Chromosomal Localization and Distribution of the Telomeric DNA in Cattle and Pigs (소, 돼지 염색체의 telomeric DNA 분포 양상)

  • Telomeres are nucleoprotein structures at the ends of chromosomes consisting of tandem repeat sequences of . (TTAGGG)n. Telomeres serve as guardians of the genome, protect individual chromosomes within the nucleus, and help in meiotic pairing of homologous chromosomes. To investigate the telomere distributions of cattle and pig chromosomes, fluorescence in situ hybridization(FISH) was carried out on metaphase spreads of in vitro fibroblast cultures from Holstein and Landrace using a human telomeric DNA repeat probe. Results indicate that the distinct double spots on both ends of chromosomes of cattle and pigs were observed. In cattle, there was a random variation in the intensity of telomere signals among chromosomes. In pigs, an interstitial telomeric signal was observed on the chromosome 6q1 of all the cells examined. According to quantitative fluorescence in situ hybridization(Q-FISH) analysis, some chromosomes had consistently much more telorneres at one end of chromosomes. In general, both species had consistently much more telomeres at q-end than p-end on most of chromosomes. The relative amount of telomeres on bovine chromosomes was higher than that on pig chromosomes. In additions, Y chromosome had the highest relative amount of telorneres in cattle and pigs.

In vitro Anti-diabetic Effects of Crude Extracts of Platycodi Radix (In vitro에서 길경 추출 분획물의 항당뇨 효과 조사)

  • Anti-diabetic effect of Platycodi radix (PR) extract fractions was determined if vitro by investigating insulin-like action, insulin sensitizing action, glucose-stimulated insulin secretion, gene expression related to ${\beta}-cell$ function and mass, and ${\alpha}$-glucoamylase suppressing action. Insulin-like activity was not promoted by the treatment of PR methanol factions in 373-L1 fibroblast. However, treatment with 0, 20 and 100% PR methanol fractions along with 1 ng/mL insulin increased insulin-stimulated glucose uptake in 373-L1 adipocytes. In addition, the treatment of 0% and 100% methanol fractions along with differentiation inducers significantly increased the differentiation of 373-L1 fibroblasts to adipocytes. These fractions may contain insulin sensitizer. The 20%, 80% and 100% methanol fractions enhanced glucose-stimulated insulin secretion in Min6 cells, insulin secreting cell line. This was related to the mechanism to promote glucose sensing and ${\beta}-cell$ proliferation, which was regulated by the induction of IRS-2, glucokinase and PDX-1 genes. As expected, 20, 80 and 100% methanol fractions increased mRNA levels of IRS-2, glucokinase and PDX-1 genes. However, PR fractions did not affect the ${\alpha}-glucoamylase$ activity in vitro. These data suggested that PR extract fractions have anti-diabetic actions through improving insulin sensitization, glucose-stimulated insulin secretion, and ${\beta}-cell$ proliferation. Therefore, PR extracts can be beneficial for anti-diabetic treatment in lean diabetic patients.

Protective Effects of Fucoidan against UVB-Induced Oxidative Stress in Human Skin Fibroblasts (자외선에 의해 유도된 인간 피부 섬유아세포의 산화 스트레스에 대한 후코이단의 보호효과)

  • Ultraviolet-B (UVB) radiation induces the formation of reactive oxygen species (ROS) and depletes stores of cellular antioxidants. Fucoidan, polysaccharides containing L-fucose and sulfate ester groups, are constituents of brown algae. In this study, the protective effects of fucoidan on UVB-induced oxidative stress in cultured human skin fibroblast HS68 cells were assessed. Pretreatment with fucoidan significantly reduced malondialdehyde (MDA) content in a dose-dependent manner. With fucoidan pretreatment at a dose of $100\;{\mu}g/ml$, the level of intracellular glutathione was increased by 21.5%, compared to UVB irradiation alone. Fucoidan significantly reduced UVB-induced ROS generation by 40.1% and 68.4% at 10 and $100\;{\mu}g/ml$, respectively, compared to UVB irradiation alone. The positive staining rates of senescence-associated $\beta$-galactosidase were reduced by 23.1% and 16.4% with 10 and $100\;{\mu}g/ml$ of fucoidan, compared to UVB irradiation alone. Fucoidan may exert a photoprotective effect against UVB radiation-induced oxidative stress.

TISSUE REACTION IN RESPONSE TO AUGMENTATION OF SOFT TISSUE DEFECTS ACCORDING TO THE DERMIS GRAFT MATERIALS IN RABBITS (연조직 결손을 일으킨 가토에서 진피 이식재에 따른 조직반응 비교)

  • The augmentation of soft tissue defects is one of the critical problems in the oral and maxillofacial surgery. Various types of graft materials, both autologous and non-autologous, have been used for the augmentation of soft tissue in the facial region. However, it is not easy to choose an ideal material for soft tissue augmentation because each has its advantages and disadvantages. An ideal graft material should meet the following criteria : it should not leave a scar at the area from which it was taken; should have less likelihood of causing infection; should feel natural after implanted; and should be not absorbed. Among the materials meeting these criteria, human dermis and artificial dermis are commonly used for clinical purposes. The present study was aimed to investigate and compare the resorption rate and the histological change following the use of the autologous dermis, the human homogenous dermis $Alloderm^{(R)}$, and the artificial dermis $Terudermis^{(R)}$ to reconstruct the soft tissue defect. Twenty mature rabbits of either sex, weighing about 2 ㎏, were used. Each rabbit was transplanted with the autologous dermis, $Alloderm^{(R)}$, and $Terudermis^{(R)}$ size $1{\times}1-cm$ at the space between the external abdominal oblique muscle and the external abdominal oblique fascia. They were then divided into 4 groups (n=5 each) according to the time elapsed after the surgery: 1, 2, 4, and 8 weeks. The resorption rate was calculated by measuring the volume change before and after the transplantation, and H-E stain was preformed to observe the histological changes. The resorption rate after 8 weeks was 21.5% for the autologous dermis, 16.0% $Alloderm^{(R)}$, and 36.4% $Terudermis^{(R)}$, suggesting that $Alloderm^{(R)}$ is the most stable while $Terudermis^{(R)}$ is the most unstable. In microscopic examinations, the autologous dermis graft was surrounded by inflammatory cells and showed foreign body reactions. The epidermal inclusion cyst was observed in the autologous dermis graft. $Terudermis^{(R)}$ and $Alloderm^{(R)}$ demonstrated neovascularization and the progressive growth of new fibroblast. The results suggest that $Terudermis^{(R)}$ and $Alloderm^{(R)}$ can be availably for substituting the autologous dermis.

Anti-obesity Effects of Wolbi-tang(越婢湯) on the Obese-mice Induced by High-fat Diet (월비탕(越婢湯)이 고지방식이(高脂肪食餌)로 유도된 비만 생쥐에 미치는 영향)

  • Objectives : In order to investigate the anti-obesity effects of Wolbi-tang(here in after referred to WBT) on the obese gene and obese inhibitory, C57BL/6 mice were induced by high-fat diet. Methods : C57BL/6 mice were divided into 5 groups(normal, only high-fat diet, high-fat diet with Reductil, high-fat diet with WBT 400, 200 mg/kg extract) and fed for 5 weeks. And observed body weight change, total cholesterol, low density lipoprotein cholesterol(LDL-cholesterol), high density lipoprotein cholesterol (HDL-cholesterol), triglyceride, glucose, leptin change, alanine transaminase(ALT), aspartate transaminase(AST), serum creatinine, the expression of ${\beta}3$-adrenergic receptor(${\beta}3AR$), leptin, uncoupling protein(UCP2) gene in 3T3-L1 adipocyte, 3T3-L1 adipocyte proliferation, histological analysis of adipose tissue and liver tissue. Results : 1. Refer to cell cytotoxicity, viability of human fibroblast cells(hFCs) showed not significant changes. 2. The amount of ALT, AST was decreased significantly in WBT 400 mg/kg, 200 mg/kg groups. The amount of creatinine showed not significant changes. 3. Body weight was decreased significantly in WBT 400 mg/kg, 200 mg/kg groups. 4. The amount of total cholesterol and triglyceride was decreased significantly in WBT 400 mg/kg, 200 mg/kg groups. LDL-cholesterol was decreased and HDL-cholesterol was increased significantly in WBT 400 mg/kg groups. 5. The amount of glucose was decreased significantly in WBT 400 mg/kg groups. 6. The amount of serum leptin was decreased significantly in WBT 400 mg/kg, 200 mg/kg groups. 7. The revelation of ${\beta}3AR$ in 3T3-L1 adipocyte was increased significantly in WBT $100{\mu}g/ml$, $50{\mu}g/ml$ groups. The revelation of leptin was decreased significantly in WBT $100{\mu}g/ml$, $50{\mu}g/ml$ groups. The revelation of UCP2 was decreased significantly in WBT $100{\mu}g/ml$ group. 8. 3T3-L1 adipocyte proliferation was decreased significantly in WBT $100{\mu}g/ml$, $50{\mu}g/ml$ groups. The size of adipocyte was decreased relative to the control group in WBT 400 mg/kg group. 9. The adipose vacuoles in liver tissue was decreased relative to the control group. Conclusions : These results suggested that WBT has inhibitory effects of obesity. WBT might be applicated on treatment of obesity and metabolic syndrome. Further studies analysing its effects were needed.

Studies on the Anti-inflammatory Activity of Paulownia coreana Uyeki Leaf Extract (오동나무 잎 추출물의 항염 효능에 관한 연구)

  • This work was carried out to investigate the anti-inflammatory effects of Paulownia coreana Uyeki on abirritant, atopy and acne skin. Paulownia coreana Uyeki has been used as a traditional medicine having anti-febrile, anti-inflammation effect in Korea, Paulownia coreana Uyeki loaves were extracted with 70% EtOH. Its superoxide anion radical scavenging activity and inhibitory effect on LPS-induced NO production were examined. The extract inhibitied the generation of NO and $PGE_2$ induced by LPS in the macrophage cell line (Raw 264.7). Consistent with the inhibitory effects on No and $PGE_2$ generation, the extract inhibited expression of iNOS and COX-2. In further study, it was found that the extract prevented $IkB-{\alpha}$ degradation, as demonstrated by western blot analysis of $IkB-{\alpha}$ protein level. However, the extract treatment did not affect cell viability at $100{\mu}g/mL$ concentration in both human skin fibroblast and Raw 264.7 cells in vitro. Thus, the present study suggests that Paulownia coreana Uyeki leaves extract have significant anti-inflammatory activity and potential as an anti-irritation material.

Synthesis and Characterization of Porous Poly(ε-caprolactone)/Silica Nanocomposites (다공성 폴리(ε-카프로락톤)/실리카 복합체의 제조 및 특성평가)

  • Poly(${\varepsilon}$-caprolactone) (PCL) nanofibers and PCL/silica membranes were synthesized by sol-gel derived electrospinning and casting, respectively. Smooth PCL nanofibers were obtained from the precursor containing N,N-dimethylformamide (DMF). PCL/silica membranes were prepared by varying the tetraethyl orthosilicate (TEOS) contents from 0 to 40 vol% to investigate the effect of silica addition on mechanical properties and cytotoxicity of the membranes. Although the strength of the membranes decreased from 12 to 8 MPa with increasing the silica content, the strength remained almost constant 7 weeks after dipping in phosphate buffered saline solution (PBS). The strength reduction was attributed to the presence of a patterned surface pores and micro-pores present in the walls between pores. The crystal structure of the membranes was orthorhombic and the crystallite size decreased from 57 to 18 nm with increasing the silica content. From the agar overlay test, the PCL/silica membranes exhibited neither deformation and discoloration nor lysis of L-929 fibroblast cells.

The Effect of Brunfelsia grandiflora Ethanol Extract on the Induction of Autophagy in Human Lung Fibroblasts (사람 폐 섬유아 세포에서 Brunfelsia grandiflora 에탄올 추출물이 Autophagy에 미치는 영향)

  • The purpose of this study was to investigate the effect of Brunfelsia grandiflora ethanol extract (BGEE) on the induction of autophagy via regulation of SIRT1 expression and p53 activation in a human lung fibroblast cell line, IMR 90. BGEE at a concentration of $5{\mu}g/ml$ or more exhibited a cytotoxic effect on IMR 90 cells. For the first time, this study showed that BGEE induces autophagy in normal human lung fibroblasts. BGEE also increased the expression level of beclin-1 at $2.5{\mu}g/ml$ or less and Atg7 at $5{\mu}g/ml$, both of which are known to be involved in the induction of autophagy. In addition, BGEE modulated the expression of other proteins related to autophagy in normal human lung fibroblasts. The expression levels of p53 and p-p53, an active form of p53, were decreased in the presence of BGEE at a noncytotoxic concentration. In contrast, the expression level of SIRT1 was increased in human lung fibroblasts treated with BGEE at a noncytotoxic concentration. Moreover, SA-${\beta}$-Gal staining, an aging marker, was reduced in the normal human lung fibroblasts treated with BGEE. These findings suggest that BGEE promotes the induction of autophagy and antiaging through the modulation of p53 and SIRT1 in human lung fibroblasts.

Anti-aging and Anti-melanogenesis Efficacy by Antioxidative Mineral-bio Water (항산화성 바이오 미네랄 활성수에 의한 항노화 및 미백효과)

  • UV irradiated skin cells produce reactive oxygen species (ROS). ROS is known to be the primary cause of skin inflammation that is eventually leading to skin aging through decrease of collagen in the dermis. In this study, we evaluated basic efficacy of anti-aging, anti-inflammation and anti-melanogenesis using two antioxidative mineral-bio waters (Mineral-bio water 1 (MIBA-W1) and Mineral-bio water 2 (MIBA-W2)). Both antioxidative mineral-bio waters reduced TNF-${\alpha}$ expression which was induced upon UV irradiation. MIBA-W 1 increased collagen synthesis from UVB irradiated fibroblast at 0.01 % concentration but MIBA-W2 shows slight, but linear increase. Stimulation of melanogenesis by ${\alpha}$-MSH treatment in the cultured B16-F1 melanoma was significantly reduced by the treatment of MIBA-W2 in a dose dependent manner. Taken together, antioxidative MIBA-W1 and 2 seem to have potential applications as functional cosmetic materials.

Antioxidative and Anti-aging Effects of Extract from Talinum paniculatum (토인삼 추출물의 항산화 및 항노화 효과)

  • Ultraviolet irradiation in the cells and skin produces reactive oxygen species (ROS), which induces the synthesis of matrix metalloproteinases (MMPs) causing skin photoaging. Using the human dermal fibroblast (HDF), we investigated the antioxidative and anti-aging effects of the extracts from Talinum paniculatum. Talinum paniculatum leaf and stem extracts (LSE) showed free radical scavenging effect by 98.45% at 500 ${\mu}g/mL$ and superoxide radical scavenging effect by 97.01% at 500 ${\mu}g/mL$ in the xanthine/xanthine oxidase system. The photoprotective potential of LSE was tested in HDF exposed to ultraviolet irradiation. It was revealed that LSE had an inhibitory effect on MMP-1 expression in UVA-irradiated HDF without any significant cytotoxicity. The treatment of UVA-irradiated HDF with LSE resulted in dose-dependent decreases in the expression levels of MMP-1 mRNA and protein. Also, UVB-induced cytotoxicity and cell death were effectively suppressed by treatment of LSE. Additionally, the senescence-associated ${\beta}$- galactosidase (SA-${\beta}$-gal) activity was decreased in the presence of LSE. These results suggest that Talinum paniculatum leaf and stem extracts (LSE) may have anti-aging effects and can be used as new functional materials against oxidative stress-mediated skin damages.
