Contact interface fiber section element: shallow foundation modeling

  • With recent growing interests in the Performance-Based Seismic Design and Assessment Methodology, more realistic modeling of a structural system is deemed essential in analyzing, designing, and evaluating both newly constructed and existing buildings under seismic events. Consequently, a shallow foundation element becomes an essential constituent in the implementation of this seismic design and assessment methodology. In this paper, a contact interface fiber section element is presented for use in modeling soil-shallow foundation systems. The assumption of a rigid footing on a Winkler-based soil rests simply on the Euler-Bernoulli's hypothesis on sectional kinematics. Fiber section discretization is employed to represent the contact interface sectional response. The hyperbolic function provides an adequate means of representing the stress-deformation behavior of each soil fiber. The element is simple but efficient in representing salient features of the soil-shallow foundation system (sliding, settling, and rocking). Two experimental results from centrifuge-scale and full-scale cyclic loading tests on shallow foundations are used to illustrate the model characteristics and verify the accuracy of the model. Based on this comprehensive model validation, it is observed that the model performs quite satisfactorily. It resembles reasonably well the experimental results in terms of moment, shear, settlement, and rotation demands. The hysteretic behavior of moment-rotation responses and the rotation-settlement feature are also captured well by the model.

Multilevel Precision-Based Rational Design of Chemical Inhibitors Targeting the Hydrophobic Cleft of Toxoplasma gondii Apical Membrane Antigen 1 (AMA1)

  • Toxoplasma gondii is an intracellular Apicomplexan parasite and a causative agent of toxoplasmosis in human. It causes encephalitis, uveitis, chorioretinitis, and congenital infection. T. gondii invades the host cell by forming a moving junction (MJ) complex. This complex formation is initiated by intermolecular interactions between the two secretory parasitic proteins-namely, apical membrane antigen 1 (AMA1) and rhoptry neck protein 2 (RON2) and is critically essential for the host invasion process. By this study, we propose two potential leads, NSC95522 and NSC179676 that can efficiently target the AMA1 hydrophobic cleft, which is a hotspot for targeting MJ complex formation. The proposed leads are the result of an exhaustive conformational search-based virtual screen with multilevel precision scoring of the docking affinities. These two compounds surpassed all the precision levels of docking and also the stringent post docking and cumulative molecular dynamics evaluations. Moreover, the backbone flexibility of hotspot residues in the hydrophobic cleft, which has been previously reported to be essential for accommodative binding of RON2 to AMA1, was also highly perturbed by these compounds. Furthermore, binding free energy calculations of these two compounds also revealed a significant affinity to AMA1. Machine learning approaches also predicted these two compounds to possess more relevant activities. Hence, these two leads, NSC95522 and NSC179676, may prove to be potential inhibitors targeting AMA1-RON2 complex formation towards combating toxoplasmosis.

Comparison of Safety Margin of Shallow Foundation on Weathered Soil Layer According to Design Methods (설계법에 따른 풍화토 지반 얕은기초의 안전여유 비교)

  • In this paper bearing capacity and safety margin of shallow foundation on weathered soil ground against shear failure by using current design method of allowable stress design (ASD), load resistance factor design (LRFD) based on reliability analysis and partial safety factor design (PSFD) in Eurocode were estimated and compared to each other. Results of the plate loading test used in construction and design were collected and analysis of probability statistics on soil parameters affecting the bearing capacity of shallow foundation was performed to quantify the uncertainty of them and to investigate the resistance bias factor and covalence of ultimate bearing capacity. For the typical sections of shallow foundation in domestic field as examples, reliability index was obtained by reliability analysis (FORM) and the sensitivity analysis on soil parameters of probability variables was performed to investigate the effect of probability variable on shear failure. From stability analysis for these sections by ASD, LRFD with the target reiability index corresponding to the safety factor used in ASD and PSDF, safety margins were estimated respectively and compared.

Evaluation of Rocking Behaviors During Earthquake for the Shallow Foundation System on the Weathered Soil Using Dynamic Centrifuge Test (동적 원심모형실험을 이용한 풍화토 지반에 놓인 얕은기초 시스템의 지진 시 회전 거동 특성 평가)

  • Rocking behavior of shallow foundation during the earthquake can reduce the seismic load of the superstructure. The dynamic centrifuge tests were performed to investigate the availability of using rocking behavior for the weathered soil condition. The centrifuge test model was composed of the weathered soil, shallow foundation and single degree of freedom structure. And the accelerations of soil, foundation and structure, and the foundation settlement were measured during the earthquake. From the test result, the seismic load of the structure for the strong earthquake input was reduced by the rocking behavior with foundation uplift and the maximum foundation settlement was less than 0.5% of the foundation width. This shows the potential that the rocking foundation concept can be used in the economical seismic design of foundation for the weathered soil in the future with additional research and verification.

  • The piled-raft foundation, a new design concept, is one of the most effective kinds of foundation for reducing settlement of structures. An alternative piled-raft system with disconnection cap and a sand cushion between the pile and raft was also investigated to compare the influence of ultimate bearing capacity and settlement. Load-settlement relation curves were used to evaluate the ultimate bearing capacity. In the numerical analyses, a plane strain elasto-plastic finite element model (Mohr-Coulomb model) was used to present the response of the piled-raft foundation.

  • This paper is analysis of stiffened opening thick plate on foundation. This paper has the object of investigating natural frequencies of opening thick plates on Pasternak foundation by means of finite element method and providing Kinematic design data for mat of building structures. In this paper, vibration analysis of rectangular opening thick plate is done by use of Serendipity finite element with 8 nodes by considering shearing strain of plate. And vibration analysis of stiffener is done by used of Timoshenko beam-column element wit 3 nodes. It is shown that natural frequencies depend on not only Winkler foundation parameter but also shear foundation parameter, opening position, opening size, stiffener size.

Vibration analysis that opening plate subjected to In-plane stress is presented in this paper. Finite element analysis of rectangular opening plate is done by use of rectangular finite element with 8-nodes. In order to analysis plate which is supported on Pasternak foundation, the Winkler foundation parameter is varied with 0, 10, 102, 103 and the shear foundation parameter is 0, 5, 10, 15. The ratio of In-plane force to critical load is applied as 0.2, 0.8, respectively. This paper analyzed varying opening Position and opening size.

  • This paper has the object of investigating dynamic stability of stiffened tapered thick plate with concentrated mass on Pasternak foundation by means of finite element method and providing kinematic design data for mat of building structures. Finite element analysis of stiffened tapered thick plate is done by use of rectangular finite element with 8-nodes. In order to analysis plate which is supported on Pasternak foundation, the Winkler foundation parameter is varied with 10, 100, 1000 and the shear foundation parameter is 5, 10, concentrated mass is $0.25m_c$, $1.0m_c$, tapered ratio is 0.25, 0.5. The ratio of In-plane force to critical load is applied as $0.4\sigma_{cr},\;0.6\sigma_{cr},\;0.8\sigma_{cr}$ respectively. This paper analyzed varying tapered ratio.
