Effect of phytol in forage on phytanic acid content in cow's milk

  • Objective: Bioactive compounds in ruminant products are related to functional compounds in their diets. Therefore, this study aimed to explore the effect of forage sources, Italian ryegrass (IR) silage vs corn silage (CS) in the total mixed ration (TMR), on milk production, milk composition, and phytanic acid content in milk, as well as on the extent of conversion of dietary phytol to milk phytanic acid. Methods: Phytanic acid content in milk was investigated for cows fed a TMR containing either IR silage or CS using 17 cows over three periods of 21 days each. In periods 1 and 3, cows were fed CS-based TMR (30% CS), while in period 2, cows were fed IR silage-based TMR (20% IR silage and10% CS). Results: The results showed that there were no differences in fat, protein, lactose, solids-not-fat, somatic cell count, and fatty acid composition of milk among the three experimental periods. There were no differences in the plasma concentration of glucose, triglycerides, total cholesterol, and nonesterified fatty acids among the three experimental periods, while the blood urea nitrogen was higher (p<0.05) in period 2. The milk phytanic acid content was higher (p<0.05) in period 2 (13.9 mg/kg) compared with periods 1 (9.30 mg/kg) and 3 (8.80 mg/kg). Also, the phytanic acid content in the feces was higher (p<0.05) in period 2 (1.65 mg/kg dry matter [DM]) compared with period 1 (1.15 mg/kg DM), and 3 (1.17 mg/kg DM). Although the phytol contents in feces did not differ among the three feeding periods, the conversion ratio from dietary phytol to milk phytanic acid was estimated to be only 2.6%. Conclusion: Phytanic acid content in cow's milk increases with increasing phytol content in diets. However, phytol might not be completely metabolized in the rumen and phytanic acid, in turn, might not be completely recovered into cow's milk. The change of phytanic acid content in milk may be positively correlated with the change of phytol in the diet within a short time.

Effect of Illuminance on Color-based Analysis of Diabetes-Related Urine Fusion Analytes on Dipstick Using a Smartphone Camera (스마트폰 카메라를 활용한 뇨시험지 당뇨병관련 융합 분석인자의 색기반 분석에 미치는 외부 조도 영향)

  • Recently, the miniaturization and digitalization for the inspection devices of point-of-care testing (POCT) are rapidly evolving. In the urine test, a lot of researches on index paper technology are being conducted because people can be self-diagnosed through visual color comparison using a urine test paper, Dipsick. The purpose of this study is to analyze the RGB values from the color changes on Dipstick Pad, which isused for urine test, using a smartphone camera. To this end, the primary, analytes in urine wasdiabetes-related parameters such as glucose, ketone body and pH, which is the most frequently tested elements, and we pursuited to quantify the changes in dipstick color caused from artificial urine containing different ranges of sugar, ketone body, and pH. In this experiment, changes in RGB values under bright and dark illuminances were compared, and changes in RGB value were monitored as a function of concentration of analytes under the ambient illumination of laboratory. As a result, color separation at the bright luminance region was good, but it did not appearat the low luminance region, and the changed profiles in RGB value under different illuminances was suggested to correct the problem of the color separation algorithm.

Effects of hematological parameters and plasma components of olive flounder, Paralichthys olivaceus by acute nitrite exposure according to water temperature (수온별 아질산 급성 노출에 따른 넙치, Paralichthys olivaceus의 혈액학적 성상 및 혈장성분의 영향)

  • Olive flounder (Paralichthys olivaceus) (Weight 110.9±17.1 g, length 22.3±1.2 cm) were exposed to waterborne nitrite at 0, 30, 60, 120, 240, 480 and 960 mg NO2-/L according to water temperature at 20℃ and 25℃ for 96 hours. The lethal concentration 50 (LC50) of olive flounder, P. olivaceus exposed to waterborne nitrite was 513.87 mg NO2-/L at 20℃ and 208.35 mg NO2-/L at 25℃, which means a significant difference in LC50 by the water temperature. Hemoglobin and hematocrit were significantly decreased by waterborne nitrite exposure. The inorganic component, plasma calcium, was significantly decreased, and the organic components such as plasma glucose and cholesterol were significantly decreased showing a similar tendency with calcium. In enzymatic components, the AST and ALP were also significantly decreased by nitrite exposure. The results of this study indicate that exposure to nitrite can affect the survival and hematological physiology of P. olivaceus, and the effect of exposure to nitrite had a significant effect on nitrite toxicity depending on the water temperature.

The association of dietary patterns with insulin resistance in Korean adults: based on the 2015 Korea National Health and Nutrition Examination Survey (한국 성인의 식사 패턴과 인슐린 저항성 간의 상관성: 2015년도 국민건강영양조사를 이용하여)

  • Purpose: This study was conducted to identify the association between insulin resistance and the major dietary patterns of Korean adults. Methods: This study used data from the 2015 Korea National Health and Nutrition Examination Survey. The subjects were 2,276 adults aged 19 to 64 years old. Based on the food frequency questionnaire data, 112 food items were reclassified into 30 food groups. The principal component analysis method was applied to identify major dietary patterns. We used homeostatic model assessment of insulin resistance (HOMA-IR) and quantitative insulin sensitivity check index (QUICKI) value as indicators of insulin resistance. The association between major dietary patterns and insulin resistance was investigated using logistic regression analysis. Results: Three major dietary patterns were identified and assigned descriptive names based on the food items with high loadings: 'healthy Korean meal pattern', 'western meal pattern', and 'white rice, alcohol, meat pattern'. As the 'white rice, alcohol, meat pattern' score increased, significant increasing trends for fasting glucose concentration and HOMA-IR and a significant decreasing trend for QUICKI were observed after adjusting for age and sex. The odds ratio of insulin resistance according to the 'healthy Korean meal pattern' and the 'western meal pattern' were not statistically significant. the 'white rice, alcohol, meat pattern' showed a significant positive association with the risk of insulin resistance after adjusting for covariates. Conclusion: These results suggest that the 'white rice, alcohol, meat pattern' is positively associated with the risk of insulin resistance. The white rice, alcohol, meat pattern was related to the high consumption of alcohol together with rice or meat. This pattern was also associated with the high intake of sodium and low intakes of vitamin C, calcium, potassium, and dietary fiber. To confirm the association, further longitudinal studies are required.

Anti-obesity and Anti-diabetes Effects of the Fermented White Jelly Fungus (Tremella fuciformis Berk) Using Lactobacillus rhamnosus BHN-LAB 76 (유산균을 이용한 흰목이 버섯의 발효를 통한 항비만과 항당뇨 효과)

  • White jelly fungus (Tremella fuciformis Berk) is effective for hypertension, diabetes, obesity, lung disease, beauty, and has been known as an elixir of life in ancient Chinese dynasty. In this study, we investigated the anti-obesity and diabetic effects of the fermented white jelly fungus extract by the bioconversion process. We fed an obesity-inducing mouse with 5% non-fermented Tremella fuciformis (TF), 2.5% fermented Tremella fuciformis (FTF), and 5% FTF containing High Fats Diet (HFD) and HFD for 8 weeks, respectively. The oral glucose tolerance test (OGTT) was performed analysis after 7 weeks of feeding and the dietary intake, food efficiency ratio, body weight, liver, epididymal fat weight, and serum insulin level were measured after 8 weeks of feeding. Also, HOMA-IR was analyzed. The concentrations of serum total cholesterol, triglycerides test was analyzed. The FTF compare with 5% TF and HFD confirmed that 5% FTF reduced body weight, tissue weight, triglycerides concentration, HOMA-IR, respectively. As a result, we confirmed that the fermented white jelly fungus has the anti-obesity effect. Finally, this study can be used a basic data for obesity treatment using fermented white jelly fungus.

Development of a Hydrogen Peroxide Sensor Based on Palladium and Copper Electroplated Laser Induced Graphene Electrode (PdCu를 전기 도금한 레이저 유도 그래핀 전극 기반의 과산화수소 측정 센서 개발)

  • In this paper, we describe the fabrication and characterization of a hydrogen peroxide ($H_2O_2$) sensor based on palladium and copper (PdCu) electroplated laser induced graphene (LIG) electrodes. $CO_2$ laser was used to form LIG electrodes on a PI film. This fabrication method allows simple control of the LIG electrode size and shape. The PdCu was electrochemically deposited on the LIG electrodes to improve the electrocatalytic reaction with $H_2O_2$. The electrochemical performance of this sensor was evaluated in terms of selectivity, sensitivity, and linearity. The physical characterization of this sensor was conducted using scanning electron microscopy (SEM) and energy-dispersive X-ray spectroscopy (EDS), which confirmed that PdCu was formed on the laser induced graphene electrode. In order to increase the sensor sensitivity, the Pd:Cu ratio of the electroplated PdCu was varied to five different values and the condition of highest amperometric current at an identical of $H_2O_2$ concentration was chosen among them. The resulting amperometric current was highest when the ratio of Pd:Cu was 7:3 and this Pd;Cu ratio was employed in the sensor fabrication. The fabricated PdCu/LIG electrode based $H_2O_2$ sensor exhibited a sensitivity of $139.4{\mu}A/mM{\cdot}cm^2$, a broad linear range between 0 mM and 16 mM of $H_2O_2$ concentrations at applied potential of -0.15 V, and high reproducibility (RSD = 2.6%). The selectivity of the fabricated sensors was also evaluated by applying ascorbic acid, glucose, and lactose separately onto the sensor in order to see if the sensor ourput is affected by one of them and the sensor output was not affected. In conclusion, the proposed PdCu/LIG electrode based $H_2O_2$ sensor seems to be suitable $H_2O_2$ sensor in various applications.

Effect of κ-Carrageenan on Sperm Quality in Cryopreservation of Canine Semen (개 정액의 냉동보존 시 κ-Carrageenan이 정자 성상에 미치는 영향)

  • This study was conducted to find out the effect that ${\kappa}-Carrageenan$ has on the properties of dog sperm when it was added to the cryoprotectant. Extender basically was contained 1.21 g Trizma base, 0.67 g citric acid, 0.4 g glucose, 0.03 g penicillin G, 0.05 g streptomycin sulfate. Extender1 was added with 0.1%, 0.2%, 0.3%, and 0.5% carrageenan, while extender2 was supplemented with glycerol. After freezing-thawing, the motility, viability, acrosome integrity, apoptosis, and ROS (reactive oxygen specifications) of sperm were measured to analyze the effects of the supplementation of carrageenan. Total Motile (TM), Rapid Progressive Motile (RPM), Medium Progressive Motile (MPM), and Immotile were measured through the CASA system after thawing in 37 degree water. Extender with 0.2% ${\kappa}-carrageenan$ ($64.26{\pm}0.49$) was significantly higher than control ($40.24{\pm}8.27$) (p < 0.05). RPMs of extender with 0.1%, 0.2% ${\kappa}-carrageenan$ ($57.64{\pm}6.34$, $56.47{\pm}1.35$) were significantly higher than the other groups (p < 0.05). Acrosome integrity was measured by dyeing to PSA-FITC with an epifluorescence microscope. Normal acrosome ratio of extender with 0.5% ${\kappa}-carrageenan$ ($61{\pm}8.03$) was higher than the other groups (p < 0.05). Apoptosis was measured with a FACSCalibur flow cytometer using FITC (FITC Annexin V Apoptosis Detection Kit). Treated groups of ${\kappa}-carrageenan$ of 0.1% ($0.81{\pm}0.05$), 0.2% ($0.85{\pm}0.05$) were significantly higer (p < 0.05) than control. Modified SYBR/PI staining was used for determination of viability and DCF staining was used for evaluation of ROS. Viability and ROS were not significantly different from other groups. In conclusion, adding a certain concentration of carrageenan to the extender of cryopreservation, carrageenan contributes to the improvement of the sperm motility, acrosome integrity and prevention of apoptosis.

Fermented cottonseed meal improves production performance and reduces fat deposition in broiler chickens

  • Objective: This study was conducted to investigate the effect of fermented cottonseed meal (FCSM) on growth performance, carcass traits, and fat deposition in white-feather broiler chickens. Methods: A total of 480 male one-day-old white-feather broiler chickens were selected randomly and divided into four groups with six replicates of 20 chickens in each. The experimental chickens were fed diets including 3%, 6%, or 9% FCSM fermented by Candida tropicalis until 42 days old. In the experiment, the chickens of the control group were fed soybean meal. Results: FCSM supplementation linearly decreased the feed conversion ratio from d 15 to 21 and d 36 to 42, respectively (p<0.05). The percentage of carcass and semi-eviscerate increased in response to dietary FCSM supplementation at d 21 (p<0.05). The percentage of eviscerated and semi-eviscerate of 3FCSM was higher than that in other groups at d 35 (p<0.05). At the age of 42 d, the percentage of carcass increased in a quadratic way among increasing FCSM in diets (p<0.05). The subcutaneous fat thickness linearly decreased with the increasing levels of FCSM at d 21 (p<0.05). Gompertz and Logistic functions provided a better fit on abdominal fat and subcutaneous fat, respectively. The best fitted equation predicted that the maximum growth rate of abdominal fat weight and subcutaneous fat thickness occurred at d 28. FCSM had no significant effects on the shape of growth curve of abdominal fat weight and subcutaneous fat thickness, but reduced the height of the curve. Birds receiving the 6FCSM diet for 21 d had smaller adipocyte surface and lower serum glucose as well as triglyceride concentration. Conclusion: FCSM is beneficial for broiler chickens as it positively affects their growth and carcass in addition to altering their fat deposition.

Evaluation of γ-Aminobutyric Acid (GABA) Production by Lactic Acid Bacteria Using 5-L Fermentor (Lactic Acid Bacteria (LAB)와 5-L 발효기를 이용한 γ-Aminobutyric Acid 생산기술 개발)

  • This study aimed to optimize gamma-aminobutyric acid (GABA) production by employing five strains of lactic acid bacteria (LAB) that were capable of high cell growth and GABA production using a modified synthetic medium. GABA production in the strains was qualitatively confirmed via detection of colored spots using thin layer chromatography. Lactobacillus plantarum SGL058 and Lactococcus lactis SGL027 were selected as the suitable strains for GABA production. The conditions of the carbon and nitrogen sources were determined as 5 g/l glucose (L. plantarum SGL058), 5 g/l lactose (L. lactis SGL027), 10 g/l yeast extract (L. plantarum SGL058), and 20 g/l yeast extract (L. lactis SGL027) for GABA production. The cell growth, monitored by optical density at 600 nm, was 5.93 for L. plantarum SGL058. This value was higher than the 3.04 produced by L. lactis SGL027 at 36 h using a 5-L fermenter. The highest concentration of GABA produced was 546.7 ㎍/ml by L. plantarum SGL058 and 404.6 ㎍/ml by L. lactis SGL027, representing a GABA conversion efficiency of (%, w/w) of 4.0% and 3.4%, respectively. The fermentation profiles of L. plantarum SGL058 and L. lactis SGL027 provide a basis for the utilization of LAB in GABA production using a basal synthetic medium.

The role of discoid domain receptor 1 on renal tubular epithelial pyroptosis in diabetic nephropathy

  • Pyroptosis, a form of cell death associated with inflammation, is known to be involved in diabetic nephropathy (DN), and discoid domain receptor 1 (DDR1), an inflammatory regulatory protein, is reported to be associated with diabetes. However, the mechanism underlying DDR1 regulation and pyroptosis in DN remains unknown. We aimed to investigate the effect of DDR1 on renal tubular epithelial cell pyroptosis and the mechanism underlying DN. In this study, we used high glucose (HG)-treated HK-2 cells and rats with a single intraperitoneal injection of streptozotocin as DN models. Subsequently, the expression of pyroptosis-related proteins (cleaved caspase-1, GSDMD-N, Interleukin-1β [IL-1β], and interleukin-18 [IL-18]), DDR1, phosphorylated NF-κB (p-NF-κB), and NLR family pyrin domain-containing 3 (NLRP3) inflammasomes were determined through Western blotting. IL-1β and IL-18 levels were determined using ELISA. The rate of pyroptosis was assessed by propidium iodide (PI) staining. The results revealed upregulated expression of pyroptosisrelated proteins and increased concentration of IL-1β and IL-18, accompanied by DDR1, p-NF-κB, and NLRP3 upregulation in DN rat kidney tissues and HG-treated HK-2 cells. Moreover, DDR1 knockdown in the background of HG treatment resulted in inhibited expression of pyroptosis-related proteins and attenuation of IL-1β and IL-18 production and PI-positive cell frequency via the NF-κB/NLRP3 pathway in HK-2 cells. However, NLRP3 overexpression reversed the effect of DDR1 knockdown on pyroptosis. In conclusion, we demonstrated that DDR1 may be associated with pyroptosis, and DDR1 knockdown inhibited HG-induced renal tubular epithelial cell pyroptosis. The NF-κB/NLRP3 pathway is probably involved in the underlying mechanism of these findings.
