One-pot Enzymatic Synthesis of UDP-D-glucose from UMP and Glucose-1-phosphate Using an ATP Regeneration System

  • Glucose-1-phosphate uridylyltransferase from E. coli K12 was used to convert uridine-5'-triphosphate and glucose-1-phosphate to UDP-D-glucose. The conversion was efficient and completed within 5 minutes under the employed conditions. In addition, thymidine-5'-monophosphate kinase and acetate kinase were proven to be non-specific, converting udridine-5'-monophosphate to uridine-5'-triphosphate with 55% conversion after 6 h, which was much slower than the production of TTP under the same conditions (complete conversion within one hour). Since these two reactions could proceed under the same conditions, a one-pot synthesis of UDP-D-glucose with ATP regeneration was designed from easily available starting materials, and conversion up to 40% by HPLC peak integration was achieved given a reaction time of 4 h.

Conversion of Glucose and Xylose to 5-Hydroxymethyl furfural, Furfural, and Levulinic Acid Using Ethanol Organosolv Pretreatment under Various Conditions

  • The objective of this study was to understand the conversion characteristics of glucose and xylose using the major monosaccharide standards for lignocellulosic biomass. The acid-catalyzed organosolv pretreatment conducted using ethanol was significantly different from the acid-catalyzed process conducted in an aqueous medium. 5-hydroxymethylfurfural (5-HMF), levulinic acid and furfural were produced from glucose conversion. The maximum yield of 5-HMF was 5.5%, at 200℃, when 0.5% sulfuric acid was used. The maximum yield of levulinic acid was 21.5%, at 220℃, when 1.0% sulfuric acid was used. Furfural was produced from xylose conversion and under 0.5% sulfuric acid, furfural reached the maximum yield 48.5% at 210℃. Ethyl levulinate and methyl levulinate were also formed from the glucose standard following the esterification reaction conducted under conditions of the combined conversion method, which proceeded under both ethanol-rich and water-rich conditions.

Optimization of Two-stage Pretreatment from Soybean Hull for Efficient Glucose Recovery

  • Soybean hull is an attractive feedstock for glucose production. To increase the glucose conversion in acid hydrolysis, a pretreatment method combined steam explosion with alkali pretreatment for soybean hull was studied. For first step pretreatment, steam explosion conditions (log Ro 2.45) were optimized to obtain maximum solid recovery and cellulose content. In the second step pretreatment, the conditions for potassium hydroxide pretreatment of steam exploded soybean hull were optimized by using RSM (response surface methodology). The optimum conditions for minimum lignin content were determined to be 0.6% potassium hydroxide concentration, $70^{\circ}C$ reaction temperature and 198 min reaction time. The predicted lignin content was 2.2% at the optimum conditions. Experimental verification of the optimum conditions gave the lignin content in similar value with the estimated value of the model. Finally, glucose conversion of pretreated soybean hull using acid hydrolysis resulted in $97.1{\pm}0.4%$. This research of two-step pretreatment was a promising method for increasing the glucose conversion in the cellulose-to-glucose process.

  • The effect of glucose concentration on the production of erythritol by Trichosporon sp. was mainly studied. The specific growth rate and production rate of erythritol gave the highest values of $0.23 h^{-1}\; and\; 4.2 g/\ell/h,\; respectively,\; on\; 100 g \; glucose/\ell$ of medium. The conversion yield of erythritol during the exponential phase and the stationary phase was constantly maintained at 19% and 51 %, respectively, while the glucose concentration in the medium varied from 100 g/$\ell$ to 400 g/$\ell$. The maximum overall erythritol conversion yield of 47% was obtained when the glucose concentration in the medium was 400 g/$\ell$. It corresponded to a 74% increase compared with the 100 g/$\ell$ glucose medium. The diauxy growth of this microbe was also observed. It grew exponentially consuming glucose, then after the second lag phase, biomass slowly increased using glycerol and erythritol.

Mass Production of Pullulan with Optimized Concentrations of Carbon and Nitrogen Sources by Aureobasidium pullulans HP-2001 in a 100-L Bioreactor with the Inner Pressure

  • Cell growth and the production of pullulan by Aureobasidium pullulan HP-2001, the UV-induced mutant of A pullulans ATCC 42023, increased with increased concentration of glucose up to 15.0% (w/v). Maximal production of pullulan in the flask scale was 27.65 g/l, when concentrations of glucose and yeast extract were 15.0 and 0.25% (w/v), respectively. Maximal conversion rate of pullulan from glucose as the carbon source was 0.37, when those of glucose and yeast extract were 5.0 and 0.15% (w/v), respectively. On the basis of total amount of pullulan, the conversion rate of pullulan from glucose, and utilization rate of glucose to cell mass and pullulan by A. pullulans HP-2001, the optimal concentrations of glucose and yeast extract for the mass production of pullulan were determined to be 10.0 and 0.25% (w/v), respectively, at which concentrations the production of pullulan and its conversion rate were 27.14 g/l and 0.27, respectively. Maximal production of pullulan with optimized concentrations of carbon and nitrogen sources by A. pullulans HP-200l in a 7-1 bioreactor was 32.12 g/l for 72 h culture, and that in a 100-1 bioreactor with the inner pressure of $0.4 kgf/cm^2$ was 36.87 g/l. Increased inner pressure of a 100-1 bioreactor resulted in a higher concentration of dissolved oxygen in the medium, which might enhance the production of pullulan by A. pullulans HP-2001.

Optimization of Concentrated Acid Hydrolysis of Waste Paper Using Response Surface Methodology

  • Waste paper stands for the major biodegradable organic fraction of most of municipal solid waste. The potential of waste paper for glucose production was investigated in this current work. The pretreatment was accomplished by first subjecting waste paper to disintegration time (30 s), followed by ink removal of disintegrated waste paper using an deinking agent. Concentrated acid hydrolysis of waste paper with sulfuric acid was optimized to maximize glucose conversion. The concentrated acid hydrolysis conditions for waste paper (disintegrated time: 30 s, deinking agent loading : 15 ml) were optimized by using central composite design and response surface methodology. The optimization process employed a central composite design, where the investigated variables were acid concentration (60~80%), loading sulfuric acid (1~5 ml) and reaction time (1~5 h). All the tested variables were identified to have significant effects (p < 0.05) on glucose conversion. The optimum concentrated acid hydrolysis conditions were acid concentration of 70.8%, loading sulfuric acid of 3.2 ml and a reaction time of 3.6 h. This research of concentrated acid hydrolysis was a promising method to improve glucose conversion for waste paper.

  • Pentachlorophenol(PCP)가 대두(大豆) 및 벼 현탁배양(縣濁培養)과 과 수도체 중에서 수용성성분(水溶性成分)으로 전환(轉換)되는 것을 경시적으로 조사하여 다음과 같은 결과를 얻었다. 1. 현탁배양에서 처리한 PCP의 50%가 수용성 대사물로의 전환되는 시간은 대두의 경우 4시간, 벼는 48시간 이내였다. 수용성 대사물의 생성속도와 양은 PCP의 cell 생육 저해 정도와 밀접한 관계가 있었다. 2. 식물 cell에서 PCP의 주(主) 대사과정(代謝過程)은 생육시기(生育時期) 및 약제(藥劑) 처리시간(處理時間)을 달리한 전환시험에서 얻은 수용성 대사물 및 glucose cunjugates 함량변화 조사와 conjugates가 가수분해 되는 성질로부터 glucose conjugates(${\beta}$-anomers)형성 반응(反應)임을 알 수 있었다. 3. Cell에서 glucose conjugates는 약제처리(藥劑處理) 초기 및 대수성장기 이전에 생성 속도가 매우 빨라 전체 수용성 대사물의 70% 이상이었으며, 점차 비가수분해성 극성성분(極性成分)으로 전환되었고, 일부는 배양액(培養液)으로 분필(分泌)되었다.

Optimization of C/N ratio for production of pullulan

  • The production of pullulan by Aureobasidium pullulans HP-2001 was investigated under various ratios of glucose as carbon source and yeast extract as the nitrogen source, Highest conversion rate (productivity) of glucose to pullulan was 40.0% when concentrations of glucose and yeast extract were 5% and 0.15%, respectively. Maximal production of pullulan was 29.3g/1 when the concentration of glucose was 8%(w/v) and that of yeast extract was 40:1. On basis of the result that production of pullulan was found in a medium which concentration of glucose as carbon source was up to 20%(w/v), Aureobasidium pullulans HP-2001 seemed to overcome the catabolite repression. Conversion rate of pullulan from 20%(w/v) of glucose was 11.1%.

Production of dTDP-4-keto-6-deoxy-D-glucose by Immobilization of dTDP-D-glucose 4,6-dehydratase

  • The dTDP-D-glucose 4,6-dehydratase from Salmonella enterica was immobilized using covalent binding to cyanogen bromide activated sepharose. The immobilized enzyme was used to produce dTDP-4-keto-6-deoxy-D-glucose, a key sugar intermediate that can be used economically to produce diverse classes of unusual sugars appended in various antibiotics. The enzyme was immobilized on the sepharose after activation with cyanogen bromide. The maximum immobilization (80.03%) was achieved after 14 h of coupling. The covalently immobilized enzyme was stable, and an average of 78.4 % conversion was achieved until 120 h of immobilization when it was repeatedly used. Similar conversion was noticed for the first batch using the enzyme entrapped-hydrogel but activity was gradually decreased in the following batches. The production of dTDP-4-keto-6-deoxy-D-glucose by using an immobilized enzyme has high potential for commercial application.

젖산 생성 감소 및 단일군항체 생산성 향상을 위한 하이브리도마의 유가배양 (Fed Batch Culture of Hybridoma for Reduction of Lactate and Higher Monoclonal Antibody Productivity)

  • 하이브리도마 세포 배양을 통한 단일 군항체 생산성을 향상시키기 위해서는 배양환경에 의해 유해 대사산물 생산이 어떤 식으로 변화되는가에 대한 이해가 필요하다 세포성장과 단일 군항체 생산을 저해시키는 대사물질 중에 하나인 젖산의 생성을 줄이기 위해, 포도당의 젖산으로의 전환율에 영향을 미치는 인자들에 대해 연구하였다. 초기 포도당 농도가 높을수록, 세포 성장이 빠를 수록 젖산으로의 전환율이 커짐을 알 수 있었다. 또한 세포농도에 따른 비성장속도 변화에 대한 모델식을 기초로 하여 포도당 소비속도만큼 포도당을 공급하는 유가배양을 통해 젖산 생성을 19% 감소시켰고, 적분세포 농도 증가를 통해 단일군항체 생산성을 41% 향상시킬 수있었다.
