This study was designed to investigate the effects of sea tangle (Laminaria japonica) extract and fucoidan components on chronic degenerative diseases. Sprague-Dawley(SD) male rats (210$\pm$5g) were fed experimental diets: Dasi-Ex group: dasima extract powder of 4.0% added to control diet; Fuco-I, II and III groups: fucoidan powder of 1, 2 and 3% added to Dasi-Ex group for 45 days. Triglyceride (TG) levels in serum were significantly lower (10~15%) in Fuco-I, II and III groups compared with control group. Total cholesterol levels were significantly decreased (7~10% and 15~ 35%) in brain mitochondria and microsomes of Fuco-II and III group compared with control group. LDL-cholesterol levels were remarkably decreased (20~30%) in Dasi-Ex and Fuco-I, II, III groups, but HDL-cholesterol levels were significantly increased (10~12%) in Fuco-II and III groups only compared with control group. The ratios of HDL/total cholesterol resulted in a marked increase (3 5~55%) in Dasi-Ex and Fuco-I, II, III groups, but atherogenic indices were remakably decreased (40~50%) in Dasi-Ex and Fuco-I, II, III groups compared with control group. Membrane fluidities were remarkably increased (45~70% and 38~42%, respectively) in brain mitochondria and microsomes of Fuco-II and III groups compared with control group. Administrations of fucoidan added to dasima effectively decreased TG, total and LDL-cholesterol, and atherogenic index, while also effectively increased HDL-cholesterol, HDL/total cholesterol ratio, and membrane fluidity, suggesting chronic degenerative diseases were very effectively prevented by the administration of fucoidan component.

ICR male mice (20$\pm$2g) were fed basic experimental diets and given free through water bottle filled with these beverages instead of water for 18 days including sociopsychological stress. Body weight gains were consistently lower in Dasi-Ex and Fuco-I,II,III groups compared with control group, expecting in a inhibitory effect of obesity. Dasi-Ex group resulted in a significant decrease of 25% in serum corticosterone (CS) secretion, while Fuco-I,IIand IIIgroups resulted in a marked decreases of 45~55% in serum CS secretion compared with control group. Noradrenaline (NA) secretions were significantly increased about 15% in Dasi-Ex group, and 20~22% in Fuco-I,II,III groups compared with control group. Significant differences in brain MHPG-SO4 levels of Dasi-Ex group could not be obtained, but Fuco-I,II,III groups resulted in a marked decreases of 20~25% in brain MHPG-SO4 ratio of brain, but Fuco-I,II and III groups resulted in a marked increases of 45~60% in NA/MHPG-SO4 ratio of brain compared with control group. These results suggest that fucoidan beverage may play a effective role in a ridding of the sociopsychological stress by pivotal anti-stress effect of fucoidan.
Rheological properties of partially purified fucoidans of Laminaria religiosa, sporophylls of Undaria pinnatifida, Hizikia fusiforme, and Sargassum fulvellum in Korea were investigated, The fucoidans produced aqueous solutions of low apparent viscosity with pseudoplastic flow behavior. The viscosity of $5\%$ solution of the fucoidan from sporophylls of U. pinnatifida was 2.2 mPa.s. The activaton energies of L. religiosa, sporophylls of U. pinnatifida, H. fusiforme and S. fulvellum were $8.269\times10^6,\;7.498\times10^6,\;7.141\times10^6,\;and\;6.677\times10^6\;J/Kg.mol$ at 1,500 1/s of shear rate, respectively. The viscosity of the fucoidan solution was increased by the addition of sugar, but that was not changed by addition of NaCl.
The anticancer and immunomodulatory activities of low molecular weight (Mw 11 kDa) fucoidan isolated from Hizikia fusiforme (H. fusiforme) via the ultrasonification extraction process were assessed in this study. Low molecular weight fucoidan improved the growth of human B and T cells, up to approximately 40% as compared to the controls (untreated) and 30% for commercially available fucoidan (Mw 150 kDa). IL-6 and TNF-$\alpha$ were secreted from human B cells at levels of $7.8\times10^{-4}$ pg/mL and $7.2\times10^{-4}$ pg/mL, respectively, and these levels were higher than the levels measured in the controls and with other high molecular weight fucoidan. It was also determined that the cytokine from human B and T cells cultivated with added fucoidan enhanced the growth of human NK cells. The fucoidan isolated from H. fusiforme showed low cytotoxicity, approximately 19%, after the addition of 1.0 mg/mL, the highest tested concentration. The growth of human lung cancer cells (A549) and human breast cancer cells (MCF-7) were inhibited by 69.8% and 83.3%, respectively. These results demonstrated that the low molecular weight fucoidan isolated from H. fusiforme has potential as a new functional food component that evidences immunomodulatory activities and anticancer activity. One of the primary positive features of this fucoidan is that low molecular weight polysaccharides can be readily handled during processing.
The radical scavenging activity of methanol extracts of seaweeds and their active compounds, alginic acid, fucoidan and phloroglucinol, were investigated under in vitro. Among methanol extracts of seaweeds (sea mustard, sea tangle, seaweed papulosa, fusiforme, sea lettuce, purple laver and chlorella), seaweed papulosa and sea tangle showed strong scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and hydroxyl radical (${\cdot}OH$). In addition, under in vitro, the scavenging activities on DPPH radical of alginic acid and fucoidan, which are active compounds of brown algae, and phloroglucinol, the active compound from Ecklonia species, were evaluated and compared. Fucoidan and phloroglucinol showed strong DPPH scavenging effect, in particular, phloroglucinol had strongest activity among the active compounds. On the other hand, alginic acid did not exert DPPH scavenging activity. From the present study, we could confirm the antioxidative activity of seaweeds and its active compounds.
Fucoidans were isolated from Hizikia fusiformis and Sargassum fulvellum and characterized on their chemical properties. Crude fucoidans were extracted at $65^{\circ}C$ for 1hr with acid solution of pH 2.0, and cetylpyridinum chloride was used for partial purification. The yields of partially purified fucoidans were $2.51\%$ for H. fusiformis, and $65\%$ for S. fulvellum, respectively. The partially purified fucoidans were separated into 3 fractions by DEAE-Sephadex A-25 ion exchange column chromatography and the major fractions were refractionated with fractional preripitation with ethanol. $60-70\%$ ethanol precipitated fraction(P-70) of H. fusiformis and $60-65\%$ ethanol precipitated fraction(P-65) of S. fulvellum turned out to be homogeneous by cellulose acetate electrophoresis and gel filtration chromatography. The molar ratios of fucose, galactose, and sulfate In the purified fucoidans were 1 : 0.66 2.74 for H. fusiformis and 1 : 0.24. 1.46 for 5. fulvellum. The averaged molecular weights of the purified fucoidans from H. fusiformis and S. fulvellum were 26,000 and 105,000, respectively.

In order to improve the durability of the PEMFC(Proton Exchange Membrane Fuel Cells) polymer membrane, a radical scavenger and a support are used. In this study, the durability of membranes containing fucoidan extracted from seaweeds and tannic acid serving as a crosslinking agent is evaluated to improve chemical and physical durability. Physical durability is evaluated by measuring tensile strength, and chemical durability is measured by Fenton experiment. Membrane and electrode assembly (MEA) is prepared and mechanical and chemical durability are measured through accelerated durability evaluation in the cell. The tensile strength measurement showed that fucoidan and tannic acid can improve the mechanical durability of the membrane by improving the strain rate and yield strength. It is shown in Fenton experiment that fucoidan acts as a radical scavenger. As a result of the accelerated durability test in the unit cell, fucoidan improved both chemical and mechanical durability, increasing the accelerated durability evaluation time by 38.1% compared to the additive-free membrane. When tannic acid is added, the durability of the polymer membrane is improved by 13.9% by improving the mechanical durability.
A fucoidanase, which was screened from the heptopancreas of Patinopectin yessoensis, was fractionated according to by ammounium sulfate precipitation and anion exchange chromatography. The crude fucoidanase activity was hydrolyzed a small amounts of fucoidan agar and not arrageenan. The crude enzyme hydrolyzed fucoidan to produce oligosaccha-rides of fucose, glucose, maltose maltotriose and maltoteraose as the reaction products.
A fucoidan, isolated from Korean Undaria pinnatifida spoprophyll (UP-F), was investigated for its immunomodulating activity on murine macrophages and splenocytes, and its activity was compared with that of fucoidan from Fucus vesiculosus (FV-F). Treatment of UP-F resulted in inhibition of the growth of murine macrophage RAW 264.7 cells, but its cytotoxicity was not observed in normal murine splenocytes. FV-F was shown to be highly cytotoxic to both immune cells, and its cytotoxic activity was higher than that of UP-F. Treatment of UP-F induced TNF-α in a dose-dependent manner from two types of macrophages, RAW 264.7 cells and murine peritoneal macrophages. The TNF-α-inducing activity of UP-F was higher than that of FV-F. UP-F also actively induced chemokines (RANTES and MIP-1α) from RAW 264.7 cells. Furthermore, treatment of UP-F gave rise to activation of murine splenocytes to produce cytokine (IL-6) and chemokines (RANTES and MIP-1α), showing significantly higher activity than that of FV-F. These results indicate that UP-F is less cytotoxic to immune cells than FV-F, and possesses immunomodulating activity to produce cytokines and chemokines from macrophages and splenocytes.

This study was conducted to investigate the effects of dietary inclusion of various additives Solid sulfur, fucoidan, and glucan on growth performance and immune responses in Juvenile olive flounder paralichthys olivaceus. Six experimental diets supplemented with : no additives (CON); 0.5 and 1.0% solid sulfur (S); 0.2 and 0.4% fucoidan (F); 0.1% glucan (G) of these additives diet on a dry-matter basis. Fish averaging $8.2{\pm}0.17g$ were fed one of six experimental diets in triplicate groups for 6 weeks. By the end of the feeding trial, weight gain, feed efficiency, specific growth rate, protein efficiency ratio, hepatosomatic index and condition factor of fish fed diet CON were significantly lower then those of fish fed the all additives diets (P < 0.05). In challenge test, fish were infected by intraperitonel injection of 0.1 ml bacterial suspension with Edwardsiella tarda per fish after the feeding trial. As a result fish fed All additive diet showed a lower cumulative mortality then did fish fed CON diet throughout the challenge test. In conclusion, these results indicated that solid sulfur, fucoidan and glucan enhanced the growth, feed efficiency and non-specific immune activity of juvenile flounder and protect the fish against microbial infections.
