Optimization of Fermentation Process and Quality Properties of Wild Grape Wine (산머루 와인의 최적 발효조건 및 품질특성)

  • Wild grapes contain various organic acids, vitamins, and inorganic salts compared to other various fruits. Their anthocyanin compounds are also known to have excellent effects in preventing cancers and heart diseases. Wild grapes are supposed to be a superior material for wine fermentation. This study was performed for searching optimum conditions for producing wild grape wines to improve their quality and taste. Four wild strains were isolated from Muscat Bailey A, ‘Kyoho’ grapevine, and wild grapes. For wild grape wine production, an optimum initial sugar concentration was $24^{\circ}Brix$ and an optimum temperature was $24^{\circ}C$. Sensory characteristics of the wild grape wine were evaluated for incense, sour taste, astringency, and sweet taste. Quality properties of the wild grape wine produced under the optimized fermentation conditions were analyzed. Total sugar, polyphenol contents, and acidity of the wild grape wine were 50.00 mg/mL, 25.30 mg/mL, and 0.95%, showing higher value than those of commercial grape wines. Amino acids, tannins, alcohol contents, and pH were 1.80 mg/mL, 1.88 mg/mL, 12%, and 3.59, respectively. The optimum fermentation conditions can be used to improve quality of the wild grape wine.

Characterization of the Effects of Different Wavelengths of Night-break Lighting on the Fruit Quality and Yield of 'Kyoho' Grapes (파장별 야간 조사에 따른 '거봉' 포도의 품질 및 생산량)

  • This study was conducted to estimate the light pollution damage caused by night-break lighting in 'Kyoho' grapes. Night irradiation was performed every night for four hours (10 p.m. to 2 a.m.) from the full bloom to the harvest stage using red, blue, and white lamps as artificial light. Fruit yield, soluble solid content, anthocyanin content, and chlorophyll a content were affected by night irradiation. The soluble solid content of the control was $17.5^{\circ}Brix$, approximately $1^{\circ}Brix$ higher than the red ($16.4^{\circ}Brix$), blue ($16.2^{\circ}Brix$), and white light treated grapes ($16.3^{\circ}Brix$). The anthocyanin content of the skin was also higher in the control at $4.08{\mu}g{\cdot}cm^{-2}$ compared to the red ($3.14{\mu}g{\cdot}cm^{-2}$), blue ($2.47{\mu}g{\cdot}cm^{-2}$), and white ($2.82{\mu}g{\cdot}cm^{-2}$) light treated samples. On the other hand, the chlorophyll a content of the control was the lowest at $0.268{\mu}g{\cdot}cm^{-2}$ as compared with the red ($0.339{\mu}g{\cdot}cm^{-2}$), blue ($0.345{\mu}g{\cdot}cm^{-2}$), and white ($0.372{\mu}g{\cdot}cm^{-2}$) light treated samples. Considering that higher soluble solid contents, higher accumulation of anthocyanin, and lower chlorophyll a contents are factors involved in fruit maturation, night irradiation may delay fruit maturation and red light treatment may result in decreased yield. Our results confirmed that night-break lighting regardless of the wavelength provoked light pollution in 'Kyoho' grapes. Therefore, fruit maturation may be poor in the presence of artificial light, including streetlamps, in neighboring vineyards.

  • The database for the oriental medicine had been existed in documentation in past times and it has been developed to the database type for random accesses in the information society. However, the aspects of the database are not so diversified and the database for the bio herbal material exists in widened type dictionary style. It is a situation that the database which handles the in-depth raw herbal medicines is not sufficient in its quantity and quality. Korean wild grape is a deciduous plant categorized into the Vitaceae and it was found experimentally that it has various medical effects. It is one of the medical materials with higher potentiality of academic study and commercialization recently because it has a bigger possibility to be applied into diverse industrial fields including the medical product for health, food and beauty. We constituted the cooperative system among the Muju cluster business group for Korean mountain wild grapes, Physiology Laboratory in Kyung Hee University Oriental Medicine and Medical Classics Laboratory in Kyung Hee University Oriental Medicine with a view to focusing on such potentiality and a database for Korean wild grapes was made a touchstone for establishing the in-depth database for the single bio medical materials. First of all, the literatures based on the North East Asia in ancient times had been categorized into the classical literature (Korean literature published by government organization, Korean classical literature, Chinese classical literature and classical literature fro Korean and Chinese oriental medicine) and modern literature (Modern literature for oriental medicine, modern literature for domestic and foreign herbal medicine) to cover the eastern and western research records and writings related to Korean wild grapes and the text-mining work has been performed through the cooperation system with the Medical Classics Laboratory in Kyung Hee University Oriental Medicine. First of all, the data for the experiment and theory for Korean wild grape were collected for the Medline database controlled by the Parliament Library of USA to arrange the domestic and foreign theses with topic for Korean wild grapes and the network hyperlink function and down load function were mounted for self-thesis searching function and active view based on the collected data. The thesis searching function provides various auxiliary functions and the searching is available according to the diverse searching/queries such as the name of sub species of Korean wild grape, the logical intersection index for the active ingredients, efficacy and elements. It was constituted for the researchers who design the Korean wild grape study to design of easier experiment. In addition, the data related to the patents for Korean wild grape which were collected from European Patent Office in response to the commercialization possibility and the system available for searching and view was established in the same viewpoint. Perl was used for the query programming and MS-SQL for database establishment and management in the designing of this database. Correlations between quality indices and consumer acceptance in environment-friendly 'Campbell early' grapes (친환경 포도의 품질 인자와 소비자 기호도의 상관성 분석)

  • This study investigated the correlation between physicochemical (color, soluble solids content (SSC), pH, titratable acidity (TA), and firmness) and sensory (appearance, taste, odor, and texture) characteristics of environment-friendly 'Campbell early' grapes to identify quality indices. For analysis, samples of similar-sized grapes were collected from five orchards. The results showed that the physicochemical characteristics of CIE $L^*$, CIE $a^*/b^*$, SSC, pH, TA, and firmness and the sensorial characteristics of color intensity, freshness of stem, odor, sourness, sweetness, and elasticity were different among groups. Correlation analysis results showed that an increase in sweetness and firmness and a decrease in sourness were associated with an increase in overall acceptance. Sourness and sweetness were positively correlated with CIE $L^*$ (r=0.88) and firmness (r=0.95), individually. In the principal component analysis results, component F1 and F2 explained 44.35% and 33.77%, respectively, of the total variance (78.12%). F1 represented firmness, sweetness, elasticity, hardness, grape odor, color intensity, sweet odor, sourness, and damage degree. F2 represented CIE $L^*$, TA, CIE $a^*$, CIE $a^*/b^*$, SSC/TA, SSC, and peel thickness. The results showed that consumer acceptance of 'Campbell early' grapes can be determined by assessing physicochemical attributes of firmness, CIE $L^*$, TA, CIE $a^*$, CIE $a^*/b^*$, SSC/TA, and SSC and various sensorial attributes including sweetness, fruit elasticity, fruit hardness, grape odor, and color intensity.

Effective extraction of antioxidantive oligomeric proanthocyanidins from mountain grape seeds

  • The interest of oligomeric proanthocyanidins(OPCs) as therapeutic agents against diseases involving radical damage is growing. Proanthocyanidins are a class of polyphenolic compounds in several plant species and are oligomers of flavan-3-ol monomer units. Polyphenols in green and black tea, grape seeds, grapes and wine have raised much attention but mountain grape seed has not been investigated intensively up to now This study investigated the total OPCs contents and the total antioxidant activity of mountain grape seeds. Total antioxidant activity using DPPH method was employed and OPCs contents were determined by means of the UV-VIS spectrophotometer. The total OPCs yield of mountain grape seeds was about 1.45 % and total antioxidant activity was 15.8 ${\mu}g/m{\ell}$.

  • Post-harvest decay, caused by Penicillium spp. is a serious problem of fruits worldwide. Morphological characteristics and molecular markers were used to characterize 22 Penicillium isolates from apples, 18 isolates from pears, 60 from oranges and 18 from grapes and 23reference isolates representing related Penicillium spp. to assess their diversity and resolve their taxonomy. Based on morphological and physiological characteristics, the isolates were grouped as identical or very similar to P. digitatum, P. italicum, P. ulaiense or very similar to P. crustosum, P. expansum, P. solitum and unidentified Penicillium spp. Based on sequence comparisons of ITS region, variable site were presented within and among the species, but there variation were not correlated with the species. Cluster analyses of AP-PCR fragment patterns using UP and L45 primer and the -tubulin gene sequence, the Penicillium species were segregated into distinct groups. Particularly. the -tubulin partial sequence data provided support for species concepts based on morphological and physiological characteristics.

  • Cepacidine A was previously isolated as a novel antifungal antibiotic from the culture broth of Pseudomonas cepacia AF2001. It exhibits a potent in vitro antifungal activity against various plant pathogenic fungi, such as Plasmopora veticola on grapes, Septoria nodorum and Fusarium culmorum on wheat, as well as Colletotrichum lagenarium on cucumbers. Accordingly, this study was conducted to evaluate the potential crop protection activity of strain P. cepacia AF2001. The strain was tested in semi-greenhouse biocontrol assays, and showed an excellent biological activity against Pythium ultimum in cotton and cucumbers; however, only a minor activity against Rhizoctonia aolani in cotton was observed. Furthermore, the nematodes Haemonchus contortus and Trichostrongylus only exhibited a moderated activity in the in vitro larval development assay with no activity in the in vivo animal model.

A Study on Relationship between Food Preferences and Personality of University Students (대학생의 기호식품과 인성과의 관계에 대한 연구)

  • The objective of this research was to examine the relationship between food preferences and personality of university students. The questionnaires were distributed to 198 university students in 2009. The data showed high correlation between personality and food preferences. According to the result of food preferences exams the most favorite food of university students was coffee, refreshing drinks, eggs, ice creams, bananas, porks, apples, chickens, weeds, grapes and food they dislike were sea cucumbers, livers, mung bean sprout, crown daisy, flat fishes, bean curd, green lavers, lotus roots, turban shells, egg plant etc. The correlation coefficient between food preferences and personality showed that male students had more dominance and sociability, and female students had more sociability and validity. On the other hand, a good diet group had higher responsibility and a poor diet group had higher sociability. Consequently, good food habits is essentially needed for the formation of desirable personality of students.

Resveratrol Induces the Apoptosis of Osteosarcoma Saos-2 Cells (레스베라트롤에 의한 골육종 Saos-2 세포의 세포고사)

  • Resveratrol, a phytoalexin found in grapes, berries, and peanuts, is one of the most promising agents for cancer prevention. Recent studies show that the antitumor activity of resveratrol occurs through p53-mediated apoptosis. This study demonstrated the mechanism that resveratrol induced apoptosis in human osteosarcoma Saos-2 cells lacking p53. Treatment of osteosarcoma Saos-2 cells with resveratrol resulted in decrease of cell viability, which was revealed as apoptosis characterized by activation of caspase-3 protease as well as cleavage of poly(ADP-ribose) polymerase (PARP) with change of mitochondrial membrane potential transition. These results suggest that resveratrol may be potentially useful to treat osteosarcoma via activation of caspase protease and mitochondrial dysfunction.
