Microcystis aeruginosa (M. aeruginosa) is a cyanobacterium species that can form harmful algal blooms in freshwater bodies worldwide. The use of pine needle extract (PNE) to control nuisance algae by allelopathic inhibition will be environmentally friendly and promising. PNE removed successfully upto 98% of M. aeruginosa at the following optimal conditions: pH 7, $25^{\circ}C$ of temperature, 100 rpm of mixing rate, 5 min of mixing time. These results was indicated that the amount of 1 g/L PNE was removed 1g dryweight/L of M. aeruginosa. The kinetic data showed substrate inhibition kinetics and maximum growth rate was obtained when the M. aeruginosa was grown in medium containing 0.5 g/L of initial concentration of PNE. Different substrate inhibition models were fitted to the kinetic data and found the Luong model was best. The model predicted kinetic parameters were in agreement with the experimental findings. The natural extract, PNE, can be a promising inhibition due to its high efficiency and low dose requirements.
Cochlodinium polykrikoides is a red-tide forming dinoflagellate that causes significant worldwide impacts on aquaculture industries and the marine ecosystem. There have been extensive studies on managing and preventing C. polykrikoides blooms, but it has been difficult to identify an effective method to control the bloom development. There is also limited genome information on the molecular mechanisms involved in its various ecophysiology and metabolism processes. Thus, comprehensive genome information is required to better understand harmful algal blooms caused by C. polykrikoides. We estimated the C. polykrikoides genome size using flow cytometry, with detection of the fluorescence of DNA stained with propidium iodide (PI). The nuclear genome size of C. polykrikoides was 100.97 Gb, as calculated by comparing its mean fluorescence intensity (MFI) to the MFI of Mus musculus, which is 2.8 Gb. The exceptionally large genome size of C. polykrikoides might indicate its complex physiological and metabolic characteristics. Our optimized protocol for estimating the nuclear genome size of a dinoflagellate using flow cytometry with PI can be applied in studies of other marine organisms.
Recently, the occurrence of harmful algae blooms from the Chattonella group has been increasing and expanding in the southern and western seas of Korea. We investigated the relationship between growth kinetics and nutrients in the harmful algae Chattonella marina and Chattonella ovata of the South Sea, Korea. As a result; high concentrations of ammonium ($30\;{\mu}M$ and above) was not effective to the growth of C. ovata, while C. marina displayed good growth at concentration of $100\;{\mu}M$. The half-saturation constant ($K_s$) of C. marina for ammonium ($2.51\;{\mu}M$), nitrate ($5.36\;{\mu}M$) and phosphate ($0.75\;{\mu}M$) was higher than C. ovata (1.85, 4.01, and $0.61\;{\mu}M$, respectively). This indicates that C. ovata can achieve higher cell densities than C. marina under lower nutrient conditions. These $K_s$ values were comparatively higher than those of diatoms and other flagellates previously reported. Therefore, our results indicate that the growth of C. marina and C. ovata is less adapted to lower nutrient environments than other competition species, such as Skeletonema costatum and Cochlodinium polykrikoides.
Due to climate change, algal blooms frequently occur not only in Korea but also around the world, and the risk of toxicity of harmful algae has recently been issued. It is known that the representative harmful algae, cyanobacteria, are caused by the intersection of three factors: water temperature, residence time, and nutrients. In this study, water quality simulation was carried out using EFDC, a three-dimensional numerical model, to analyze the variations in water quality due to the decrease of residence time according to the opening of Yeongju Dam in Naeseong-Cheon. In fact, the concentration of chlorophyll-a in Yeongju Dam in the summer of 2021 was significant, exceeding the 'algae warning' for a long time based on the previous algae warning system. On the other hand, as a result of performing the simulation under the condition that the dam gate was completely opened, the concentration of chlorophyll-a was mostly reduced below the 'algae warning' level during the simulation period. It was confirmed that reducing the residence time by restoring the flow of Naeseong-Cheon is a way to immediately reduce algae in Yeongju Dam.

Harmful algal blooms (HABs), commonly called red tides, are caused by some toxic phytoplanktons, and have made massive economic losses as well as marine environmental disturbances. As an effective and environment-friendly strategy to control HAB outbreaks, biological methods using marine bacteria capable of killing the harmful algae or algicidal extracellular compounds from them have been given attention. A new member of the $\gamma$-Proteobacteria, Hahella chejuensis KCTC 2396, was originally isolated from the Korean seashore for its ability to secrete industrially useful polysaccharides, and was characterized to produce a red pigment. This pigment later was identified as an alkaloid compound, prodigiosin. During the past several decades, prodigiosin has been extensively studied for its medical potential as immunosuppressants and antitumor agents, owing to its antibiotic and cytotoxic activities. The lytic activity of this marvelous molecule against Cochlodinium polykrikoides cells at very low concentrations ($\sim$l ppb) was serendipitously detected, making H. chejuensis a strong candidate among the biological agents for HAB control. This review provides a brief overview of algicidal marine bacteria and their products, and describes in detail the algicidal characteristics, biosynthetic process, and genetic regulation of prodigiosin as a model among the compounds active against red-tide organisms from the biochemical and genetic viewpoints.

We investigated the ranges of total cellular microcystins content of cyanobacterial blooms collected in Korean lakes and rivers from 2005 to 2009. The amount and composition of microcystins of Korean cyanobacteria varied depending on the sampling water bodies and dominant cyanobacterial genera. Toxic cyanobacterial cell numbers equivalent to $1{\mu}g$ MCYSTs/L using total cellular microcystin content of Korean cyanobacteria were in the range of 2,348 to 66,980,638 cells/mL. Only four samples among forty nine samples showed less cell numbers than current criterion of Harmful Algae Alert System, 5,000 cells/mL indicating current criterion do not reflect properly the microcystins content of Korean cyanobacteria. Anabaena and Aphanizomenon spp. showed three to six times higher cell numbers equivalent to $1{\mu}g$ MCYSTs/L than Microcystis spp. To propose criteria of Harmful Algae Alert System for Korean toxic cyanobacteria, we calculated about 50% selective geometrical means of cyanobacterial cell numbers equivalent to $1{\mu}g$ MCYSTs/L in order of toxic content. The proposed criteria for Microcystis, Oscillatoria, Anabaena, and Aphanizomenon spp., are 10,000, 20,000, 40,000, and 80,000 cells/mL, respectively.
We developed a biologically-derived substance naphthoquinone (NQ) derivate for the eco-safe mitigation of harmful cyanobacteria blooms such as Microcystis and Dolichospermum. NQ was reacted with various substituents ($R_n$) to produce different NQ derivatives. We tested a total of 92 algicidal compounds based on the algicidal activity of Microcystis and Dolichospermum. 22 compounds of NQ were selected as candidates (algicidal activity >80% at $1{\mu}M$). Among them, NQ 40 compound showed the highest algicidal activity of 99.6% and 100% at the optimal concentration of $1{\mu}M$ on Microcystis and Dolichospermum, respectively. No algicidal effects of NQ 40 ($1{\mu}M$) were observed against non-target algae such as Stephanodiscus, Cyclotella and Peridinium. According to the results of acute eco-toxicity assessment, the $EC_{50}$ values of NQ 40 compound for Selenastrum capricornutum and Daphnia magna were 3.2 and $14.5{\mu}M$, respectively, and the $LC_{50}$ for Danio rerio was $15.7{\mu}M$. In addition, for D. magna chronic eco-toxicity assessment, no toxicity toward survival, growth and reproduction was observed. Therefore, we suggested the NQ 40 ($1{\mu}M$) compound as an alternative eco-safe algicidal substance to effectively mitigate harmful cyanobacteria blooms.

Monitoring of cyanobacteria bloom in reservoir systems is important for water managers responsible of water supply system. Cyanobacteria affect the taste and smell of water and pose considerable filtration problems at water use places. Harmful cyanobacteria bloom in reservoir have significant economic impacts. We develop a new method for estimating the cyanobacteria bloom using Landsat TM and ETM+ data. Developed model was calibrated and cross-validated with existing in situ measurements from Daecheong Reservoir's Water Quality Monitoring Program and Algae Alarm System. Measurements data of three stations taken from 2004 to 2012 were matched with radiometrically converted reflectance data from the Landsat TM and ETM+ sensor. Stepwise multiple linear regression was used to select wavelengths in the Landsat TM and ETM+ bands 1, 2 and 4 that were most significant for predicting cyanobacteria cell number and bio-volume. Based on statistical analysis, the linear models were that included visible band ratios slightly outperformed single band models. The final monitoring models captured the extents of cyanobacteria blooms throughout the 2004-2012 study period. The results serve as an added broad area monitoring tool for water resource managers and present new insight into the initiation and propagation of cyanobacteria blooms in Daecheong reservoir.

Red tide is a temporary natural phenomenon involving harmful algal blooms (HABs) in company with a changing sea color from normal to red or reddish brown, and which has a bad influence on coast environments and sea ecosystems. The HABs have inflicted massive mortality on fin fish and shellfish, damaging the economies of fisheries for almost every year from 1990 in South Korea. There have been many studies on red tide due to increasing damage from red tide on fishing and aquaculture industry. However, internal study of automatic red tide image classification is not enough. Especially, extraction of matching center features for recognizing algae image object is difficult because over 200 species of algae in the world have a different size and features. Previously studies used a few type of red tide algae for image classification. In this paper, we proposed the red tide image recognition method using NMF and revison of rotation angle for enhancing of recognition of red tide algae image.
Mastoparan B (MPB), an antimicrobial cationic peptide isolated from the venom of the hornet Vespa basalis, is a basic amphipathic α-helical peptide composed of fourteen amino acid residues. In this study, we have investigated the algicidal effect of MPB against harmful algae blooms (HABs) casative Alexandrium tamarense, Chattonella marina, Cochlodinium polykrikoides and Gymnodinium catenatum. The algicidal effect of MPB showed in the concentration of 31.3 $\mu{g}$/mL to 500 $\mu{g}$/mL against 4 HAB species and observed cell lysis or cell ecdysis by microscopy. MPB reacted more sensitive to C. marina and C. polykrikoides than A. tamarense and G. catenatum. The algicidal study of MPB against HABs will provides much insight into development of new algicidal substances.
