Bisphenol A Exposure and Childhood Obesity (Bisphenol A 노출과 소아비만)

  • Over the a few decades, the incidences of obesity and associated metabolic syndrome diseases have been increased dramatically and resulted in a global health crisis. Recent findings suggest endocrine disrupting chemicals (EDCs) as an obesogen, because they disrupt normal development and homeostatic controls over adipogenesis and energy balance in obesity. Furthermore, risk of cardiovascular disease and mortality is elevated among those who were obese during childhood. Thus, we focused on etiology of obesity in children and performed biological monitoring of bisphenol A (BPA), which is a broadly exposed EDC in environment. Study subjects were age and sex-matched obese and normal children in Seoul (N=52; age, $8.67{\pm}1.46$ years). Exposure levels of BPA were analyzed with HPLC/FLD as a conjugated form in urine. As results, ranges of urinary BPA were 0~54.38 ${\mu}g/g$ creatinine (median, 4.57 ${\mu}g/g$ creatinine). Levels of urinary BPA were 1.7 fold higher in the obese children than those in the controls (medians of obese and control children, 7.31 and 4.25 ${\mu}g/g$ creatinine, respectively, p=0.22). In the near future, enlarge scaled studies should be performed to confirm the risk of BPA for obesity.

Single Toxicity Evaluation of the Polygonati Rhizoma Preparata with Benzo[a]pyrene Contents in ICR Mice (구증황정(九蒸黃精)의 벤조피렌 함량과 마우스 단일투여 독성실험)

  • The object of this study was two. One was if Polygonati Rhizoma preparata had a benzo(a)pyrene, the other was to evaluate the single dose toxicity of 9 repetitive steaming and fermenting processed Polygonati Rhizoma, dried root parts of Polygonati Rhozoma preparata extract, in male and female mice. We measured a content of benzo(a)pyrene in Polygonati Rhozoma preparata using a method with HPLC/FLD. And for single dose toxicity, aqueous extracts of Polygonati Rhozoma preparata (EPP; Yield = 35.4 %) was administered to female and male ICR mice as an oral dose of 2,000, 1,000 and 500 mg/kg (body weight) according to the recommendation of Korea Food and Drug Administration (KFDA) Guidelines. Animals were monitored for the mortality and changes in body weight, clinical signs and gross observation during 14 days after dosing, upon necropsy; organ weight and histopathology of 12 principle organs were examined. As results, we could not find any mortality, clinical signs, and changes in the body and organ weight except for slight soft feces sporadically detected in EPP treated male mice at 1 day after administration. In addition, no EPP-treatment related abnormal gross findings and changes in histopathology of principle organs were detected except for some sporadic accidental findings. The results obtained in this study suggest that benzo(a)pyrene was not existed in Polygonati Rhozoma preparata and the 50% lethal dose and approximate lethal dose of EPP aqueous extracts in both female and male mice were considered as over 2,000 mg/kg, the limited highest dosage recommended by KFDA Guidelines. However, it also observed that the possibilities of digestive disorders, like soft feces when administered over 500 mg/kg of EPP aqueous extracts in the present study.

Analysis of low molecular organic compounds produced during the spoilage of dairy cattle compound feed (착유우용 배합사료의 부패과정 중 발생하는 저분자 유기화합물의 분석)

  • In this study, we analyzed for the changes of low organic compounds during 4 weeks incubation though inoculation of harmful microorganisms on commercial feed. Two percent of overnight cultures of Exiguobacterium acetylicum, Acinetobacter calcoaceticus, Aspergillus flavus and Fusarium graminearum were inoculated in feed, respectively. After adjusting moisture level to 50% for the promotion of feed spoilage, pH was decreased to 4.58~5.03 and microorganism was ranged to $6{\sim}10log_{10}CFU/g$. The compounds were compare between aflatoxin G1 producing feeds and aflatoxin G1 non-producing feeds. Aflatoxins G1 were detected by the immunoaffinity column clean-up method with HPLC-FLD, and were confirmed in samples incoulated by Aspergillus flavus and Acinetobacter calcoaceticus. Koiganal II, cyclohexanol and butadien-one were detected from samples (the non-sterilized inoculated feed) by Aspergillus flavus and Acinetobacter calcoaceticus. Respectively as aflatoxin G1 pre-detected substance, Koiganal II, cyclohexanol and butadien-one may be useful substance for the pre-detection of aflatoxin G1.

Anthocyanin Profiling and Radical Scavenging Activity of Selected Pigmented Rice Varieties

  • BACKGROUND: Anthocyanins have been recognized as health-enhancing substances due to their antioxidant activity, anti-inflammatory, anticancer, and hypoglycemic effects. The objective was to identify anthocyanins-rich rice grains for the development of functional foods and/or functional food colorants METHODS AND RESULTS: Rice grains of one black and three red-hulled rice varieties were extracted with acidified 80% aqueous methanol. The antioxidant activity of the methanolic extracts was screened on TLC plates and in an in vitro assay using DPPH (1, 1-diphenyl-2-picrylhydrazyl) as a free radical source. Red-hulled rice varieties exhibited higher antioxidant activity (88%, 1 mg/mL) than black rice (67%, 1 mg/mL). Among the red-hulled varieties tested, rice variety SSALBYEO54 (901452) was the most active (72%, 0.5 mg/mL). Rice extracted anthocyanin compounds were analyzed by HPLC-DAD-FLD and LC-MS/MS. Red-hulled varieties comprised cyanidin-3-glucoside in addition to ferulic acid esters, apigenin and kaempferol glycosides. CONCLUSION(s): Anthocyanins identified in the black rice variety were cyanidin-7-O-galactoside, cyanidin-3-Oglucoside, cyanidin-3'-O-glucoside, cyanidin-5-O-glucoside, cyanidin-3, 7-O-diglucoside, cyanidin-3, 5-O-diglucoside and peonidin-4'-O-glucoside. The results of this study show that the black rice (IT212512) and red-hulled rice variety SSALBYEO54 (901452) contain notable antioxidant activity for potential use in nutraceutical or functional food formulations.

Changes in γ-Aminobutyric Acid of Bitter Melon (Momordica charantia L.) with Different Cultivation Regions, Harvest Time and Maturation Stages, with Method Validation (재배지역, 수확시기 및 숙기에 따른 여주의 γ-Aminobutyric Acid 함량 변화 및 분석법 검증)

  • This study aimed to investigate the changes in the ${\gamma}$-aminobutyric acid (GABA) content of bitter melon (Momordica charantia L.) cultivated from different regions, with different harvest times and at various maturation stages. Methods for observing the changes in GABA content were validated by determining the specificity, linearity, limit of detection (LOD), limit of quantification (LOQ), and precision and accuracy using the HPLC-FLD system. Results showed high linearity in the calibration curve with a coefficient of correlation ($R^2$) of 0.9999. The LOD and LOQ values for GABA were 0.29 and $0.87{\mu}g/mL$, respectively. The relative standard deviations for intra- and inter-day precision of GABA were less than 5%. The recovery rate of GABA was in the range of 98.77% to 100.50%. The average content of GABA was 0.93 mg/g and Cheongju showed highest GABA content of 1.88 mg/g. As the time of harvest increased from May to September, the GABA content decreased from 1.56 to 0.86 mg/g. Also, maturation of the bitter melon fruit was associated with a decreased in GABA content.

Analytical method of aflatoxins in edible oil and infant-children foods (식용유지와 영유아식품 중 아플라톡신 분석방법)

  • Aflatoxins are secondary metabolites of the molds of Aspergillus flavus and Aspergillus parasiticus. They are highly carcinogenic compounds and can affect a wide range of vegetable commodities such as cereals (especially corn), nuts, peanuts, fruits and oil seeds, in the field and during storage. In fact, oilseeds are often stored for weeks in conditions that promote the mould growth, and the possible consequent presence of aflatoxins in oilseeds can lead to their transfer in oil. In addition, aflatoxins can be found as a natural contaminant in multi-cereals and beans making baby food for infants and young-children. The objective of this study was to validate the liquid extraction method or develop an analytical method for edible oil and infant-children foods. Therefore, this study developed condition of extract for aflatoxins ($B_1$, $B_2$, $G_1$ and $G_2$) in edible oil using a high performance liquid chromatography with florescence detector (HPLC/FLD). Aflatoxins were extracted from edible oil samples by means of MSPD (Matrix solid phased dispersion), utilizing $C_{18}$ as dispersing material and purified by using immunoaffinity column. The gression line coefficients were above 0.999. The recoveries for aflatoxins ranged from 85.9 to 93.0%, and relative standard deviations were below 5.7%. The new developed method of aflatoxins effectively enhanced recoveries by using MSPD-Immunoaffinity column compared with liquid extraction. The analytical method for liquid extraction of aflatoxin was appropriate for infant-children food. Reviewing the current method, the recoveries of aflatoxins ($B_1$, $B_2$, $G_1$ and $G_2$) were 89.5~92.3%.

Exposure Assessment of Total Aflatoxin in Foods (식품중 총 아플라톡신의 노출량 평가)

  • A survey of total aflatoxin levels was conducted on 565 food samples (cereals, nuts, etc) collected in commercial markets. The determination of aflatoxins ($B_{1}$, $B_{2}$, $G_{1}$ and $G_{2}$) was performed using HPLC with fluorescence detector. The Limit of Detections (LODs) of the B group and G group were 0.05 ng/g and 0.07 ng/g, respectively. In addition, recoveries of rice, peanut butter, and red pepper flour were satisfactory. Total aflatoxin was detected 27 samples(4.8%) out of 565 samples. Incidence ratios in cereals, nuts, processed products, and other foods were 0.2, 0.4, 3.0 and 1.2%, respectively, but aflatoxin was not detected in pulse and dried fruits. The daily intake of total aflatoxin using food intakes was 0.04 ng/kg bw/day.

Monitoring of Pesticide Residues in the Certificated Organic and Pesticide-free Cereals and Root Vegetables (유기 및 무농약 인증 곡류와 근채류 중 잔류농약 모니터링)

  • This study was carried out to investigate characteristics of the pesticide residues in environment friendly cereal grains and root vegetables. The samples, 747 the environment friendly agricultural products were collected from supermarkets and retail stores in eight major cities in May and August 2012. Residues of 245 pesticides in the samples collected were analyzed by multiresidue methods using GC and HPLC. No pesticide residues were detected in all the samples, representing the environment friendly cereal grains and root vegetables were produced according to the guideline for prohibition of use of pesticides during crop cultivation.

Determination of Polycyclic Aromatic Hydrocarbons in Sesame Oils Derived from Sesame Seeds of Different Places of Origins (원산지가 다른 참깨로 제조한 참기름에서의 polycyclic aromatic hydrocarbons 함량 분석)

  • Polycyclic aromatic hydrocarbon(PAH) contents were evaluated in sesame oils from sesame seeds of different origins and in commercial samples using HPLC with fluorescence detection. The sesame seeds, which had been harvested from India, China, and Korea, were roasted at $250^{\circ}C$ for 25 min, and the commercial sesame oils were purchased from a local market. The recoveries for eight PAHs spiked into the sesame oils ranged from 80.2 to 99.2%. The mean levels of total PAHs in the sesame oils harvested from China, Korea, and India were 3.97, 1.57, and 1.20 ${\mu}g$/kg, respectively. The PAH contents in the commercial sesame oils ranged from 0.79 to 2.15 ${\mu}g$/kg.

Influence of Polycyclic Aromatic Hydrocarbons Formation in Sesame Oils with Different Roasting Conditions (참깨의 볶음 조건이 참기름 중 polycyclic aromatic hydrocarbons 생성에 미치는 영향)

  • Polycyclic aromatic hydrocarbons (PAHs) are environmental carcinogenic compounds that arise by several means including food processing methods such as smoking and direct drying and cooking. This study examined the concentration of PAHs in sesame oils with various roasting temperatures (190, 220 and $250^{\circ}C$), methods (direct heating vs. indirect hot air heating), and times (5, 10, 15, 20 and 25 min). The PAHs in the sesame oils were analyzed using liquid-liquid extraction and solid-phase clean up (Florisil), followed by HPLC with fluorescence detection. According to the results, mean levels of total PAHs increased when the sesame oils were roasted at increasing temperatures and times. The sesame oil roasted at $250^{\circ}C$ for 25 min had the highest mean value of total PAHs (4.66 ${\mu}g$/kg). The results of this study suggest that the indirect hot air roasting method decreased PAH formation during sesame oil processing.
