Effect of Irrigation Methods for Reducing Drainage on Growth and Yield of Paprika (Capsicum annuum 'Coletti') in Rockwool and Cocopeat Culture (배액절감형 양액공급 방법이 파프리카(Capsicum annuum 'Coletti') 생육과 수량에 미치는 영향)

  • This study was carried out to investigate the effect of irrigation methods for reducing a drainage on the growth and yield in rockwool (Grodan co.) and cocopeat (chip : dust = 50 : 50 included fiber) culture. The nutrient solution was irrigated by $100J{\cdot}cm^{-2}$-100 mL, $50J{\cdot}cm^{-2}$-45 mL, $50J{\cdot}cm^{-2}$-40 mL, $50J{\cdot}cm^{-2}$-35 mL ($100{\sim}50J{\cdot}cm^{-2}$-100~35 mL, Nutrient solution 100~35 mL was irrigated per plant when the accumulated radiation was $100{\sim}50J{\cdot}cm^{-2}$). The drain rates per plant of 100-100, 50-45, 50-40, 50-35 were 26.3%, 8.8%, 6% 4.4% and 23.1%, 7.5%, 5% 3.4% in rockwool and cocopeat slabs. The water contents and EC of 100-100 and 50-45 were managed by the 55~70%, $3.0{\sim}5.0dS{\cdot}m^{-1}$ which were good condition for paprika culture in rockwool and cocopeat slabs, while those of 50-40 and 50-35 were managed by beyond 50%, $4.5{\sim}9.5dS{\cdot}m^{-1}$. The plant height, number of branches and leaf size of 100-100 and 50-45 were similarly increased while those of 50-40 and 50-35 were decreased. The fruit size and weight of 50-40 and 50-35 were small and light, while those of 100-100 and 50-45 were similarly big and heavy. The marketable fruits of 100-100 and 50-45 treatments were similarly more by 9.7~9.8 in rockwool and 8.8~8.9 in cocopeat, while the unmarketable fruits, the small and blossom end rot fruits were increased in 50-40 and 50-35 treatments. The yield of 100-100 and 50-45 treatments were similarly high.

Skin Protection Effect of Grape Pruning Stem Extract on UVB-induced Connective Tissue Injury (포도전정가지 추출물이 UVB로 유도된 결합 조직 손상에 미치는 피부 보호 효과)

  • This study aimed to analyze the contents of rutin, procyanidin B3, quercetin, and kaempferol, known to have antioxidant, anti-inflammatory, and anti-carcinogenic effects, among the polyphenol types contained in grape pruning stem extracts (GPSE). It utilized grape stems discarded after harvest to measure the effects of GPSE on skin moisture, inhibition of skin cell proliferation, and anti-inflammatory activity on the damaged skin of HR-1 mice induced with ultraviolet B (UVB), and to verify the applicability of GPSE as a material for functional food and functional cosmetics. The polyphenol was extracted from grape pruning stems with 80% EtOH, and then the extract was used while storing at $-20^{\circ}C$, after filtering, concentrating, and freeze-drying it. The content of an active ingredient of GPSE was analyzed using high performance liquid chromatography (HPLC). From 53 kg of the grape pruning stem specimen, 2.34 kg of the EtOH fraction extracts were extracted to achieve a 4.42% yield ratio. Analysis of the active ingredients showed 0.28 mg/g of procyanidin B3, 12.81 mg/g of rutin, 0.51 mg/g of quercetin, and 8.24 mg/g of kaempferol. After UVB irradiation on the dermis, to confirm the degree of inhibition of collagen synthesis, we examined the protein expression of MMP-9 using immunohistochemical staining. The results of this study confirm the existence of active polyphenol types, such as rutin, kaempferol, quercetin, and procyanidin B3, in GPSE. Moreover, the study found that GPSE has anti-collagenase effects and it decreases the effects of UV damage on skin barrier function. GPSE is a functional ingredient with a potential for skin protection effects, and it has high utilization potential as an ingredient for functional cosmetics.

Physical Properties of Pan Bread Made with Various Amounts of Squeezed Danggui Leaf (Angelica acutiloba Kitagawa) Juice (당귀잎 착즙액을 이용하여 제조한 식빵의 물리적 특성)

  • Danggui leaf (Angelica acutiloba Kitagawa) has numerous dietary fiber and healthy functional properties such as anti-inflammatory activity. However, it is usually discarded after its roots are harvested. For wide application of danggui leaf, squeezed danggui leaf juice was used for making pan bread. Treatments included imported wheat flour (IMWF) and domestic wheat flour (DOWF) as a control, 1% soluble solid contents of squeezed danggui leaf juice instead of wheat flour (1% SDLJ), 2% SDLJ, 3% SDLJ, and 4% SDLJ. In the fermentation expansion, dough volume was not significantly different between DOWF and 1% SDLJ, whereas it decreased according to increased amount of squeezed danggui leaf juice. Regarding physical properties, springiness and cohesiveness decreased according to increased amount of squeezed danggui leaf juice and longer storage period. Gumminess, brittleness, and hardness increased according to increased amount of squeezed danggui leaf juice, although these were not statistically different among IMWF, DOWF, and 1% SDLJ after making pan bread. Principal component analysis (PCA) was performed to assess the correlation between storage period and physical properties. In the PCA, variance proportion of principal component (PC) 1 was 0.87 while that of PC 2 was 0.10. Further, 1% SDLJ showed similar results as DOWF and INWF after making pan bread and after 3 days of storage. In conclusion, use of 1% SDLJ is desirable for making squeezed danggui leaf juice.

The Effect of Maturity Stage and Particle Length of Sorghum-Sudan Hybrid on the Quality of Silage (Sorghum-Sudan Hybrid의 생육시기와 절단길이가 Silage의 품종에 미치는 영향)

  • The purpose of this study is to examine the effect of maturity stages and particle length on quality of silage made from Sorghum-sudan hybrid (Pioneer 988). The silage was made at heading, milky, dough and yellow ripe stage and cut 1.0, 2.5, 4.0cm at each stage. The quality of the silage was evaluated on the bases of the chemical compositions, content of organic acids, pH and $NH_3-N$ of the silage, and also the correlation between main compositions of the silage. The results were summarized as follows; 1. The moisture content was decreased with advancing maturity, but dry matter content was increased. Crude protein was decreased with advancing maturity at the level of 12.55 percentageat heading stage. Crude fiber was tended to increase. 2. The moisture content of silage was markedly decreased with advancing maturity (p<0.01). 3. The pH value of silage was the lowest when particle length was 2.5cm, and there was no significant difference between particle lengths. At the yellow ripe stage, the pH value was the lowest (3.53) and at the dough stage, the pH value was the highest (4.59) (p<0.01). 4. The rate of $NH_3-N$ to total-N was the highest (16.3%) at heading stage, the lowest (9.2%) at the dough stage. 5. The organic acid contents was not uniformly fixed at the particle length, but the highest quality silage was produced at yellow ripe stage as was not produced butyric acid and plenty of lactic acid was contented. 6. The correlations between pH and lactic acid (r=0.719), pH and total acid (r=-0.716), butyric acid and Flieg's score (r=0.872) were negative, respectively (p<0.01). And those between lactic acid and total acid (r=0.990), moisture and $NH_3-N$ (r=0.767) were positive, respectively (p<0.01).

Effects of Supplemental Synbiotics Composed of Anaerobic Bacteria, Yeast and Mold on the Change of Chemical Composition and Fermentation Characteristics of Total Mixed Ration for Cattle (혐기성 박테리아, 효모 및 곰팡이로 제조된 synbiotics 첨가 축우용 완전혼합사료의 성분 변화 및 발효 특성에 미치는 영향)

  • In order to investigate the effects of synbiotics on change of chemical composition and fermentation characteristics of total mixed ration (TMR), eight TMRs fermented by synbiotics composing the anaerobic microbes (bacteria, yeast, mold) were alloted to the experimental treatments. Treatments were composed of untreated synbiotics(US), bacterial synbiotics (BS), yeast synbiotics (YS), mold synbiotics (MS), bacterial and mold synbiotics (BMS), yeast and mold synbiotics (YMS), bacterial and yeast synbiotics (BYS), and bacterial, yeast and mold synbiotics (BYMS). After 7 days of anaerobic fermentation, fermented-TMRs were exposed to air during 1, 3, 5, 7, 14 and 21 days. One hundred forty four (8 treatments ${\times}$ 6 exposing days ${\times}$ 3 replications) fermented- TMRs were manufactured by vinyl bag sized of 43 cm by 58 cm. The results obtained were as follows. Moisture contents of the fermented TMRs anaerobically ranged from 41% to 45%, and was similar to those of basal TMRs. As results of anaerobic fermentation, the concentration of crude protein was decreased by 11.7% to 14.8% in the untreated sample, while was rather increased by 11% when the TMR was fermented with BMYS. And also BMYS treatment showed decreases by 32% for crude fiber, 15.5% for NDF and 26.1% for ADF. Internal temperature of fermented-TMRs was highest at 7 day of exposing in the air. The pH of fermented-TMR juice was significant difference betweentreatments after 7 day of exposing in air, and that of BMS was highest at 14 day after exposing in air (P<0.05). Acid buffering capacity was increased in proportion to the exposing day of TMR, and peaked at 7 or 14 days after exposing. Ammonia concentration of fermented-TMRs was highest at 5 day after exposing in the air. Individual volatile fatty acid of fermented-TMR juice was very low level in all treatments. Although BMYS treatment to TMR inclined to increase in crude protein and decrease in fibers, but there were no positive effects on the fermentation characteristics after exposing in the air by supplementation of anaerobic synbiotics to TMR.

Characteristics of Chungkookjang that Enhance the Flavor and GABA Content in a Mixed Culture of Bacillus subtilis MC31 and Lactobacillus sakei 383 (Bacillus subtilis MC31와 Lactobacillus sakei 383의 혼합배양으로 향상된 풍미와 GABA 함량을 지닌 청국장의 품질 특성)

  • Chungkookjang has several functional properties, such as fibrinolytic activity, anticancer effects, and antioxidant effects. However, children do not like Chungkookjang because of its foul odor. A mixed culture of Bacillus subtilis MC31 and Lactobacillus sakei 383 was used to improve the production of GABA in Chungkookjang and its flavor. Most of the foul odor of Chungkookjang was removed. The slime content and viscosity of Chungkookjang fermented in the mixed culture were similar to those of commercial Chungkookjang when B. subtilis MC31 and Lactobacillus sakei 383 were inoculated in a 1:1 ratio. The maximum GABA content was obtained when Chungkookjang was fermented with B. subtilis MC31 and L. sakei 383, which was fermented at $37^{\circ}C$ for 72 hr. During the period of fermentation, the viable cell number of B. subtilis MC31 reached a peak (log 9.13 CFU/g) at six days, and L. sakei 383 reached a peak (log 6.78 CFU/g) at two days. The moisture, crude ash, crude protein, crude fat, and crude fiber contents were 61.71%, 2.05%, 17.54%, 8.36%, and 1.95%, respectively. The amino-type nitrogen content of Chungkookjang fermented by B. subtilis MC31 and L. sakei 383 was less than Chungkookjang fermented by B. subtilis MC31 alone. The ammonia-type nitrogen and reducing sugar content of the Chungkookjang fermented by B. subtilis MC31 and L. sakei 383 were higher than that of steamed soybean. The glutamic acid and GABA content detected with an amino acid analyzer were 1.40 mg/g and 0.47 mg/g, respectively. These results suggest that fermentation with B. subtilis MC31 and L. sakei 383 in a 1:1 ratio removes more of the foul odor and increases the GABA content compared with single fermentation.

Nutritional Value of Soybean Meal from Various Geographic Origin and Effect of Their Dietary Supplementation on Performance of Broilers (원산지가 다른 대두박의 영양적 가치평가와 육계의 생산성에 미치는 영향)

  • This study was carried out to investigate the nutritional value of soybean meal (SBM) from various geographic origins and the effects of their dietary supplementation on performance of broiler chickens. Nutritional value of dehulled SBM originating from USA, and non-dehulled SBM from India (IND), Argentina (ARG) and Korea (KOR) were evaluated by analyzing chemical composition, urease activity (UA) and KOH solubility, and determining true metabolizable energy (TME), nitrogen corrected TME (TMEn) and true amino acid availability (TAAA). The contents of crude protein ranged from 45.43% (ARG) to 48.47% (USA) and those of crude fiber varied widely from 3.48% (USA) to 7.12% (IND). The measurements of lysine varied from 2.79% (IND) to 3.09% (USA) and those of methionine from 0.56% (IND) to 0.65% (USA). The values of TMEn varied from 2986.6 kcal/kg (IND) to 3228.9 kcal/kg (USA) and the averages of TAAA were from 91.61% (IND) to 92.27% (USA). UA was found to be from 0.02 (ARG) to 0.04 (KOR, USA) and those of KOH solubility from 73% (ARG) to 84% (USA). A total of four hundred 20-days-old male broiler chicks were divided into four groups and fed with isocaloric and isonitrogenous experimental diets containing 27.5% of SBM and same amounts of lysine and sulfur amino acids for 15 days. Final body weight and body weight gain were the highest in birds fed with SBM from USA and lowest in birds fed with SBM from IND although the differences were not statistically significant. The feed/gain in chicks fed diet containing SBM from USA was significantly improved (p<0.05) compared to those of the other groups. There were no significant differences in carcass characteristics and the concentration of total cholesterol in serum among the treatments. The results of in vitro assay and bioassay agreed with the performance of broiler chicks, and thus there were close correlation between the broiler performance and the measured nutritive values of SBM. In conclusion, dehulled SBM from USA was superior to non- dehulled SBM from ARG and IND with regard to nutritive values.

Effects of Sowing Mixed Legume Forage and Applying Cattle Manure on the Productivity and Organic Hanwoo Feeding Capacity of Whole Crop Wheat (콩과 사료작물의 혼파와 우분의 시용이 총체 밀의 생산성과 유기한우 사육능력에 미치는 영향)

  • This study was to assess the effects of sowing mixed legume forage and applying fermented cattle manure on the productivity and feed value of whole crop wheat (WCW) as a representative of winter crops and also to estimate feeding capacity of Korean native cattle (Hanwoo) per unit area by defining optimal application levels of cattle manure for more fertile soil and manure recycling for the purpose of reduced environmental pollution and efficient production of organic forage contributable to production of organic animal products. Sowing mixed legume forage significantly (p<0.05) increased yields of dry matter (DM), crude protein (CP) and total digestible nutrients (TDN) for WCW compared to WCW alone sowing, but there were no differences between mixed sowing treatments. The yields of DM, CP and TDN were significantly (p<0.05) higher in 100~150 kg N/ha. The CP and acid detergent fiber (ADF) contents were highest in mixed sowing with hairy vetch, followed by sowing mixed forage pea and WCW alone (p<0.05). There were no differences in CP among cattle manure application levels. As nitrogen application level increased, ADF content increased (p<0.05), but TDN content decreased (p<0.05). Feeding capacity of organic Hanwoo (head/ha) weighing 450 kg of body weight with 400 g of daily gain significantly (p<0.05) increased due to mixed sowing with legume forage, and with increasing application levels of fermented cattle manure, feeding capacity significantly (p<0.05) increased. The results indicated that mixed-sowing of WCW and legume forage not only reduced nitrogen application level of fermented cattle manure by over 50 kg per ha but also increased their feed value and productivity, and feeding capacity. This would be contributable to diversity of forage resources as well as production of organic animal product by creating low cost and high quality of forage.

A Studies on the Chemical Composition and in vitro Biological Activities of a Hot Water Extracts of Gastrodia elata (천마추출물의 성분분석 및 in vitro 생물활성에 관한 연구)

  • A hot water extract was prepared from the artificially grown Gastrodia elata to investigate its chemical composition and various in vitro biological activities as an effort to develop G. elata as health/functional food materials. The contents of crude protein, ash, fat, fiber, moisture and total sugar were 5.4, 2.6, 3.6, 3.3, 8.1 and 77% (w/w), respectively. The extract of G. elata had greater amount of potassium (1,150 mg/100 g) than phosphorus (300 mg/100 g). Dose-dependence against human carcinoma (Hep3B, MCF-7, A549 and AGS) were observed from 0.2 mg/ml to 1.0 mg/ml. Especially, the treatment of 1.0 mg/ml extracts showed the highest cytotoxicity with 83% against gastric carcinoma (AGS). The extracts showed weak antimicrobial activities against Bacillus subtilis and Pseudomonas aeruginosa, but practically no antimicrobial activity against the other microorganisms tested. The effect of ${\alpha}$-glucosidase inhibition was 64% at the concentration of 1.0 mg/ml. The inhibitory effect of angiotensin converting enzyme (ACE) of the extract in the range of $0.2{\sim}1.0mg/ml$ showed $63{\sim}89%$, and the highest ACE inhibition was 89% at the concentration of 1.0 mg/ml of extracts. The highest activity of glutathion S-transferase (GST) was 221 % at the concentration of 1.0 mg/ml of the G. elata extracts. These results suggest that G. elata may be used as health/functional food materials.

Physicochemical Properties, Free Sugar and Volatile Compounds of Korean Citrons Cultivated in Different Areas (산지별 유자의 이화학적 특성, 유리당 및 향기성분)

  • Physicochemical properties, free sugar and volatile flavor compounds of citrons harvested in different cultivation areas, such as Geoje, Goseong, Goheung, and Namhae, were analyzed and compared. Total weight of citron, ranging from 107.97 g to 154.86 g, was significantly different according to producing area. Weight ratio of flesh to whole citron was higher than one of peel in citron produced from Geoje, Goheung-improved, or Namhae-native. Citron size was the highest in Namhae-native citron, but the color of peel was significantly the higher in Goheung-native and its improved citron. In the peel of citron, hardness of citron produced from Geoje was the strongest (2337.13 $cm/kg^2$) whereas citron produced from Goseong (1592.38 $cm/kg^2$) showed the softest hardness. Moisture content was 85.35~87.81 g/100 g and ash was contained below 0.8 g/100 g in flesh of all samples. Although crude fiber content in Geoje ctiron was significantly the lowest in the peel, peel was 1.88~2.60 times higher than flesh. The order of sugar content seemed to be fructose> glucose> sucrose in all citrons, and content of fructose or glucose was higher in peel than flesh. Content of total sugar was 8.44~12.19 g/100 g in citron, and contents of improved citrons from Goheung and Namhae were significantly higher than those of native citrons. In addition, major volatile compounds were dl-limonene (59.52~74.30%) and $\gamma$-terpinene (5.60~7.88%) among 15 kinds of volatile compounds identified from citron peel. These results suggested that physicochemical properties and volatile flavor compounds of citrons showed some differences with regard to producing areas.
