Sophoricoside was isolated as the inhibitor of IL-5 bioactivity from Sophora japonica (Leguminosae). To develope as novel anti-allergic drug, kinetic study was performed in rats. Serum concentration of sophoricoside was measured by gas chromatography-mass spectrometry (GC/MS) in male Sprague-Dawley rat (250${\pm}$10g, n=5) after oral administration of sophoricoside (100mg/kg). The recovery of sophoricoside after extraction and concentration was above 95 % from rat serum. Between-day precision(relative standard deviation 2.2-2.8%) and within-day precision(2.0-12.1%) were determined from replicate analysis of a spiked control and incurred serum sample. The detection limits of sophoricoside in this serum was approximately 0.1 ng/mL. The Pharmacokinetic parameters were derived from the noncompartmental analysis. The C$\_$max/(3.56${\pm}$0.34 $\mu\textrm{g}$/mL) value for sophoricoside in male rat was observed at 7.6 h. The elimination half-life(t$\_$1/2/) of sophoricoside was approximately 4.47 h, the mean residence time (MRT) averaged 10.75 h, the total body clearance (Cl) averaged 0.0042 mL/min/kg. and the area under the serum concentration-time curve (AUC$\_$0-$\infty$/) was 24.93 $\mu\textrm{g}$$.$hr/mL.

The present study numerically investigates the aerodynamic characteristics of two-dimensional wings in the vicinity of the ground by solving two-dimensional steady incompressible Navier-Stokes equations with the turbulence closure model of the realizable k-${\varepsilon}$ model. Numerical simulations are performed at a wide range of the normalized ground clearance by the chord length ($0.1{\leq}h/C{\leq}1.25$) for the angles of attack ($0^{\circ}{\leq}{\alpha}{\leq}10^{\circ}$) in the prestall regime at a Reynolds number (Re) of $2{\times}10^6$ based on free stream velocity $U_{\infty}$ and the chord length. As the physical model of this study, a cambered airfoil of NACA 4406 has been selected by a performance test for various airfoils. The maximum lift-to-drag ratio is achieved at ${\alpha}=4^{\circ}$ and h / C = 0.1. Under the conditions of ${\alpha}=4^{\circ}$ and h / C = 0.1, the effect of the Reynolds number on the aerodynamic characteristics of NACA 4406 is investigated in the range of $2{\times}10^5{\leq}Re{\leq}2{\times}10^9$. As Re increases, $C_l$ and $C_d$ augments and decreases, respectively, and the lift-to-drag ratio increases linearly.
본 연구에서는 다른 host cell 에 비하여 여러 가지 장점을 가지고 있는 Methylotrophic yeast 중 Pichia pastoris와 Hans-enula polymorpha의 fed batch 실험을 통하여 유전자 재조합 단백질 발현최적조건을 구하여 각 균주의 유전자 재조합 albumin 발현 최적화 연구를 수행하였다. 글리세롤이 두 균주의 promoter의 AOX 1과 MOX promoter repression에 미치는 영향을 확인한 바 H. polymorpha가 P. pastoris보다 promoter repression이 심함을 알 수 있었다. 두 균주의 promoter를 induction시키는 최적 메탄올 농도는 P. pastoris의 경우 메탄올 8g/L, H. polymorpha의 경우는 13 g/L임을 알 수 있었다. 또한 메탄올에 의한 induction시기는 두 균주 모두 O.D. 4 정도되는 exponential growth stage에서 메탄올을 첨가하는 경우가 초기 세포 성장단계에 메탄올을 첨가한 경우에 비해 약 20% 정도 높아짐을 확인하였다. 두 균주의 재조합 albumin 발현에 미치는 pH의 영향을 조사하였는데, p. pastoris의 경우 pH 5에서 가장 높은 albumin 생산성을 보여 약 300mg/L albumin을 발현하였고, H. polymorpha 의 경우 pH 5와 6에서 최대 약 180 mg/L의 albumin을 발현하였지만 pH 8에서는 이의 절반 수준에 그쳤다. 두 균주의 최적 fed-batch 방법을 확인하는 실험을 수행하였는데 P. pastoris의 경우의 최적 fed-batch 방법은 mixed feeding은 바람직하지 않고 글리세롤 배지를 공급하여 세포를 성장시킨 후 글리세롤 공급을 멈추고 바로 메탄올로 전환하는 방법을 효과적이며, H. polymorpha의 경우 비성장속도 제어를 통한 글리세롤 공급으로부터 메탄올 공급으로의 단계적 전환방법이 균주의 albumin 발현에 큰 영향을 준다는 것을 알 수 있었다. 이 방법을 통하여 두 균주의 고농도 배양 실험을 수행한 결과 P. pastoris의 경우는 O.D. 300에서 약 4.7g albumin/L를 발현하였다. 이와같은 결과를 바탕으로 산업체에서 methlotrophic yeast를 이용한 상업화를 계획함에 있어서 host로서의 균주를 선택할 수 있는 기본 자료를 제공함과 아울러 균주가 선택된 후에 그 균주를 이용한 재조합 단백질 최적화 방법을 제공하여 줄 것으로 생각된다.

A robust controller for a 6 DOF magnetically levitated fine manipulator is presented. The proposed controller consists of following two parts : a model reference controller (MRC) and a H$_{\infty}$ controller (HIC). First, the MRC stabilizes the motion of the manipulator. Then, the motion of the manipulator follows that of the reference model. Second, the HIC minimizes errors generated from the MRC due to noise and disturbance since the HIC is a kind of robust controller. The experiments of position control and tracking control are carried out by use of the proposed controller under the conditions of free disturbances and forced disturbances. Also, the experiments using PID controller are carried out under the same conditions. The results from above two controllers are compared to investigate the control performances. As the results, it is observed that the proposed controller has similar position accuracy but better tracking performances comparing to the PID controller as well as good disturbance rejection effect due to the robust characteristics of the controller. In conclusion, it is verified that the proposed controller has the simple control structure, the good tracking performances and good disturbance rejection effect due to the robust characteristics of the controller..
Quantitative Feedback Theory (QFT) is one of most effective methods of robust controller design and can be considered as a suitable method for systems with parametric uncertainties. Particularly it allows us to obtain controllers less conservative than other methods like $H_{\infty}$ and ${\mu}$-synthesis. In QFT method, we transform all the uncertainties and desired specifications to some boundaries in Nichols chart and then we have to find the nominal loop transfer function such that satisfies the boundaries and has the minimum high frequency gain. The major drawback of the QFT method is that there is no effective and useful method for finding this nominal loop transfer function. The usual approach to this problem involves loop-shaping in the Nichols chart by manipulating the poles and zeros of the nominal loop transfer function. This process now aided by recently developed computer aided design tools proceeds by trial and error and its success often depends heavily on the experience of the loop-shaper. Thus for the novice and First time QFT user, there is a genuine need for an automatic loop-shaping tool to generate a first-cut solution. In this paper, we approach the automatic QFT loop-shaping problem by using an algorithm involving Linear Programming (LP) techniques and Genetic Algorithm (GA).

In this paper we want to overcome the major drawback of QFT method which is lack of an automatic method for loop-shaping step of the method so we focus on the following problem: Given a nominal plant and QFT bounds, synthesize a controller that achieves closed-loop stability and satisfies the QFT boundaries. The usual approach to this problem involves loop-shaping in the frequency domain by manipulating the poles and zeros of the nominal loop transfer function. This process now aided by recently developed computer aided design tools proceeds by trial and error and its success often depends heavily on the experience of the loop-shaper. Thus for the novice and First time QFT user, there is a genuine need for an automatic loop-shaping tool to generate a first-cut solution. Clearly such an automatic process must involve some sort of optimization, and while recent results on convex optimization have found fruitful applications in other areas of control theory we have tried to use LMI theory for automating the loop-shaping step of QFT design.
An add-on type output regulator is proposed in this paper. By an add-on controller we mean an additional controller which operates harmonically with a pre-designed one. The role of the add-on controller is to reject a sinusoidal disturbance of unknown magnitude and phase but with known frequency. Advantages of the proposed controller include that (1) it can be used only when the performance of disturbance rejection needs to be enhanced, (2) when it is turned on or off, unwanted transient can be avoided (i.e., bumpless transfer), (3) it is designed for perfect disturbance rejection not just for disturbance reduction, (4) ability for perfect rejection is preserved even with uncertain plant model. This design may be promising for optical disc drive (ODD) systems in which disc eccentricity results in a sinusoidal disturbance. For ODD systems, the sensitivity function obtained by the pre-designed controller, which may have been designed by the lead-lag, $H_{\infty}$, or DOB (disturbance observer) technique, does not change much with the add-on controller except at the frequency of the disturbance. Since the add-on controller does the job of rejecting major eccentricity disturbance, the gain of the pre-designed controller does not have to be too high.
POD (proper orthogonal decomposition) is applied to turbulent leading-edge separation bubble to extract coherent structures. A two-dimensional leading-edge separation bubble is simulated by discrete-vortex method, where a time-dependent source forcing is incorporated. Based on the wealth of numerical data, POD is applied in a range of the forcing amplitude ( $A_{o}$ = 0, 0.5, 1.0 and 1.5) and forcing frequency (0 .leq. $f_{F}$H/ $U_{\infty}$.leq. 0.3). It is demonstrated that the structures of POD have noticeable changes with local forcings. In an effort to investigate the mechanism of decreasing reattachment length, dynamic behaviors of the expansion coefficients and contributions of the eigenfunctions of POD are scrutinized. As the forcing amplitude increases, the large-scale vortex structures are formed near the forcing amplitude increases, the large-scale vortex structures are formed near the separation point and the flow structures become more organized and more regular, accompanying with the reduction of reattachment length. By further inverstigation of POD global entropy, it is seen that the reattachment length is closely linked to the degree of organization of the flow structures.es.s.
