Comparison of Normalization Methods for Defining Copy Number Variation Using Whole-genome SNP Genotyping Data

  • Precise and reliable identification of CNV is still important to fully understand the effect of CNV on genetic diversity and background of complex diseases. SNP marker has been used frequently to detect CNVs, but the analysis of SNP chip data for identifying CNV has not been well established. We compared various normalization methods for CNV analysis and suggest optimal normalization procedure for reliable CNV call. Four normal Koreans and NA10851 HapMap male samples were genotyped using Affymetrix Genome-Wide Human SNP array 5.0. We evaluated the effect of median and quantile normalization to find the optimal normalization for CNV detection based on SNP array data. We also explored the effect of Robust Multichip Average (RMA) background correction for each normalization process. In total, the following 4 combinations of normalization were tried: 1) Median normalization without RMA background correction, 2) Quantile normalization without RMA background correction, 3) Median normalization with RMA background correction, and 4) Quantile normalization with RMA background correction. CNV was called using SW-ARRAY algorithm. We applied 4 different combinations of normalization and compared the effect using intensity ratio profile, box plot, and MA plot. When we applied median and quantile normalizations without RMA background correction, both methods showed similar normalization effect and the final CNV calls were also similar in terms of number and size. In both median and quantile normalizations, RMA backgroundcorrection resulted in widening the range of intensity ratio distribution, which may suggest that RMA background correction may help to detect more CNVs compared to no correction.

Effects of Single Nucleotide Polymorphism Marker Density on Haplotype Block Partition

  • Many researchers have found that one of the most important characteristics of the structure of linkage disequilibrium is that the human genome can be divided into non-overlapping block partitions in which only a small number of haplotypes are observed. The location and distribution of haplotype blocks can be seen as a population property influenced by population genetic events such as selection, mutation, recombination and population structure. In this study, we investigate the effects of the density of markers relative to the full set of all polymorphisms in the region on the results of haplotype partitioning for five popular haplotype block partition methods: three methods in Haploview (confidence interval, four gamete test, and solid spine), MIG++ implemented in PLINK 1.9 and S-MIG++. We used several experimental datasets obtained by sampling subsets of single nucleotide polymorphism (SNP) markers of chromosome 22 region in the 1000 Genomes Project data and also the HapMap phase 3 data to compare the results of haplotype block partitions by five methods. With decreasing sampling ratio down to 20% of the original SNP markers, the total number of haplotype blocks decreases and the length of haplotype blocks increases for all algorithms. When we examined the marker-independence of the haplotype block locations constructed from the datasets of different density, the results using below 50% of the entire SNP markers were very different from the results using the entire SNP markers. We conclude that the haplotype block construction results should be used and interpreted carefully depending on the selection of markers and the purpose of the study.

해양온도차발전용 반경류 터빈의 설계 및 해석 (Design and Analysis of a Radial Turbine for Ocean Thermal Energy Conversion)

  • 해양온도차발전용 터빈의 효율과 크기를 파악하기 위해 R134a를 작동유체로 하고 출력 5 kW인 반경류형 터빈의 설계가 수행되었다. 터빈입구온도 $25^{\circ}C$, 출구 정압 4.9 bar, 질량유량 1.16 kg/s 로 설정하고 평균유동해석을 수행하여 터빈의 회전수와 주요 치수를 결정하였다. 이들을 바탕으로, 3 차원 터빈 모델을 구축하였으며, 도출된 터빈회전수 12,820 rpm에 대하여 전산유체역학(CFD) 소프트웨어 ANSYS CFX를 이용하여 볼류트와 노즐을 포함하는 터빈 내부 유동장 특성과 효율이 조사되었다. 80%이상의 터빈 효율이 적정 범위 내의 노즐 안내깃 수(10-15 개)에서 제시되었으며, 가장 높은 터빈 효율은 15 개의 안내 깃에서 나타났다.

Genetic Association Study of $THR{\beta}$Polymorphisms with Obesity in Korean Population

  • The growing problem of obesity is associated with numerous medical conditions. Several studies have reported that activation of thyroid hormone receptor beta $(THR{\beta})$ is involved in lipid metabolism and thermogenesis. To identify the relationship between the $THR{\beta}$ gene and obesity, we genotyped eighty two single nucleotide polymorphisms (SNPs) in the gene using the Affymetrix array chip in 209 overweight/obese and 155 normal subjects in Korean population. Of the eighty two polymorphisms, the seven SNPs exhibited a significant association with overweight/obesity in three alternative models (codominant, dominant, and recessive models; P<0.05 after adjusting for age and sex) were rs826221 (+267878 T>C), rs4858604 (+186399 A>G), rs1158265 (+200152 T>C), rs1868575 (+206031 G>A), rs1700939 (+238467 T>A), rs1505301 (+241933 T>C), and rs1924768 (+126491 T>C). During haplotype analysis using HapAnalyzer software, 2 haplotypes (block 13: TTAT; block 15: CTGC) containing significant polymorphisms (rs1700939 +238467 T>A and rs4858604 +186399 A>G) were detected to be significantly different. The results suggest that the $THR{\beta}$ gene may be associated with overweight/obesity in Korean population.

버드나무류 (Salix spp.)의 계절학적 특성과 주요 기상요인 상관분석 (Correlation Analysis between Phenology of Salix spp. and Meteorological Factors)

  • The objective of this study was to analyze correlation between phenological characteristics of Salix spp. and meteorological factors in the Upo wetlands. Phenology of Salix subfragilis Andersson and Salix chaenomeloides Kimura was monitored from 2007 to 2012. Meteorological variables were monitored by Korea Meteorological Administration (Hap-chon). Average date of flowering, fruiting, seed dispersion was 86, 113, 136 days for S. subfragilis and 112, 140, 164 days for S. chaenomeloides as Julian days. Flowering of S. subfragilis and S. chaenomeloides were correlated with daily mean air temp. in March (r=-0.92, r=-0.85, p<0.05). Fruiting of S. subfragilis was correlated with total precipitation between Jan and March of previous year (r=-0.90, p<0.01), however, the fruiting of S. chaenomeloides was highly correlated with max. temp. in Jan of previous year (r=0.99, p<0.01). Seed dispersion of both species is correlated with min. temp. in Feb. Phenology monitoring will contribute to understanding Salix spp. response against climate change.

폐업한 자영업자의 재성과 대운·세운분석을 통한 진로상담 가능성 탐색 (A Study on Searching Possibility of Career Counseling through Meungri-Saju Analysis, Wealthy Structures, Deawoon, Saewoon for Failed Self-employeed)

  • 이 연구의 목적은 폐업한 자영업자의 재성과 대운 세운분석을 통해 명리학을 진로상담에 적용하여 그 가능성을 탐색하는데 있었다. 사업적성에 해당하는 재성(財星)의 강 약을 통해 Holland 흥미 적성검사를, 10년의 환경인 대운(大運)과 1년의 환경인 세운(歲運)에서 일간(日干)에게 가장 필요한 용신(用神)의 합(合) 형(刑) 충(?)의 변화로 인한 길(吉) 흉(凶)을 통해 Super가 언급한 개인의 처해진 환경을 파악하였다. 또한 직업궁을 알아 볼 수 있는 월지(月支)의 변화를 살펴 직업변동의 가능성을 확인할 수 있었다. 본 연구를 통해 폐업한 자영업자의 재성은 약한 사주이며, 대운 세운에서 용신이 합 형 충 되어 흉하게 작용하였고 그로 인해 폐업할 수밖에 없었다는 결과를 얻었다. 이러한 연구 성과를 통해 현행 서양검사지 위주의 진로상담에 대한 문제점을 보완하면서 흥미 적성뿐만 아니라, 개인의 환경까지 아우를 수 있는 진로상담을 위한 명리학의 적용가능성을 탐색할 수 있었다.

한국인에 있어서 PEPT1 유전자(exon 5 및 16)의 단일염기변이 빈도 및 일배체형 분석 (Haplotype Analysis and Single Nucleotide Polymorphism Frequency of PEPT1 Gene (Exon 5 and 16) in Korean)

  • The aim of this study was to investigate the frequency of the SNPs on PEPT1 exon 5 and 16 and to analyze haplotype frequency on PEPT1 exon 5 and 16 in Korean population. A total of 519 healthy subjects was genotyped for PEPT1, using pyrosequencing analysis and polymerase chain reaction-based diagnostic tests. Haplotype was statistically inferred using an algorithm based on the expectation-maximization (EM). PEPT1 exon 5 G381A genotyping revealed that the frequency for homozygous wild-type (G/G), heterozygous (G/A) and homozygous mutant-type (A/A) was 30.4, 53.4 and 16.2%, respectively. PEPT1 exon 16 G1287C genotyping revealed that the frequency for homozygous G/G, heterozygous G/C and homozygous C/C type was 88.8, 10.0 and 1.2%, respectively. Based on these genotype data, haplotype analysis between PEPT1 exon 5 G381A and exon 16 G1287C using HapAnalyzer and PL-EM has proceeded. The result has revealed that linkage disequilibrium between alleles is not obvious (|D'|=0.3667).

추적식 수상 태양광발전 구조물의 시공 및 안전성 평가 (Installation and Safety Evaluation of Tracking-type Floating PV Generation Structure)

  • Pultruded glass fiber reinforced polymeric plastic (PFRP) and FRP member manufactured by sheet molding compound (SMC) have superior mechanical and physical properties compared with those of conventional structural materials. Since FRP has an excellent corrosion-resistance and high specific strength and stiffness, the FRP material may be highly appreciated for the development of floating-type photovoltaic (PV) power generation system. In this paper, advanced floating PV generation system made of PFRP and SMC is designed. In the design, it includes tracking solar altitude by tilting photovoltaic arrays and tracking solar azimuth by spinning structures. Moreover, the results of the finite element analysis (FEA) are presented to confirm stability of entire structure under the external loads. Additionally, installation procedure and mooring systems in the Hap-Cheon Dam are discussed and the measurement of strain under the actual circumstances is conducted for assuring stability of actually installed structures. Finally, by comparison with allowable stress, appropriate safety of structure is confirmed to operate the system.

『장진요편(藏珍要編)』의 선혈특성(選穴特性)에 관한 연구 (A Research on the character of selecting acupoints in Jangjinyopyeon(藏珍要編))

  • Objective : Jangjinyopyeon(藏珍要編, JJYP) was published in 1894. The purpose of the research is to know the character of JJYP, and to know JJYP inherited the tradition of Chosun(朝鮮) acupuncture which put emphasis on diagnosis and cure of Jangbu(臟腑) through meridian system. Method : First, identify the bibliographic information of the Mimeograph version of Original JJYP. After then, count and classify the provision by selected acupoints. Analyzed the result in succession. Conclusion : From the research of the mimeograph version of Original JJYP, come to following conclusion. 1. The pen name of author is Songgye(松溪), which means the region of his origin. 2. JJYP used 49 acupoints in total. It covers all regular meridian system except the regular Meridian of Hand Soeum[手少陰心經] for heart organ would never be injured in Neijing(內經) 3. JJYP suggested Eight Confluent points[八脈交會穴] as major acupoints in the end of book. But actually they were selected as the acupoints of belonging Jangbu(臟腑) and various symtoms. 4. Oegwan(外關) and Sameumgyo(三陰交) were maximum frequency in selecting of all acupoints, which means JJYP value controling and covering whole body. 5. Generally, Reinforcing and reducing manipulations concentrated one way, but Hugye(後谿) used each manipulation equally, which means JJYP regards tonification or purgation highly in Hugye(後谿). 6. Most acupoints of high frequency were Nak-point[絡穴], Won-point[原穴], Hap-point[合穴], which supposed to be directly connected to Jangbu(臟腑). So, we can see JJYP continue the tradition of Chosun acupuncture which put emphasis on Jang image[藏象] of meridian.

  • We have studied the blending effects of Diene NF 35 R and Alfin 1530 at various blending ratios, 100/0, 70/30, 50/50, 30/70, 0/100, and of carbon black HAF-HS and ISAF-HS at various compounding ratios of 45 PHR, 55 PHR, 65 PHR, for tyre tread rubber. As the results, it was found that; 1. For tyre tread rubber, as the blending ratio, AR 1530/Diene NF 35R, indicated 70/30, the physical properties we examined were most excellent. 2. Excellent result was obtained in the case of carbon black compounding ratio of 55 PHR. The compounding of ISAF-HS made better result than that of HAF-HS for tensile strength, but the compounding of HAF-HS made better result than that of ISAF-HS for tearing strength and best result for abrasion quantity. 3. Heat buildup obtained from compounding carbon black HAF-HS indicated low temperature than that from compounding carbon black HAP-HS. As the compounding amount of carbon black increased, and as the blending amount of AR 1530 decreased, the heat buildup increased. 4. Carbon black was more efficient to AR 1630 than io Diene NF 35 R. 5. In the physical properties, mooney viscosity and mooney scorch time, as the compounding amount of carbon black increased, the values of mooney viscosity increased, but that of mooney scorch time had a max. point at the compoundiug amount of carbon black, 55 PHR.
