RP-HPLC를 이용한 반하백출천마탕에서 12종 지표성분의 동시분석 (Simultaneous analysis of 12 kind maker components in Banhabaekchulchhonma-tang by RP-HPLC)

  • 반하백출천마탕은 다양한 약리활성을 가진 전통한방처방이다. 본 연구는 한방처방의 품질관리를 위한 기초자료로서, 고성능액체 크로마토그래피 (RP-HPLC)을 이용한 반하백출천마탕에서 12종 지표성분 Homogentisic acid, Hesperidin, Naringin, Alisol A, Atractylenolide II, Atractylenolide III, Ginsenoside Rg1, Formononetin, Gastrodin, Berberine, Palmatine, 6-gingerol를 동시분석 하였다. 표준액은 상업용 $C_{18}$ 역상 컬럼과 이동상으로 물(0.1% TFA)과 아세토나이트릴을 사용하여 기울기용매조성법인 유속은 1.0mL/min, 주입부피는 $10{\mu}L$, 컬럼오븐 온도 $40^{\circ}C$, 파장 200, 220, 280, 340nm에서 수행되었다. 이 결과 모든 표준성분의 검량선은 우수한 직선성 $r^2$ >0.999으로 나타내었다. 또한 검출한계(LOD)와 정량한계(LOQ)는 $0.012{\sim}0.878{\mu}g/mL$$0.009{\sim}0.290{\mu}g/mL$, 정밀성 intra-day와 inter-day는 0.07~1.21%및 0.20~0.90%, 회수율은 97.17~108.40%, 각각의 함량은 매우 미미한 변화를 타나내었다. 이 연구결과는 실험적으로 입증된 동시분석법의 확립에 따라 반하백출천마탕의 품질평가를 효율적으로 보여주었다.

신령버섯 자실체 메탄올 추출물의 항산화, 항당뇨 및 Nitric Oxide의 저해 효과 (Antioxidant, Anti-diabetic, Anti-cholinesterase, and Nitric Oxide Inhibitory Activities of Fruiting Bodies of Agaricus brasiliensis)

  • 신령버섯(Agaricus brasilliensis)는 식용과 약용으로 사용되고 있는 담자균류에 속하는 버섯이다. 본 연구에서는 신령버섯의 자실체를 메탄올과 열수를 이용해 추출한 물질의 항산화, 항당뇨 및 nitric oxide 생성 저해 효과에 대한 실험을 수행하였다. 고속액체 크로마토그래피를 이용해 이 버섯 자실체 추출물의 페놀성 화합물을 분석한 결과 gallic acid, procatechuic acid, chlorogenic acid, (-)-epicatechin, vanillin, rutin hydrate, naringin, quercetin, formononetin, biochanin-A 등 총 10종류의 페놀성 화합물이 확인되었다. 항산화 효과 실험에서 메탄올 추출물의 DPPH radical 소거능은 양성대조군으로 사용한 BHT에 비해 낮았지만 비교적 효과가 우수하였으며, 철 이온 제거 항산화 효과는 메탄올 추출물이 양성대조군인 BHT에 비해 모든 실험 농도에서 유의성 있게 높았다. 환원력의 경우 이 버섯의 추출물은 양성대조군에 비해 모두 모든 농도에서 효과가 낮게 나타났다. 항당뇨 실험에서 ${\alpha}$-glucosidase에 대한 메탄올추출물의 저해효과는 1.0 mg/mL와 2.0 mg/mL의 농도에서 각각 54.48%와 78.43%를 보여 양성대조군인 acarbose의 51.77%과 81.81%에 비해 조금 낮았다. 염증저해 효과 실험에서 RAW 264.7 대식세포가 배양되고 있는 배지에 신령버섯 자실체의 메탄올과 열수추출물을 각각 전 처리 한 후 염증 매개 물질인 LPS를 투여하여 메탄올과 열수 추출물의 NO 생성 저해효과를 조사한 결과 추출물의 농도가 증가함에 따라 대식세포에 생성된 NO의 양이 감소하는 경향을 나타내어 높은 농도에서 저해효과가 우수한 것으로 나타났다. 따라서 신령버섯의 자실체에는 항산화, 항당뇨, 및 항염증 효과를 나타내는 천연물질이 함유되어 있어서 이를 섭취할 경우 건강에 유용한 것으로 생각되었다.

Inhibitory Effects of Flavonoids from Spatholobus suberectus on Sortase A and Sortase A-Mediated Aggregation of Streptococcus mutans

  • Seven flavonoids were isolated from Spatholobus suberectus via repetitive column chromatography and high-performance liquid chromatography. The chemical structures of these compounds were identified by spectroscopic analysis and comparison with values reported in the literature. Among the flavonoids tested, 7-hydroxy-6-methoxyflavanone (1) and formononetin (4) exhibited strong inhibitory activity against Streptococcus mutans SrtA, with $IC_{50}$ values of 46.1 and $41.8{\mu}M$, respectively, but did not affect cell viability. The onset and magnitude of inhibition of saliva-induced aggregation in S. mutans treated with compounds 1 and 4 were comparable to the behavior of a srtA-deletion mutant without treatment.

Garlic Fermentation by Lactic Acid Bacteria

  • Garlic has been used for condiments and also for medicines to cure various diseases since ancient times. Many studies on the processing of garlic have been published, however, few of them were related with fermentation because of the antimicrobial action of the garlic. In this study, to conduct garlic fermentation, 4 lactic acid bacteria (LAB) strains with growth abilities in garlic medium were selected. Addition of various nitrogen, carbon, and mineral sources generally did not improve the growth of experimental strains during garlic fermentation except for Lactobacillus casei KFRI 704 by yeast extract and Lactococcus lactis subsp. cremoris ATCC 19257 by mineral sources. High performance liquid chromatography (HPLC) analysis of 32 phenolic compounds during fermentation showed that formononetin was decreased time dependently. The concentrations of volatile compounds and alliin did not change during fermentation. The results of this study would provide the basic understanding of garlic fermentation by selected strains of LAB.

Quantitative Analysis of Isoflavones and Lignans in Sea Vegetables Consumed in Korea Using Isotope Dilution Gas Chromatography-Mass Spectrometry

  • The phytoestrogens including isoflavones (genistein, daidzein, biochanin A, formononetin, and glycitein), coumestrol, and lignans (secoisolariciresinol, matairesinol, and anhydrosecoisolariciresinol) were quantified in edible sea vegetables from Korea. Sea vegetable samples were collected based on domestic consumption data. After hydrolysis of phytoestrogen glycosides in prepared samples, aglycones of phytoestrogens were extracted with diethyl ether and analyzed with isotope dilution gas chromatography-mass spectrometry in selected ion monitoring mode (ID-GC-MS-SIM). Total samples included 19 samples representing eight species. Most of the samples showed rather low concentrations, ranging from not determinated to $79.2\;{\mu}g/kg$ for isoflavones and from 106.4 to $694.8\;{\mu}g/kg$ for lignans. The daily intake of phytoestrogen from sea vegetables, estimated from the present data and domestic consumption data, was about $0.13\;{\mu}g/day$ for isoflavones and $2.0\;{\mu}g/day$ for lignans. When we compared these results with those from legumes, sea vegetables would not be considered the major source of phytoestrogens in the Korean diet.

Lactobacillus plantarum 발효에 의한 갈근탕의 생물 전환 성분 연구 (Bioconversion Constituents of Galgeun-tang Fermented by Lactobacillus plantarum)

  • Galgeun-tang (GGT) is a traditional medicinal formula that is widely prescribed to treat cold, asthma, and hives in Korea. Fermented herbal medicines can be made more effective than normal herbal medicines by increasing the absorption and bioavailability of the active compounds. In this study, we fermented Galgeun-tang to produce bioconversion constituents using Lactobacillus plantarum (GGT144), and found that four peaks were decreased, three peaks were increased and two new peaks appeared in the HPLC-DAD chromatogram. After HPLC-DAD-guided fractionation of the newly-appearing compounds (1 and 5) and the increased (6, 7, and 9) compounds, the structure of the compounds was determined using NMR and MS. Using this approach the compounds were identified to be pyrogallol (1), daidzein (5), liquiritigenin (6), cinnamyl alcohol (7), and formononetin (9), respectively. In addition, the decreased compounds were identified to be daidzin (2), liquiritin (3), ononin (4), and cinnam aldehyde (8) using HPLC-DAD analysis with standard compounds. The high performance liquid chromatography method was used to quantify the nine constituents in GGT and GGT144. All calibration curves of the standard compounds displayed excellent linearity with a $R^2$ > 0.9968.

Assessment of Antioxidant and Phenolic Compound Concentrations as well as Xanthine Oxidase and Tyrosinase Inhibitory Properties of Different Extracts of Pleurotus citrinopileatus Fruiting Bodies

  • Cellular damage caused by reactive oxygen species has been implicated in several diseases, thus establishing a significant role for antioxidants in maintaining human health. Acetone, methanol, and hot water extracts of Pleurotus citrinopileatus were evaluated for their antioxidant activities against ${\beta}$-carotene-linoleic acid and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals, reducing power, ferrous ion-chelating abilities, and xanthine oxidase inhibitory activities. In addition, the tyrosinase inhibitory effects and phenolic compound contents of the extracts were also analyzed. Methanol and acetone extracts of P. citrinopileatus showed stronger inhibition of ${\beta}$-carotene-linoleic acid compared to the hot water extract. Methanol extract (8 mg/mL) showed a significantly high reducing power of 2.92 compared to the other extracts. The hot water extract was more effective than the acetone and methanole extracts for scavenging DPPH radicals. The strongest chelating effect (92.72%) was obtained with 1.0 mg/mL of acetone extract. High performance liquid chromatography analysis detected eight phenolic compounds, including gallic acid, protocatechuic acid, chlorogenic acid, ferulic acid, naringenin, hesperetin, formononetin, and biochanin-A, in an acetonitrile and hydrochloric acid (5 : 1) solvent extract. Xanthine oxidase and tyrosinase inhibitory activities of the acetone, methanol, and hot water extracts increased with increasing concentration. This study suggests that fruiting bodies of P. citrinopileatus can potentially be used as a readily accessible source of natural antioxidants.

Anti-Helicobacter pylori Compounds from Maackia amurensis

  • Eight isoflavonoid compounds were isolated from the EtOAc fraction of Maackia amurensis which had shown the highest anti-Helicobacter pylori activity among the fractions, using medium pressure liquid chromatography and recrystallization. Based on the spectroscopic data including $^1H$-NMR, $^{13}C$-NMR, HMBC and MS data, the chemical structures of the isolates were determined to be (-)-medicarpin (1), afromosin (2), formononetin (3), tectorigenin (4), prunetin (5), wistin (6), tectoridin (7) and ononin (8). Anti-H. pylori activity of each compound was evaluated with broth dilution assay. As a result, (-)-medicarpin (1), tectorigenin (4) and wistin (6) showed anti-H. pylori activity. (-)-Medicarpin (1) exhibited the most potent growth inhibitory activity against H. pylori with the minimal inhibitory concentration $(MIC)_{90}$ of $25{\mu}M$, and tectorigenin (4) with $MIC_{90}$ of $100{\mu}M$ ranked the second. This is the first study to show the anti-H. pylori activity of M. amurensis, and it is suggested that the stem bark of M. amurensis or the EtOAc fraction or the isolated compounds can be a new natural source for the treatment of H. pylori infection.

Antioxidant Activities and Tyrosinase Inhibitory Effects of Different Extracts from Pleurotus ostreatus Fruiting Bodies

  • We evaluated the antioxidant activity and tyrosinase inhibitory effects of Pleurotus ostreatus fruiting bodies extracted with acetone, methanol, and hot water. The antioxidant activities were tested against $\beta$-carotene-linoleic acid, reducing power, 1,1-diphenyl-2-picrylhydrazyl free radical scavenging activity, and ferrous chelating ability. Furthermore, phenolic acid and flavonoid contents were also analyzed. The methanol extract showed the strongest $\beta$-carotene-linoleic acid inhibition as compared to the other exracts. The acetone extract (8 mg/mL) showed a significantly high reducing power of 1.54 than the other extracts. The acetone extract was more effective than other extracts for scavenging on 1,1-diphenyl-2-picrylhydrazyl radicals. The strongest chelating effect (85.66%) was obtained from the acetone extract at 1.0 mg/mL. The antioxidant activities of the extracts from the P. ostreatus fruiting bodies increased with increasing concentration. A high performance liquid chromatography analysis detected seven phenolic compounds, including gallic acid, protocatechuic acid, chlorogenic acid, naringenin, hesperetin, formononetin, and biochanin-A in an acetonitrile and 0.1 N hydrochloric acid (5 : 1) solvent extract. The total phenolic compound concentration was $188{\mu}g$/g. Tyrosinase inhibition of the acetone, methanol, and hot water P. ostreatus extracts increased with increasing concentration. The results revealed that the methanol extract had good tyrosinase inhibitory ability, whereas the acetone and hot water extracts showed moderate activity at the concentrations tested. The results suggested that P. ostreatus may have potential as a natural antioxidant.

β-glucosidase 활성 균주 발효에 의한 황기 Isoflavonoid 성분변화 및 생리활성 (The Isoflavonoid Constituents and Biological Active of Astragalus Radix by Fermentation of β-glucosidase Strains)

  • Background: In this study, the radix of Astragalus membranaceus Bunge extract fermented by Saccharomyces cerevisiae, Weissella cibaria, and Pediococcus pentosaceus to increase the levels of isoflavonoid aglycone contents. Methods and Results: In order to change the in isoflavonoids, we fermented the radix of A. membranaceus extracts with microorganisms that have β-glucosidase activity. Besed on the β-glucosidase activity, we selected three strains, Weissella cibaria, Pediococcus pentosaceus, and Saccharomyces cerevisiae. HPLC analysis revealed that the levels of isoflavonoid aglycones were increased in all fermentation cases, and the extracts fermented by S. cerevisiae showed the highest levels of isoflavonoid aglycones. We evaluated the antioxidant activity, anti-wrinkle effects and whitening effects of the S. cerevisiae-fermented extracts using the DPPH assay, tyrosinase inhibition activity assay, and collagenase inhibition activity assay. We confirmed higher activity in S. cerevisiae-fermented extracts than in control, with the half maximal inhibitory concentration (IC50) value of 565.1 ± 59.1 ㎍/㎖ in DPPH radical scavenging activity, tyrosinase inhibition rate of 78.4 ± 0.9%, and collagenase inhibition rate of 83.8 ± 1.1%. Conclusions: We selected three stains of microorganisms showing high β-glucosidase activity, W. cibaria, P. pentosaceus and S. cerevisiae. Isoflavonoid glycones in the radix of A. membranaceus were converted to isoflavonoid aglycones by fermentation. In addition, the fermented radix of A. membranaceus exhibited antioxidant activity, anti-wrinkle effect, whitening effect and radical scavenging activity.
