Characteristics and Antibiotic Susceptibility of Imipenem-Resistant Clinical Isolates Producing Carbapenemase (Carbapenemase를 생산하는 imipenem 내성 세균의 특성 및 항생제 감수성)

  • Imipenem-resistant bacteria were isolated from clinical specimens taken from hospitalized patients in Suncheon, Korea. Fifty-four isolates were phylogenetically analyzed based on 16S rRNA gene and gyrB gene sequence comparisons. Isolates were affiliated with Pseudomonas aeruginosa (30 strains; 55.6%), Acinetobacter baumannii (21; 38.9%), Enterobacter hormaechei (2) and Pseudomonas putida (2). Twenty-two isolates produced metallo-$\beta$-lactamase (MBL); 12 Acinetobacter baumannii strains, 7 Pseudomonas aeruginosa strains, 2 P. putida strains and 1 Enterobacter hormaechei strain. Antibiotic susceptibility of the isolates was determined using the disc diffusion method and Vitek system. Strains producing metallo-$\beta$-lactamase (type IMP & VIM) were more resistant to antibiotics ceftazidime, aztreonam, amikacin and gentamicin than to strains producing OXA and SHV type of $\beta$-lactamase.

  • Recent observations of Sloan Digital Sky Survey (SDSS) for the ugriz filters have provided the largest and the most homogeneous photometric database. Especially, the ugriz systems have been used in many telescopes, such as SDSS, CFHT, and Gemini, and will be applied next generation large telescopes. In addition, many stellar evolution models, like as PADOVA, BaSTI and DSEP, have introduced theoretical isochrones in ugriz system, to apply the isochrones into the photometry of stars in globular clusters. In this study, we transformed Yonsei-Yale($Y^2$) isochrones to ugriz photometric system and fit the isochrones to the (g-r, r), (g-i, r), and (r-i, r) CMDs of 13 globular clusters (Chun et al. 2009, Clem et al. 2008, An et al. 2008). We found that the derived ages from $Y^2$-isochrones are 1~3Gyr younger and the estimated distance moduli are 0.3mag larger than the values inferred from the other isochrones in r-band. Also, the E(B-V) are 0.02~0.04mag less than those estimated from the other models. The ages of each globular cluster estimated from $Y^2$-isochrones are 12~14Gyr and distance moduli show good agreement with previous studies. From this result, we confirmed the availability of the $Y^2$-isochrones in gri filters. However, it is a problem which will improve that E(B-V) values obtained from $Y^2$-isochrones are too small. Finally, the result of this study is expected to be used research of globular cluster in ugirz photometric system.

Antagonistic Assay of Bacillus spp. for Eco-friendly Biological Control of Melon Powdery Mildew (멜론 흰가루병 친환경 생물적 방제를 위한 Bacillus속 균의 길항력 평가)

  • Melon powdery mildew, caused by Podosphaera fusca, is one of the serious diseases of melon plant in Korea. In this study, we evaluated the effect of selected antagonistic bacteria on the inhibition of mycelial growth of various plant pathogens, and control of melon powdery mildew. Based on the 16S rDNA and gyrA gene sequences, the selected antagonistic bacteria, M09, M70, and M99-1, were identified as Bacillus velezensis. These bacteria not only inhibited the mycelial growth of 47~69% in various plant pathogens, but also significantly reduced the incidence of powdery mildew. The three strains selected in this study could be used as potential biological control agents for various plant diseases as well as melon powdery mildew.

STANDARD STELLAR MODELS; $\alpha$ CEN A AND B

  • The standard stellar models for $\alpha$ Cen A and B have been constructed without resorting to the arbitrary constraint of the Solar mixing length ratio. Assuming that the chemical compositions and the ages of the two stars are the same, series of models have been constructed. Using the observational constraints, [Z/X], we were able to constrain the number of the 'possible' models. We find that utilizing the observational constraints of [Z/X] the best models for $\alpha$ Cen system are with the initial Z = 0.03, X = 0.66$\~$0.67. In particular, the primary and the secondary stars may have the same mixing length ratio 1.6$\~$1.7, which is the same as that of the calibrated Solar model. And, the age of the system is about 5.4 Gyr. Finally, the large spacing of the p-modes is predicted to be 104 $\pm$ 4$\mu$Hz for $\alpha$ Cen A.

BVI PHOTOMETRIC STUDY OF THE OLD OPEN CLUSTER RUPRECHT 6

  • We present a BV I optical photometric study of the old open cluster Ruprecht 6 using the data obtained with the SMARTS 1.0 m telescope at the CTIO, Chile. Its color-magnitude diagrams show the clear existence of the main-sequence stars, whose turn-off point is located around $V{\approx}18.45mag$ and $B-V{\approx}0.85mag$. Three red clump (RC) stars are identified at V = 16.00 mag, I = 14.41 mag and B - V = 1.35 mag. From the mean $K_s-band$ magnitude of RC stars ($K_s=12.39{\pm}0.21mag$) in Ruprecht 6 from 2MASS photometry and the known absolute magnitudes of the RC stars ($M_{K_S}=-1.595{\pm}0.025mag$), we obtain the distance modulus to Ruprecht 6 of $(m-M)_0=13.84{\pm}0.21mag$ ($d=5.86{\pm}0.60kpc$). From the ($J-K_s$) and (B - V ) colors of the RC stars, comparison of the (B - V ) and (V - I) colors of the bright stars in Ruprecht 6 with those of the intrinsic colors of dwarf and giant stars, and the PARSEC isochrone fittings, we derive the reddening values of E(B - V ) = 0.42 mag and E(V - I) = 0.60 mag. Using the PARSEC isochrone fittings onto the color-magnitude diagrams, we estimate the age and metallicity to be: $log(t)=9.50{\pm}0.10(t=3.16{\pm}0.82Gyr)$ and $[Fe/H]=-0.42{\pm}0.04dex$. We present the Galactocentric radial metallicity gradient analysis for old (age > 1 Gyr) open clusters of the Dias et al. catalog, which likely follow a single relation of $[Fe/H]=(-0.034{\pm}0.007)R_{GC}+(0.190{\pm}0.080)$ (rms = 0.201) for the whole radial range or a dual relation of $[Fe/H]=(-0.077{\pm}0.017)R_{GC}+(0.609{\pm}0.161)$ (rms = 0.152) and constant ([Fe/H] ~ -0.3 dex) value, inside and outside of RGC ~ 12 kpc, respectively. The metallicity and Galactocentric radius ($13.28{\pm}0.54kpc$) of Ruprecht 6 obtained in this study seem to be consistent with both of the relations.

Comparative Transcriptome Analysis of Caryophyllene-Treated Helicobacter pylori

  • Helicobacter pylori (H. pylori) establishes long-term infections associated with severe gastric diseases such as peptic ulceration and gastric cancer. Exposure to an antibacterial agent can help regulate the expression levels of its pathogenic genes. In this study, we analyzed the transcriptional changes in H. pylori genes induced by β-caryophyllene. We used next-generation sequencing (NGS) to analyze RNA expression changes, and reverse transcription-polymerase chain reaction (RT-PCR) was performed as required to verify the results. The NGS results showed that 30 out of 1,632 genes were expressed differentially by β-caryophyllene treatment. Eleven genes associated with DNA replication, virulence factors, and T4SS components were significantly downregulated. RT-PCR confirmed that treatment reduced the expression levels of 11 genes. RT-PCR showed the reduced expression of 11 genes (dnaE, dnaN, holB, gyrA, cagA, vacA, secA, flgE, virB2, virB4, and virB8) following β-caryophyllene treatment. These results suggest that β-caryophyllene can modulate various H. pylori pathogenic determinants and be a potential therapeutic agent for H. pylori infection.

Selection of Bacillus amyloliquefaciens CC110 for Biological Control of Cucumber Downy Mildew Caused by Pseudoperonospora cubensis (오이 노균병의 생물적 방제를 위한 Bacillus amyloliquefaciens CC110균주 선발)

  • In order to select antagonists for biological control of downy mildew of cucumber, 126 bacteria were isolated from cucumber plants collected from several locations in Korea. Among them, Five isolates were selected as potential biocontrol agents of cucumber downy mildew using a leaf disc bioassay method. In preventive and curative effect tests, the isolate CC110 was found to be effective to control downy mildew on cucumber showing diseased area by 0% whereas that of control was 15.0~18.0%. A bacterium isolate CC110 was identified as Bacillus amyloliquefaciens subsp. plantarum based on phylogenetic analysis using gyrB gene sequence. The culture liquid of isolate CC110 in TSB media were more effective for the control of the disease than those cultured in LB, NB, and KB media in leaf disk bioassay. when undiluted liquid, two-fold, five-fold diluted culture broth, and undiluted liquid, two-fold, five-fold diluted filtrate of isolate CC110 in TSB media were treated, diseased area of cucumber powdery mildew were 0%, 3.0%, 8.0%, 0%, 4.0% and 7.0%, respectively, whereas diseased area in the control was 21.0%. In the cucumber seedling tests, when the culture broth of isolate CC110 in TSB media was treated, diseased area were 35.0%, whereas that of control was 82.0%. When B. amyloliquefaciens CC110 was treated four times at five-day interval in the vinylhouse test, the control effect of cucumber downy mildew was higher than that treated three at seven-day interval.

Isolation and Identification of Antagonistic Bacterium Active against Sclerotinia sclerotioum Causing Sclerotinia Rot on Crisphead Lettuce (결구상추 균핵병균(Sclerotinia sclerotioum)에 대한 길항세균의 분리 및 동정)

  • The fungus genus Sclerotinia contains a number of important plant pathogens. Vegetable growers in our country are probably most familiar with Sclerotinia sclerotiorum, the causes of sclerotinia rot on crisphead lettuce. S. sclerotiorum has a wide host range which can include lettuce as well as crops such as broccoli, cabbage, carrots, celery, beans, peppers, potatoes, stocks, and tomato. Some fungicides, including benomyl, are effective in some crops, but not all. So, we isolated a antagonistic bacteria that are active on sclerotinia rot caused by S. sclerotiorum and that can be used to control it. About 702 strains had been isolated from soil around plant roots in the field. Ten strains showed strong antifungal activity against S. sclerotiorum. In pot test for antagonistic activity, A-7 strain showed high control value against the pathogen when compared with others. The strain was, therefore, selected as a biocontrol candidate against sclerotinia rot and its biochemical properties and 16S rDNA sequence was analyzed. The A-7 strain was highly related to Bacillus subtilis and B. amyloliquefaciens. To confirm precise identification, we had performed gyr A gene sequences analysis. Its sequence had 96% similarity with B. amyloliquefaciens. Consequently, the isolate was identified as B. amyloliquefaciens A-7.

Bacterial Spot Disease of Green Pumpkin by Pseudomonas syringae pv. syringae (Pseudomonas syringae pv. syringae에 의한 애호박 세균점무늬병)

  • A pathogen that causes a new disease on green pumpkin in the nursery and the field was characterized and identified. Symptoms of the disease on green pumpkin were water soaking lesions and spots with strong yellow halo on leaf, brown lesion on flower, and yellow spot on fruit. The bacterial isolates from the leaf spot were pathogenic on the 8 curcubitaceae crop plants, green pumpkin, figleaf gourd, wax gourd, young pumpkin, zucchini, cucumber, melon, and oriental melon, whereas they did not cause the disease on sweet pumpkin and watermelon. They were Gram-negative, rod shape with polar flagella, fluorescent on King's B agar and LOPAT group 1a by LOPAT test. Their Biolog substrate utilization patterns were similar to Pseudomonas syringae pv. syringae's in Biolog database. Phylogenetic trees with 16S rRNA gene sequences and multilocus sequence typing (MLST) with nucleotide sequences of 4 housekeeping genes, gapA, gltA, gyrB, rpoD and those of P. syringae complex strains in the Plant Associated and Environmental Microbes Database (PAMDB) showed that the green pumpkin isolates formed in the same clade with P. syringae pv. syringae strains. The clade in MLST tree was in the genomospecies 1 group. The phenotypic and genotypic characteristics suggested that the isolates from green pumpkin lesion were P. syringae pv. syringae.

Selection of Stable Reference Genes for Real-Time Quantitative PCR Analysis in Edwardsiella tarda

  • Edwardsiella tarda is a gram-negative pathogenic bacterium in aquaculture that can cause hemorrhagic septicemia in fish. Many secreted proteins have already been identified as virulent factors of E. tarda. Moreover, since virulent phenotypes are based on the expression regulation of virulent genes, understanding the expression profile of virulent genes is important. A quantitative RT-PCR is one of the preferred methods for determining different gene expressions. However, this requires the selection of a stable reference gene in E. tarda, which has not yet been systematically studied. Accordingly, this study evaluated nine candidate reference genes (recA, uup, rpoB, rho, topA, gyrA, groEL, rpoD, and 16S rRNA) using the Excel-based programs BestKeeper, GeNorm, and NormFinder under different culture conditions. The results showed that 16S rRNA was more stable than the other genes at different culture growth phases. However, at the same culture time, topA was identified as the reference gene under the conditions of different strains, different culture media, and infection, whereas gyrA was identified under the condition of different temperatures. Thus, in experiments, the expression of gapA and fbaA in E. tarda was analyzed by RT-qPCR using 16S rRNA, recA, and uup as the reference genes. The results showed that 16S rRNA was the most suitable reference gene in this analysis, and that using unsuitable reference genes resulted in inaccurate results.
