Antioxidant, Antimicrobial, and Cytotoxic Activities of Ovotransferrin from Egg White

  • The antioxidant, antimicrobial, and cytotoxic activities of ovotransferrin were investigated in vitro. The antioxidant capacity of ovotransferrin was evaluated using the 2,2-Diphenyl-1-picryl hydrazyl (DPPH) radical scavenging method, antimicrobial effects using the agar well diffusion method, and cytotoxicity using the 3-(4,5-dimethylthizol-2-yl)-2,5-diphenylatetetrazolium bromide (MTT) assay. The DPPH radical-scavenging capacity of ovotransferrin at 1 mg/mL level reached approximately 60% after 48 h of reaction. The antimicrobial effects of ovotransferrin against common food-borne pathogens, Staphylococcus aureus KCCM 32395, Bacillus cereus KCCM 40935, Listeria monocytogenes ATCC 15313, Escherichia coli O157:H7 ATCC 43895, and Helicobacter pylori HpKCTC 26695 were dose dependant. Gram-positive bacteria was more sensitive to ovotransferrin than gram-negative bacteria. Ovotransferrin showed stronger antimicrobial effect against L. monocytogenes than other gram-positive bacteria tested. The cytotoxicity of ovotransferrin was evaluated in human cancer cell lines, various tissue origins, including the larynx (Hep-2), stomach (AGS), lung (SK-MES-1), liver (HepG2), breast (MCF-7), cervix (HeLa), and colon (HT-29). Ovotransferrin displayed relatively high cytotoxicity (${\leq}60%$ inhibition effects) at 40 mg/mL. At lower concentrations (${\leq}10mg/mL$), however, ovotransferrin cytotoxic effects were not significant in all cancer cell lines tested. These results indicated that ovotransferrin has potential to be used as an antioxidant or antimicrobial agent in foods or a pharmaceutical agent against cancers.

애기땅빈대 추출물의 지방산 조성 및 인체 암세포 증식 억제 효과 (Fatty Acid Composition and Antiproliferative Activity of Extracts from Euphorbia Supina)

  • 본 연구에서는 다양한 약효가 있다고 알려진 애기땅빈대(Euphorbia supina)의 추출물 및 분획물의 지방산 조성을 분석하고 인체 암세포 증식 억제효과에 대하여 살펴보았다. A+M과 MeOH 추출물의 지방산 조성 패턴은 유사했고 A+M과 MeOH 추출물은 각각 53.4% 및 42.1%의 18:3n-3를 함유하였다. 분획물들 중 85% aq. MeOH는 가장 높은 함량의 18:3n-3를 나타내었다. 애기땅빈대에 의한 암세포 증식 억제효과는 MeOH 추출물과 비교했을 때 A+M 추출물에 의한 억제효과가 높았으며 AGS 인체 위암세포에 대한 억제효과가 높았다. 분획물들 중에서는 n-Hexane과 85% aq. MeOH 분획물들에 의한 증식 억제효과가 높았다. 따라서 애기땅빈대의 항암 활성 성분은 n-Hexane과 85% aq. MeOH 분획물들에 함유되어 있는 것으로 여겨지며 향후 정제하여 규명할 필요가 있다고 사료된다.

토후박 추출물의 항산화 및 항당뇨 활성 (Antioxidant and Anti-diabetes Activity of Extracts from Machilus thunbergii S. et Z.)

  • Machilus thunbergii was an important medicinal resource and distributed widely in China. In this study, the bioactivities of dichloromethane fraction (DF) and water fraction (AF) from Machilus thunbergii methanol extract were investigated. Total phenolic contents of DF and AF were 57.90 mg Gal/g and 189.92 mg Gal/g, and total flavonoid contents were 17.34 mg Que/g and 58.38 mg Que/g respectively. The $EC_{50}$ for DPPH radical scavenging activity of DF and AF were $24.37\;{\mu}g/m{\ell}$ and $2.10\;{\mu}g/m{\ell}$. Reducing power and hydroxyl radical scavenging activities of AF were higher than those of DF. The $\alpha$-glucosidase inhibitory activity of AF ($IC_{50}\;=\;1.13\;{\mu}g/m{\ell}$) was higher than that of DF ($IC_{50}\;=\;5.34\;{\mu}g/m{\ell}$). The cell viability was showed that only the DF had anti-proliferation effect on human cancer cell HT-29. These results suggested that both the DF and AF extract of Machilus thunbergii were potential materials for anti-diabetes and functional food for their radical scavenging activity.

Microencapsulation of Probiotic Lactobacillus acidophilus KBL409 by Extrusion Technology to Enhance Survival under Simulated Intestinal and Freeze-Drying Conditions

  • The probiotic Lactobacillus acidophilus KBL409 was encapsulated with alginate (Al) and alginate-chitosan (Al/Chi) through extrusion method. The sizes and zeta potentials of microspheres were measured to confirm encapsulation. To evaluate the protective effect of microspheres against gastrointestinal fluids, all the samples were exposed to simulated gastric fluids (SGFs, pH 1.5) at $37^{\circ}C$ for 1 or 2 h, followed by incubation with simulated intestinal fluids (SIFs, pH 6.5) for 2 h. The mucoadhesive ability of microspheres was evaluated using the intestinal epithelial cell line HT29-MTX. To extend the shelf-life of probiotics, lyoprotectants such as disaccharide and polysaccharide were mixed with free or encapsulated cells during the freeze-drying process. The size of the microspheres demonstrated a narrow distribution, while the zeta potentials of Al and Al/Chi-microspheres were $-17.9{\pm}2.3$ and $20.4{\pm}2.6mV$, respectively. Among all the samples, Al/Chi-encapsulated cells showed the highest survival rate even after exposure to SGF and SIF. The mucoadhesive abilities of Al and Al/Chi-microspheres were higher than 94%, whereas the free L. acidophilus showed 88.1% mucoadhesion. Ten percent of sucrose showed over 80% survival rate in free or encapsulated cells. Therefore, L. acidophilus encapsulated with Al and Al/Chi-microspheres showed higher survival rates after exposure to the gastrointestinal tract and better mucoadhesive abilities than the free cells. Also, sucrose showed the highest protective effect of L. acidophilus during the freeze-drying process.

Physiological Characteristics of Lactobacillus casei Strains and Their Alleviation Effects against Inflammatory Bowel Disease

  • Lactobacillus casei, one of the most widely used probiotics, has been reported to alleviate multiple diseases. However, the effects of this species on intestinal diseases are strain-specific. Here, we aimed to screen L. casei strains with inflammatory bowel disease (IBD)-alleviating effects based on in vitro physiological characteristics. Therefore, the physiological characteristics of 29 L. casei strains were determined, including gastrointestinal transit tolerance, oligosaccharide fermentation, HT-29 cell adhesion, generation time, exopolysaccharide production, acetic acid production, and conjugated linoleic acid synthesis. The effects of five candidate strains on mice with induced colitis were also evaluated. The results showed that among all tested L. casei strains, only Lactobacillus casei M2S01 effectively relieved colitis. This strain recovered body weight, restored disease activity index score, and promoted anti-inflammatory cytokine expression. Gut microbiota sequencing showed that L. casei M2S01 restored a healthy gut microbiome composition. The western blotting showed that the alleviating effects of L. casei M2S01 on IBD were related to the inhibition of the NF-κB pathway. A good gastrointestinal tolerance ability may be one of the prerequisites for the IBD-alleviating effects of L. casei. Our results verified the efficacy of L. casei in alleviating IBD and lay the foundation for the rapid screening of L. casei strain with IBD-alleviating effects.

Expression of ssrA in non-pathogen-induced adaptation in the oral cavity through signal exchange with oral pathogens

  • Introduction: This study was conducted to evaluate ssrA expression resulting from adaptation of Escherichia coli (E. coli) to oral pathogens through signal exchange. Materials and Methods: Human cell lines Hep2 and HT29, wild-type E. coli (WT K-12), ssrA knock-out E. coli (${\Delta}K$-12), and Scleropages aureus (S. aureus) were used. A single culture consisting of Hep2, HT29, WT K-12, and ${\Delta}K$-12, and mixed cultures consisting of Hep2 and WT K-12, Hep2 and ${\Delta}K$-12, WT K-12 and S. aureus, ${\Delta}K$-12 and S. aureus, and Hep2, WT K-12, and S. aureus were prepared. For HT29, a mixed culture was prepared with WT K-12 and with WT K-12 and S. aureus. Total RNA was extracted from each culture with the resulting expression of ssrA, nuclear factor kappa-light-chain-enhancer of activated B cells (NF-${\kappa}B$), and p53 was evaluated by Reverse transcription polymerase chain reaction (RT-PCR). Results: The expression of ssrA in a single culture of WT K-12 was lower than that observed in the mixed culture of WT K-12 with S. aureus. Greater ssrA expression was observed in the mixed culture of WT K-12 with Hep2 than in the single culture of WT K-12. The expression of NF-${\kappa}B$ was higher in the mixed culture of Hep2 with ${\Delta}K$-12 than that in the mixed culture of Hep2 with WT K-12, and was lowest in the single culture of Hep2. The expression of ssrA was higher in the mixed culture of WT K-12 with Hep2 and S. aureus than in the mixed culture of WT K-12 with Hep2. Conclusion: These results suggest that ssrA plays an important role in the mechanism of E. coli adaptation to a new environment.

인체 암세포 성장 및 DNA 합성 억제에 미치는 된장 분획물의 영향 (Effect of Solvent Fractions from Methanol Extract of Doenjang on Inhibition of Growth and DNA Synthesis of Human Cancer Cells.)

  • 본 연구는 된장의 항암효과를 검토하기 위해서 된장 메탄올 추출물과 그 분획물들에 의한 여러 인체 암세포들의 성장 억제와 DNA합성 저해 실험을 행하였다. AGS 인체 위암세포를 이용하여 6일간 배양하여 된장의 메탄을 추출물 $200{\mu}g/ml$를 처리한 결과 $32\%$의 억제효과를 나타내었고 된장 메탄올의 분획물들 중 디클로로메탄층이 가장 높았으며 $200{\mu}g/ml $ 농도로 첨가했을 때 $90\%$의 위암세포 성장을 저해시켰고 에틸아세테이트 분획물을 동일 첨가농도로 투여했을 때 $62\%$의 저해효과를 보였다. 인체 간암세포인 Hep 3B의 경우, 첨가농도 $200{\mu}g/ml$에서 메탄올 추출물은 $51\%$로 암세포 증식을 억제하였으나 디클로로메탄, 에틸아세테이트 분획물의 저해효과가 각각 $89\%$, $86\%$로 매우 높았다. 인체의 결장암세포인 HT-29 세포의 경우, 첨가농도 $200{\mu}g/ml$에서 메탄올 추출물, 디클로로메탄, 에틸아세테이트 분획물은 각각 $84\%$,$91\%$, $71\%$로 암세포 증식을 억제하였다. MC-63 인체의 골육암 세포의 경우, 첨가농도 $200{\mu}g/ml$에서 메탄올 추출물은 $33\%$로 낮은 저해효과를 보였으나 메탄올 추출물의 분획물들 중에는 디클로로메탄 분획물이 $96\%$로 저해효과가 가장 높았으며, 다음으로 에틸아세테이트 분획물이 $71\%$로 암세포 증식을 억제시켰다 된장의 메탄올 추출물 중에서 암세포 증식 억제 효과에서 가장 컸었던 디클로로메탄 분획물과 에틸아세테이트 분획물을 AGS 위암세포에 투여한 2일 후에 세포내의 DNA 합성에 미치는 영향을 측정한 결과, 된장의 디클로로메탄 분획물 $50{\mu}g/ml $, $100{\mu}g/ml$, $200{\mu}g/ml$ 농도에서 각각 $ 66\%$, $73\%$, $94\%$로 DNA 합성이 감소되었고 된장의 에틸아세테이트 분획물 $50{\mu}g/ml$, $100{\mu}g/ml$,$200{\mu}g/ml $ 농도에서 각각 $57\% $, $93\%$, $95\%$의 DNA 합성 저해효과를 나타내었다. 인체 간암세포의 경우, 된장의 디클로로메탄 분획물은 $200{\mu}g/ml$ 농도에서 $ 80\% $의 높은 저해효과를 보였고 에틸아세테이트 분획물은$ 200\mug/ml $ 농도에서 $ 64\% $의 저해효과가 나타내었다. 이상의 얻어진 결과로 된장의 메탄올 추출물과 그 분획물은 여러 인체 암세포의 증식을 억제하고 DNA 합성도 저해하여 in vitro 상에서 암예방 효과 및 항암효과가 있는 것으로 추정된다.

콩 종류별 항돌연변이 및 암세포 증식 억제 효과 비교 (Comparison of Effect of Various Types of Soybeans on Mutagenicity and Growth of Human Cancer Cell Lines)

  • 본 연구에서는 대두와 검정콩에 속하는 흑태, 서목태 및 서리태의 항돌연변이 및 암세포 증식 억제 활성을 비교하였다. Ames test를 이용한 항돌연변이 실험에서 간접돌연변이원인 $AFB_1$에 대해 모든 종류의 콩 메탄올 추출물들은 농도 의존적으로 돌연변이 억제 효과가 증가하였다(p<0.05). 대두 메탄올 추출물보다는 검정콩들인 흑태, 서목태 및 서리태 메탄올 추출물에 의한 효과가 높은 경향을 나타내었다. 검정콩 중에서 약콩이라고 불리는 서목태에 의한 항돌연변이 효과가 높아 첨가농도 5 mg/plate일 때 82%의 효과를 나타내었다. 직접 돌연변이원인 MNNG에 대한 각 종 콩 메탄올 추출물들의 항돌연변이성 실험을 한 결과, $AFB_1$에 대한 효과와 유사하게 검정콩중 서목태에 의한 효과가 높았으며 첨가농도 2.5 및 5 mg/plate일 때부터 각각 58% 및 61%로 돌연변이를 억제시켰다. 인체 암세포를 이용하여 대두 및 검정콩 메탄올 추출물들에 의한 암세포 증식 억제 효과를 검토한 결과, 모든 종류의 콩 메탄올 추출물(첨가농도 1 mg/ml)을 인체 위암세포(AGS)에 처리했을 때 50% 이상의 암세포 증식 억제 효과를 나타내었고 대두보다는 검정콩에 의한 효과가 높았으며 흑태, 서목태 및 서리태 메탄올 추출물은 각각 61%, 70% 및 65%의 암세포 증식 억제효과를 나타내었다. 인체 결장암세포(HT-29)의 경우, 검정콩 메탄올 추출물에 의한 효과가 대두보다 높은 경향을 나타내었으나 서목태 메탄올 추출물을 제외하고는 유의적 차이는 살펴 볼 수가 없었다. 인체 간암세포(Hep 3B)에 의한 증식 억제효과는 이상의 암세포에 대한 효과보다 다소 낮았으나 흑태, 서목태 및 서리태 메탄올 추출물은 첨가농도 1 mg/ml에서 각각 51%, 59% 및 52%의 저해효과를 나타내어 여기서도 서목태에 의한 억제효과가 높았다. 따라서 본 연구의 결과로부터 Ames test를 이용한 항돌연변이 및 인체 암세포 증식 억제 실험에서 검정콩 메탄올 추출물에 의한 억제 효과가 높았고 특히 소립 검정콩에 해당하는 서목태에 의한 생리활성이 우수하였으므로 검정콩에 많이 함유된 색소에 의한 효과라고 추정되며 여기에 대한 향후 연구가 필요하다.

Novel SIRT Inhibitor, MHY2256, Induces Cell Cycle Arrest, Apoptosis, and Autophagic Cell Death in HCT116 Human Colorectal Cancer Cells

  • We examined the anticancer effects of a novel sirtuin inhibitor, MHY2256, on HCT116 human colorectal cancer cells to investigate its underlying molecular mechanisms. MHY2256 significantly suppressed the activity of sirtuin 1 and expression levels of sirtuin 1/2 and stimulated acetylation of forkhead box O1, which is a target protein of sirtuin 1. Treatment with MHY2256 inhibited the growth of the HCT116 (TP53 wild-type), HT-29 (TP53 mutant), and DLD-1 (TP53 mutant) human colorectal cancer cell lines. In addition, MHY2256 induced G0/G1 phase arrest of the cell cycle progression, which was accompanied by the reduction of cyclin D1 and cyclin E and the decrease of cyclin-dependent kinase 2, cyclin-dependent kinase 4, cyclin-dependent kinase 6, phosphorylated retinoblastoma protein, and E2F transcription factor 1. Apoptosis induction was shown by DNA fragmentation and increase in late apoptosis, which were detected using flow cytometric analysis. MHY2256 downregulated expression levels of procaspase-8, -9, and -3 and led to subsequent poly(ADP-ribose) polymerase cleavage. MHY2256-induced apoptosis was involved in the activation of caspase-8, -9, and -3 and was prevented by pretreatment with Z-VAD-FMK, a pan-caspase inhibitor. Furthermore, the autophagic effects of MHY2256 were observed as cytoplasmic vacuolation, green fluorescent protein-light-chain 3 punctate dots, accumulation of acidic vesicular organelles, and upregulated expression level of light-chain 3-II. Taken together, these results suggest that MHY2256 could be a potential novel sirtuin inhibitor for the chemoprevention or treatment of colorectal cancer or both.

함초 분획물의 in vitro에서의 암세포 성장억제 및 Quinone Reductase 활성 유도 효과 (Growth Inhibitory and Quinone Reductase Induction Activities of Salicornia herbacea L. Fractions on Human Cancer Cell Lines in vitro)

  • 본 연구에서는 염생식물의 하나인 함초를 각 용매별로 분획하여 암세포 성장억제 효과와 quinone reductase(QR) 활성 유도 효과를 보았다. 간암 세포인 HepG2, 대장암 세포주인 HT-29 그리고 유방암 세포주인 MCF-7을 이용하여 암세포 성장 억제 효과를 실험한 결과 간암세포주인 HepG2에서 제일 효과가 컸고 사용한 3종의 암 세포주 모두 SHMM층에서 월등히 높은 암세포 성장 억제 효과를 나타내었다. 그리고 암 예방 효과를 알아보기 위하여 3종의 암세포 중 유일하게 quinone reductase를 가지고 있는 인체 간암세포주인 HepG2를 이용하여 QR 활성 유도 효과를 측정한 결과 역시 SHMM층에서 유의적으로 QR 활성 유도 효과가 높게 나타났다. 본 실험 결과에서 함초의 methanol 분획층인 SHMM층에서 암세포 성장 억제 효과와 QR 유도 활성 효과가 높게 나타났으며 이 분획층에 유효한 생리활성 물질이 함유되어 있을 가능성이 높을 것으로 추정된다. 본 연구를 통하여 함초를 이용한 암예방 기능성 식품을 개발할 수 있는 가능성이 보이며, 특히 함초의 methanol 분획층에 대한 집중적인 연구가 요구된다.
