The study was performed to establish an efficient screening method for resistant cucumber to Fusarium oxysporum f. sp. cucumerinum. The isolate KR5 was identified as F. oxysporum f. sp. cucumerinum based on molecular analyses of ITS and TEF genes and host-specificity test on cucurbits including melon, oriental melon, cucumber, and watermelon. Then four cucumber and two rootstock cultivars showing different resistance degrees to the Fusarium wilt pathogen KR5 were selected. And development of Fusarium wilt of the six cultivars according to several conditions, including incubation temperature after inoculation, inoculum concentration, root wounding, and growth stages of seedlings, was investigated. Disease severity of Fusarium wilt on the resistant cultivars was changed with incubation temperatures after inoculation. The resistant cultivars showed the higher resistance when inoculated plants were kept at 25 or $30^{\circ}C$ than at $20^{\circ}C$. Among four different growth stages of the seedlings, seven-day-old seedling represented the most difference of resistance and susceptibility to Fusarium wilt. From above results, we suggest that an efficient screening method for resistant cucumber to F. oxysporum f. sp. cucumerinum is to dip the non-cut roots of seven-day-old seedlings in spore suspension of $1.0{\times}10^6-1.0{\times}10^7$ conidia/ml and to transplant the seedling into a non-infected soil, and then to incubate the inoculated plants in a growth room at $25^{\circ}C$ for 3 weeks to develop Fusarium wilt.
Five formae speciales of Fusarium oxysporum in Korea were examined using RFLP analysis to find the possibility for classification and analyze genetic variations. DNAs from F. oxysporum f. sp. lycopersici, cucumerinum, fragariae, garlic and sesami were used with three recombinant probes such as pFC46, pFC52 and pFC57. Distinct differences among five formae speciales of this fungus were detected in RFLP band patterns based on southern hybridization of genomic DNA using each recombinant clone, which was a repetitive copy probe. Strains belong to four formae speciales could be very stable in genetic variation except f. sp. sesami which has more variation than the others based on the RFLP analysis. They formed their own cluster which has high similarity within the same formae specialis resulted from the UPGMA analysis for genetic relationship analysis and each cluster represented its own formae specialis. The method using three recombinant DNA probes could be a good tool for classification of formae speciales in F. oxysporum.
Endophytic fungi have received much attention as plant growth promoters as well as biological control agents against many plant pathogens. In this study, 30 endophytic fungal species, isolated from various plants in China, were evaluated using in vitro dual culture assay against Fusarium oxysporum f. sp. cucumerinum, causing wilt in cucumber. The results of the present study clearly showed that all the 30 endophytic fungal isolates were highly capable of inhibiting the mycelial colony growth of Fusarium oxysporum f. sp. cucumerinum with inhibition % over 66% as compared to control treatments. Among all of them, 5 isolates were highly effective such as, Penicillium sp., Guignardia mangiferae, Hypocrea sp., Neurospora sp., Eupenicillium javanicum, and Lasiodiplodia theobromae, respectively. The Penicillium sp. and Hypocrea sp. were highly effective as compared to other isolates. From in vitro results 10 best isolates were selected for greenhouse studies. The results of the greenhouse studies showed that among all of them 3 endophytic fungal isolates successfully suppressed wilt severity when co-inoculation with pathogen Fusarium. oxysporum f. sp. cucumerinum. The endophytic fungi also enhanced plant growth parameters of the host plants, the antagonistic fungal isolates increased over all plant height, aerial fresh, and dry weight as compared to control.
Fusarium wilt caused by Fusarium oxysporum f. sp. cucumerinum has caused high damage in cucumber under greenhouse condition. This disease was suppressed $30\~55\%$ by application of organic matters compared with natural cropping soils. The suppression effect was the highest in the mushroom humic compost and fowl excretion matter among the various organic matters, varying with kinds of organic matters and degrees of humic resolvability. There was a slight difference in severity of fusarium wilt between sterilized organic matters and soils. The disease occurrence was delayed more in the nonsterilized organic matters and soils than in the sterilized ones. At 30 days after inoculation of F. oxysporum, numbers of Actinomycetes, fungi and bacteria were considerably increased, whereas F. oxysporum was decreased in the organic matter amended-soils compared with natural control soils.
In order to find out effect on the inorganic and organic compounds as a soil amendment to control Fusarium wilt of cucumber caused by Fusarium oxysporum f. sp. cucumerinum, this study was conducted during the last two years from 1993 to 1995. According to add 14 inorganic chemicals (1%, w/v) including $Al_2(SO_4)_3$ individually in vitro, these chemicals were confirmed their suppression, and especially Alum, CaO and $Al_2(SO_4)_3$ suppressed not only $20.9{\sim}25.0$ percent on mycelial growth of the fungus, but also inhibited $72.8{\sim}97%$ on conidial germination. $Ca(NO_3)_2$ suppressed mycelial growth only, while KCl, $K_2SO_4,\;NH_4NO_3$, and Urea suppressed conidial germination. The 7 chemicals were finally seleced. Composted pine bark (CPB) suppressed definely more than 90% on conidial germination in the different extract concentration (2,5 and 10%), although mycelial growth on extract medium of CPB and milled alfalfa leaves (MAL) were not remarkable. The antagonist Trichoderma sp. (Tr-3) mixed with an amended soil (1%, w/w) containing composted pine bark showed a good mycelial growth to compete the causal fungus. And the antagonist Pseudomonas sp. (7-1-3) was also confirmed its antagonistic ability with culture filterate. It is known that a CPB soil amendment mixed with the two antagonists (1%, w/w) controlled almost completely Fusarium wilt of cucumber in greenhouse pots and a field experiment. It is therefore expected that biocontrol on Fusarium wilt of cucumber by a soil amendment can be applied to farmmer's fields.
The intergenic spacer(IGS) sequence of Fusarium oxysporum have been reported to provide reliable information concerning intraspecific variation and phylogeny of fungal species. The eleven strains of Fusarium oxysporum and its formae speciales belonging to section Elegans were compared with sequencing analysis. The direct sequencing of partial IGS was carried out using PCR with primer NIGS1(5'-CTTCGCCTCGATTTCCCCAA-3')/NIGS2(5'-TCGTCGCCGACAGTTTTCTG-3') and internal primer NIGS3(5'-TCGAGGATCGATTCGAGG-3')/NIGS4(5'-CCTCGAATCGATCCTCGA-3'). A single PCR product was found for each strain. The PCR fragments were sequenced and revealed a few within species polymorphisms at the sequence level. The size of partial IGS sequencing of F. oxysporum was divided into three groups; $526{\sim}527$ bp including F. o. f. sp. chrysanthemi, cucumerinum, cyclaminis, lycopersici, and fragariae; $514{\sim}516$ bp including F. o. f. sp. lilii, conglutinans, and raphani; 435 bp for F. o. f. sp. cucumerinum from Korea. Sequence analysis of PCR products showed that transitions were more frequent than transversions as well as the average numbers of substitution per site were range 0.41% to 3.54%.
Bacteria and fungi antagonistic to Fusarium oxysporum f. sp. cucumerinum Owen were effectively isolated with each of modified Triple Layer Agar (TLA) technique from rhizosphere soil where cucumber had been grown healthily in plastic film house. Three predominant bacterial isolates selected were identified as Pseudomonas fluorescens, and P. putida, Serratia sp. and three fungal isolates were Gliocladium sp. Trichoderma harzianum, and T. viride. Antagonistic bacteria inhibited $26-45\%$ of germination and $41-56\%$ of germ tube elongation of microconidia of F. oxysporum f. sp. cucumerinum on Water Agar (WA). P. fluorescens was the strongest inhibitor. Several my co parasitisms were observed on dual culture of WA between antagonistic fungi and F. oxysporum f. sp. cucumerinum such as coiling, penetration, overgrowing, and lysis. Mycelial lysis of the pathogen was the most severe at pH 4.6, followed by 3.6, 5.6 and 6.6 of the medium in decreasing order. At pH 6.6, mycelia of the pathogen were not conspicuously damaged, however, the antagonistic fungi formed abundant chlamydospores especially Gliocladium sp. T. harzianum revealed the most excellent antagonism in vitro.
This study was carried out to isolate of antagonistic bacterium against Pythium ultimum and Rhizoctonia solani AG-4, causal pathogens of vegetables damping-off. Total of 600 strains were isolated from soil and plait roots. The isolates were screened for antagonism against Pythium ultimum and Rhizoctonia solani AG-4. One strain, named YJ-37, was sellected for detained study among those microoganisms screened. It was identified as Bacillus ehimensis based on morphological and physiological characterisitics according to the Bergey's mannual of systematic bacteriology, Sherlock system of Microbial ID Inc. and 16S rDNA sequences methods. Furthermore Bacillus ehimensis YJ-37 showed antifungal activities against Alternaria altrata, Collectotrichum gloeosporioides, Didymella bryoniae, Fusarium moniliforme, Fusarium oxysporum, F. oxysporum cucumerinum, F. oxysporum niveum, Gloeosporium sp., Glomerella sp., G. cingulata, G. lagenaria, Penicillium digitatum, P. italicum, Phytophthora capsici, Sclerotinia sclerotiorum, and Stemprhylium solani.
The growth of germ tube of Fusarium oxysporum f. sp. cucumerinum was remarkably inhibited on the water agar treated with 100ppm of Fe-EDDHA, a synthetic iron chelating agent, whereas germination rate of microconidia did not show much differences compare with that of non treated water agar. Both of the germination and the germ tube elongation of microconidia were suppressed significantly in King's B agar by the bacterial siderophores produced by Pseudomonas putida. The highest germination of the chlamydospores was obtained in the soil added with $0.25\%$ of glucose plus $0.05\%$ of asparagine. The chlamydospores of cucumber wil fungus germinated about $14\%$ in rhizosphere soil of 2 day-old cucumber seedlings within 48 hours, and the germination was enhanced notably in rhizosphere soil of 10 day-old seedling. But the rates of germination was not increased according to cucumber growth age after 10 day-old seedling. The effect of P. putida and Fe-EDDHA on the germination on chlamydospores in conducive soil was not pronounced in the non-rhizosphere soil added with nutrient. However, the germination was suppressed significantly both in rhizosphere soil and in rhizosphere soil added with nutrient. The suppression of chlamydospore germination was greater in the bacteria inoculated soil than that in Fe-EDDHA treated soil.
