitive to growth inhibition by DK-5-62 via apoptosis, as evidenced by activation of ERK/p38/Bcl-2 family signaling, as well as alteration in caspase-9 and caspase-3.

The Relationship of Dietary Heavy Metal Intake with Serum Trace Elements in College Women Living in Choong-Nam Area

  • The purpose of this study was to study the intake of heavy metals such af arsenic, lead and cobalt and the relationship of dietary heavy metals with serum iron, topper, and zinc, which play important roles in hematopoiesis, in healthy college women living in Choongnam Korea, where we have detected heavy metals (As, Pb, Co) in some marine products in previous studies. The nutritional status of the subjects (35 women) was evaluated by anthropometric measurements, 24-hr dietary recall for 3 days. And 3-day diets (by weighing method) and blood were collected to analyze As, Pb, Co, Fe, Cu, Zn, Hb, Hct, and MCHC. The mean age, height, weight, and BMI were 20 years, 158 cm, 55 kg and 22.42 kg/$m^2$, respectively. The mean daily energy intake was 85.85% of RDA for Koreans. The ratio of energy from carbohydrate, protein, and fat was 60 : 24 : 16. The mean daily intake of heavy metals (As, Pb, Co) was 1.77 mg/day, 75.21 $\mu$g/day and 21.12 $\mu$g/day. And the mean daily intake of iron, copper, and Zinc concentrations were 97, 68, and 92% of normal values. The mean serum heavy metals (As, Pb, Co) were 16.14 $\mu$g/dl, 4.32 $\mu\textrm{g}$/dl and 0.02 $\mu$g/dr, respectively Mean blood levels of Fe, Cu, Zn, Hb, Hct, and MCHC were at normal levels. Dietary heavy metals except Co were not significantly different from serum Fe, Cu, Zn and Hb, Hct, and MCHC. However, there was a tendency toward lower serum concentration of Fe, Hb, Hct, and MCHC in the subjects with higher heavy metals (As) intake. Among heavy metals, only dietary Co showed a significant negative correlation with Hb (p< 0.001) and Hct (p < 0.001).

Emodin-Provoked Oxidative Stress Induces Apoptosis in Human Colon Cancer HCT116 Cells through a p53-Mitochondrial Apoptotic Pathway

  • Emodin, a natural anthraquinone isolated from the traditional Chinese medicine Radix rhizoma Rhei, can induce apoptosis in many kinds of cancer cells. This study demonstrated that emodin induces apoptosis in human colon cancer HCT116 cells by provoking oxidative stress, which subsequently triggers a p53-mitochondrial apoptotic pathway. Emodin induced mitochondrial transmembrane potential loss, increase in Bax and decrease in Bcl-2 expression and mitochondrial translocation and release of cytochrome c to cytosol in HCT116 cells. In response to emodin-treatment, ROS increased rapidly, and subsequently p53 was overexpressed. Pretreatment with the antioxidant NAC diminished apoptosis and p53 overexpression induced by emodin. Transfecting p53 siRNA also attenuated apoptosis induced by emodin, Bax expression and mitochondrial translocation being reduced compared to treatment with emodin alone. Taken together, these results indicate that ROS is a trigger of emodin-induced apoptosis in HCT116 cells, and p53 expression increases under oxidative stress, leading to Bax-mediated mitochondrial apoptosis.

제주조릿대의 인간 암세포 증식 저해와 자연사멸 효과 (Antiproliferative and Apoptotic Effects of Sasa quelpaertensis Nakai in Human Cancer Cells)

  • 본 연구에서는 제주도 한라산에 광범위하게 자생하는 제주조릿대의 항암 제제로써의 이용 가능성을 평가하기 위하여 6개 암세포(A549, MCF-7, HepG-2, Hela, HCT116, A375)를 대상으로 세포주기 교란 작용 및 자연사멸 효과를 탐색하였다. MTT 분석 결과 제주조릿대가 다양한 암세포의 증식을 효과적으로 저해하였으며, sub-G1기의 증가와 DNA 분절로 인한 자연사멸 증가에 산화질소가 연관성이 있었다. 이와 별개로 제주조릿대는 세포주기의 장애를 야기하여 암세포의 생장을 억제하는 것으로 나타나 상기의 결과들로 예측하여 볼 때 제주조릿대를 항암 활성을 지닌 소재로 활용 가능할 것이며, 향후 정확한 자연사멸기전 규명을 위한 연구가 진행되어야 할 것이다.

PRP4 Kinase Domain Loss Nullifies Drug Resistance and Epithelial-Mesenchymal Transition in Human Colorectal Carcinoma Cells

  • We have investigated the involvement of the pre-mRNA processing factor 4B (PRP4) kinase domain in mediating drug resistance. HCT116 cells were treated with curcumin, and apoptosis was assessed based on flow cytometry and the generation of reactive oxygen species (ROS). Cells were then transfected with PRP4 or pre-mRNA-processing-splicing factor 8 (PRP8), and drug resistance was analyzed both in vitro and in vivo. Furthermore, we deleted the kinase domain in PRP4 using Gateway™ technology. Curcumin induced cell death through the production of ROS and decreased the activation of survival signals, but PRP4 overexpression reversed the curcumin-induced oxidative stress and apoptosis. PRP8 failed to reverse the curcumin-induced apoptosis in the HCT116 colon cancer cell line. In xenograft mouse model experiments, curcumin effectively reduced tumour size whereas PRP4 conferred resistance to curcumin, which was evident from increasing tumour size, while PRP8 failed to regulate the curcumin action. PRP4 overexpression altered the morphology, rearranged the actin cytoskeleton, triggered epithelial-mesenchymal transition (EMT), and decreased the invasiveness of HCT116 cells. The loss of E-cadherin, a hallmark of EMT, was observed in HCT116 cells overexpressing PRP4. Moreover, we observed that the EMT-inducing potential of PRP4 was aborted after the deletion of its kinase domain. Collectively, our investigations suggest that the PRP4 kinase domain is responsible for promoting drug resistance to curcumin by inducing EMT. Further evaluation of PRP4-induced inhibition of cell death and PRP4 kinase domain interactions with various other proteins might lead to the development of novel approaches for overcoming drug resistance in patients with colon cancer.

어혈처방 3종이 지방전구세포 분화에 미치는 영향 (Anti-adipogenesis Effects of 3 Herbal Formula on Blood Stasis)

  • Objectives : Blood stasis (BS) is related to be caused by blood circulation and stagnation which are cancer, atherosclerosis and hyperlipidemia in traditional medicine. We extracted 3 kinds of BS formula; Seogakjihwang-tang (SGT), Tonggyuhawlhyul-tang (THT), Hyulbuchukeo-tang (HCT). This study was conducted to investigate whether the 3 kinds of herbal formula extracts have inhibitory efficacy association with anti-adipogenesis. Methods : To investigate the anti-adipogenesis, we used the mouse fibroblast cell line, 3T3-L1 which differentiated into adipocytes in response to insulin, IBMX and dexamethasone (MDI). Cytotoxicity of herbal formula extracts were examined by CCK-8 kit. Intracellular lipid droplets were detected by Oil-Red-O staining. Triglyceride (TG) and leptin were measure using elisa kit. Results : The yield of water extracts was 14.62% (SGT), 21.27% (THT), 20.02% (HCT). Lipid accumulation was reduced significantly by 3 kinds of herbal formula compared to control. Especially, THT and HCT decreased lipid droplet, respectively at all concentration. The TG and leptin were also inhibited by 3 kinds of herbal formula. The IC50 of TG were $280.51{\mu}g/m{\ell}$ (SGT), $52.62{\mu}g/m{\ell}$ (THT), $313.99{\mu}g/m{\ell}$ (HCT). The IC50 of leptin were $348.76{\mu}g/m{\ell}$ (SGT), $164.02{\mu}g/m{\ell}$ (THT), $257.00{\mu}g/m{\ell}$ (HCT). THT was better than other herbal formula on anti-adipogenesis. Conclusion : kinds of herbal formula inhibited adipogenesis in MDI-induced 3T3-L1 adipocytes, as indicated by the significant reduction in TG and leptin concentration without cytotoxicity. Therefore, 3 kinds of herbal formula for BS might act as a therapeutic agent for preventing lipid diseases, such as obesity and atherosclerosis.

Replication Association Study between RBC Indices and Genetic Variants in Korean Population

  • Hemoglobin (Hb) concentrations and hematocrit (Hct) values can be changed by factors such as erythrocyte production, destruction, and bleeding. In addition, variants in the protein expression involved in the amount of red blood cells that determine Hb metabolism or Hct value can increase susceptibility to complex blood diseases. Previous studies have reported significant single nucleotide polymorphisms (SNPs) by applying a genome-wide association study (GWAS) on Hb levels and Hct values in European population. In this study, we confirmed whether the significant SNPs are replicated in Koreans. In previous studies, 26 and 18 SNPs with a significant correlation Hb and Hct were identified in Korean genotype data, and 21 and 12 SNPs were selected, respectively. The SNPs of PRKCE (rs10495928), TMPRSS6 (rs2235321, rs5756505, rs855791) were significantly associated with Hb (P<0.05). In the association analysis of Hct, the SNPs of HBS1L (rs6920211, rs9389268, rs9483788), PRKCE (rs4953318), SCGN (rs9348689) and TMPRSS6 (rs2413450) genes showed a significant correlation (P<0.05). Replicated SNPs and not replicated SNPs showed the difference of genetic distance calculated by Fst. The replicated SNPs with a significant correlation showed similar allele frequencies, whereas the not replicated SNPs showed a large difference in allele frequency. All replicated SNPs with significant correlations had Fst values less than 0.05, indicating that the genetic distance between the groups was close. On the other hand, the not replicated SNPs showed that the Fst value was 0.05 or more and the genetic distance was relatively large.
